Efeito do uso de uma dose adicional de prostaglandina F2α durante o protocolo de IATF à base de estradiol e progesterona na fertilidade de vacas holandesas em lactação em anestro

Orientador: José Eduardo Portela Santos / Resumo: The objectives of these experiments were to determine the effects of a second prostaglandin F2α (PGF2α) dose in an estradiol (E2) and progesterone (P4) based timed AI (TAI) protocol on LH pulsatility, pre-ovulatory follicle characteristics, and pregnancy per AI (P/AI) in anestrous lactating Holstein cows. In experiment 1, 2,011 Holstein cows had their estrous cycles synchronized and ovaries scanned by ultrasound to determine if a corpus luteum (CL) was present at the time of protocol initiation (d-12) and on the day of PGF2α (d-4). Cows without CL on d-12 and d-4 were classified as anestrous (n = 454) and submitted to the following TAI protocol: d -12 or -11: two intravaginal P4 devices and estradiol benzoate (EB); d -4 PGF2α and withdrawal of one P4 device; d -2 estradiol cypionate (ECP) and withdrawal of the second P4 device; on d 0, TAI was performed. On d -4, cows were randomly assigned to one of four treatments: one dose of PGF2α on d -4 and 9 days with a P4 device (1PGF9d, n = 116); two doses of PGF2α, the first on d -4 and the second on d -2 and 9 days with a P4 device (2PGF9d, n = 115); one dose of PGF2α on d -4 and 10 days with a P4 device (1PGF10d, n = 111) or two doses of PGF2α, the first on d -4 and the second on d -2 and 10 days with a P4 device (2PGF10d, n = 112). Análisis del factor de crecimiento diferencial 9 (GDF-9) y la proteína morfogenética ósea (BMP-15) durante el desarrollo de folículos antrales en ovarios caninos mediante citometría de flujo

Memoria para optar al Título Profesional de Médico Veterinario / El ciclo estral de la hembra canina presenta algunas características únicas al ser comparadas con otras especies de mamíferos domésticos. Sin embargo, poco se conoce de las características del desarrollo folicular en relación a otras especies. El Factor de Crecimiento Diferencial 9 (GDF-9) y la Proteína Morfogenética Ósea 15 (BMP-15), son miembros de la superfamilia de Factores de Crecimiento Transformantes Beta (TGF-β), y han demostrado un rol fundamental en el desarrollo folicular en muchas especies. El objetivo de este estudio fue analizar la presencia de ambas proteínas en células foliculares proveniente de folículos antrales del ovario canino mediante citometría de flujo. Se obtuvieron células de la granulosa y del cúmulo a través de aspiración y raspado de folículos antrales de distintos niveles de desarrollo y en distintas etapas del ciclo estral. Las células se fijaron e incubaron con anticuerpo anti GDF-9 humano y anti BMP-15 de ratón (1:100) y anticuerpos secundarios conjugados con FITC y PerCP (1:500), respectivamente. Al análisis por citometría se hizo un Gate discriminatorio por tamaño y complejidad en el Dot plot inicial y adicionalmente se discriminó con marcadores de CD45 para leucocitos y yoduro de propidio (IP) para eritrocitos y debris en los histogramas correspondientes. Los resultados se analizaron con ANOVA y regresión lineal. La expresión de GDF-9 disminuyó (P<0.05) a medida que avanzó el desarrollo folicular en Anestro y Proestro/Estro, pero aumentó (P<0.05) en Diestro. BMP-15 aumentó (P<0.05) su presencia en folículos antrales más desarrollados en la etapa de Anestro, disminuyendo (P<0.05) en Proestro/Estro al alcanzar un mayor nivel de desarrollo. Estas proteínas por tanto, se expresaron en las células foliculares de caninos en distintas etapas del ciclo estral y de manera diferente durante el desarrollo folicular, donde ambas podrían estar relacionadas con las características particulares. / The estrous cycle of the bitch presents unique features when compared to other domestic mammals, however, little is known about the characteristics of the follicular development compared with other species. The Growth Differential Factor 9 (GDF-9) and Bone Morphogenetic Protein 15 (BMP-15) are members of the Transforming Growth Factor (TGF-β) superfamily and have demonstrated an important role during the follicular development of