Evaluation of a modified paleolithic dietary intervention for the treatment of relapsing-remitting multiple sclerosis

Improvements in fatigue and quality of life observed in primary and secondary progressive multiple sclerosis (MS) patients adhering to a modified Paleolithic dietary intervention (MPDI), nutritional supplementation, exercise, and neuromuscular electrical stimulation regime are hypothesized to be due primarily to the effect of diet. However, no research has been conducted evaluating effects of the dietary intervention alone thus, the purpose of this research was to evaluate a MPDI in the treatment of Relapsing-Remitting Multiple Sclerosis (RRMS). We tested effects of the MPDI in seventeen men and women (mean age: 36.3 ±4.7 years) with neurologist-verified RRMS. Nine subjects (one male) were randomized to a "usual care" (control) group and eight subjects (one male) were taught the MPDI. Both groups adhered to their assigned protocol for three months. Significant improvement was seen in Fatigue Severity Scale (FSS, p=0.03), Multiple Sclerosis Quality of Life-54 Physical Health (MSQOL-P, p=0.03), and Mental Health (MSQOL-M, p=0.02) scores from baseline in MPDI subjects compared to controls. Increased vitamin K serum levels (p=0.02) were also observed in MPDI subjects at three months compared to controls. Significantly reduced time to complete 9-Hole Peg Test (9-HPT) with the dominant hand (p=0.02) was also observed. Our results indicate trends for improved non-dominant 9-HPT (p=0.05), Metabolic Equivalent Tasks (METs, p=0.08), and 25-Foot Walk (25-FW, p=0.09) scores from baseline in MPDI subjects compared to controls. A Paleolithic diet may be useful in the treatment and management of MS, by reducing perceived fatigue, increasing mental and physical quality of life, increasing exercise capacity, and improving hand and leg function. The MPDI may also reduce inflammation as evidenced by increased vitamin K serum levels.

Acute neural adaptations to resistance training performed with low and high rates of muscle activation

Training is associated with specific neural adaptations. Skill training has been associated with increases in corticospinal tract excitability, leading to long term adaptations within motor cortex. Neural adaptations associated with strength training are less well established. It is not known how they are affected by volume, intensity, rate of muscle activation, or rest period. We evaluated the acute neural adaptations to a single session of strength training with the goal of evaluating the influence of rate of muscle activation on neural adaptations at the cortical and spinal levels. Thirty subjects participated in a single session of maximal, isometric knee extensions with the right leg. The training consisted of 4 sets of 5 contractions, with sets separated by approximately 5 minutes. Subjects were randomized into a high rate of muscle-activation group (Ballistic), a low rate of muscle-activation group (Ramp), and a Control group that did all testing but no training. Cortical spinal tract excitability was assessed using transcranial magnetic stimulation, spinal excitability was assessed using peripheral nerve stimulation, and inhibition of motor cortex was assessed using short-interval intracortical inhibition. In addition, we measured changes in motor performance. These same measures were assessed 24 hours later. Results showed that an acute strength training session is associated with a depression in resting cortical spinal tract excitability, but no change in active excitability. This change was immediate, taking place after just 2 contractions. Training was also associated with an increase in the excitability of the monosynaptic reflex circuit within the spinal cord during muscle activation, but not at rest. After 24 hours, cortical motor tract excitability had returned to normal, but intracortical inhibition was decreased from the original measure. Subjects from all groups increased maximum rate of torque development from Day 1 to Day 2. These results indicate that strength-training is associated with neural adaptations, though the adaptations were different than those for skill training. Understanding these adaptations will allow coaches and clinicians to better design programs to optimize the strength potential of the nervous system along with that of the muscles.

