Min studie undersöker en nyligen införd en-till-en-satsning i en svensk gymnasieskola, med tonvikt på upplevda utmaningar och möjligheter, liksom förhoppningar inför framtiden. Den metod jag valt är kvalitativa intervjuer med fem lärare, var och en slumpmässigt vald från varje utbildningsprogram skolan erbjuder. Studiens primära resultat är att lärarna verkar nöjda med satsningen, och tror den kommer att leda till stora fördelar i framtiden. De tror den kommer att leda till mer effektiva lektioner. Det finns dock en farhåga att vissa elever inte kommer att klara av att motstå frestelsen att använda sitt nya verktyg som en slags digital distraktion, och att de kommer att hamna efter som ett resultat av detta. Det finns också reservationer gällande vissa delar av implementationen. Informanterna hade gärna sett tidiga utbildningsinsatser för att lyfta alla lärare till en nivå där de hade kunnat utnyttja det nya systemet fullt ut. De önskar också att alla hårdvarukomponenter hade fungerat som de ska redan från början. Vissa viktiga delar har varit oanvändbara i nästan ett år. Dessa problem hade kunnat undvikas med bättre planering. Tidiga utbildningsinsatser, med tester av all nödvändig hårdvara, hade lett till en avsevärt bättre upplevelse för alla. / My study aspires to assess a recently implemented one-to-one computer initiative in a Swedish upper secondary school, with emphasis on perceived challenges and possibilities, as well as hopes for the future. The method chosen involves qualitative interviews with five teachers, randomly selected from each programme the school offers. The primary result of this study is that teachers seem satisfied with the initiative, and believe it will lead to great benefits in the future. They believe it will lead to more efficient lessons. There is a fear however, that some students will be unable to resist the temptation to use their new tool as a means of digital distraction, and that they will fall behind as a result. Also, there are reservations about parts of the implementation. The interviewees wish there had been early educational efforts to bring all teachers up to a level where they could use the system fully. They also wish all hardware components had worked as intended from the start. Some important parts have been unusable for almost a year. These problems could have been avoided with better planning. Early educational efforts, including testing of all necessary hardware, would have made for a substantially better experience for all.

The Spruce Point Site (DjKq-1) : A late woodland community pattern and cultural assemblage from Northwestern Ontario and their relationships within the Selkirk compositeRajnovich, M. Grace N. 27 March 2014 (has links)
The Spruce Point Site (DjKq-1) in the northwestern sector of Lake of the Woods,
Ontario, is a Selkirk site dated through ceramic seriational analysis to the sixteenth or
seventeenth century A.D. It is rare for two reasons. First, it is a single component among
a plethora of multi-component sites with collapsed stratigraphy which
characterize the major portion of the Lake of the Woods prehistoric record. This
factor allows for the first analysis and description of Lake of the Woods Selkirk
community patterns, material remains, especially ceramics, and adaptive strategy free from
stratigraphic interference. The assemblage is represented by ceramics and lithics akin
to the Winnipeg River Complex and a faunal sample indicating an
exploitation pattern using an unspecialized strategy and varied resources. Second,
the site has remains of two house structures, unreported elsewhere for the Selkirk
Composite, that are similar in floor plan and size to earlier Laurel structures and later Cree
houses reported in the ethnographic literature. House style and geographic location,
plus the faunal assemblage indicate the site was occupied during the summer and was chosen for
its varied animal, plant and lithic resources.

The Apostle Paul and homosexuality : a socio-historical study / Petrus Hendrik BothaBotha, Petrus Hendrik January 2004 (has links)
Like many other people I have thought about the biblical understanding of homosexuality.
Because of my previous study on sexual purity before marriage and the exegetical work
done on key biblical passages for that study, I thought I understood where the real issues
lay regarding the subject of homosexuality. This study is an attempt to contribute to the
current discussion in the Dutch Reformed Church in South Africa on the topic of
homosexuality.
In bringing this study to its final format I have received the indispensable help of others.
First and foremost I wish to express my gratitude to my promoter, Professor Fika J van
Rensburg. I am indebted to him beyond what mere words could convey in terms of
gratitude. Our academic relationship spans five years and has developed into a warm
friendship. It has been to my matchless benefit to be associated with him. I have profited

Roles of the DOG-1 and JRH-1 helicase-like proteins in DNA repair in Caenorhabditis elegansYouds, Jillian L. 05 1900 (has links)
Helicases perform vital roles in the cell by unwinding D N A to make it accessible for the
essential processes of replication, transcription and repair. In Caenorhabditis elegans, the DOG-
1 helicase-like protein is required for polyG/polyC-tract (G/C-tract) maintenance, as dog-l
animals have a mutator phenotype characterized by deletions that initiate in G/C-tracts. DOG-1
may unwind secondary structures that form in polyguanine D N A during lagging strand
replication. In order to more completely understand the role of dog-1, genetic interactors were
identified, dog-1 functionally interacts with the him-6/BLM helicase. Absence of
recombinational repair-implicated proteins in the dog-1 background, including HIM-6/BLM,
RAD-51, BRD-1/BARD1 and HIM-9/XPF, as well as the trans-lesion synthesis polymerases
polKMD po/7 increased the frequency of animals with G/C-tract deletions, indicating that these
pathways are important mechanisms for repair at G/C-tracts in the absence of DOG-1. These
data support the hypothesis that persisting D N A secondary structures can cause replication fork
stalling, which can be resolved by deletion-free or deletion-prone mechanisms.
DOG-1 has highest sequence identity to human BR1P1/FANCJ, which is mutated in
patients from the Fanconi Anemia (FA) subgroup J. D N A damage sensitivity experiments
indicated that, like chicken F A N C J cells, dog-1 mutants were not significantly sensitive to DNA
damage from X-ray or UV-irradiation, but were extremely hypersensitive to the D N A interstrand
cross-linking agent UVA-activated trimethylpsoralen. Thus, DOG-1 appears to have a
conserved role in cross-link repair and is the C. elegans F A N C J homolog. Characterization of
the dog-1/FANCJ-relatsd helicase, Jrh-1, revealed that mutants for this putative helicase are
moderately sensitive to cross-linking agents, dog-1 jrh-1 double mutants displayed a synthetic
lethal phenotype characterized by excessive recombination intermediates and mitotic catastrophe
in the germline. However, absence of JRH-1 did not have any effect on G/C-tract deletions,
indicating that JRH-1 does not have a redundant function with DOG-1 at G/C-tracts. Absence of
JRH-1 reduced the fitness of eTl and nTl translocation hétérozygotes, but not translocation
homozygotes, jrh-1 was synthetically lethal with him-6/BLM and with the endonuclease mus-81,
suggesting a possible role for JRH-1 in regulating the balance between different types of repair.