many species. The aim of this study was to analyze the presence of both proteins in the follicular cells from canine ovarian antral follicles by flow cytometry. Granulosa and cumulus cells were obtained by aspiration and scraping of antral follicles from different sizes and stages of the estrous cycle. Cells were fixed and incubated with first antibodies against human GDF-9 and mouse BMP-15 (1:100) and second antibodies conjugated with FITC and PerCP (1:500), respectively. A size and complexity discriminatory gate was used for the cytometryc analysis in the initial dot plot and, additionally, a CD45 marker for leukocyte and Iodum Propide for erythrocyte and debris discrimination in the corresponding histograms. The results were analyzed with ANOVA and lineal regression. GDF-9 expression decreased (P<0,05) during follicular development in Anestrous and Proestrous/Estrous, but increased during Diestrous (P<0,05). BMP-15 expression increased (P<0,05) during follicular development in Anestrous, but decreased in roestrous/Estrous (P<0,05) compared to small and medium sizes. A égua é classificada como poliéstrica estacional, o que implica suspensão da atividade reprodutiva cíclica usualmente a partir da metade do outono, na maioria destes animais. Esta resposta é devida à diminuição gradual no fotoperíodo natural, considerado como fator primário na sazonalidade reprodutiva. A ausência de ciclos ovulatórios durante os meses de inverno e a inconsistência da fase de ressurgência atingem não só os criadores de raças que procuram o nascimento de potros mais cedo na temporada reprodutiva oficial, mas principalmente aqueles que empregam a técnica da transferência de embriões. A obtenção de ovulações ao longo do ano possibilitaria a utilização reprodutiva ótima de éguas doadoras e receptoras. A fim de verificar a ciclicidade reprodutiva durante outono e inverno, dezessete éguas em uma propriedade do sul do Brasil foram distribuídas ao acaso em dois grupos. De quinze de Janeiro a doze de Setembro de 2009, os animais do grupo Tratamento foram submetidos a fotoperíodo diário fixo de 15 horas. Duas vezes por semana, o trato reprodutivo de todos os animais foi avaliado por ultrassonografia transretal, para verificação da presença de folículos dominantes e corpo lúteo. Dosagem de progesterona foi utilizada para confirmação de anestro. Oitenta e cinco por cento dos animais submetidos ao tratamento mantiveram ciclicidade reprodutiva, enquanto oitenta por cento dos animais controle entraram em anestro de inverno. Houve aumento na duração dos ciclos, devido a fatores endógenos, nutricionais e ambientais. Os resultados demonstram que o tratamento foi efetivo para obtenção de ciclos ovulatórios durante os meses de inverno. / The mare is classified as a seasonal poliestric, which means the suspension of reproductive activity usually from the first half of autumn, most of the mares. This is a response to the gradual decline in the natural photoperiod, considered as a primary factor of reproductive seasonality. The absence of ovulatory cycles during winter months and the inconsistency of the resurging phase strike both breeders who aim foals’ birth early in the official breeding season and those who employ the embryo transfer technique. Achieving ovulation throughout the year would enable the optimal reproductive use of donors and recipient mares. In order to check autumn and winter cyclicity, seventeen mares on a farm in southern Brazil were randomly assigned into two groups. From January 15th to September 12th 2009, animals in Treatment group were subjected to a fixed 15 hours daily photoperiod. Twice a week, the reproductive tract of all animals was examined by transrectal ultrasonography, searching for dominant follicles and presence of corpora lutea. Progesterone levels were used to confirm winter anestrus. 85% mares undergoing treatment maintained reproductive activity, while 80% of Control mares showed winter anestrus. There were changes in lenght of cycles, due to endogenous, nutritional and environmental factors. Results demonstrate that the treatment was effective to attain ovulatory cycles during winter months.