CRISPR Genetic Editing: Paths for Christian Acceptance and Analysis of In Vivo and In Vitro Efficiency

With advancements in CRISPR-cas9 broadening the potential paths for clinical usage of genetic editing, conversations about genetic editing have grown to outside simply scientific communities and into mainstream conversations. This study focuses specifically on Christian discourse of genetic editing and locates four major tensions for many Christians when they think about genetic editing: beginning of life, Creator-human relationship, imago Dei, and stewardship. With these major concerns in mind, I identify epigenetics, somatic cell genetic editing, and in vivo genetic editing research as important research paths to pursue as they can potentially produce techniques that more Christian individuals would feel comfortable using. I pursue one of these paths and conclude with an experimental proposal for an analysis of in vivo and in vitro CIRSPR-Cas9 efficiency in regards to on- and off-target rates.

A COMPARATIVE STUDY OF SLEEP, DIURNAL PATTERNS, AND EYE CLOSURE BETWEEN THE HOUSE MOUSE (MUS MUSCULUS) AND AFRICAN SPINY MOUSE (ACOMYS CAHIRINUS)

To understand the function and origins of sleep, sleep needs to be studied across many different species. Although it is well conserved throughout mammals, 95% of papers are restricted to just three species, Homo sapiens, Mus musculus, and Rattus norvegicus. We aimed to characterize sleep and wake in a Murid rodent Acomys cahirinus in greater detail alongside the well-studied laboratory house mouse (Mus musculus) and wild M. musculus using a well validated, non-invasive, piezoelectric system for sleep and activity monitoring. We confirmed A. cahirinus, M. musculus, and wild M. musculus to be primarily nocturnal, but with clearly distinct behavioral patterns. Specifically, the activity of A. cahirinus sharply increases right at dark onset, which is common in nocturnal species, but surprisingly, decreases sharply just one hour later. Using infra-red camera recordings in single and group cage conditions, we found that A. cahirinus is more active early in the night period than late night period in single and group cages, and this decreased activity in the latter half of the night is much greater compared to M. musculus. In order to better understand these differences in activity, we investigated the sleep architecture of A. cahirinus using electroencephalogram (EEG) recordings. Our data show that A. cahirinus have a few key differences in sleep from M. musculus. A. cahirinus have significantly longer daily sleep periods and exhibit a much higher amount of REM sleep. A. cahirinus are awake at dark onset, but sleep more than M. musculus after the middle of the night. Most strikingly, A. cahirinus do not close their eyes virtually at all while sleeping, day or night. In order to test whether the sleep patterns of A. cahirinus are affected by or responsive to different light input, we set up a light flashing experiment during the daytime. While sleep amount did not change significantly during light flashing, A. cahirinus spent significantly less time in REM compared to baseline. In contrast, M. musculus had no difference in REM sleep percentage. Histological studies showed A. cahirinus have thinner retinal layers, but much thicker corneas than M. musculus. Electroretinography (ERG) results, specifically b-wave amplitudes, are significantly different between these two species. While eye closure and sleep have not been systematically studied across mammals, our observation is clearly a rare behavior. This raises further questions about A. cahirinus sleep architecture, the adaptive value of eyes open sleep to A. cahirius and whether they may have limited visual processing even during normal sleep.

Regulatory T Cells Promote Breast Cancer Progression Through Inhibiting Classical Activation of Macrophages

Transient ablation of regulatory T cells has been shown to be effective at hindering tumor growth and metastasis in murine breast cancer model. Based on our lab’s previous work, we have demonstrated that NK cells and CD8+ cytotoxic T cells were not required for the protective effect of Treg cell ablation. However, we also reported that CD4+ helper T cells and IFN-γ were required for the protective effect of Treg cell ablation. Furthermore, we observed that CD11B+ cells responded to Treg ablation therapy by up-regulating target genes of IFN-γ. Therefore, this study aimed to investigate the connection between the myeloid cell compartment and IFN-γ signaling after regulatory T cell ablation therapy. Through a combination of conditional knockout mouse models, cellular fate mapping experiments, adoptive transfers, and co-injection experiments, we demonstrated that tumor-associated macrophages (TAMs), derived from the bone marrow via CCR2/CCL2 axis, were responsible for the therapeutic effect of regulatory T cell ablation. In addition, we determined that IFN-γ signaling was required for the TAMs to mediate the protective phenotype seen after regulatory T cell ablation. Furthermore, based on our findings, we developed a genetic signature based on TAMs from treated or untreated tumors that had a predictive value for patient survival. Thus, our findings indicated a strong connection between IFN-γ release, classical activation of TAMs, and depletion of regulatory T cells, and taken together, our data could offer potential clinical strategies to mimic regulatory T cell ablation.

