Aerobic Biodegradability of Methyl tert-Butyl Ether(MTBE)

Contamination of groundwater supplies by gasoline and other petroleum-derived hydrocarbons released from underground or aboveground storage tanks is a serious and widespread environmental problem. Corrosion, ground movement, and poor sealing can cause leaks in tanks and associated piping. Petroleum hydrocarbons contain methyl tertiary-butyl ether (MTBE) (a fuel oxygenate), benzene, toluene, ethylbenzene, and xylene isomers (BTEX), the major components of gasoline, which are hazardous substances regulated by many nations. MTBE possesses all the characteristics of a persistent compound in the subsurface: high solubility, low volatility, low sediment sorption, and resistance to biodegradation. The objectives of this study were to (1) evaluate the biodegradibility of MTBE under aerobic conditions, and (2) assess the potential of using the aerobic bioremediation technique to clean up aquifers contaminated by MTBE. In this study, microcosms were constructed to determine the feasibility of biodegrading MTBE by intrinsic microbial consortia (aquifer sediments) under aerobic and aerobic cometabolic conditions. In the cometabolic microcosms, propane, ethanol, and BTEX were applied as the primary substracts to enhance the biodegradation of MTBE. The inocula used in this microcosm study were aquifer sediments collected from the contaminated-zones of a petroleum-hydrocarbon (including MTBE) contaminated site. Microcosms were constructed with nutrient medium (or site groundwater), sediments, and MTBE solution in 70-mL serum bottles sealed with Teflon-lined rubber septa. MTBE was analyzed using purge-and-trap instrument following gas chromatography (GC)/flame ionization detector (FID). Results show that the indigenous microorganisms were able to biodegrade MTBE under aerobic conditions using MTBE as the sole primary substrate. Microcosms with site groundwater as the medium solution show higher MTBE biodegradation rate. This indicates that site groundwater might contain some trace minerals or organics, which could enhance the MTBE biodegradation rate. Results show that the addition of BTEX would also enhance the MTBE removal. However, no significant MTBE biodegradation was observed in microcosms with propane and ethanol as the primary substrates. This reveals that the supplement of the second carbon source might inhibit the degradation of MTBE due to the preferential removal of some organics over MTBE. Results from the microcosm study suggest that aerobic biodegradation plays an important role on the MTBE removal. Intrinsic bioremediation is a feasible technology to remediate the studied MTBE-contaminated site.

Biodegradation of 2,4-Dinitrotoluene in the Waste Streams of a Munitions Plant

Wastewater from the manufacture of propellants typically contains 2,4-dinitrotoluene (DNT), a suspected animal carcinogen. Previous studies have indicated that DNT is aerobically biodegradable. However, inconsistent removal of DNT during aerobic treatment has been observed at a munitions wastewater treatment plant, necessitating the use of activated carbon pre-treatment. The objective of this study was to evaluate the effect of nutrient and cosubstrate amendments on the rate and extent of DNT removal. Addition of ethanol (100-500 mg/l) and phosphate (0.8-3.3 mg/l) significantly accelerated the rate of aerobic DNT (0.3-5.6 mg/l) biodegradation. Addition of phosphate alone also increased the rate of DNT degradation, but to a lesser degree. The presence of ethyl ether, another substrate commonly found in munitions plant wastewater, had comparatively little effect on the rate of DNT removal. Interruptions in the DNT manufacturing process can result in DNT being absent from the munitions plant wastewater for extended periods. The effect of such interruptions was evaluated in semi-continuously operated reactors, fed daily with phosphate-amended wastewater, at a hydraulic residence time of 3 days. DNT removal resumed without a lag even after it was absent from the feed for periods up to 15 days. During aerobic biodegradation of DNT, reduction to 4-amino-2-nitrotoluene and 2-amino-4-nitrotoluene was consistently observed, with reduction at the para position predominating. The highest level of aminonitrotoluene formation was 23% of the total DNT degraded. Aminonitrotoluene isomers were consumed shortly after they formed in the semi-continuously operated reactors, confirming the potential for degradation of these metabolites. Although the aminonitrotoluene isomers are not currently regulated, their presence in treated munitions wastewater is a concern due to possible toxicity. / Master of Science

Fate of phytosterols in pulp and paper wastewater treated in a simulated aerated stabilization basin