Fotoperíodo artificial sobre a atividade reprodutiva de éguas durante a transição outonal. / Artificial photoperiod on reproduction activity of mares during autumn transition

O presente experimento verificou se o fotoperíodo artificial iniciado no verão retarda o início da transição outonal e nos casos de ocorrência do anestro determinar sua forma de início. Foram utilizadas no experimento 13 éguas com idades variando entre 4 e 10 anos. Todas as éguas no início do experimento encontravam-se ciclando, com presença de corpo lúteo funcional ou folículo dominante. Antes do início do experimento os animais foram divididos em dois grupos, Grupo Luz (GL) com 08 animais e Grupo Controle (GC) com 05 animais, buscando equilibrar a idade e a condição corporal das éguas nos diferentes grupos. A partir do dia 23 de fevereiro de 2006 as éguas do GL foram submetidas a um fotoperíodo artificial de 15 horas de luz e de 9 horas de escuridão, e as éguas do GC foram mantidas sob fotoperíodo natural. Em relação ao início do anestro e à condição corporal não foram observadas diferenças entre o GL e o GC. Dentre os 13 animais utilizados no experimento todos entraram em anestro. Não se observaram diferenças entre os dois grupos de tratamento em relação ao número de dias entre o início do tratamento e a última ovulação e os inícios do tratamento e do anestro, bem como entre o número de dias de duração do último corpo lúteo. Entretanto observou-se diferença no grupo luz onde 75% das éguas apresentaram diestro prolongado enquanto que no grupo controle isto ocorreu em somente 40% das éguas. O anestro inicia por persistência lútea seguida de inatividade folicular ou por inatividade folicular após um diestro regular. O fotoperíodo artificial iniciado a partir da segunda metade do verão não impediu o início do anestro. Entretanto, a maioria das éguas submetidas ao fotoperíodo artificial iniciou o anestro após uma persitência lútea. / The present study aimed to verify if the artificial photoperiod started during summer retards the beginning of autumn transition and if anestrus occurs, to determine the way it begins. There were used 13 mares with ages varying between 4 and 10 years. All the mares were cycling in the beginning of the study, having either a functional corpus luteum or a dominant follicle. Before starting the experiment, the animals were divided in two groups: Light Group (LG) with 08 animals and Control Group (CG) with 05 animals, trying to equilibrate age and body condition of the different group mares. After February 23rd, 2006, the mares of LG were submitted to artificial photoperiod of 15 hours light and 9 hours dark, and the mares of CG were kept under natural photoperiod. Concerning the beginning of anestrus and body condition, no differences were observed between LG and CG. Among the 13 animals used in the study, all entered anestrus period. No differences were observed between the treatment groups about the number of days between the beginning of the treatment and the last ovulation, and the beginning of treatments and anestrus, as well as the number of days of duration of the last corpus luteum. However, differences were found in the light group, where 75% of the mares presented a prolonged diestrus while in the control group it only happened in 40% of the mares. The anestrus begins by luteal persistency followed by follicular inactivity or by follicular inactivity after a regular diestrus. The artificial photoperiod initiated after the second half of the summer did not affect the beginning of the anestrus. However, most of the mares submitted to artificial photoperiod initiated anestrus after a luteal persistency.

Assim, avaliaram-se as implicações práticas do uso de marcadores de fertilidade para o direcionamento da suplementação com progesterona exógena após a inseminação e melhoria dos programas de IATF em bovinos de corte. Esta tese foi dividida em dois capítulos. No primeiro capítulo, estudou-se o efeito da presença ou não de CL no início do protocolo, do índice de escore corporal (IEC), da expressão do estro e do diâmetro do folículo pré ovulatório sobre a taxa de concepção de fêmeas bovinas da raça Nelore submetidas a um programa de IATF. Observou-se que não houve efeito da presença ou não de CL no início do protocolo e do IEC nas taxas de concepção. Entretanto, animais que expressaram estro entre a retirada do implante intravaginal e a inseminação artificial e tiveram maior diâmetro do folículo pré-ovulatório no momento da inseminação, tiveram maior taxa de concepção após a IATF. O segundo capítulo estudou o efeito da suplementação com acetato de melengestrol (MGA) após a IATF sobre a taxa de concepção de fêmeas bovinas da raça Nelore de forma geral e de acordo com a ciclicidade durante o protocolo, o diâmetro do folículo pré-ovulatório no momento da inseminação e a expressão do estro entre a retirada do implante intravaginal e a inseminação. Observou-se que houve efeito positivo do tratamento com MGA sobre a taxa de concepção em relação aos animais não suplementados. O tratamento com MGA melhorou a taxa de