Developing a high temperature, oxidation resistant molybdenum-silica composite

A new powder processing approach to produce oxidation resistant molybdenum alloys for high temperature use has been developed. Oxidation protection is provided by fine dispersion of silica glass particles within a molybdenum matrix. As the molybdenum oxidizes, the glass is exposed and melts to form a self-healing protective oxide coating. Additionally, homogeneously dispersed Mo5SiB2 and/or Mo2B provide boria upon oxidation which reduces glass viscosity and allows flowing glass to coat the surface while remaining solid internally. This is similar to the oxidation protection used in Mo-3Si-1B (wt%) systems; however embedding the glass directly into the Mo matrix and eliminating the Mo3Si (A15) phase provides the same volume of glass at lower volume fractions of brittle phases and also without embrittling Si impurities in solution in Mo. Additionally the glass composition can be tailored for different applications and different temperatures beyond that achievable in Mo-Si-B based systems. A variety of microstructures, compositions and additional components for improved oxidation protection are also explored, and mechanisms of the oxidation protection are discussed.

Metab-Immune analysis of the non-obese diabetic mouse

Type 1A diabetes mellitus or T1D is a chronic disease characterized by T cell mediated destruction of the insulin producing β cells in the islets of Langerhans. The classical symptoms include high glucose levels in urine and blood, polyuria, and polydipsia. Complications associated with T1D include blindness, amputations, and end-stage renal disease, and premature death. The non-obese diabetic (NOD) mouse, first described in 1980, is widely used as a model organism for T1D. T1D disease in the NOD mouse shares a number of similarities to human T1D including dependence on genetic and environmental factors. More than 30 disease associated gene regions or loci (termed insulin dependent diabetes, or Idd, loci) have been associated with T1D development in NOD. For some of these Idds, the corresponding region in human has been linked to the development of T1D in human. T1D, both in humans and mice, is recognized as a T cell mediated disease. However, many studies have shown the importance of both the metabolome and the immune system in the pathogenesis of the disease. Appearance of autoantibodies in the serum of patients is the first sign of pathogenesis. However, molecular and cellular events precede the immune attack on the β-cell immunity. It has been shown that patients who developed T1D have an altered metabolome prior to the appearance of autoantibodies. Although much is known about the pathogenesis of T1D, the contribution of the environment/immune factors triggering the disease is still to be revealed.  In the present study both metabolic and immune deviations observed in the NOD mouse was analyzed. Serum metabolome analysis of the NOD mouse revealed striking resemblance to the human metabolic profile, with many metabolites in the TCA cycle significantly different from the non-diabetic control B6 mice. In addition, an increased level of glutamic acid was of the most distinguishing metabolite. A detailed bioinformatics analysis revealed various genes/enzymes to be present in the Idd regions. Compared to B6 mice, many of the genes correlated to the metabolic pathways, showed single nucleotide polymorphism (SNP), which can eventually affect the functionality of the protein. A genetic analysis of the increased glutamic acid revealed several Idd regions to be involved in this phenotype. The regions mapped in the genetic analysis harbor important enzymes and transporters related to glutamic acid. In-vitro islet culture with glutamic acid led to increased beta cell death indicating a toxic role of glutamic acid specifically towards insulin producing beta cells. In the analysis of the immune system, B cells from NOD mice, which are known to express high levels of TACI, were stimulated with APRIL, a TACI ligand. This resulted in enhanced plasma cell differentiation accompanied with increased class switching and IgG production. NOD mice have previously been shown to react vigorously to T-dependent antigens upon immunization. In this study we confirmed this as NOD mice showed an enhanced and prolonged immune response to hen egg lysozyme. Thus, serum IgG levels were significantly increased in the NOD mice and were predominantly of the IgG1 subtype. Immunofluorescence analysis revealed increased number of germinal centers in the NOD mice. Transfer of purified B and T cells from NOD to an immune deficient mouse could reproduce the original phenotype as seen in the NOD mice.     Collectively, this thesis has analyzed the metabolomics and immune deviations observed in the NOD mice.