Phytosterols are steroid chemicals produced by plants for the purposes of membrane function and hormone production. Phytosterols can cause endocrine disruption in aquatic species at very low concentrations and are suspected of contributing to endocrine disruption linked to pulp and paper effluent. Wastewater from the pulp and paper industry is often treated biologically in aerated stabilization basins (ASBs) that expose phytosterols to a range of redox zones. Phytosterol removal in ASBs varies and stigmasterol has even been shown to increase across the treatment system. Little is known about the microbial processes that occur within ASBs and their effect on phytosterol removal. The objective of this research was to assess the biotransformation potential of phytosterols in a simulated ASB treatment system and to improve understanding of the processes that occur within the various redox zones and their impact on the removal of phytosterols. To assess the biotransformation of phytosterols under aerobic conditions, three assays were conducted using a stock aerobic culture fed with pulp and paper wastewater. The assays tested three conditions: phytosterols present as a sole added carbon source, phytosterols with dextrin as an added carbon source, and phytosterols with ethanol as a solubilizing agent and added carbon source. Phytosterol biotransformation was found to be limited by low phytosterol solubility. When solubilized, phytosterol removal occurred in two phases: an initial near-linear removal, followed by accelerated removal during the culture's stationary stage, possibly due to the release of extracellular cholesterol oxidase. The anoxic and anaerobic biotransformation of phytosterols was examined through a series of three semi-batch cultures maintained under nitrate-reducing, sulfate-reducing and fermentative/methanogenic conditions, all developed from stock cultures fed with pulp and paper wastewater. Phytosterol removal was significant in the nitrate-reducing culture, although microbial activity and phytosterol removal declined in later stages. Phytosterol removal was also observed in the sulfate-reducing culture, although there was a significant lag period before removal occurred. No phytosterol removal was observed in the fermentative/methanogenic culture. Phytosterol biotransformation was also examined in the context of a lab-scale ASB fed continuously with pulp and paper wastewater. The steady-state ASB effluent and sediment characteristics were examined over three hydraulic retention times (HRTs). Effluent quality was not significantly affected by a change in HRT but sediment characteristics were significantly affected and, at shorter HRTs, phytosterols accumulated in the sediment. Wastewater bioassays demonstrated the release of phytosterols during the breakdown of solids. This research improves the understanding of biological processes within ASBs and their effect on phytosterol removal.

For the aerobic degradation study the soil sample was divided into two groups, one was fertilised. The two groups of soil in the experimental bottles were treated to varying amount of methane in pairs. DCE and TCE were added to all samples while PCE was found in the contaminated soil. Both aerobic and anaerobic experiments were conducted. For aerobic study air was added to all bottles to serve as electron acceptor (oxygen). It was observed that all the samples showed a very small amount of methane consumption while the fertilised soil samples showed more oxygen consumption. For the chlorinated compounds the expected degradation could not be ascertained since the control and experimental set up were more or less the same.</p><p>For the anaerobic biodegradation study soil slurry was made with different media i.e. basic mineral medium (BM), BM and an organic compound (lactate), water and sulphide, phosphate buffer and sulphide and phosphate buffer, sulphide and ammonia. To assure anaerobic conditions, the headspace in the experimental bottles was changed to N2/CO2. As for the aerobic study all the samples were added DCE and TCE while PCE was found in the contaminated soil. The sample without the soil i.e. the control was also given PCE. It was observed that there was no clear decrease in the GC peak area of the pollutants in the different media. The decrease in GC peak area of the pollutants could not be seen, this may be so because more susceptible microorganisms are required, stringent addition of nutrients and to lower the risk of the high concentration of PCE and petroleum products in the soil from Startvätten AB.</p>

Etude de l'effet des processus diagénétiques sur les alcénones : impact sur les estimations de paléotempératures / Effects of diagenetic process on alkenones : impact on palaeotemperature estimations