concepção dos animais em anestro, mas não influenciou na taxa de concepção dos animais que estavam ciclando. Melhorou a taxa de concepção independente da expressão ou não de estro, entretanto, em relação ao diâmetro do folículo pré-ovulatório no momento da inseminação, apenas os animais com DFOL >14mm tiveram efeito positivo da suplementação com MGA em relação a taxa de concepção. Os resultados indicam que o diâmetro do folículo pré-ovulatório no momento da inseminação e a expressão de estro após o protocolo de sincronização foram associados a melhores taxas de concepção de fêmeas bovinas da raça Nelore, sendo importantes marcadores de fertilidade, podendo ser utilizados para escolha de estratégias após a IATF que melhorem as taxas de concepção. Além disso, foi possível obter maiores taxas de concepção através da suplementação com MGA após a IATF em fêmeas bovinas da raça Nelore lactantes e a avaliação da ciclicidade ovariana previamente ao protocolo de IATF mostrou ser um marcador importante na escolha dos animais a serem suplementados com progestágeno, assim como o marcador expressão do estro. / The objective of this study was to evaluate possible fertility markers and the association with exogenous progesterone supplementation after insemination on the design rate of Nelore cattle in TAI programs. Thus, the practical implications of the use of fertility markers for the targeting of exogenous progesterone supplementation after insemination and improvement of TAI programs in beef cattle. This thesis was divided into two chapters. In the first chapter, the effect of the presence or not of CL at the beginning of the protocol, the body condition score (BCS), the estrus expression and the preovulatory follicle diameter on the conception rate of bovine Nelore breed submitted to an TAI program. It was observed that there was no effect of the presence or not of CL at the beginning of the protocol and of the BCS at conception rates. However, animals that expressed estrus between intravaginal implant withdrawal and artificial insemination and had a larger preovulatory follicle diameter at the time of insemination, had a higher conception rate after TAI. The second chapter studied the effect of supplementation with melengestrol acetate (MGA) after TAI on the conception rate of Nelore cattle in general and according to the cyclicity during the protocol, the diameter of the preovulatory follicle in the moment of insemination and estrus expression between intravaginal implant withdrawal and insemination. It was observed that there was a positive effect of MGA treatment on conception rate over non-supplemented animals. MGA treatment improved the conception rate of animals in anestrous, but did not influence the design rate of the animals that were cycling. However, in relation to the diameter of the preovulatory follicle at the time of insemination, only animals with DFOL> 14 mm had a positive effect of supplementation with MGA in relation to conception rate. The results indicate that the diameter of the preovulatory follicle at the time of insemination and the expression of estrus after the synchronization protocol were associated with better conception rates of Nelore cattle, being important fertility markers and could be used to choose of strategies after TAI what improve conception rates. In addition, it was possible to obtain higher conception rates through the supplementation with MGA after the TAI in lactating Nelore cattle and the evaluation of the ovarian cyclicity prior to the TAI protocol proved to be an important marker in the choice of animals to be supplemented, as the expression marker of estrus.

A égua é classificada como poliéstrica estacional, o que implica suspensão da atividade reprodutiva cíclica usualmente a partir da metade do outono, na maioria destes animais. Esta resposta é devida à diminuição gradual no fotoperíodo natural, considerado como fator primário na sazonalidade reprodutiva. A ausência de ciclos ovulatórios durante os meses de inverno e a inconsistência da fase de ressurgência atingem não só os criadores de raças que procuram o nascimento de potros mais cedo na temporada reprodutiva oficial, mas principalmente aqueles que empregam a técnica da transferência de embriões. A obtenção de ovulações ao longo do ano possibilitaria a utilização reprodutiva ótima de éguas doadoras e receptoras. A fim de verificar a ciclicidade reprodutiva durante outono e inverno, dezessete éguas em uma propriedade do sul do Brasil foram distribuídas ao acaso em dois grupos. De quinze de Janeiro a doze de Setembro de 2009, os animais do grupo Tratamento foram submetidos a fotoperíodo diário fixo de 15 horas. Duas vezes por semana, o trato reprodutivo de todos os animais foi avaliado por ultrassonografia transretal, para verificação da presença de folículos dominantes e corpo lúteo. Dosagem de progesterona foi utilizada para confirmação de anestro. Oitenta e cinco por cento dos animais submetidos ao tratamento mantiveram ciclicidade reprodutiva, enquanto oitenta por cento dos animais controle entraram em anestro de inverno. Houve