[pt] Este trabalho teve como objetivo principal avaliar as
representações sociais,
elaboradas por mulheres que tiveram câncer de mama, sobre a
doença, sua
etiologia e tratamento. Desenvolvemos teoricamente o tema a
partir de quatro
perspectivas: biológica, psicossomática, psicossocial e
multifatorial. Realizamos
uma pesquisa de campo, de natureza qualitativa, estudando
10 casos de mulheres
que tiveram câncer de mama com idades entre 35 e 50 anos,
que se submeteram à
mastectomia, podendo ter feito ou não a reconstrução
mamária, e que
participavam como membros de uma associação de apoio a
mulheres com câncer
de mama da cidade de Niterói, Rio de Janeiro. Como
instrumento desta pesquisa,
utilizamos um questionário identificador e entrevistas
semidirigidas, aplicados a
tais mulheres individualmente e, depois, a seus familiares,
com base em roteiro
pré-elaborado. Para avaliação dos dados obtidos, empregamos
a análise de
discurso intra-sujeito e intersujeito. Na primeira,
buscamos identificar, em cada
caso, a percepção das entrevistadas sobre a doença, sua
etiologia e as repercussões
do tratamento oncológico em sua identidade feminina. Na
segunda, construímos
10 categorias de análise. Os resultados revelaram que o
câncer é apreendido pelas
mulheres como um risco e ameaça à vida, que a retirada da
mama afeta,
majoritariamente, a identidade corporal e feminina, e que o
adoecimento provoca
muitas mudanças em suas vidas. / [en] The prime purpose of this paper is to assess the social
representations
created by women who have had breast cancer, and to discuss
the disease, its
etiology and treatment. We developed the theme
theoretically, based on four
perspectives: biological, psychosomatic, psychosocial and
multi-factorial. We
performed field research of a qualitative nature, studying
ten cases of women who
had breast cancer in the 35 to 50 age group and had
undergone a mastectomy,
could have done mammary reconstruction or not, and who
participated as
members of a support association to women with breast
cancer in Niterói city, Rio
de Janeiro State. As an instrument of this research, we
used an identifying
questionnaire and semi-focused interviews with those women
individually and
later with their relatives, based on a previously prepared
script. In order to assess
the obtained data, we used intra-subject and inter-subject
discourse analysis. In the
former, in each case we looked to identify the
interviewee s perception of the
disease, its etiology and the repercussions of cancer
treatment on her female
identity. In the latter, we created ten analytical
categories. The results showed that
women are apprehensive of cancer as life threatening and a
risk, that breast
removal in most cases affects the corporal and female
identity, and that
contracting the disease causes many changes in their lives.

[pt] A presente tese analisa, em uma perspectiva de múltiplos ciberespaços, performances identitárias de Vitória e Glória, mulheres em tratamento de câncer de mama que, através de suas interações online, constroem-se como vítimas ao descobrirem a doença e passam a agir como protagonistas em redes sociais, na luta contra o câncer. Sendo assim, os objetivos deste estudo são: (i) analisar as performances identitárias de duas mulheres com câncer de mama em diferentes sites; e (ii) compreender de que forma as participantes se constroem como protagonistas em redes sociais ao longo do tratamento da doença. A perspectiva teórica da pesquisa situa-se em estudos sobre as performances identitárias na narrativa e no discurso multimodal, no protagonismo discursivo e social e na teoria do posicionamento. A metodologia da pesquisa é qualitativa e interpretativa de cunho netnográfico. Os dados consistem de vídeos, entrevistas e narrativas das participantes que estão registradas em páginas públicas do Facebook, do YouTube, de três sites institucionais e de um blog. As análises indicam que: (i) há mudanças de performances identitárias e de posicionamento das participantes nas redes sociais, sinalizando transformação das trajetórias de vida ao longo de seus tratamentos – do ser comum ao protagonismo; (ii) as participantes ressignificam suas experiências de dor e lamentação, em uma perspectiva de discurso como ação social via discursos de emoção, esperança, triunfo e engajamento político; (iii) o agenciamento é direcionado aos interlocutores nas redes sociais, enquanto posicionamento político e identitário de enfrentamento do câncer. Nesse sentido, como contribuição social, esta tese sinaliza que, na sociedade contemporânea, as redes sociais se tornam espaços férteis de resistência e de empoderamento pessoal e de outros usuários, e espaços de metamorfose de lagarta a borboleta em relação ao câncer. / [en] The present thesis analyzes, from a multi-sited perspective, the identity performances of Vitória and Glória, women in treatment of breast cancer who, through their online interactions, construct themselves as victims when they discover the disease and begin to act as protagonists in social networks in the fight against cancer. Thus, the objectives of this study are: (i) to analyze the identity performances of two women with breast cancer in different websites; and (ii) to understand how the participants construct themselves as protagonists in social networks throughout the treatment of the disease. The theoretical perspective of the research is located in studies on the identity performances in narrative and multimodal discourse, in the discursive and social protagonism and in the theory of the positioning. The methodology of the research is qualitative and interpretative of a netnographic nature. The data consist of videos, interviews and narratives of the participants that are registered in public pages of Facebook, YouTube, three institutional websites and one blog. The analysis indicate that: (i) there are changes in identity performances and in the positioning of participants in social networks, signaling the transformation of life trajectories throughout their treatments - from being ordinary to protagonism; (ii) participants reassign their experiences of pain and lamentation, from a discourse perspective as social action through discourses of emotion, hope, triumph and political engagement; (iii) agency is directed to interlocutors in social networks, as a political and identity positioning for coping with cancer. In this sense, as a social contribution, this thesis indicates that, in contemporary society, social networks become fertile spaces of resistance and personal/users empowerment, and spaces of metamorphosis from caterpillar to butterfly in relation to cancer.