ADJACENT CHANNEL INTERFERENCE MEASUREMENTS WITH CPFSK AND FQPSK-B SIGNALS

International Telemetering Conference Proceedings / October 22-25, 2001 / Riviera Hotel and Convention Center, Las Vegas, Nevada / This paper will present measured data in an adjacent channel interference (ACI) environment for both filtered continuous phase frequency shift keying (CPFSK) and Feher’s patented quadrature phase shift keying (FQPSK-B) [1]. The quantity measured was bit error probability (BEP) versus signal energy per bit to noise power spectral density ratio (E(b)/N(o)). The interferers were either CPFSK or FQPSK-B signals. The results presented in this paper will be for bit rates of 5 Mb/s, one interferer 20 dB larger than desired signal, various channel spacings, and two different telemetry receivers. The ACI test effort will collect data sets at several bit rates and with one and two interferers. The results will be useful to system designers and range operators as they attempt to maximize the number of Mb/s that can be simultaneously transmitted in the telemetry bands.

Progress in Global Air Traffic Management (GATM) Avionics System Test at the Air Force Flight Test Center

In many parts of the world today the continuing growth of commercial air traffic is running up against limits brought on by overuse of aviation resources. Air corridors in Europe and on transoceanic air routes are operating at maximum capacity. Civil Aviation Authorities (CAAs) are working these challenges on two levels—near-term incremental improvements and long-term visionary changes. Each country has a CAA; ours being the Federal Aviation Administration (FAA). Near-term solutions focus on better utilization of resources such as air space and frequency spectrum and improved performance of air traffic control facilities. Long-term visionary changes, such as free flight, could fundamentally change the current civil aviation business process model. CAA policies and standards are driving near-term improvements and migration toward long-term objectives. This initiative is referred as Communication Navigation Surveillance/Air Traffic Management (CNS/ATM). Implementation of the U.S. military’s vision, Global Reach/Global Power, requires the ability to rapidly deploy armed forces to major regional conflicts anywhere in the world, and to sustain these forces for as long as it takes to resolve these conflicts. To achieve this goal and accomplish rapid deployments while at the same time minimizing costs, the Air Force has adopted a solution that makes extensive use of CNS/ATM. The Air Force calls its initiative Global Air Traffic Management (GATM). Air Force aircraft equipped with GATM avionics will be able to use CNS/ATM capabilities such as reduced vertical separation minimum (RVSM), 8.33 kHz data links, automatic dependent surveillance - broadcast (ADS-B), and global communication networks. These capabilities make possible improved flight safety, lower fuel costs, and quicker turn times. The Air Force Flight Test Center supports the GATM initiative by providing Air Traffic Control (ATC) Communications Test Facilities and Avionic System Test (ACTFAST) capabilities to support aircraft modification programs.

B lymphocyte activation and exhaustion in chronic HIV : novel surrogate markers of generalised immune activation and selective modulation of aberrant B cell responses using vasoactive intestinal peptide (VIP)

