Participação das vias alfa 2 - adrenérgica, dopaminérgica e gabaérgica no controle da secreção de GH no período pré-púbere de novilhas da raça nelore /

Orientador: Guilherme de Paula Nogueira / Banca: Cláudia Maria Bertan Membrive / Banca: Ciro Moraes Barros / Resumo: Foi avaliada a variação na concentração de GH em resposta a agonistas ou antagonistas de neurotransmissores durante o período pré-púbere em 10 novilhas da raça Nelore. A administração de um antagonista gabaérgico (picrotoxina, 0,18 mg/kg, I.V., amostras a cada 15 min por 10 h) aumentou a concentração de GH aos 10 meses de idade, mas não aos 14 e 17 meses. Tanto no grupo Picrotoxina, como no Controle a concentração média de GH foi maior aos 10 meses do que aos 17 meses de idade. O grupo picrotoxina apresentou aos 17 meses maior área de concentração de GH que o controle. O grupo picrotoxina apresentou aos 14 meses maior área total de picos que o controle. Aos 10 e 14 meses no grupo Picrotoxina houve maior amplitude de picos do que aos 17 meses. Bloqueando o receptor (D2) dopaminérgico (sulpiride, 0,59 mg/kg, SC, amostras a cada 15 min por 10 h) houve aumentou na concentração de GH aos 8 e 12 meses, mas não aos 16 meses de idade, comparado ao grupo controle. No grupo Sulpiride a concentração média de GH foi maior aos 8 e 12, comparado com os 16 meses de idade. Em relação à área total de secreção, área total de picos, e amplitude máxima de picos de secreção de GH não houve diferença entre os grupos. Aos 8 meses o grupo Sulpiride apresentou menor tempo, porém aos 12 meses o tempo foi maior para o primeiro pico em relação ao controle. O tempo para o primeiro pico foi menor aos 8 e 16 meses que aos 12 meses no grupo Sulpiride. No grupo controle aos 12 meses o tempo para o primeiro pico foi menor que aos 8 e 16 meses. Aos 16 meses o grupo Sulpiride apresentou maior freqüência de picos que o Controle. A aplicação de um estimulador alfa-adrenérgico (clonidina, 10 µg/kg, I.V., amostras a cada 15 min por 4 h) aumentou a concentração média de GH aos 8 e 15 meses em relação ao grupo Controle e em alguns intervalos aos 12 meses... A gabaergic antagonist (picrotoxin, 0.18 mg/kg,I.V., samples every 15 min for 10h) promoted an increase on GH concentration at 10 months of age but not at 14 and 17 mo. Inside Sulpiride group time was lower at 8 and 16 month compared to 12. In the control group time for the first peak was lower at 12 than 8 and 16 months. At 16 month the Sulpiride group had higher peak frequency than Control. O objetivo deste estudo foi determinar os efeitos da aplicação de hCG no quarto dia após a inseminação a tempo fixo (IATF) sobre o tamanho do corpo lúteo (CL) existente, a indução de CL acessórios, a concentração de progesterona (P4) sérica e taxa de prenhez de vacas de corte em lactação. Nos três experimentos, vacas de corte multíparas (n=569), cruza Bos taurus, com período pós-parto entre 45 e 70 dias, foram sincronizadas com a administração de 2mg de benzoato de estradiol i.m. e a inserção de um dispositivo intravaginal contendo 0,750g de P4 (Dia -9). Sete dias após, administraram-se 150 μg de D-cloprostenol e 0,5 mg de cipionato de estradiol, i.m., no momento da retirada do dispositivo (Dia -2). Sessenta vacas de corte em lactação foram divididas aleatoriamente em dois grupos: hCG (n=30) tratadas com 1500UI i.m. de hCG e Controle (n=30) injetadas com 1,5 mL de solução salina i.m. no dia 4 depois do estro (Dia 0). Realizaram-se coletas sanguíneas dos animais para mensuração da concentração sérica de P4 nos Dias 4, 7, 10 e 14 do ciclo estral. Para acompanhamento da dinâmica ovariana, sessenta animais foram divididos aleatoriamente nos grupos hCG (n=30) ou Controle (n=30) e receberam o mesmo protocolo hormonal como citado anteriormente. No Dia 0, os ovários foram examinados por ultrassonografia transretal para determinar o diâmetro do folículo ovulatório; no Dia 