Les alcénones constituent une classe de cétones insaturées à longue chaîne (C35 à C41)synthétisées par un nombre limité d’haptophytes. La proportion des alcènones en C37 di et triinsaturéesvarie en fonction de la température de croissance de l’haptophyte. A partir de cette caractéristique et de leur ubiquité dans l’environnement marin, un indice nommé '37U K =[C37 :2] / ([C37 :2 + C37 :3]) est utilisé depuis la fin des années 1980s comme paléomarqueur des températures des eaux de surface.Des processus de dégradation biotique et abiotique sélectifs, jusqu’alors partiellement ignorés,peuvent entraîner des biais significatifs (du fait de la perte préférentielle des alcénones les plus insaturées) dans les valeurs de paléotempératures estimées à partir de l’ '37U K . Ce travail a été entrepris dans le but d'étudier l'impact de ces processus diagénétiques sur les alcénones et d'évaluer leur importance dans l'environnement marin.Durant la première partie de ce travail, nous avons isolé et identifié diverses souches bactériennes à partir de cultures d’Emiliania huxleyi, que nous avons testées pour leur capacité à dégrader les alcénones. La souche Dietzia maris s'est révélée capable de dégrader sélectivement les alcénones. Cette dégradation sélective fait intervenir une époxydation initiale des doubles liaisons des alcénones, qui est vraisemblablement induite par une monoxygénase ayant une plus grande affinité pour la double liaison en position w29 et peut conduire à une augmentation des valeurs de l' '37U K de l’ordre de +0,10 (soit une surestimation des températures de +3°C).L'impact de ces processus de dégradation (biotique et abiotique) in situ s'est révélé plus oumoins significatif selon les zones géographiques considérées. En mer Méditerranée,l'augmentation des valeurs de l' '37U K (0,43 à 0,55) s'explique essentiellement par une forte autoxydation des alcénones lors de leur sédimentation. La détection d'alcénones stéréomutées à la surface du sédiment nous a permis d'estimer que les processus de stéréomutatio npouvaient entraîner un biais dans les valeurs de l' '37U K de +0,05 dans cette région. En Alaskaainsi que dans le Pacifique équatorial, les biais observés résultent essentiellement d’une dégradation bactérienne sélective des alcénones (+0,7 à +2,4°C et +2°C, respectivement).Lors de nos analyses de matériel particulaire provenant de l’océan Pacifique équatorial, nos observations ont mis en évidence une relation entre l’état de photo-oxydation des cellules phytoplanctoniques sénescentes et l’état physiologique des bactéries qui leur sont associées.Un transfert d’oxygène singulet (1O2) des phytodétritus aux bactéries entraînerait un déclin important de la croissance bactérienne et limiterait ainsi considérablement la biodégradation.Dans notre étude, ce transfert d’1O2 s’est révélé plus efficace dans les particules en suspension(forte photo-oxydation des bactéries) que dans les particules prélevées par trappes (forte biodégradation du phytodétritus). Cette différence s’expliquerait par l’abondance de particules riches en silice dans les particules prélevées par trappes (dominées par des agglomérats de diatomées) dont le caractère polaire réduirait la durée de vie de l’1O2.Nos résultats confirment que les processus de (i) dégradation bactérienne sélective, (ii)d'autoxydation et (iii) de stéréomutation peuvent introduire des bais significatifs dans les reconstructions de paléotempératures et des moyens de corriger les biais résultant de cette diagenèse ont été proposés afin d'améliorer les reconstructions de paléotempératures basées sur cet outil. / Alkenones constitute a class of long-chain unsaturated ketones (C35 to C41) synthesized by alimited number of haptophytes. The proportion of C37 di- and tri-unsaturated alkenones varies according to the growth temperature of the haptophytes. From this characteristic and theubiquity of alkenones in the marine environment, an index named '37U K = [C37: 2] / ([C37: 2 + C37:3]) is used since the late 1980s as paleomarker of sea surface temperatures. Selective biotic and abiotic degradation processes, previously ignored in the literature, canlead to significant biases (due to the preferential loss of the more unsaturated alkenones) in paleotemperature values estimated from the '37U K . This work was undertaken to estimate theimpact of diagenetic processes on alkenones in the marine environment.During the first part of this work, we isolated and identified various bacterial strains from cultures of Emiliania huxleyi, which were tested for their ability to degrade alkenones. Thestrain Dietzia maris sp. S1 appeared to be able to degrade selectively di- and tri-unsaturatedalkenones. This selective degradation involves an initial epoxidation of alkenone doublebonds, which is probably induced by a monooxygenase showing a greater affinity for the w29double bond and leads to increases of the '37U K values ranging from +0.05 to +0.10 units(corresponding to an overestimation of temperatures of 1.5 - 3°C).The impact of these biotic and abiotic degradation processes in situ was more or lesssignificant depending on the area considered. In Mediterranean Sea, increasing values of '37U Kwith depth (0.43 to 0.55) seemed to mainly result from an intense autoxidation of alkenones.The detection of stereomutated alkenones in surface sediments also attested to the importanceof these processes in this region (increased in '37U K values of +0.05 units). In contrast, in Alaska and Equatorial Pacific, the biases observed (+0.7 to +2.4°C and +2°C, respectively) appeared to be mainly induced by selective bacterial degradation of alkenones.Analyses of particulate matter from the Equatorial Pacific Ocean, revealed a relationshipbetween the state of photo-oxidation of senescent phytoplankton cells and the physiologicalstate of associated bacteria. A transfer of singlet oxygen (1O2) from phytodetritus to thebacteria may induce damages in bacteria and thus significantly limit biodegradation. This transfer of 1O2 appeared to be more effective in suspended particles (high photo-oxidation ofbacteria and preservation of phytodetritus) than in the sinking particles (weak photo-oxidationof bacteria and high biodegradation of phytodetritus). These differences were attributed to theabundance of particles rich in silica in sinking particles (dominated by agglomerates ofdiatoms), whose the polar character could reduce lifetime of 1O2.Our results confirm that the process of (i) selective bacterial degradation, (ii) autoxidation and(iii) stereomutation may introduce significant biases in the reconstruction ofpaleotemperatures. Some tools were proposed to correct some of these biases and thus toimprove the paleotemperature reconstructions based on alkenones