Thesis (MScMedSc)--Stellenbosch University, 2015. / ENGLISH ABSTRACT: Introduction: Chronic HIV-1 infection is characterized by immune activation and dysregulation of immune homeostasis, which impacts on multiple immune cell types. The B-cell compartment, which plays an important role in the producing neutralizing antibodies, is also dysregulated in HIV- 1 infection. In this study we investigated peripheral blood B-cell subset distribution, and changes in expression of cellular activation, inhibition, and apoptosis signaling markers in both untreated chronic HIV-1 infected individuals and healthy uninfected controls. The neuropeptide immune modulator, vasoactive intestinal peptide (VIP) is known to selectively down-regulate activation of CD4+ T-cells in various disease settings including HIV-1, however to our knowledge, no studies have investigated the effect of VIP inhibition on B-cell activation. Materials & Methods: A total of 21 HIV+ve (CD4 count >250 cells/µl), and 19 HIV-ve individuals were recruited from the Emavundleni voluntary testing and counseling clinic in Crossroads, Western Province, South Africa. Whole blood was stained to distinguish B-cell subsets (activated memory (AM: CD21-CD27+), resting memory (RM: CD21+CD27+), mature naïve (MN: CD21+CD27-), or tissue-like memory (TLM: CD21loCD27lo). In addition expression of markers of B-cell activation (CD126, CD86, CD38, CD284, CD287), inhibition (CD72, CD85j, CD300a, CD305, CD307d), and apoptosis signaling (CD95), was assessed ex vivo by flow cytometry (BD FACSCanto II). For determination of functional responsiveness isolated B-cells (RosetteSep, Stemcell Technologies) were cultured for 18h (37°C, 5%CO2) without stimulation or stimulated with TLR ligands (LPS or R848). Stimulation experiments were also performed in the presence or absence of VIP. Results: Chronic HIV-1 infection affected B-cell subset distribution. The percentage (%) of TLM was increased by 59.24%, and %RM was decreased by 22.73% (both p<0.01). Total expression of the VIP receptor VPAC2 was decreased by 47.35% (p=0.0296). Subsets had a mixed phenotype ex vivo; HIV infection upregulated CD38 (by 59.56%, p=0.0004), CD72 (by 60.70%, p=0.0396), CD307d (by 68.63%, p=0.0015) on AM, while RM B-cells had increased expression of TLR4 (by 107.04%, p=0.0057) and TLR7 (by 208.14%, p=0.0199). TLM B cells (i.e. exhausted phenotype) displayed upregulated TLR7 (by 550%, p=0.0128) and CD307d (by 72.40% p=0.045) expression. MN B-cells had increased CD72 expression (by 70.98%, p=0.0026). R848 upregulated CD86 expression by 42.20% on AM (p<0.01), and by 56.06% on RM B-cells (p<0.01), which was significantly downregulated with VIP inhibition (both p<0.05). Similarly, CD95 expression on RM, TLM, and MN B-cells increased by 31.10% (p<0.001), 21.46% (p<0.01), and 39.92% (p<0.01) with R848 stimulation respectively, which was also significantly downregulated with VIP inhibition. Conclusion: These data indicate that B-cells in untreated HIV infection display increased levels of activation, and also the potential for increased susceptibility to apoptosis as evidenced by increased FAS (CD95) expression. VIP significantly down-regulated markers of activation, inhibition, and apoptosis signaling. Dysregulation of B-cells is thus apparent in asymptomatic stable chronic HIV-1 infection, which may impact on both inefficient neutralizing antibody production and hypergammaglobulinemia. The ability of VIP to prevent stimulationassociated marker upregulation may indicate that VIP is a potential therapeutic agent. Its immuno-modulatory properties were demonstrated to limit B-cell hyperactivation, and selectively down-regulate apoptosis and mark it out for further investigation. / AFRIKAANSE OPSOMMING: Inleiding: Immunaktivering en ongekoppelde immuun-homeostase is kenmerke van chroniese MIVinfeksie. Merkers van aktivering (CD126, CD86, CD38, CD284, CD287), inhibisie (CD72, CD85j, CD300a, CD305, CD307d), en apoptose signalering (CD95) is via vloeisitometrie (BD FACSCanto II) op B-selle ex vivo en ook op geïsoleerde B-selle (RosetteSep, Cell Technologies) ondersoek. Vir die bepaling van funksionele responsiwiteit, geïsoleerde B-selle (RosetteSep, StemCell Technologies) was vir 18h (37°C, 5%CO2) gekweek, sonder stimulasie of gestimuleer met TLR ligande (LPS of R848). Stimulasie eksperimente het ook in die teenwoordigheid of afwesigheid van VIP plaasgevind. Resultate: Chroniese MIV-1 infeksie het B-sel subset verspreiding geraak. Die persentasie (%) van TLM is verhoog deur 59,24%, en% RM het met 22.73% afgeneem (beide p <0,01). Totale uitdrukking van die VIP reseptor VPAC2 het met 47,35% afgeneem (p = 0,0296). An exploration into the influence of servicescape cues on perceptions of counterfeit products

