The main concern of this thesis is examining the affects and causes of traffic oriented design on the social generation of boulevards in terms of communication. For this purpose, boulevard concept and its historical evaluation are explained. In addition, communication is used as a tool for understanding, combining and examining the social and technical structure of boulevard.
In this respect, this thesis aims to explore the boulevard concept with the spatial communication measures. To provide empirical evidence, a chart of categories of communication is prepared in terms of human and machine interposed aspects of communication. The activities of relation along the boulevard are examined by transversal and longitudinal movements with the help of the behavioral sciences and proxemics and the theoretical relation between boulevard and communication is applied on the case of The Atatü / rk Boulevard, Ankara. It is observed from the findings that, longitudinal movement prior to situated activity is increased, the transversal movement prior to random activity is decreased. This causes the loss of human aspects along the boulevard, which is designed for human.

Visualization could be defined as the graphical presentation of information, in which the main aim is to improve the user&rsquo / s perception. In all phases of the disaster management, decision makers come across huge data sets with spatio-temporal content. It is hard to deal with these sets in order to find answers to the main question of &ldquo / How can we decrease the losses due to disasters?&rdquo / , which is at the core of the disaster management concept. To furnish this aim, disaster risk information has to be transparent and clearly stated to the public, decision makers and disaster managers. This might be more sophisticated than the calculation of the risk.
Taking precautions before a disaster to reduce the causalities and lossess engendered by natural disasters is relatively cheaper, and more importantly, better than cure. To achieve enhanced preparations for all kinds of disasters, visualization is quite an important tool for decision support and risk communication. The basic aim of this research is to propose a conceptual framework, with the consideration of all stakeholders related to the disaster management issue to have a better risk communication, and to guide the design, implementation and integration of the 3D urban modeling tools into disaster risk visualization. Moreover, an empirical methodology is also developed for the generation of visualization solutions through the design, and employment of the tool for disaster management framework. The proposed framework has three main phases .These are the definition of visualization components, object representation, and needs assessment. A new LoD hierarchy with indoor is proposed to visualize all the possible 3D urban disaster situations in the first phase. Then, a decision rule with eight attributes is proposed in the second phase to establish a link between the hazard type and the LoD needed in a 3D urban model for visualization. This decision rule is applied in a proposed three-level hierarchycal structure. The assessed objects of these three levels are urban, sub-urban zone and building. Moreover, a method to define the needed sub-urban zone is proposed. Finally, different 3D urban modelling methods are analyzed to define the data and process needs of possible 3D urban disaster visualization situations.
Two natural hazard cases are studied within the scope of this dissertation to assess the operability of the proposed framework. These implementations involve one earthquake and one tsunami case. Special attention is paid to finding one specific sample for two modelling viewpoints, namely static and dynamic. The first applications of the proposed framework with all the related features prove quite promising.

1,25(OH)2D3 and Initial Regulation of Smad2/3 Activity in PC-3 Prostate Cancer CellsStahel, Anette January 2009 (has links)
<p>The vitamin D metabolite 1,25(OH)2D3 has long been known to inhibit growth of prostate cancer cells and this mainly through a VDR-mediated pathway controlling target gene expression, resulting in cell cycle arrest, apoptosis and differentiation. Another major way in which 1,25(OH)2D3 inhibits cell growth in prostate cancer is via membrane-initiated steroid signalling, which triggers activation of signal cascades upon steroid binding to a receptor complex, leading to induction of genes regulating cell growth, proliferation and apoptosis. The main prostate cancer inhibiting membrane-initiated route is the TGFβ signalling pathway, elicited by the protein TGFβ. Two other important proteins downstream in this cascade are Smad2 and Smad3. In this study the early effects of 1,25(OH)2D3 on activated Smad2/3 levelsin PC-3 prostate cancer cells were examined. PC-3 cells were incubated for 3, 5, 10, 30 and 60 minutes as well as 38 hours both together with 1,25(OH)2D3 of the concentrations 10-10 and 10-7 M and without. Western Blots were then performed on supernatants from the cells treated followed by treatment of the membranes with primary antibodies against phosphorylated Smad2/3 C-terminal linker regions, alkaline phosphatase conjugated secondary antibodies and finally visualization with BCIP/ NBT tablets. As the downstream cascade protein JNK is a proposed activator of Smad2/3, this procedure was also repeated with a JNK inhibitor. This is a follow-up to an earlier study which examined the influence of 1,25(OH)2D3 on TGFβ levels using the same doses and time points and which found that 1,25(OH)2D3 initially lowered the level of active TGFβ, then increased it. The results of this study indicated a 1,25(OH)2D3 mediated induction of the same pattern in the levels of active Smad2 and 3, both with and without JNK inhibitor. The results did not indicate that 1,25(OH)2D3 activates the Smad2/3 C-terminal linker region via the JNK pathway.</p>