An Investigation of the effects of increased tidal inundation, competition, and facilitation on salt marsh systems

The low-lying topographic nature of salt marshes makes plants in these communities particularly vulnerable to increased salinity and inundation exposure associated with sea level rise. Both increased salinity and inundation have been cited as major causes of reduced plant performance and survival in marsh and areas fringing marsh. In addition to limitations imposed by physical stress, interspecific interactions have also been shown to mediate the performance and survival of salt marsh and salt marsh fringing species. The Stress Gradient Hypothesis (SGH) postulates that species interactions shift from competitive to facilitative as stress levels increase and predicts that (a) the frequency and intensity of facilitative interactions increase as conditions become more stressful for plants and (b) the strength of competitive interactions increases as abiotic stress levels diminish. The SGH has been rigorously tested to examine how both the frequency and intensity of species interactions change under varying physical stress levels. Studies conducted in salt marsh systems have shown facilitation to be as strong of a driving force as competition in influencing plant performance and survival and have shown that while competition appears to be the pervasive force in the less physically stressful terrestrial zones fringing salt marshes, facilitation influences the performance and survival of species in harsher marsh areas. Under conditions of sea level rise, it remains unclear if the nature of interspecific interactions would shift as stress levels change. This research endeavors to examine the interplay between abiotic stresses and biotic interactions under conditions of increased salinity and inundation exposure. The first study presented here investigated the effects of increased inundation and soil salinity associated with sea level rise on four salt marsh fringing species, and assesses how competition and facilitation impact survival of salt marsh fringing plant survival under these changing conditions. All plant species experienced reduced growth and photosynthetic inhibition below their current distributional positions, both in the presence and absence of neighboring above ground vegetation. The findings also signal a potential shift in the nature of interspecific interactions from competition to facilitation to neutral as plants begin to experience increased salt and inundation exposure. The second study aimed to disentangle the effects of increased soil salinity and increased soil moisture on four salt marsh fringing species, and to examine the effects of plant neighbors. The results showed that fringe plants exposed to increased inundation experienced a two-fold reduction in performance and survival over 750 g pure salt addition, suggesting that inundation may be a more important limiting factor than salinity with rising sea levels. Landward transplants at the forest-fringe margin exposed to lower soil salinity and decreased inundation exhibited a three-fold increase in performance and survival when compared to controls. Neighbor manipulation studies, which consisted of trimming neighboring vegetation to ground level, again suggested that interspecific interactions in salt marsh fringing species may shift from competitive to facilitative with climate-induced sea level rise. Overall, our findings suggest that salt marsh fringing species may not be able to tolerate changing conditions associated with sea level rise and their survival may hinge on their ability to migrate towards higher elevations. The final experiment tested the Stress Gradient Hypothesis and investigated the relative importance of facilitation and competition in a salt marsh system under varying stress levels. This study also ascertained whether salt or inundation exposure is the primary influence on salt marsh plant performance and survival. As in previous studies, our findings suggest that many salt marsh plants don't require, but merely tolerate harsher abiotic conditions. The results showed that plants at higher elevations were depressed by strong competitive pressure from neighboring fringe species while plants at lower elevations benefited from the presence of neighbors. Collectively, the results of these studies indicate that species interactions are an integral driver of plant distribution in salt marsh communities. Furthermore, our findings indicate that changing stress levels may not always result in a shift in the nature of interspecific interactions. These studies have endeavored to show that the interplay between competition and facilitation interacts with physical processes to determine the growth and performance of both fringe and marsh plant species. The paucity of studies examining the roles of species interactions and changing abiotic stress levels on multiple salt marsh and salt marsh fringing species warrants the need for additional research. The responses of salt marsh and salt marsh fringing species to sea level rise can not only serve as very valuable and sensitive indictors of climate change, but will also aid in predicting the future location of the marsh-fringe-forest ecotone, which is predicted to shift inland as sea levels continue to rise.