Purpose The purpose of this research is to examine the role of servicescape theory in the counterfeit context and explore the extent to which servicescape cues influence perceptions of counterfeit products. Literature Following extensive examination of the current literature surrounding counterfeit activity it was discovered that counterfeits can be sold in a variety of environments; market stalls and car boots sales through to the legitimate retail environments. In the instances where the counterfeit has been integrated into the legitimate retail environment, weaknesses in the supply chain are usually to blame. These occurrences can be a major concern for both brands and consumers as they pose not only a risk to brand image but also a threat to consumer safety. Much of the current literature which explores consumer perceptions of counterfeit products concentrates on tangible product attributes and their influence. This research expands the current knowledge by examining further influential factors in the form of environmental cues. As a means of discussing the various elements which constitute a retail environment, the concept of servicescape is incorporated and analysed into the literature discussion. Following a comprehensive exploration of the various cues that may be present within a retail environment, the extent to which these cues influence consumer behaviour is explored. Further to this, as a means of understanding the ways consumers generate perceptions of counterfeit products, the processes of sensation and perception are analysed. Methodology The methodology chapter contained within this thesis considers both the philosophical positioning and the data collection methods used by this research. The philosophical positioning of the researcher is one of a constructivist-interpretive nature. Focus groups in conjunction with photo elicitation were the core data collection methods used. This combination of methods allowed an excellent opportunity for discussion and insights to be gathered and emotions to be recorded surrounding the issues of counterfeiting, servicescape and perception formation. Findings The findings which were identified by this research contribute extensively to the existing knowledge regarding counterfeiting and servicescape. The key themes highlighted the influence of human variables on perceptions of counterfeit products. Within this theme were a number of subsidiary themes including the influence of image, socio-demographics, other individuals within the counterfeit purchase environment, customer characteristics, human/social crowding and the influence of staff in the counterfeit purchase environment. In addition to this, levels of privacy also appeared to be an influential cue amongst participants in relation to their perceptions of product authenticity. Levels of spatial crowding were also an influential factor used by the research participants as a means of forming perceptions regarding product authenticity. From examination of the data, it was also made apparent that branding categorisation within a counterfeit purchase environment was particularly influential. Finally, servicescape permanency was noted to be a key theme throughout the focus group discussions. It appeared that a purchase environment‟s level of permanency was a key influencer when determining whether or not it sold counterfeit products.

Images of otherness : on the problem of empathy and its relevance to literary moral cognitivism

If the possible ends of art criticism are taken to include not only the provision of a detailed evaluation of the artwork, but, cognately, an elaboration upon how one has been, or believes oneself to have been, changed by a particular artistic encounter, then the very praxis of art criticism stands to benefit from a theoretical elucidation of the possible nature of the subjective transformations that may flow from the critical appreciation of art. We are entitled to enquire, in particular, into the conditions under which, and indeed the extent to which, such putative change at the personal level can be explicated in moral epistemological terms. It is pertinent in this context also to investigate the phenomenal character of the experiences that have been operative and their essential structures; to enquire, in short, into the phenomenology of the transformative artistic encounter. In this thesis, the bearing, in particular, of intersubjectivity upon the content and modalities of disclosure in a literary context will be investigated. It will be shown how an understanding of the relevance of intersubjectivity to the phenomenology of literary experience can inform an assessment of the claims of literary aesthetic moral cognitivism. Yet the intention to clarify the connection between literary experience and intersubjectivity also requires reflection upon what it is in the first place to encounter someone else, and to apperceive a foreign subjectivity and its motivations. For this reason, the contributions of Edmund Husserl and Edith Stein to the investigation of the phenomenology of empathy will be discussed and evaluated. This discussion will in turn be shown to be of assistance in clarifying the role of the imagination in the apperception and comprehension of another person’s mental life. The thought of Jean Starobinski will prove to elucidate the question of why the insights of the phenomenological tradition are highly pertinent to the investigation of literary experience, and to the development, in particular, of a conception of an imagined ‘Other’ who is (in a sense that will be clarified) embedded within the literary text, a person, that is, to whom one might coherently refer as the “implied author”. For reasons which will emerge in the course of this study, it will be argued that authentic empathy, in its fulfilling explication (in the Steinian sense), is given to the empathising consciousness in the manner of a semblance, and, consonantly, that the phenomenological structure of authentic empathy is characterised in its mature phases by an homological relation to pictureconsciousness. The epistemological significance of literature’s capacities for moral suggestion will be explicated principally in terms of the unfolding of values within the human personality, and in terms of the disclosure of the phenomenal character and structures of virtuous experience. It will be explained why the structure of empathy has implications for the aesthetic value of literature. The question of the relation between aesthetic and ethical value will be clarified. In this context, it will be argued on phenomenological grounds that the appresentation of moral virtue in an implied author could contribute to the aesthetic value of a literary work, although it will also be shown that implied authorial moral virtue could conflict irremediably with other qualities like moral doubt and uncertainty, which may themselves be important sources of aesthetic value. For this reason, the thesis will conclude by challenging the ethicist view that an aesthetically relevant ethical flaw in a literary work must count as an aesthetic flaw.

