Temperature responsive polymer brushes with clicked rhodamine B: synthesis, characterization and swelling dynamics studied by spectroscopic ellipsometry

Here, we report on a new temperature responsive polymer brush system with a terminal “click” functionality. Bifunctionalized poly(N-isopropylacrylamide) (PNiPAAm) with distinct functional end groups was synthesized by atom transfer radical polymerization (ATRP) and grafted to a modified silicon substrate. The presence of the active terminal alkyne functionality is validated using an azide-modified rhodamine B (N3-RhB) via copper(I) catalyzed alkyne–azide cycloaddition (CuAAC). The optical properties and swelling dynamics of an N3-RhB modified PNiPAAm brush are analyzed in dry state and in situ by VIS-spectroscopic ellipsometry (SE). The best-fit results are obtained using a Gaussian oscillator model and are confirmed by UV/VIS-spectroscopy. Conjugated Polymer Brushes (Poly(3-hexylthiophene) brushes): new electro- and photo-active molecular architectures

The aim of the present work was to screen the main methods for the synthesis of conjugated polymers for their suitability in the preparation of conductive polymer brushes. The main focus was put on the grafting of intrinsically soluble substituted regioregular polyalkylthiophenes because of their excellent optoelectronic properties. The resulting polymer films were characterized and their optoelectrical properties studied. For the first time, a synthesis of conductive polymer brushes on solid substrates using “grafting-from” method was performed. The most important, from my opinion, finding of this work is that regioregular head-to-tail poly-3-alkylthiophenes – benchmark materials for organic electronics - can be now selectively grafted from appropriately-terminated surfaces to produce polymer brushes of otherwise soluble polymers - the architecture earlier accessible only in the case of non-conductive polymers. In particular, we developed a new method to grow P3ATs via Kumada Catalyst Transfer Polymerization (KCTP) of 2-bromo-5-chloromagnesio-3-alkylthiophene. Exposure of the initiator layers to monomer solutions leads to selective chain-growth polycondensation of the monomers from the surface, resulting into P3AT brushes in a very economical way. The grafting process was investigated in detail and the structure of the resulting composite films was elucidated using several methods. The obtained data suggests that the grafting process occurs not only at the poly(4-bromstyrene) (PS-Br)/polymerization solution interface, but also deeply inside the swollen PS-Br films, penetrable for the catalyst and for the monomer The grafting process was investigated in detail and the structure of the resulting composite film was elucidated using ellipsometry, X-ray Photoelectron Spectroscopy (XPS), Rutherford backscattering spectroscopy (RBS), and Conductive atomic force microscopy (C-AFM). The obtained data suggests that the grafting process occurs not only at the poly(4-bromostyrene), PS-Br/polymerization solution interface, but also deeply inside the swollen PS-Br film, which is penetrable for the catalyst and the monomer. The process results in an interpenetrated PS-Br/P3HT network, in which relatively short poly(3-hexylthiophene), P3HT grafts emanate from long, cross-linked PS-Br chains. A further method investigated during our work was to covalently graft regioirregular P3HT to substrates modified by macromolecular anchors using oxidative polymerization of 3HT with FeCl3. P3HT layers with variable thicknesses from 30 nm up to 200 nm were produced using two steps of polymerization reaction. The P3HT obtained by oxidative polymerization had always an irregular structure, which was a result of the starting monomer being asymmetric, which is undesired for electronic applications. The third method for the production of conductive polymer brushes was to graft regioregular poly(3,3''-dioctyl-[2,2';5',2'']terthiophene) (PDOTT) by electrochemical oxidative polycondensation of symmetrically substituted 3,3''-dioctyl-[2,2';5',2'']terthiophene (DOTT). A modification of the supporting ITO electrode by the self-assembled monolayers (SAMs) of compounds having polymerizable head-groups with properly adjusted oxidative potentials was found to be essential to achieve a covalent attachment of PDOTT chains. The polymer films produced show solvatochromism and electrochromism, as well as the previous two methods. After polymerization, the next step towards building organic electronic devices is applying the methods obtained in nano- and microscale production. Block copolymers constitute an attractive option for such surface-engineering, due to their ability to form a variety of nanoscale ordered phase-separated structures. However, block copolymers containing conjugated blocks are less abundant compared to their non-conjugated counterparts. Additionally, their phase behaviour at surfaces is not always predictable. We demonstrated in this work, how surface structures of non-conductive block copolymers, such as P4VP-b-PS-I, can be converted into (semi)conductive P4VP-b-PS-graft-P3HT chains via a surface-initiated polymerization of P3HT (Kumada Catalyst Transfer Polymerization (KCTP) from reactive surface-grafted block copolymers. This proves that our method is applicable to develop structured brushes of conductive polymers. We believe that it can be further exploited for novel, stimuli-responsive materials, for the construction of sensors, or for building various opto-electronic devices. The methods developed here can in principle be adapted for the preparation of any conductive block copolymers and conductive polymers, including other interesting architectures of conductive polymers, such as block copolymers, cylindrical brushes, star-like polymers, etc. To this end, one needs to synthesize properly-designed and multi-functional Ni-initiators before performing the polycondensation.