Applicability of the Integrative Workplace Health Management (IWHM) Model in Taiwan

Over the past three decades, globalisation and rapid technological advances have fundamentally changed socio-economic structure and have widespread impacts on the nature of work and workplace health (U. Beck, 2000; Bertucci & Alberti, 2004; Bhalla, 1996; Chu & Dwyer, 2002). They have led to fierce global competition, altered the nature of work and exposed employees to new health risks (Chu & Dwyer, 2002; Stitzel & Jarvisalo, 1997). Fierce market competition demands enterprises and industries to build new competencies, restructure and improve productivity and efficiency. Associated with these changes are increased work pressures, weakening commitment to occupational health and safety, and many negative impacts on workers’ health (G. Breucker, 2006; Missler & Theuringer, 2003; WHO, 2003). Evidence shows that the world is facing growing burden of work related fatalities, injuries and diseases, particularly a dramatic increase of work stress. As part of the global community, Taiwan has also encountered similar workplace challenges in the rapidly changing environment. It also has to confront with increasing costs from the burden of work-related injuries and diseases and to find appropriate ways to deal with the serious problems (Council of Labour Affairs Taiwan, 2003, 3005; IOSH, 2002). As the success of organisations relies on having well-qualified, motivated and healthy employees, it is essential to seek effective means to protect and promote the health of the working population (ENWHP, 2005). The integrative workplace health promotion (IWHM) model emerging in the 1990s, is regarded as a comprehensive means to address multiple determinants of health and promote employee health (G. Breucker, 2006; Chu, 2003b; WHO-WPRO, 1999). There have been a great number of international successful examples demonstrating the benefits from implementing the IWHM programs (ENWHP, 2002; Chu, Breucker, Harris, & et al., 2000). In response to these workplace challenges and new occupational health risks, Taiwan has followed the international trend to initiate a series of workplace health promotion (WHP) programs since 2001 (Bureau of Health Promotion Taiwan, 2006). However, a preliminary study revealed that many WHP projects in Taiwan have met with difficulties in encouraging employee participation and sustainable development of the programs. These results were not surprising as the majority of the WHP programs in Taiwan tended to narrowly focused on physical activities, weight control and smoking cessation, while ignoring employee needs and problems relevant to specific workplace concerns (Bureau of Health Promotion, 2003; Hsu, Chang, Peng, & Chen, 2002; Hsu, Chen, & Wu, 2004). In this regard, the comprehensive IWHM which aims to meet employee health needs and improve organisational environment may present an effective means for Taiwan to address complex workplace health issues and to create healthy and sustainable workplaces. This research aims to investigate the applicability of the IWHM model in Taiwan to deal with workplace health problems. Apart from reviewing literature and relevant case studies from international communities and in Taiwan, this study conducted a small scale pilot study and a comprehensive needs assessment at selected workplaces in Taiwan as an experiment to test the applicability of the IWHM model. In practice, this research project targeted the civil servants working at the Liming governmental community in Taichung, Taiwan as the research subject. It involves a combination of qualitative and quantitative methods for data collection and analysis. A variety of data collection techniques including in-depth interviews, focus groups, participant observations, secondary data analysis and questionnaire surveys are used to investigate the workplace health and safety problems and the staff’s health needs. A triangulation technique is used to compare and contrast the different sources of information. The findings indicated that the IWHM model is applicable in the Liming community in terms of feasible methodological approaches using a needs-based program development and implementation process, and positive program outcomes such as stair improvement to prevent falls and the establishment of long-term health management, meaningful participation, practical program activities and holistic evaluation. For potential program sustainability, this project has involved employee representatives in program implementation and taking in charge of the ongoing program activities, and has managed to establish interdepartmental partnership to address common workplace health issues. Even though this research project has not made great progress, it has been moving toward meeting the project objectives and the organisations’ needs. From the Liming experience, this research provided recommendations for future development of WHP in Taiwan including: 1) develop national policies to define clear responsibilities in WHP development at different levels; 2) establish appropriate capacity building and training programs; 3) develop practical guidelines and tools tailored to suit Taiwan’s local conditions and needs; 4) set up effective evaluation and quality management system; 5) provide a professional or technical support team to assist workplaces or industries with the development and implementation of WHP programs.

