Bowdichia virgilioides is popularly known as "sucupira-preta", and used in folk medicine for the treatment of various diseases such as inflammation, diabetes, arthritis and skin diseases. This study was undertaken to evaluate the antinociceptive and anti-inflammatory effects of crude aqueous extract of the bark of Bowdichia virgilioides (EABv) in different experimental models. When administered orally, the EABv reduced the nociception by acetic acid, its effects continued for at least 4 h, a phenomenon which was not reversed by pretreatment with naloxone. Moreover, the extract did not change in the time of latency measured in the model of thermal nociception (hot plate). Thus, the EABv reaches its antinociceptive effects by a pathway independent of the opioid system and with peripheral action. These antinociceptive effects were maintained when even when the extract was prepared from bark collected in different months of the year, indicating that the antinociceptive compounds does not suffer from seasonal influences. The EABv give by i.p. route reaches antinociceptive effects in a concentration ten times lower than the extract administered orally. The EABv was unable to interfere with plasma leakage induced by carrageenan, but was effective in inhibited neutrophilia at 4 h after stimulation. In allergic inflammation, the extract inhibited not only the anaphylactic edema, but also the eosinophilia by a mechanism that seems to depend on the protein production of IL-5 and CCL11. The EABv inhibited the release of histamine after antigenic stimulation of the sensitized tissue in vitro, suggesting a direct effect on the mast cells degranulation. In the model of pellet cotton granuloma formation, the EABv inhibited both the formation of granulomatous tissue on the lymphoproliferative response in lymph nodes. Furthermore, the EABv inhibited the leukocytosis induced by zymosan and the innate immune response shown by the decrease in the rate of phagocytosis by leukocytes. In addition, the EABv not revealed any toxic effect associated with its use. Together, our results support the popular use of the aqueous extract of the bark of B. virgilioides based on their analgesic, anti-inflammatory, immunosuppressive and anti-alergic effects, associated with a low toxicity. / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Bowdichia virgilioides é conhecida popularmente como sucupira-preta , sendo empregada na medicina popular para o tratamento de diferentes agravos, tais como: doenças inflamatórias, diabetes, artrite e dermatoses. No presente estudo avaliamos o efeito antinociceptivo e antiinflamatório do extrato aquoso bruto da casca de Bowdichia virgilioides (EABv) utilizando diferentes modelos experimentais. Quando administrado por via oral, o EABv além de reduzir a nocicepção induzida pelo ácido acético, manteve seus efeitos por no mínimo 4 h, fenômeno que não foi revertido pelo tratamento prévio com naloxona. Além disso, o extrato não interferiu no tempo de latência avaliada no modelo de nocicepção térmica (placa quente). Assim, o EABv atinge seus efeitos antinociceptivos por uma via independente do sistema opióide e com ação periférica. Tais efeitos antinociceptivos foram mantidos mesmo quando o extrato foi preparado com cascas coletadas em diferentes meses do ano, indicando que a(s) substância(s) responsável (eis) por este efeito biológico não sofrem influência sazonal. Vale destacar que o EABv administrado por via intraperitoneal atingiu efeitos antinociceptivos em concentração dez vezes menor do que o extrato administrado por via oral. O extrato não foi capaz de interferir com o acúmulo de proteína induzido pela injeção intratorácia de carragenina, porém foi eficaz em suprimir neutrofilia no tempo de 4 h após estímulo. Na inflamação alérgica, o extrato inibiu além do edema anafilático, a eosinofilia pleural, sendo que este influxo celular parece depender da expressão das mensagens IL-5 e CCL11. Na ativação tecidual por antígeno in vitro, o EABv inibiu a liberação de histamina após estimulação antigênica, sugerindo efeito direto sobre a desgranulação de mastócitos. No modelo de formação de granuloma, o EABv suprimiu tanto a formação do tecido granulomatoso quanto a resposta linfoproliferativa nos linfonodos braquiais. Além disso, o EABv inibiu a leucocitose induzida por zimosan, bem como a resposta imune inata revelada pela redução na taxa de fagocitose pelos leucócitos. Em adição, o EABv não revelou nenhum efeito tóxico associado ao seu uso. Em conjunto, nossos resultados sustentam o uso popular do extrato aquoso da casca de B. virgilioides tendo por base seus efeitos antinociceptivo, antiinflamatórios, antialérgicos e imunossupressores, associada a ausência de toxicidade.