Micro- and nanostructured polymer brushes on diamond can be directly prepared by carbon templating and amplification of the latent structures by photografting of a broad variety of vinyl monomers such as styrenes, acrylates and methacrylates. Even template structures with lateral dimensions as small as 5 nm can be selectively amplified and defined polymer brush gradients of a variety of functional polymers are realizable by this technique. Furthermore, conjugation with a model protein (GFP) results in protein density gradients of high loading and improved chemical stability. Les populations neuronales ont la capacité de s’organiser en réseaux qui produisent spontanément différents patterns d’activité électrique coordonnée. Au travers de leur activité synchrone, les réseaux immatures combinent les informations génétiques et les influences environnementales et contrôlent plusieurs processus neuro-développementaux dont la plasticité synaptique. Pendant le développement prénatal et post-natal précoce, ces activités électriques synchrones peuvent être générées au sein des cortex sensoriels eux-mêmes, des structures sous-corticales ou être évoquées par l’activité des organes sensoriels, elle-même spontanée ou provoquée par les stimuli sensoriels. Dans des travaux antérieurs, nous avons montré qu’un pattern EEG typique du prématuré, le « Delta-brush » (DB), qui associe une onde lente et des oscillations rapides, pouvait être évoqué dans les cortex primaires sensori-moteur, visuel et auditif par les mouvements spontanés et par les stimuli sensoriels correspondants. L’objectif de la présente étude, dédiée aux réponses corticales aux stimuli auditifs (click), était de préciser les caractéristiques spatio-temporelles des DBs évoqués ainsi que leur rapport avec des potentiels évoqués auditifs corticaux. Pour cela, les enregistrements EEG ont été réalisés en haute résolution (32 électrodes) chez 30 nouveau-nés prématurés de 30 à 38 semaines d’âge gestationnel sans risque neurologique et la position des électrodes a été recalée sur des IRM 3D acquises chez d’autres prématurés représentatifs des âges étudiés. L’analyse de population a montré une augmentation significative de la puissance spectrale après le stimulus dans toutes les bandes de fréquence allant du delta à gamma et située au niveau de la partie moyenne et postérieure du lobe temporal. Ces réponses du cortex temporal avaient une prédominance droite, étaient plus amples dans le sommeil calme et diminuaient en puissance avec l’âge. Le moyennage des réponses EEG a révélé que la composante lente du DB était une onde lente négative de grande amplitude qui culminait dans les régions temporales moyenne et postérieure à environ 550 et 700 ms respectivement. L’analyse temps-fréquence a confirmé la présence d’oscillations rapides dont les oscillations gamma, à partir du pic de l’onde lente et cohabitant avec cette dernière durant environ 700 ms. Ces résultats suggèrent que le DB évoqué par les stimuli auditifs correspond en fait à la composante lente tardive du potentiel évoqué auditif cortical du prématuré et qu’il regroupe des oscillations dans toutes les bandes de fréquence, gamma y compris, fréquence identifiée ici pour la première fois en réponse à un stimulus sensoriel chez le prématuré humain. Nous avons obtenu des résultats préliminaires similaires aussi dans une autre modalité sensorielle ; l’activité oscillatoire du DB évoqué contribue donc probablement à la maturation des cartes corticales et représente un biomarqueur potentiel du fonctionnement normal des cortex sensoriels chez le prématuré. / At the early developmental stages, during the third trimester of gestation in humans and the first post-natal weeks in rodents, sensory neocortical areas reveal similar patterns of spontaneous correlated neuronal activity. In vitro and in vivo experiments indicate that these spontaneous activities are generated from neuronal networks in the cerebral cortex, in subcortical structures or in the sensory periphery (retina, limb jerks, whiskers). Spontaneous, periphery-driven and also sensory evoked activity is relayed to the developing cerebral cortex via the thalamus and the neocortical subplate, which amplifies the afferent sensory input. The patterns of sensory evoked activity were extensively studied in rodents, but in humans their spatiotemporal dynamics still remain elusive. In humans this developmental process happens during the second half of gestation: the major growing afferents from the thalamus spread within the transient subplate zone, relocate in the cortical plate, and form functional synapses with both transient and permanent neuronal populations. Characteristic immature activity patterns of “delta-brushes” (DBs) have been reported in the preterm temporal cortex following auditory stimuli. However, the spatiotemporal dynamics of these auditory-evoked DBs remain elusive. Here, we explored the electrophysiological responses evoked by click stimuli using 32-electrode EEG recordings in thirty premature infants from 30 to 38 postmenstrual weeks (PMW) of age. Electrodes position was digitalized and registered to 3D reconstructions of preterm heads and brains computed from MRI images of other age-matched groups. Population power spectrum analysis within the 2 seconds after stimulation revealed significant increase in all frequency bands from delta to gamma, located on the middle and posterior temporal regions with a right predominance and higher power increase in the quiet sleep. Time-frequency wavelet analysis also showed fast oscillations including gamma that begin at the peak of the delta waves and co-occur with it during a period of around 700 ms. Power of auditory evoked responses significantly decreased from 30 to 38 WPM in delta to alpha bands. These are the first report of gamma oscillations in preterm sensory evoked responses. Furthermore, average cortical auditory evoked potentials (CAEP) (processed with a mean reference and a 0.16 Hz high-pass filter) revealed high amplitude delta negative waves peaking successively from the middle to posterior temporal regions at around 550 and 700 ms. Altogether these results suggest that the auditory-evoked DBs in premature infants are a slow late component of the CAEP covering temporal regions and grouping fast oscillations notably gamma oscillations.