aumento na duração dos ciclos, devido a fatores endógenos, nutricionais e ambientais. Os resultados demonstram que o tratamento foi efetivo para obtenção de ciclos ovulatórios durante os meses de inverno. / The mare is classified as a seasonal poliestric, which means the suspension of reproductive activity usually from the first half of autumn, most of the mares. This is a response to the gradual decline in the natural photoperiod, considered as a primary factor of reproductive seasonality. The absence of ovulatory cycles during winter months and the inconsistency of the resurging phase strike both breeders who aim foals’ birth early in the official breeding season and those who employ the embryo transfer technique. Achieving ovulation throughout the year would enable the optimal reproductive use of donors and recipient mares. In order to check autumn and winter cyclicity, seventeen mares on a farm in southern Brazil were randomly assigned into two groups. From January 15th to September 12th 2009, animals in Treatment group were subjected to a fixed 15 hours daily photoperiod. Twice a week, the reproductive tract of all animals was examined by transrectal ultrasonography, searching for dominant follicles and presence of corpora lutea. Progesterone levels were used to confirm winter anestrus. 85% mares undergoing treatment maintained reproductive activity, while 80% of Control mares showed winter anestrus. Fotoperíodo artificial sobre a atividade reprodutiva de éguas durante a transição outonal. / Artificial photoperiod on reproduction activity of mares during autumn transition

O presente experimento verificou se o fotoperíodo artificial iniciado no verão retarda o início da transição outonal e nos casos de ocorrência do anestro determinar sua forma de início. Foram utilizadas no experimento 13 éguas com idades variando entre 4 e 10 anos. Todas as éguas no início do experimento encontravam-se ciclando, com presença de corpo lúteo funcional ou folículo dominante. Antes do início do experimento os animais foram divididos em dois grupos, Grupo Luz (GL) com 08 animais e Grupo Controle (GC) com 05 animais, buscando equilibrar a idade e a condição corporal das éguas nos diferentes grupos. A partir do dia 23 de fevereiro de 2006 as éguas do GL foram submetidas a um fotoperíodo artificial de 15 horas de luz e de 9 horas de escuridão, e as éguas do GC foram mantidas sob fotoperíodo natural. Em relação ao início do anestro e à condição corporal não foram observadas diferenças entre o GL e o GC. Dentre os 13 animais utilizados no experimento todos entraram em anestro. Não se observaram diferenças entre os dois grupos de tratamento em relação ao número de dias entre o início do tratamento e a última ovulação e os inícios do tratamento e do anestro, bem como entre o número de dias de duração do último corpo lúteo. Entretanto observou-se diferença no grupo luz onde 75% das éguas apresentaram diestro prolongado enquanto que no grupo controle isto ocorreu em somente 40% das éguas. O anestro inicia por persistência lútea seguida de inatividade folicular ou por inatividade folicular após um diestro regular. O fotoperíodo artificial iniciado a partir da segunda metade do verão não impediu o início do anestro. Entretanto, a maioria das éguas submetidas ao fotoperíodo artificial iniciou o anestro após uma persitência lútea. / The present study aimed to verify if the artificial photoperiod started during summer retards the beginning of autumn transition and if anestrus occurs, to determine the way it begins. There were used 13 mares with ages varying between 4 and 10 years. All the mares were cycling in the beginning of the study, having either a functional corpus luteum or a dominant follicle. Before starting the experiment, the animals were divided in two groups: Light Group (LG) with 08 animals and Control Group (CG) with 05 animals, trying to equilibrate age and body condition of the different group mares. After February 23rd, 2006, the mares of LG were submitted to artificial photoperiod of 15 hours light and 9 hours dark, and the mares of CG were kept under natural photoperiod. Concerning the beginning of anestrus and body condition, no differences were observed between LG and CG. Among the 13 animals used in the study, all entered anestrus period. No differences were observed between the treatment groups about the number of days between the beginning of the treatment and the last ovulation, and the beginning of treatments and anestrus, as well as the number of days of duration of the last corpus luteum. However, differences were found in the light group, where 75% of the mares presented a prolonged diestrus while in the control group it only happened in 40% of the mares. The anestrus begins by luteal persistency followed by follicular inactivity or by follicular inactivity after a regular diestrus. The artificial photoperiod initiated after the second half of the summer did not affect the beginning of the anestrus. However, most of the mares submitted to artificial photoperiod initiated anestrus after a luteal persistency.