Αντικείμενο της παρούσας Διατριβής Διπλώματος Ειδίκευσης είναι η κατά βάση πειραματική διερεύνηση της αποτελεσματικότητας περίσφιγξης ορθογωνικών υποστυλωμάτων με μεγάλο λόγο πλευρών, ενισχυμένων με μανδύες ινοπλισμένων πολυμερών και σύνθετων υλικών ανόργανης μήτρας. Το πειραματικό πρόγραμμα διεξήχθη στο Εργαστήριο Μηχανικής και Τεχνολογίας Υλικών του Τμήματος Πολιτικών Μηχανικών του Πανεπιστημίου Πατρών.
Το πρόγραμμα αυτό, περιελάμβανε δύο σειρές δοκιμίων. Η πρώτη περιελάμβανε έξι δοκίμια και η δεύτερη δέκα. Η κατηγοριοποίηση σε σειρές έγινε με βάση το λόγο πλευρών των δοκιμίων. Έτσι, την πρώτη σειρά αποτέλεσαν δοκίμια με λόγο πλευρών (3:1), ενώ τη δεύτερη δοκίμια με λόγο πλευρών (4:1). Ένα δοκίμιο από κάθε σειρά δοκιμάσθηκε χωρίς ενίσχυση και αποτέλεσε μέτρο σύγκρισης για όλα τα υπόλοιπα. Τρία δοκίμια από κάθε σειρά ενισχύθηκαν με τρείς στρώσεις FRP και με θυσάνους. Ακόμα, ένα δοκίμιο από κάθε σειρά ενισχύθηκε με δύο στρώσεις FRP, χωρίς χρήση θυσάνου. Επίσης, δύο υποστυλώματα από τη δεύτερη σειρά ενισχύθηκαν με δύο στρώσεις FRP, θυσάνους και δύο επιπρόσθετες στρώσεις FRP τύπου U, στις δύο μικρές πλευρές. Τέλος, ένα δοκίμιο από κάθε σειρά ενισχύθηκε με τέσσερις στρώσεις TRΜ, ενώ άλλο ένα με τέσσερις στρώσεις TRΜ και με θυσάνους. / The subject of this thesis is the experimental investigation of the effectiveness of confining rectangular columns with large aspect ratio, reinforced with fiber reinforced polymers and with tensile reinforced mortars. The experimental program was conducted at the Laboratory of Engineering and Technology of Materials in Civil Engineering, University of Patras.
This program is consisted of two sets of samples. The first included six small scale columns and the second ten. The categorization in series was based on the aspect ratio of the specimens. So specimens with aspect ratio (3: 1) were included in the first series, while the second included specimens with aspect ratio (4: 1). One specimen from each series was tested without any reinforcement and became the comparison for all the rest. Three specimens from each series were reinforced with three layers of FRP and anchors. Still, a sample from each series was reinforced with two layers of FRP, without use of anchors. Also, two columns of the second series of amplified with two layers FRP, anchors and two additional layers of FRP type U, in the two smaller sides. Finally, a sample from each series was reinforced with four layers TRM, while another one to four layers TRM and anchors.

The role of Vitamin D metabolic enzymes in bone development and repair /Naja, Roy Pascal. January 2008 (has links)
The CYP27B1 enzyme that synthesizes 1alpha,25-(OH) 2D, is expressed in chondrocytes, suggesting that local production of 1alpha,25-(OH)2D could play an autocrine or paracrine role in the differentiation of these cells. To test this hypothesis, we have engineered mutant mice that do not express the Cyp27b1 gene in chondrocytes. This led to increased width of the hypertrophic zone of the growth plate at E15.5, increased bone mass in neonatal long bones, and increased expression of the chondrocytic differentiation markers Indian Hedgehog and PTH/PTHrP receptor. VEGF mRNA levels were decreased, accompanied by decreased PECAM-1 immunostaining, suggesting a delay in vascularization. We have also engineered mice overexpressing a Cyp27b1 transgene in chondrocytes. The transgenic mice showed a partial mirror image phenotype compared to the tissue-specific inactivation model. These results support an autocrine/paracrine role of 1alpha,25-(OH) 2D in endochondral ossification and chondrocyte development in vivo. / The CYP24A1 enzyme is involved in the catabolic breakdown of 1alpha,25-(OH)2D but also synthesizes the 24R,25-(OH) 2D metabolite. Studies in chicken suggest a role for 24R,25-(OH) 2D in fracture repair. We induced stabilized transverse mid-diaphysial fractures of the tibia in four-month-old wild-type and Cyp24a1-deficient mice. Examination of the callus sections showed delayed hard callus formation in the homozygous mutant animals compared to wild-type littermates. RT-qPCR showed perturbed levels of type X collagen transcripts in mutant mice at 14 and 21 days post-fracture, reflecting the delayed healing. Rescue of the impaired healing by subcutaneous injection of 24R,25-(OH)2D3 normalized the histological appearance of the callus, static histomorphometric index and type X collagen mRNA expression, while 1alpha,25-(OH)2D 3 did not. These results show that Cyp24a1 deficiency delays fracture repair and strongly suggest that vitamin D metabolites hydroxylated at position 24, such as 24R,25-(OH)2D, play an important role in the mechanisms leading to normal fracture healing.