4, o diâmetro do FD e presença do CL e no Dia 7 para mensurar a área luteal do CL e a presença de CL acessório. Quinhentas e sessenta e nove vacas (hCG, n= 269 e Controle, n= 300) foram inseminadas a tempo fixo 52-56 horas depois da retirada dos implantes de P4 e tratadas no Dia 4, conforme o protocolo descrito. O diagnóstico de prenhez foi realizado por ultrassonografia 30 dias após a IATF. Os resultados observados, mostraram que vacas tratadas com hCG no Dia 4 apresentaram maiores concentrações sérica de P4 no Dia 7, comparadas com as do grupo Controle (4,45 vs. 3,37ng/mL, respectivamente; p<0,05), mas níveis semelhantes de P4 nos Dias 10 e 14. Os animais do grupo hCG apresentaram CL com maior (p<0,01) área luteal no Dia 7, em relação ao grupo Controle (3,52 cm² vs. 2,66 cm², respectivamente) e uma incidência de 29,6 % de indução da ovulação do FD presente no Dia 4. Observou-se uma taxa de prenhez maior (p=0,071) no grupo tratado com hCG (53,9%) comparando-se com as vacas não tratadas (46,3%). Conclui-se que a administração de hCG no quarto dia do ciclo estral promove o aumento da área do CL, melhora função luteal, pode induzir a formação de CL acessório e tende a aumentar a taxa de prenhez de vacas de corte em lactação. / The aim of this study was to determine the effects of injection of hCG on the fourth day after the TAI on the size of the existing corpus luteum (CL), the induction of CL accessories, the concentration of progesterone (P4) levels and the possibility of increasing the rate of pregnancy of lactating beef cows subjected to synchronization of estrus and ovulation. Multiparous beef cows (n= 569), Bos taurus crossbreed with postpartum between 45 and 70 days were synchronized by administration of 2mg of estradiol benzoate im and the insertion of an intravaginal device containing 0,750g of P4 (Day -9). Seven days later, were administered 150mg of D- cloprostenol 0.5 mg of estradiol cypionate, im, at the time of device removal (Day -2). Sixty lactating beef cows were randomly divided into two groups: hCG (n= 30) treated with im 1500UI hCG and Control (n= 30) injected with 1.5 ml of saline im on day 4 after estrus (Day 0). There were blood collections of animals for measurement by radioimmunoassay of serum P4 on Days 4, 7, 10 and 14 of the estrous cycle. To evaluate the ovarian dinamics, sixty animals (hCG, n= 30 and Control, n = 30) received the same hormonal protocol and were uniformly distributed into groups according to the diameter of the ovulatory follicles on day 0 (estrus). On Day 0, the ovaries were examined by transrectal ultrasonography to determine the diameter of the ovulatory follicle, on Day 4, the diameter of the dominant follicle (DF) and the presence of CL on Day 7 to measure the area of the luteal CL and the presence of accessory CL. Five hundred and sixty-nine cows (hCG, n= 269 and Control, n= 300) were inseminated at a fixed time 52 to 56 hours after removal of the P4 devices treated on Day 4, according to the protocol described previously. The diagnosis of pregnancy were performed by ultrasonography 30 days after TAI. The results demonstrate that cows treated with hCG on Day 4 showed higher serum concentrations of P4 on Day 7, compared with the control group (4.45 vs. 3.37ng/mL, respectively, (p<0.05), but similar levels of P4 on Days 10 and 14. The animals of hCG group had greater (p<0,01) luteal area on Day 7, compared to the Control group (3.52cm² vs. 2.66cm², respectively) and a 29.6% incidence of induction DF ovulation present on Day 4. We observed a higher rate of pregnancy (p= 0.071) in the group treated with hCG (53.9%) comparing with untreated cows (46.3%). We conclude that administration of hCG on the fourth day of the estrous cycle increase the CL area, improves luteal function, can induce the formation of accessory CL and tends to increase the pregnancy rate of lactating beef cows.