Quadrivalent HPV vaccine and the risk of type 1 diabetes mellitus in grade 8 girls: A population based cohort studyWalsh, Erica 29 May 2012 (has links)
Background:
Vaccines have been hypothesized in the etiology of autoimmune diseases including type 1 diabetes. There are cases of diabetes reported in the Vaccine Adverse Event Reporting System (VAERS) following the administration of the human papillomavirus (HPV) vaccine, however this potential association has yet to be investigated. The objective of this thesis was to determine whether there is an association between immunization against HPV and the development of type 1 diabetes mellitus in grade 8 girls eligible for Ontario’s vaccination program.
Methods:
A retrospective, population-based, cohort study of girls residing within an Ontario health unit and eligible for the province’s publicly funded school-based HPV vaccination program between 2007 and 2010 was executed using provincial administrative health databases and the Immunization Recording Information System (IRIS) database. To control for known, unknown and unmeasured time-independent confounders, a self-controlled case series analysis was conducted. The relative incidence and 95% confidence interval were estimated using conditional Poisson regression.
Results:
The study cohort was comprised of 3465 girls with a mean age of 13.2 years at cohort entry (range 12.7 to 13.6 years). The mean duration of follow up was 2.7 years and ranged from 1.6 to 3.6 years. The proportion of girls who received at least one dose of the qHPV vaccine during the observation period was 58.3% (n=2020). During the study follow-up 15 cases of new onset type 1 diabetes were observed, six of which were classified as etiologically exposed to the qHPV vaccine. Using an indefinite risk window, immunization with the qHPV vaccine was not associated with an increased risk of developing type 1 diabetes (age and season-adjusted RI 0.15; 95% CI 0.02-1.32).
Conclusions:
The results of this thesis regarding the risk of type 1 diabetes following immunization with the qHPV vaccine are inconclusive as a consequence of the small number of cases identified. However, the random distribution of cases across time and across exposure status suggests that there is no association. REGULATION OF THE GABPβ PROMOTER AND THE ROLE OF NRF-1 IN MAMMARY EPITHELIAL MORPHOGENESISLamparter, CHRISTINA 07 September 2012 (has links)
Decreased expression of the tumor suppressor gene BRCA1 (breast cancer 1, early onset) is frequently observed in sporadic breast tumors, with the decrease not attributed to mutations or hypermethylation of the promoter. A critical regulator of the BRCA1 promoter is the GA-Binding Protein (GABP), a heterotetramer of GABPα and GABPβ. Previous analysis of the GABPβ promoter revealed a regulatory multi-protein complex containing Nuclear Respiratory Factor 1 (NRF-1), which was aberrant in SK-BR-3 cells, resulting in decreased GABPβ and BRCA1 expression. To identify the unknown co-regulators of the NRF-1-containing complex at the GABPβ promoter, an immobilized-template assay containing the complex binding site was utilized. Despite optimization of binding and elution conditions through variations in salt content, Mg2+ concentration and pH, non-specific DNA-binding proteins were present in the column eluate. Further experimentation is therefore required to distinguish between non-specific proteins and complex co-activators. As this complex containing NRF-1 is also able to modulate BRCA1, a regulator of luminal progenitor differentiation, a defect in NRF-1 or complex co-activators could contribute to the abnormal morphogenesis and differentiation of BRCA1-deficient tumors. We therefore investigated the role of NRF-1 in mammary cell morphogenesis and its link to the GABP/BRCA1 pathway. As both GABPβ and NRF-1 are known regulators of nuclear-encoded mitochondrial proteins, this association also provides a link between mitochondrial metabolism and breast differentiation, both of which are frequently disrupted in breast cancer. An inducible lentiviral system was used to generate NRF-1 knockdown cell lines to examine its effect on morphogenesis. Growth in 3D culture resulted in abnormal cell structures having impaired cell polarization and lumen formation. In monolayer culture, prolonged NRF-1 knockdown did not result in decreased BRCA1 or GABP expression. However, these cells did display notable mitochondrial dysfunction, accompanied by the downregulation of several NRF-1 target genes involved in mitochondrial biogenesis including Tfam and cytochrome c. These results suggest a role for NRF-1 in mediating mammary morphogenesis through maintenance of functional mitochondria. Further investigation into the role of the NRF-1-containing complex at the GABPβ promoter during differentiation might also provide insight into the altered cell metabolism and differentiation observed in cancer cells. / Thesis (Master, Biochemistry) -- Queen's University, 2012-09-06 14:39:04.852