Απομάκρυνση ξενοβιωτικών ουσιών κατά τη βιοσταθεροποίηση της ιλύος και άλλες αερόβιες διεργασίες

Τις τελευταίες δεκαετίες η συνεχώς αυξανόμενη παγκόσμια δραστηριότητα, λόγω της έντονης αστικοποίησης και βιομηχανοποίησης, έχει σαν αποτέλεσμα την παραγωγή σημαντικών ποσοτήτων αποβλήτων. Μεταξύ αυτών είναι και η ιλύς, δηλαδή το ημιστερεό υπόλειμμα που παράγεται από τις μονάδες επεξεργασίας αστικών και βιομηχανικών αποβλήτων. Μολονότι η ιλύς αντιμετωπίζεται ως απόβλητο που πρέπει να διατεθεί, είναι στην πραγματικότητα ένας σοβαρά υποτιμημένος πόρος. Πιο συγκεκριμένα, η ιλύς διαθέτει πολύτιμα θρεπτικά συστατικά, ενώ παράλληλα έχει και υψηλή θερμική αξία, ιδιότητες που την καθιστούν κατάλληλη για ένα μεγάλο εύρος χρήσεων. Ωστόσο, η ιλύς είναι φορέας ρύπων, όπως οι παθογόνοι μικροοργανισμοί και οι ξενοβιοτικές ουσίες, η ύπαρξη των οποίων βρίσκεται στο επίκεντρο των ενδιαφερόντων των ερευνητών. Ανάμεσα στις διάφορες διεργασίες που έχουν εφαρμοσθεί μέχρι σήμερα τόσο για την επεξεργασία της ιλύος, όσο και για την απομάκρυνση των ξενοβιοτικών παραγόντων από το περιβάλλον, οι βιολογικές διεργασίες φαίνεται να υπερτερούν αισθητά έναντι των άλλων. Στόχος της παρούσας διατριβής ήταν η εφαρμογή μιας βιολογικής μεθόδου, της βιοσταθεροποίησης, για την επεξεργασία της ιλύος. Η ολοκλήρωση της μελέτης της βιοσταθεροποίησης έγινε με την ανάπτυξη ενός μαθηματικού μοντέλου ικανό να περιγράφει τη διεργασία. Παράλληλα, ερευνήθηκε η δυναμικότητα της διεργασίας αυτής στη βιοαποκατάσταση της ιλύος από το ξενοβιοτικό περιεχόμενό της. Επιπρόσθετα, μελετήθηκε η απομάκρυνση των ξενοβιοτικών ουσιών, μέσω της βιοαποδόμησής τους από αυτόχθονες μικροβιακούς πληθυσμούς της ιλύος, σε άλλα αερόβια περιβάλλοντα. Η εφαρμογή της διεργασίας της βιοσταθεροποίησης πραγματοποιήθηκε σε κατάλληλη πειραματική διάταξη που σχεδιάστηκε και υλοποιήθηκε για το σκοπό αυτό. Η διάταξη αυτή επέτρεπε τον πλήρη έλεγχο και τη σωστή ρύθμιση της διεργασίας. Η ολοκλήρωση των πειραμάτων της λιπασματοποίησης έδειξε ότι η διεργασία αυτή μπορεί να εφαρμοσθεί ως μέθοδος επεξεργασίας και σταθεροποίησης της ιλύος, αρκεί να ρυθμιστούν κατάλληλα καθοριστικές φυσικές και χημικές παράμετροι. Τα αποτελέσματα έδειξαν, επίσης, ότι η διεργασία δεν παρεμποδίζεται από την παρουσία υψηλών αρχικών συγκεντρώσεων ξενοβιοτικών ουσιών στην ιλύ, ενώ αντίθετα οδηγεί στην απομάκρυνσή τους. Τα ποσοστά απομάκρυνσης ήταν ιδιαίτερα υψηλά προκειμένου για τις επιφανειοδραστικές ενώσεις και αρκετά σημαντικά για το φθαλικό εστέρα και τους πολυκυκλικούς αρωματικούς υδρογονάνθρακες. Φαίνεται, λοιπόν, ο πολυποίκιλος μικροβιακός πληθυσμός της βιοσταθεροποίησης να συμβάλλει στην ουσιαστική, αν όχι πλήρη, απομάκρυνση των ξενοβιοτικών ουσιών κατά τη διάρκεια της διεργασίας και να οδηγεί στην παραγωγή σταθερού και ασφαλούς τελικού προϊόντος. Ωστόσο, υψηλά ποσοστά απομάκρυνσης των παραπάνω ξενοβιοτικών ουσιών κατέδειξαν και τα πειράματα που πραγματοποιήθηκαν σε αερόβιες υγρές μικτές καλλιέργειες. Ενδεχομένως, η μικροβιακή συνεργασία να παίζει τον καθοριστικό ρόλο στη βιοαποδόμηση αυτών των ουσιών. Η παρούσα μελέτη θα μπορούσε να συνεισφέρει αποτελεσματικά στην ανάπτυξη μιας