Effects of body condition, body weight and calf removal on productive and reproductive characteristics of extensive beef cattle in Mozambique

The objectives of the present research were to study the factors that influence postpartum reproductive characteristics of suckling beef cows in extensive production systems in Mozambique and to develop new management strategies to improve their reproductive efficiency. The effects and interactions between post-partum BW, BCS, age and parity number on plasma concentrations of estradiol, progesterone, creatinine, urea and cortisol around oestrus and the related conception rates of Bos indicus cows in extensive production system were analysed as well as the minimum BCS at the beginning of breeding to maximise the subsequent conception rates. Thirty-five days prior to the breeding season cows were synchronised using Crestar. During the second oestrus after synchronisation, 18 blood samples were collected per animal for hormonal analysis, from 24 hr before oestrus to 24 hr after oestrus. The hormonal pattern of estradiol and progesterone around oestrus were similar to that observed in Bos taurus cows under intensive conditions. Conception rates of cows in the experimental group were 90.5%. Better results on estradiol pattern and conception rates were related to a BCS of ≥ 2.5 and it was thus concluded that the post-partum management of extensive Bos inducus cows should aim to achieve at least a BCS of 2.5 at the beginning of the breeding season. Twelve-hour and 48-hr calf removal were conducted separately to evaluate their effects on conception rates of Bos indicus beef cows in extensive production systems and to quantify the related effects on calf-weaning weights. The 12-hr calf removal was performed from 45 days post-partum to the beginning of the breeding season, and the 48-hr calf removal was performed preceding the onset of the breeding season. It was concluded that 12-hr calf separation at night enhance the energy balance (3%), increases the conception rates (80%) and improves the calf-weaning weights, whereas 48-hr calf removal increases conception rates (76%) and does not affect calf weaning weights. Avaliação genômica em bovinos da raça Gir de Brasil e Colômbia /

Estudos genômicos entre populações são de interesse para identificar regiões genômicas importantes e aplicar na seleção de caraterísticas de produção. As corridas de homozigose (ROH) são regiões homozigotas contíguas do genoma, utilizadas na identificação de genes associados a características de interesse econômico, bem como na obtenção dos coeficientes de endogamia. A interação genótipo ambiente (IGA), também representa papel importante no estudo de populações de bovinos e na seleção dos melhores reprodutores para os diferentes ambientes. Com isto, os objetivos do presente estudo foram analisar o comprimento e a distribuição das ilhas ROH do genoma, com identificação dos gene presentes, além de avaliar a acurácia da imputação utilizando diferentes painéis comerciais e a interação genótipo ambiente da produção de leite em bovinos da raça Gir do Brasil e Colômbia. Na avaliação das ilhas ROH foram utilizados dados genotípicos de 173 animais selecionados para produção de carne e 291 animais selecionados para produção leiteira e obtidos os resultados via programa Plink. Análise da acurácia utilizando diferentes painéis de SNPs (GGP Bovine 30K, GGP indicus 35K e HD 777K) de 464 animais do Brasil e da Colômbia foram avaliados e os resultados comparados via correlação simples (CS) e taxa de concordância (CR). Genomic studies between populations are of interest to identify important genomic regions and apply in the selection of production traits. Runs of Homozygosity (ROH) are contiguous homozygous regions of the genome, used to identify genes associated with characteristics of economic interest, as well as to obtain inbreeding coefficients. The environment genotype (IGA) interaction also plays an important role in the study of bovine populations and in the selection of the best breeders for different environments. Thus, the objectives of the present study were to analyze the length and distribution of the ROH islands of the genome, with identification of the present genes, in addition to assessing the accuracy of imputation using different commercial panels and the environment genotype interaction of milk production in cattle of the Gir breed from Brazil and Colombia. IMPROVING LIVESTOCK CLIMATIC ADAPTATION THROUGH GENOMICS