The method of system approach is utilized to find out common problems of those existing solutions and to form an integrative solution. A model for this integrative solution is built under PowerSIM environment and the main objective of the model is to simulate the energy balance between power supply and demand. Finally, a brief discussion mainly focuses on the social and economic impact of the solution is made under PEBOSCA framework.

Planning for Reuse and Redevelopment of Inner City Blighted Contaminated Industrial Sites

Inner city blighted industrial sites are primarily associated with the general phenomenon of deindustrialization within the post-industrial inner city that is caused by spatial and functional restructuring of the industrial activity. Blighted industrial sites are vacant, obsolete, or underutilized industrial buildings, facilities and other related industrial functions and areas. Such blighted areas may have physical, functional, social, economic, and environmental impact problems both within the site, as well as on surrounding properties. Brownfield sites represent a severe form of blighted sites which include real or perceived environmental contamination. Brownfield redevelopment requires environmental clean-up to acceptable regulatory standards which may result in a costly process. The general research goal is to achieve an understanding of this complex problem context as well as to establish the pertinent planning framework for redevelopment of blighted industrial sites within the Canadian inner city. The research process includes three parts. Part One includes an extensive preliminary literature review of brownfield redevelopment case studies in Canada, United States, and the United Kingdom. It also includes an outline of general planning theory and related interdisciplinary theories. The preliminary findings of literature review reveal a multiple component interactive problem context that indicates a need for an integrative planning framework addressing multiple problem components. The preliminary research findings for the planning framework are further studied and examined in four empirical case studies included in Part Two. The specific research objectives and research questions address three constituent parts of the planning framework including the nature of the problem context, potential policy directions, and the planning process. The central research question is “what is the appropriate planning framework and approach for brownfield redevelopment given a multiple component interactive problem context? Part Three synthesizes the findings of Parts One and Two, which highlights the proposed planning framework for brownfield redevelopment, including an outline of major problems and policy directions based on impact evaluation by key participants in the empirical case studies, as well as outlining the main characteristics of the planning process. Some of the main problems and issues include site contamination and related legal liabilities, cost of site remediation that may exceed property value, stakeholders’ conflicting objectives, social stigma associated with brownfields, clarity and consistency of the environmental approval process as, community support. The proposed policy directions also represent multiple components and they are mainly as follows: • To prepare an inventory of brownfield sites, • To establish public-private partnership for project financing especially for site remediation, • To develop self-financing mechanism (like TIF/TIEF) to finance cost of site remediation, • To establish a redevelopment authority that is directly responsible for the process, • To foster public-private-community collaboration and partnership, • To secure accessibility of local residents to newly provided opportunities (like jobs), • To adopt multiple-component integrative planning framework to link major problem components and planning sub-processes The research findings also highlight the planning process being manifested at two main poles including the project developer and his consulting team, the public approval authority, in addition to community residents and interest groups that are involved in the public consultation process. The development approval process is the common organizational set-up and interface for stakeholders’ involvement in the process. The main characteristics of the proposed planning process include multiple-component multi-disciplinary context consisting of interactive planning sub-processes within each component, multi-level spatial contexts, involvement of multi-stakeholders with conflicting objectives and vision, incrementally adaptive, critical time and timing context, and mixed-rationality comprehensive planning vision. This research asserts the need for addressing the multiple components of environmental, physical, economic, social, and political planning without prior bias or predominance to any of these components. This also asserts the need for multi-stakeholder public-private-community collaboration and partnership.