Die zusätzliche Modifizierung der Polymerbürsten mit Elektronenstrahlen ermöglicht die Generierung von Gradienten und erweitert die Anwendungsmöglichkeiten in vielfältigen Bereichen der interdisziplinären Forschung, wie z.B. in Hinblick auf das Lab-on-Chip Design oder die Biomedizin. / Polymer brushes represent sensitive coatings with thicknesses of few nanometres and are interesting for interdisciplinary scientific research, e.g. for intelligent lab-on-chip design or biotechnology. Based on the collected data can be further analyzed and innovation of the carbon brush.

Entwicklung von neuartigen thermoresponsiven Oberflächenbeschichtungen auf der Basis von Poly-2-oxazolinen

Im Rahmen dieser Arbeit wurden für die Herstellung neuartiger funktioneller Oberflächenbeschichtungen Polymerbürstensysteme basierend auf linearen, thermoresponsiven Poly-2-oxazolinen (POX) entwickelt und ausführlich charakterisiert. Als Grundbaustein für die Herstellung von POX-Bürsten wurden drei endfunktionalisierte thermoresponsive POX mittels kationisch ringöffnender Polymerisation synthetisiert, wobei jeweils 2-Cyclopropyl-2-oxazolin (cPrOX) als thermoresponsive Wiederholungseinheit und 2-Methyl-2-oxazolin (MeOX) als hydrophiles Comonomer zu verschiedenen Anteilen verwendet wurden. Mittels VIS-spektroskopischen Trübungsmessungen wurde für alle POX in Wasser ein LCST-Entmischungsverhalten mit einem reversiblen und scharfen Phasenübergang nachgewiesen, wobei die Trübungstemperaturen stark von der Polymerhydrophilie sowie der Molmasse und der Polymerkonzentration in Lösung abhingen. Die Herstellung der POX-Polymerbürsten auf der Basis der synthetisierten POX erfolgte über einen „grafting to“-Ansatz, bei welchem die Polymere in einem thermisch initiierten Prozess über ein funktionalisiertes Kettenende kovalent an ein Substrat angebunden wurden. Als Hauptmethode zur Charakterisierung der physikochemischen Eigenschaften der hergestellten Schichten, insbesondere deren temperaturabhängiges Schaltverhalten in Wasser, wurde spektroskopische Ellipsometrie verwendet. Zusätzlich kamen Rasterkraftmikroskopie, Kontaktwinkel und Quarzkristallmikrowaage mit Dissipationsaufzeichnung zum Einsatz. Im Gegensatz zum sehr scharfen, diskontinuierlichen Phasenübergang der POX in Lösung konnte für alle POX-Bürsten ein kontinuierlicher Übergang von einem gestreckten Bürstenzustand bei niedrigen Temperaturen in einen kollabierten Zustand bei hohen Temperaturen bestimmt werden, wobei das Quellvermögen und die temperaturabhängigen Quellkurvenverläufe durch die Polymereigenschaften und die Bürstenparameter beeinflussbar waren. Durch die Kombination eines POX mit Polyacrylsäure (PAA) als zweite Komponente konnten zudem neuartige binäre Polymerbürstensysteme hergestellt werden, welche ein komplexes pH- und temperaturabhängiges Schaltverhalten sowie ein steuerbares Proteinadsorptionsvermögen in Abhängigkeit von der Bürstenpräparationsmethodik, der POX-PAA-Zusammensetzung sowie der Temperatur und dem pH-Wert der umgebenden Pufferlösung aufwiesen. Zur ausführlichen Charakterisierung der Schichteigenschaften wurde neben den bereits benannten Methoden der spektroskopischen Ellipsometrie, Rasterkraftmikroskopie und der Kontaktwinkelmessung auch Röntgenphotoelektronenspektroskopie genutzt. Abschließende Zelladhäsionsexperimente mit humanen mesenchymalen Stammzellen auf den bürstenmodifizierte Oberflächen unter physiologischen Bedingungen offenbarten deutliche Unterschiede in der Affinität der Zellen zur Adhäsion auf POX-Homopolymerbürsten in Abhängigkeit der Bürstenhydrophilie. Darüber hinaus konnte die Zelladhäsion auch auf POX-PAA-Mischbürstensystemen gesteuert werden.

Además, cuentan con una afinidad por productos con tendencia ecológica y de cuidado personal. Se considera que estos usuarios tienen la disposición para pagar el valor agregado que BioSmile ofrece.El plan de marketing se enfocará en los potenciales clientes de BioSmile a través de campañas y promociones, las cuales se harán en redes sociales y ferias ecológicas para tener ventas de manera mensual.