<p> As the effects of climate change become more evident, the development of effective strategies for improving livestock climatic adaptation and the long-term sustainability of animal food production have become key priorities around the world, including in the US. Together with nutrition, infrastructure, and management practices, genetically improving animals is an effective and lasting alternative to simultaneously improve productive efficiency and climatic adaptation of animals. Genetic improvement requires basic understanding of the genomic architecture of the indicator traits of interest and the availability of large-scale datasets. Understanding the role of evolution and selection (both natural and artificial) on shaping animal genomes is of paramount importance for the optimization of breeding programs and conservation of genetic resources. In addition, properly quantifying environmental stress and individual animal responses to thermal stress are still important challenges in breeding programs. Thus, the identification of optimal statistical methods and traits that better capture key biological mechanisms involved in the heat stress response has the potential to enable more accurate selection for thermal tolerant individuals. Therefore, this thesis aimed to investigate complementary topics related to thermal tolerance in livestock species based on genomic information. A total of 946 genotypes from 34 cattle breeds, as well as Datong yak (<em>Bos grunniens</em>) and Bali (<em>Bos javanicus</em>) populations, adapted to divergent climatic conditions, were used to investigate the genetic diversity and unravel genomic regions potentially under selection for thermal tolerance, with a focus on Chinese local cattle breeds and yak. Different signature of selection analyses and a comprehensive description of genetic diversity in 32 worldwide cattle and Datong yak populations was presented. Moderate genetic diversity was observed within each Chinese cattle population. However, these results highlighted the need to adopt strategies to avoid further reduction in the genetic diversity of these populations. Several candidate genes were identified as potentially under selection for thermal tolerance, and important biological pathways, molecular functions, and cellular components were identified, which contribute to our understanding of the genetic background of thermal tolerance in <em>Bos</em> species. Secondly, 8,992 genotyped individuals were used to provide a comprehensive description of genotype-by-environment interaction effects, defining optimal environmental variables based on public weather station data, and critical periods to evaluate heat tolerance for various reproduction, growth, and body composition traits in US Large White pigs. The period of 30 days before the measurement date was suggested to analyze genotype-by-environment interaction for off-test weight, muscle depth, and backfat thickness. While for number of piglets weaned and weaning weight, the suggested period ranged from the last trimester of gestation until weaning. This same population was used to access the genomic predictive ability of heat tolerance based on routinely-measured traits and explore candidate regions involved in the biological mechanisms that underlie heat stress response in pigs. Genotype-by-environment interaction was identified for most of the traits evaluated, and moderate (>0.36 ± 0.05) breeding values prediction accuracy were achieved using genomic information. Lastly, various behavioral, anatomical, and physiological indicators of heat stress were measured in a population of 1,645 multiparous Large White x Landrace lactating sows. This dataset was used to identify the best statistical models and estimate genomic-based genetic parameters for 23 indicators of heat stress, including automatically-measured vaginal temperature, skin surface temperatures, respiration efficiency, respiration rate, panting score, body condition scores, hair density, body size, and ear measurements. All the traits evaluated are heritable, with heritability estimates ranging from 0.04 ± 0.01 to 0.40 ± 0.09. The genetic correlations among these traits ranged from -0.49 (between repeated records of vaginal temperature measured at 0800 hours and caliper body condition score) to 1.0 (between repeated records of vaginal temperature measured at 0800 hours and single record of vaginal temperature measured at 0800 hours; and between repeated records of vaginal temperature measured at 1200 hours and single record of vaginal temperature measured at 1200 hours). These findings indicate that genetic progress for thermotolerance in pigs can be achieved through direct indicators of heat stress in selection schemes. However, special attention is needed due to complex relationship between these traits as evidenced by their genetic correlations. In conclusion, this thesis provides important information to be used when designing breeding strategies for improving thermal tolerance in cattle and pigs, important genomic regions and metabolic pathways that are important for understanding the biological mechanism regulating thermal tolerance, as well as future directions for investigations in the area of livestock climatic adaptation.</p>

Fusion of bovine fibroblasts to mouse embryonic stem cells: a model to study nuclear reprogramming

