Estudo da imunidade celular e humoral em potros recem-nascidos

Orientador: Euripedes Ferreira / Dissertação (mestrado) - Universidade Federal do Parana, Setor de Ciencias Agrarias / Resumo: Para um estudo dos parâmetros normais das condições imunológicas de potros PSI recém-nascidos até um seguimento de 120 dias, realizou-se a colheita e processamento de sangue periférico, para posterior obtenção de soro e linfócitos do mesmo. Para estabelecer níveis de imunoglobulinas da amostragem do soro obtido dos animais hígidos ( 10), realizou-se a determinação da proteína total, eletroforese de proteínas, e imunodifusão&o radial simples, verificando-se que os potros recém-nascidos são agamaglobulinêmicos ao nascer, apresentando apenas uma pequena quantidade de IgM. Já às 24 horas, as IgG, M e A dos potros apresentam níveis semelhantes aos do soro da égua, sendo que em todo o período de observação ( 120 dias) cada uma das imunoglobulinas tem um comportamento diferente. Os níveis de IgG após 24 horas até ás 72 horas permanecem em torno de 3450,6mg/dl, caindo ao final do primeiro mês para 2017,5 mg/dl, enquanto que no segundo, no terceiro e no quarto mês ficam em torno de 1406 mg/dl. A IgM dos potros, ao nascer, está em torno de 19,60 mg/dl indo a 46,26 mg/dl nas primeiras 72 horas, sofrendo um pequeno decréscimo ao primeiro mês ( 40,52 mg/dl ), estabilizando-se ao término do período de observação em torno de 81,78 mg/dl. Já a IgA, inexistente ao nascer, apresenta-se em torno de 75 mg/dl nas primeiras 72 horas, revelando uma queda no primeiro mês (42,4 mg/dl), finalizando o período de observação com 77,07 mg/dl. Para estabelecer os parâmetros normais das subpopulações linfocitárias T e B, usou-se técnicas de marcadores de membranas. Quanto aos linfócitos T, com a formação de rosetas com eritrócitos heterógenos, determinou-se que os mesmos, apresentanse em torno de 3,4% a 10,8% em todo período de observação. Já os linfócitos B mostraram-se, através de técnicas de fluorescência, em porcentagens altas nos primeiros dias de vida (aproximadamente 37,95%) e após o 3Qq dia os valores estabilizaram-se entre 10,7% a 18,1%. / Abstract: The present study was undertaken in order to stablish the normal immunological parameters, under normal conditions, of foal as newborn until 120 days of life. The humoral immunity was assessed throug the assay of total protein in the serum, by electrophoresis and simple radial immunedifusion. It was found that newborn foal has agammaglobulinemia soon after birth, with a very low level of IgM. However, after 24 hours, the serum levels of IgG, IgA and IgM are similar as the serum levels of its mother. The behavior of different immunoglobulins in the subsequent 120 days is quite different for each immunoglobulin. The IgG level between 24 and 72 hours is around 3450.6mg/dl, in the first month, falling to 2017.5mg/dl and by the second and third months is maintained at 1406m/dl. The IgM level at birth is around 19.60mg/dl, raising to 46.26mg/dl in the first 24 hours, falling to 40.52mg/dl by the first month and, again, raising and stabilising at 81.78mg/dl at 120 days. The IgA, not present at birth, raises to 75mg/dl in the next 72 hours, falling to 42.4mg/dl in the first month and. stabilising at 77.07mg/dl by the 120 days. The T and B lymphocytes are determined by using specific surface marker of each sub-population. The T lymphocytes were identified through rosette formation, using guinea pig erythrocytes. The T lymphocytes ranged from 3.4% to 10.8% during the different periods of this study. The B lymphocytes were identified by detecting immunoglobulins on its surface. Antihorse immunoglobulin conjugated with fluorescein was employed. A very high percentage of B lymphocytes (37.95%) was detected in the first days of life and after one month the percentage ranged between 10.7 and 18,1%