αξιόπιστης μεθόδου για τη διαχείριση της ιλύος. Συνολικά, η λιπασματοποίηση μπορεί να αποδειχθεί ένα χρήσιμο εργαλείο για την επιθυμητή βιώσιμη ανάπτυξη της κοινωνίας μας, αφού αφ’ ενός μπορεί να συμβάλλει στη λύση του περιβαλλοντικού προβλήματος της διαχείρισης της ιλύος που παράγεται από τις εγκαταστάσεις επεξεργασίας λυμάτων, που την απασχολεί έντονα, και αφ’ ετέρου μπορεί να εξουδετερώσει το παθογόνο περιεχόμενο και να εξαλείψει τις τοξικές ξενοβιοτικές ουσίες. / The continuously increased human activity of the last few decades, because of the intense urbanization and industrialization, has resulted in the production of a serious amount of wastes. Among these is sewage sludge, which is the semi-solid residue produced by municipal and industrial waste treatment plants. Although sludge is considered as a waste product which must be disposed, it is actually an underestimated resource. Specifically, sludge contains valuable nutrients and high thermal value making sludge appropriate for a wide range of uses. Nevertheless, the existence of pathogens and xenobiotics in sludge has attracted the interest of scientific community. Among various methods that have been used for sludge treatment, as well as the removal of xenobiotics from environmental systems, biological methods are advantageous. The aim of the present work was to assess the sewage sludge treatment by a biological process, which is closed-vessel composting. This study involved also the development of a mathematical model capable of describing the composting process. The efficiency of bioremediation of xenobiotics content during sludge composting was also studied. Finally, the xenobiotics removal was investigated through biodegradation using sludge indigenous microbial populations in other aerobic environments. A suitable experimental configuration was designed and constructed in order to study the composting process. The composting system was designed for complete control and regulation of the process. Based on the results of the composting experiments, it can be concluded that composting is an efficient method allowing for treating and stabilization of sewage sludge, provided that critical physicochemical parameters are adjusted. Results revealed also that composting process is not inhibited by the presence of high initial concentration of xenobiotics. On the contrary, composting results in removal of xenobiotics. With reference to surfactants (LAS.NP and NPEO), the removal percentages were particularly high, while the relevant percentages for phthalate ester and polycyclic aromatics hydrocarbons were also significant. It seems, then, that the variable microbial population of the composting mixture contributes to the elimination of xenobiotic compounds and results in a safe and stable final product. The experiments which were conducted in aerobic mixed liquid cultures showed also high xenobiotic removal percentages. Probably, the microbial consortium could play a significant part in biodegradation of xenobiotic compounds. The present study contributes to the development of a reliable method of sewage sludge treatment.