The cells from the inner cell mass (ICM) of an early embryo have the potential to differentiate into all the different cell types present in an adult organism. Cells from the ICM can be isolated and cultured in vitro, becoming embryonic stem cells (ESCs). ESCs have several properties that make them unique: they are unspecialized, can self-renew indefinitely in culture, and given the appropriate cues can differentiate into cells from all three germ layers (ecto-, meso-, and endoderm), including the germline, both in vivo and in vitro. Induced pluripotent stem cells (iPSCs) can be generated from adult, terminally differentiated somatic cells by transient exogenous expression of four transcription factors (Oct4, Sox2, Klf4, and cMyc; OSKM) present normally in ESCs. It has been shown that iPSCs are equivalent to ESCs in terms of morphology, gene expression, epigenetic signatures, in vitro proliferation capacity, and in vitro and in vivo differentiation potential. However, unlike ESCs, iPSCs can be obtained from a specific individual without the need for embryos. This makes them a promising source of pluripotent cells for regenerative medicine, tissue engineering, drug discovery, and disease modelling; additionally, in livestock species such as the bovine, they also have applications in genetic selection, production of transgenic animals for agricultural and biomedical purposes, and species conservancy. Nevertheless, ESC and iPSC lines that meet all pluripotency criteria have, to date, only been successfully produced in mice, rats, humans, and non-human primates. In the first part of this dissertation, we attempted reprogramming of three types of bovine somatic cells: fetal fibroblasts (bFFs), adult fibroblasts (bAFs), and bone marrow-derived mesenchymal stem cells (bMSCs), using six different culture conditions adapted from recent work in mice and humans. Using basic mouse reprogramming conditions, we did not succeed in inducing formation of ESC-like colonies in bovine somatic cells. The combination of 2i/LIF plus ALK5 inhibitor II and ascorbic acid, induced formation of colony-like structures with flat morphology, that occasionally produced trophoblast-like structures. These trophoblast-like vesicles did not appear when an inhibitor of Rho-associated, coiled-coil containing protein kinase 1 (ROCK) was included in the medium. We screened for expression of exogenous OSKM vector with RT-PCR and found upregulation of OSKM vector 24h after Dox was added to the medium; however, expression was sharply decreased on day 2 after Dox induction, and was not detectable after day 3. In a separate experiment, we induced reprogramming of bFF and bAFs using medium supplemented with 50% of medium conditioned by co-culture with the bovine trophoblast CT1 line. These cells expressed both OCT4 and the OSKM vector 24h after Dox induction. However, similar to our previous observations, both markers decreased expression until no signal was detected after day 3. In summary, we were unable to produce fully reprogrammed bovine iPSCs using mouse and human protocols, and the exact cause of our lack of success is unclear. It is possible that a different method of transgene expression could play a role in reprogramming. However, these ideas would be driven by a rather empirical reasoning, extrapolating findings from other species, and not contributing in our understanding of the particular differences of pluripotecy in ungulates. Our inability to produce bovine iPSCs, combined with the only partial reprogramming observed by others, justifies the need for in depth study of bovine pluripotency mechanisms, before meaningful attempts to reprogram bovine somatic cells to plutipotency are made. Therefore, we focused on getting a better understanding of bovine nuclear reprogramming. This would allow us to rationally target the specific requirements of potential bovine pluripotent cells. Cell fusion is a process that involves fusion of the membrane of two or more cells to form a multinucleated cell. Fusion of a somatic cell to an ESC is known to induce expression of pluripotency markers in the somatic nucleus. In the second part of this dissertation, we hypothesized that fusion of bFFs to mouse ESCs (mESCs) would induce expression of pluripotency markers in the bFF nucleus. We first optimized a cell fusion protocol based on the use of polyethylene glycol (PEG), and obtained up to 11.02% of multinucleated cells in bFFs. Next, we established a method to specifically select for multinucleated cells originated from the fusion of mESCs with bFFs (heterokaryons), using indirect immunofluorescence. With this in place, flow cytometry was used to select 200 heterokaryons which were further analyzed using RNA-seq. We found changes in bovine gene expression patterns between bFFs and heterokaryons obtained 24h after fusion. Focusing on the bovine transcriptome, heterokaryons presented upregulation of early pluripotency markers OCT4 and KLF4, as well as hypoxia response genes, contrasted with downregulation of cell cycle inhibitors such as SST. The cytokine IL6, known to increase survival of early embryos in vitro, was upregulated in heterokaryons, although its role and mechanism of action is still unclear. This indicates that the heterokaryon cell fusion model recapitulates several of the events of early reprogramming, and can therefore be used for further study of pluripotency in the bovine. The cell fusion model presented here can be used as a tool to characterize early changes in bovine somatic nuclear reprogramming, and to study the effect of different reprogramming conditions on the bovine