The Effect of Intermediate Filament Inhibitors on Steroidogenesis and Cytoskeleton in Y-1 Mouse Adrenal Tumor Cells

When Y-1 mouse adrenal tumor cells were treated with sodium orthovanadate, an intermediate filament (IF) inhibitor in BHK21-F cells, there was no change in the amount of 20α-dihydroprogesterone produced. A neurofilament inhibitor, β, β'-iminodipropionitrile (IDPN), enhanced the ability of Y-1 cells to produce steroid in response to ACTH by acting on the plasma membrane. Electron microscopy of Y-1 cells extracted with Triton X-100 revealed that both vanadate and IDPN caused the aggregation of cytoskeletal and granular structures in the perinuclear area. The steroidogenic effects of IDPN suggest that the perinuclear aggrergation of cytoskeletal structures may result from the detachment of IF from the plasma membrane, while the reason for the cytoskeletal changes by vanadate is unknown.

Orientador: Marcus Antonio Rossi Feliciano / Coorientador: Wilter Ricardo Russiano Vicente / Coorientador: Sofia Borin Crivellenti / Banca: Eliandra Antônia Pires Buttler / Banca: Lenadro Zuccolotto Crivellenti / Resumo: As glândulas adrenais exercem papel fundamental na manutenção da homeostasia tanto em humanos, como nos animais, logo a avaliação ultrassonográfica dessas glândulas deve ser rotina, uma vez que pode auxiliar no diagnóstico de uma dada afecção, principalmente o hiperadrenocorticismo, doença relativamente comum principalmente nos cães, cujo diagnóstico baseia-se na avaliação do eixo hormonal hipotálamo-hipófise-adrenal. Quanto às outras doenças, como neoplasias, cistos, hiperplasias, dentre outras, o diagnostico se torna desafiador, pois depende de métodos invasivos de alto risco. Logo, este estudo buscou a aplicabilidade de técnicas de imagem avançadas e não invasivas, como a elastografia ARFI (quantitativa e qualitativa) e a ultrassonografia Doppler, com o objetivo de determinar os valores de normalidade ainda não relatados. Os resultados obtidos na avaliação ultrassonográfica modo-B nos cães saudáveis, mostraram hipoecoica em relação a gordura adjacente, homogêneas, contorno liso, formato típico e dimensões dentro dos valores de normalidade. A avaliação Doppler determinou os índices vasculares da artéria frênico-abdominal da glândula adrenal esquerda: velocidade sistólica 31,34 ± 5,79 cm/s e velocidade diastólica 9,54±3,03 cm/s, e da glândula adrenal direita: velocidade sistólica: 34,25±8,18 cm/s e velocidade diastólica 10,72±3,75 cm/s. Portanto, este estudo conseguiu com êxito demostrar a aplicabilidade destas técnicas na avalição das glândulas adrenais, mostrando-se promis... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The adrenals glands have an important role in maintaining homeostasis in humans and animals, so the ultrasonography evaluation of these glands should be routine, since it may help in the early diagnosis of a particular condition, as hyperadrenocorticism, a relatively common disease mostly in dogs, whose diagnosis is based on the evaluation of the hypothalamic-pituitary-adrenal hormonal axis. Other diseases such as tumors, cysts, hyperplasia, among others, the diagnosis becomes challenging because it depends on high-risk invasive methods. Therefore, this study sought the application of advanced imaging techniques and non-invasive, such as elastography ARFI (quantitative and qualitative) and Doppler ultrasound, in order to determine normal values not reported yet. The results obtained in the ultrasound B-mode assessment in healthy dogs, were within the normal range. The Doppler evaluation determined the vascular indices of the phrenic-abdominal artery of the left adrenal gland: systolic velocity 31.34 ± 5.79 cm / s and diastolic velocity 9:54 ± 3:03 cm / s, and right adrenal gland: systolic velocity: 34.25 ± 8.18 cm / s speed diastolic 10.72 ± 3.75 cm / s, hitherto not described in the veterinary literature. Foram incluídas no estudo 21 crianças pré-puberes (8M/I3F) entre 2,1 e 10,2 anos de idade, portadoras de Hiperplasia Congênita das Supra-renais por deficiência de 21-hidroxilase forma clássica perdedora de sal, que estavam em acompanhamento no Ambulatório de Endocrinologia Pediátrica do HCIUNICAMP. Realizou-se um questionário e um exame antropométrico, chamado de avaliação antropométrica atual, que incluiu as medidas do peso, estatura, perímetro braquial e dobras cutâneas (tricipital, bicipital, subescapular e suprailíaca). Foi avaliada a composição corporal por bioimpedância. Foram obtidas do prontuário dos pacientes as seguintes informações: idade ao diagnóstico; peso ao nascimento, peso e comprimento no início do tratamento, com um e dois anos de idade; resultados laboratoriais de 170H e androstenediona; dose de hidrocortisona presérita e altura dos pais. Os pacientes foram classificados de acordo com seu IMC em obesos (escore z acima do percentil 95), sobrepesos (escore z entre os percentis 85 e 95) e normais (escore z entre os percentis 5 e 85). Cigarette Residues Affect Steroidogenesis in Cultured Y-1 Mouse Adrenal Tumor Cells

This study (1) quantitatively compared steroid production in cultured Y-l mouse adrenal tumor cells exposed to Camel and Carlton-smoke derived residues, and (2) localized the effects in the cell. Basal steroid production was increased by Camel residues but not by Carlton, while ACTH stimulation was interfered with by both residues. Camel basal stimulation was comparable to that of cAMP, and was abolished by Cytochalasin D. The stimulation was also comparable to that of cholera toxin, which activates adenyl cyclase. Results indicate that residue components dissolve in the membrane stimulating adenyl cyclase at a point similar to or before that utilized by cholera toxin for its stimulating effect.