[en] SYNTHESIS OF THE 11A-N-ARYLSULFONIL-TETRAHYDRO-5H-BENZO[A]CARBAZOLES WITH ANTITUMORAL ACTIVITY / [pt] SÍNTESE DE 11A-N-ARILSULFONIL-TETRAHIDRO-5H-BENZO[A]CARBAZÓIS COM AÇÃO ANTITUMORALJOSEANE ALVES MENDES 11 April 2017 (has links)
[pt] Este trabalho apresenta a síntese e caracterização de 11-tosil-6,6a,11, 11-tetraidro-5H-benzo[a]carbazois análogos ao LQB223, um composto previamente sintetizado em nosso grupo de pesquisa que tem ação anticâncer e antileishmanial in vitro e ação antimalarial in vitro e in vivo. A etapa chave da reação para a obtenção dos 5-metoxi-11a-N-arilsulfonil-tetrahidro-5H-benzo[a]carbazol (97a), 6-metoxi-11a-N-arilsulfonil-tetrahidro-5H-benzo[a]carbazol (97b) e 7-metoxi-11a-N-arilsulfonil-tetrahidro-5H-benzo[a]carbazol (97c) envolve uma reação de aza-arilação de Heck catalisada por paládio na presença de carbonato de prata e acetona entre o N-tosil-o-iodoanilina (55b) e dihidronaftalenos substituídos (100a, 100b, e 100c), preparados previamente a partir de tetralonas comerciais (84a, 84b, 84c). Os compostos 5-hidroxi-11a-N-arilsulfonil-tetrahidro-5H-benzo[a]carbazol (98a), 6-hidroxi-11a-N-arilsulfonil-tetrahidro-5H-benzo[a]carbazol (98b) e o 7-hidroxi-11a-N-arilsulfonil-tetrahidro-5H--benzo[a]carbazol (98c) foram obtidos através da desproteção das metoxilas com BBr3 em rendimentos quantitativos. Todos os compostos foram avaliados quanto a ação anticâncer em linhagens de leucemia que apesentam o fenótipo MDR lucena e FEPS e em linhagem de leucemia K562 e nas linhagens de câncer de mama MCF-7, que expressa o receptor de estrogênio, e MDA-MD231.Dentre os compostos preparados, o composto 6-metoxi-11a-N-arilsulfonil-tetrahidro-5H-benzo[a]carbazol (97b) foi tão ativo quanto o LQB223 frente a ação antileucêmica nas linhagens K562, lucena e FEPS, entretanto foi mais eficaz que o LQB223 nas linhagens de MCF-7 (IC50 igual 5,93 mais ou menos 3,58 MiM) e MDA-MD231 (IC50 igual 8,29 mais ou menos 2,08 MiM), apresentando um nível baixo de toxicidade para as células sadias. Por outro lado, o composto 6-hidroxi-11a-N-arilsulfonil-tetrahidro-5H-benzo[a]carbazol (98b), apesar de ser muito ativo em linhagens de câncer mama, possui um alto nível de toxicidade para células sadias e composto 7-hidroxi-11a-N-arilsulfonil-tetrahidro-5H-benzo[a]carbazol (98c) foi eficaz em MDA-MD231 (IC50 igual 4,84) porém ainda não foi avaliado quanto a toxicidade em células sadias. Os dados sugerem que os compostos oxigenados 97b e 98c direciona a ação para o câncer de mama. / [en] This work describes the synthesis and characterization of the 11a-N-arylsulfonil-tetrahydro-5H-benzo[a]carbazoles analogous to LQB223, a previously synthesized compound by our research group that has anti-cancer and antileishmanial activity in vitro and antimalarial activity in vitro and in vivo. The key step of the reaction for obtaining the 5-methoxy-11a-N-arylsulfonil-tetrahydro-5H-benzo[a]carbazole (97a), 6-methoxy-11a-N-arylsulfonil-tetrahydro-5H-benzo[a]carbazole (97b) and 7-methoxy-11a-N-arylsulfonil-tetrahydro-5H-benzo[a]carbazole (97c) is the palladium catalyzed Heck aza-arylation reaction in the presence of silver carbonate and acetone from N-tosyl-o-iodoaniline (55b) and substituted dihydronaphthalenes (100a, 100b and 100c), previously prepared from commercial tetralones (84a, 84b and 84c). The compounds 5-hydroxy-11a-N-arylsulfonil-tetrahydro-5H-benzo[a]carbazole (98a), 6-hydroxy-11a-N-arylsulfonil-tetrahydro-5H-benzo[a]carbazole (98b) and 7-hydroxy-11a-N-arylsulfonil-tetrahydro-5H-benzo[a]carbazole (98c) were obtained by deprotection with BBr3 in yields quantitative. All compounds were evaluated for anticancer activity in K562, FEPS and Lucena leukemia cell lines, which exhibit the MDR phenotype, and in breast cancer cell lines MCF-7, which expresses the estrogen receptor, and MDA-MD231.Among the compounds prepared, the compound 97b is as active as LQB223 for the antileukemic activity on K562 lines, and Lucena FEPs, however it is more effective than the LQB223 in MCF-7 (IC50 equal 5.93 plus/minus 3.58 MiM) and MDA-MD231 (IC50 equal 8.29 plus/minus 2.08 uM) breast cancer lines, with low level of toxicity to healthy cells. The compound 98b presents similar activity to 97b but is toxic. The compound 98c is effective in MDA-MD231 (IC50 equal 4,84) but, level of toxicity to healthy cells was not test yet. The reports suggest that the oxygenation pattern in the 3 position directs to the anticancer activity to the breast cancer lines.