transcriptome. / Ph. D. / The cells of an early embryo have the potential to give rise to any cell type found in the adult body. When these cells are transferred to a culture dish and kept under the right conditions, they become Embryonic Stem Cells (ESCs), and they retain the same developmental potential as the original embryonic cells they were derived from. In 2006, researchers in Japan found that it is possible to “reprogram” the cells of an adult individual (for example, fibroblast skin cells taken from a biopsy) to an embryonic state, by forcing the cells to express extra copies of genes that are normally active in embryos. These reprogrammed cells are called induced Pluripotent Stem Cells (iPSCs), and similarly to ESCs, they also have the potential to produce any cell type found in an adult organism. Lines of iPSCs from livestock species have possible applications in agriculture, species conservancy, biomedical industry, and veterinary and human health. Unfortunately, for reasons that are to date not fully understood, the technology to produce iPSCs has, so far, only worked in mice, rats, humans, and non-human primates. We first attempted to produce bovine iPSCs by adapting methods and conditions used to derive iPSCs in mice and humans. We observed partial reprogramming of bovine cells, but were ultimately not able to produce true bovine iPSCs. This suggests that the bovine requires alternative/additional factors to induce reprogramming in adult cells. However, not knowing exactly what conditions or reagents will induce the reprogramming process in the cow, we decided to take a different approach. We focused on trying to understand how nuclear reprogramming works in the bovine. This would allow us to rationally target the specific requirements of potential bovine pluripotent cells. It is known that the fusion (“merging”) of an adult cell with a stem cell, causes the adult cell to change its gene expression pattern to resemble a stem cell. We therefore fused mouse ESCs with bovine fibroblasts, and observed changes in bovine gene expression pattern as early as 24 hours after fusion. The gene expression changes observed resemble those found during early reprogramming of human and mouse iPSCs, and are accompanied by silencing of fibroblast specific genes. This suggests that our cell fusion model recreates the changes that happen during reprogramming, and can therefore be used as a tool to better understand pluripotency in the cow. The cell fusion method described in this dissertation can in theory be adapted to other species; by fusing somatic cells from other species to mouse ESCs, this model can be used to find species specific relevant pluripotency genes.

Cette thèse propose l’examen des textes de théorie de l’art et de critique d’art portant sur la peinture en France, aux XVIIe et XVIIIe siècle, suivant l’angle de la vraisemblance. De nombreuses réflexions sur la légitimité de la critique de ceux qui ne pratiquent pas la peinture ont précédé la naissance de la critique d’art, et la question fait toujours débat au XVIIIe siècle. Or, la notion de vraisemblance, présente dans le discours des théoriciens (peintres ou non) du point de vue de la réalisation des œuvres, manifeste la prise en compte d’un regard porté sur le tableau, c’est-à-dire du point de vue de la réception : est-ce que l’ « on » peut croire à ce que le tableau représente ? Ce que nous souhaitons montrer est le rôle crucial que joue la vraisemblance dans la légitimation de la critique d’art. Si un spectateur, qui n’est pas peintre, n’est pas à même de juger la qualité du dessin ou du coloris dans une œuvre, il est en droit de dire si ce que le tableau lui présente est vraisemblable ou non.Notre étude cherche à comparer les différentes acceptions que le terme de vraisemblance prend sous la plume des théoriciens et des critiques, afin d’une part de mettre en évidence que cette notion est considérée des deux côtés comme une règle en peinture, et de préciser son évolution dans les discours au XVIIIe s. / This thesis offers to examine theory of art and art criticism texts about painting in 17th and 18th c. France, via the angle of verisimilitude. Numerous thoughts upon the legitimacy of the criticism of those who do not practice painting have preceded the birth of art criticism, and the debate is still going on during the 18th c. However, the notion of verisimilitude, present in art theory discourses (by painters or not) about the creation of an art piece, manifests that the perception of the painting by someone looking at it is taken into account, this time from the point of view of reception: can “one” believe what the painting shows? We aim at proving the crucial role of verisimilitude in giving art critic legitimacy. If a spectator – who is not a painter – has no legitimacy to judge the quality of drawing or colour in an art piece, he can say whether the painting is credible or not.We compare and study the several acceptations of verisimilitude in theorists and critics’ writings in order to highlight that the notion is considered as a rule of painting on both sides, and analyse its evolution in 18th c. discourses.