The radical humanism of Erich Fromm : a re-appropriation

This thesis attempts to advance the underappreciated thought of Erich Fromm as both a crucial contribution to twentieth century intellectual history and a potentially pivotal point from which to transcend current theoretical impasses. In particular, I argue that Fromm’s radical humanism can participate in the rejuvenation of contemporary social theory, which is still largely constrained by the dual reductionism of positivism and poststructuralism, and that a return to it will encourage renewed theorising of, and empirical engagement with, the connections that obtain between the ‘psychological’ and the ‘social’, the ‘essential’ and the ‘constructed’, and the ‘is’ and the ‘ought’. I try to show that Fromm’s qualified essentialism and ethical normativism are sensible, viable, and desirable, and that they, coupled with his analytic social psychology, which is based on his underlying humanism and elaborated through a unique fusion of Marx and Freud, provide the basis for the development of practical strategies to realise humanism in the world. Perhaps above all, I try to show that there is a deceptive complexity and sophistication to Fromm’s ideas, which are all too often taken as simple and naïve.

Characterisation of cell markers, cytokines and transcription factors involved in B cell responses from cartilaginous fish

Cartilaginous fish are the most ancient lineage to possess an adaptive immune system however nothing is known about the processes involved in the development, maintenance or proliferation of B cells in this group. In this thesis studies were undertaken to explore whether transcription factors, cytokines and cell surface markers that are involved in B cell immune responses in mammals are also present in cartilaginous fish. Through sequence database mining forty-one genes involved in B cell immune biology were found; of these twelve were B cell transcription factors. The presence of important B cell transcription factors, such as EBF1, pax5, E2A, Blimp1, PU.1 and Bcl6 suggests that B cells in cartilaginous fish probably undergo a similar developmental pathway as those in mammals. Eight cytokines, eleven cytokine receptors and four B cell surface markers were also identified, including CD40L, BAFF and CD79α that were further studied in this thesis. I cloned CD40L and BAFF from cartilaginous fish, both members of the tumour necrosis factor family and found cartilaginous fish BAFF genes have an extra exon that forms a unique α-helix insertion, and which may impact on receptor binding. The importance of all three molecules for the adaptive immune response was indicated by relatively high expression in shark immune tissue, such as spleen, gut-associated lymphoid tissue, gill and the Leydig organ, and the fact that their expression could be induced by immunostimulants, such as pokeweed mitogen. The finding that CD79α expression significantly correlates with those of immunoglobulin heavy-chains and the co-expression of CD79α and IgM on the B cell surface indicate that as in mammals CD79α in cartilaginous fish is expressed by B cells and associates with surface-bound immunoglobulin to form the active B cell receptor. Thus CD79α may serve as a pan B cell marker in future immunological studies in cartilaginous fish.

Dissertation (PhD)--University of Stellenbosch, 2007. / ENGLISH ABSTRACT: This dissertation describes how the isolation and characterisation of ovine liver cytochrome b5 was accomplished by referring to the following goals achieved in this study: - The optimisation of the isolation and purification procedure for ovine liver microsomal cytochrome b5 in order to obtain sufficient material for aggregation and immunological studies. - The removal of the membrane binding domain of cytochrome b5 by means of tryptic digestion to establish the role of the carboxyl terminal in ovine cytochrome b5 aggregation. - The raising of antibodies against both the trypsin truncated and intact forms of cytochrome b5 to study the aggregation of the protein. - The investigation into the influence of purified cytochrome b5 on steroidogenesis in ovine adrenal microsomes. / AFRIKAANSE OPSOMMING: Die isolering en karakterisering van skaaplewersitochroom b5, soos beskryf in hierdie proefskrif, is uitgevoer deur die volgende doelwitte suksesvol af te handel: - Die optimalisering van die prosedure vir die suksesvolle isolering en suiwering van skaaplewersitochroom b5 ten einde genoegsame hoeveelhede van die suiwer proteïen te hê vir die bestudering van die aggregasie van die proteïen sowel as ‘n immunologiese studie. - Die verwydering van die membraanbindingsdomein van sitochroom b5 om die invloed van die karboksielterminaal op die aggregering van die proteïen te bestudeer. - Die gebruik van sowel die tripties gesnyde as die intakte vorms van sitochroom b5 om ‘n immuunrespons in hase op te wek vir die verkryging van sitochroom b5 spesifieke anti-liggame. - Die gebruik van die gesuiwerde proteïene om die invloed van sitochroom b5 op adrenale steroïdogenese te bestudeer.
