Atividade endectocida de uma nova alternativa terapêutica (S-cifenotrina, Butóxido de piperonila, D-tetrametrina e Ivermectina) em cães

Quatro experimentos foram conduzidos para avaliação terapêutica da nova associação, comparativamente à formulações adquiridas no mercado. Para avaliação carrapaticida e pulicida foram conduzidos dois experimentos. No primeiro ensaio, 21 cães naturalmente infestados por Rhipicephalus sanguineus, selecionados por meio de duas contagens consecutivas, foram randomizados e sorteados em três grupos de sete animais cada. No segundo ensaio, 21 cães selecionados foram randomizados e sorteados, também, em três grupos de sete animais cada. Infestações artificiais foram realizadas nos dias -4, -2, 6, 13, 20, 27 e 34 com Ctenocephalides felis felis (100 pulgas) e nos dias -1, 6, 13, 20, 27 e 34 com R. sanguineus (30 carrapatos). Nos dois experimentos a nova formulação foi comparada à associação D-fenotrina 78,125%+ piriproxifen 2,575%. A eficácia sarnicida da nova formulação experimental, comparativamente à selamectina 12%, foi avaliada em 15 cães naturalmente infestados por S. scabiei var. canis. Raspados cutâneos e avaliações clínicas (regressão das lesões de pele) foram realizados, em todos os cães experimentais. Para avaliação anti-helmíntica, 24 cães foram selecionados por meio de exames coprológicos e distribuídos em três grupos de oito animais cada. As contagens de ovos de nematódeos por grama de fezes (OPG) foram realizadas nos dias -3, -2, -1 (seleção dos animais) e 1, 3, 7, 10 e 14 pós-tratamento. A nova formulação experimental foi comparada à Selamectina 12%.Todos os dados foram avaliados estatisticamente, e as inferências devidamente extraídas... / Utilizing of the appropriate experimental design, was evaluated the anti-parasite activity of a new formulation, composed of two piretroids (S-cifenotrin and D-tetrametrin), piperonil butox and ivermectinn. Four experiments had been lead for therapeutical evaluation of the new association, comparativily to the formulation acquired in the market. For the evaluation against ticks and flea, two experiments had been lead. In the first assay, 21 dogs naturally infested by Rhipicephalus sanguineus, selected by means of two consecutive countings, were shuffted and randomly put into three groups of seven animals each. In the second assay, 21 selected dogs were allocated randomly, also, in three groups of seven animals each. Artificial infestations had been carried through in days -4, -2, 6, 13, 20, 27 and 34 with Ctenocephalides felis felis (100 fleas) and in days -1, 6, 13, 20, 27 and 34 with R. sanguineus (30 ticks). In the two experiments the new formulation was compared with the association D-fenotrin 78.125%+ piriproxifen 2.575%. The scabies effectiveness of the new experimental formulation, comparativily to selamectin 12%, was evaluated in 15 dogs naturally infested by S. scabiei var. canis. Cutaneous scrapings and clinical evaluations (regression of the skin injuries) had been carried through, in all the experimental dogs. For antihelmintic evaluation, 24 dogs had been selected by means of the coprology examinations and distributed in three groups of eight animals each. The egg countings of nematodes for gram of fezes (EPG) had been carried through in days -3, -2, -1 (election of the animals) and 1, 3, 7, 10 and 14 post-treatment. Infecção experimental de Amblymma cajennense, Amblyomma ovale e Rhipicephalus sanguineus (Acari> Ixodidae) com Hepatozoon canis (Apicomplexa: Hepatozoidae)

Rhipicephalus sanguineus é considerado seu vetor biológico, entretanto, é possível que existam outros vetores. No Brasil, H. canis é descrito principalmente em áreas rurais, onde cães podem ser infestados por outras espécies, incluindo Amblyomma spp. O objetivo do presente trabalho foi infectar experimentalmente diferentes espécies de carrapatos com H. canis, a partir de cães naturalmente infectados. Para isso, foram utilizadas ninfas de A. cajennense e R. sanguineus e adultos de A. cajennense e A. ovale. Após o ingurgitamento dos espécimes em animal infectado, foi realizada pesquisa de formas evolutivas do protozoário na hemolinfa, intestino e hemocele dos ixodídeos, por técnicas citológicas e histológicas. Alguns carrapatos foram submetidos ao diagnóstico molecular pela técnica de PCR. Somente em duas fêmeas de A. cajennense foram observadas estruturas morfologicamente semelhantes a Hepatozoon spp. e uma fêmea de A. ovale apresentou oocistos na hemocele. Além disso, duas fêmeas de A. ovale e uma de A. cajennense foram positivas na PCR. Entretanto, não é possível caracterizar essas espécies como vetores de H. canis, pois foram usados muitos exemplares nas infecções experimentais e poucos foram capazes de se infectar com o parasita. É provável que R. sanguineus tenha pouca importância epidemiológica na transmissão de H. canis no Brasil, pois este estudo, assim como pesquisas anteriores, demonstraram que a espécie foi incapaz de se infectar experimentalmente com o protozoário / Hepatozoon canis is a hemoparasite that afflicts dogs in several regions of the world. Rhipicephalus sanguineus is considered to be the biologic vector of this agent, although other species may act as possible vectors. In Brazil, H. canis is described mainly in rural areas, where dogs are infested by other species, such as Amblyomma spp. The aim of this study was to transmit experimentally H. canis to different species of ticks, using naturally infected dogs. For this purpose A. cajennense and R. sanguineus nymphs and A. cajennense and A. ovale adults were used. After the repletion of specimens fed on a naturally infected animal, we searched for development forms of the protozoa in hemolymph, gut and hemocoel of the ticks by cytological and histopathologic examinations. Some ticks were also examined by molecular detection (PCR). Only two female ticks of the species A. cajennense contained structures morphologically similar to Hepatozoon spp. and oocysts were recovered from the hemocoel of an A. ovale female. In addition, two females of A. ovale and one female of A. ovale were positive through PCR. However, our results are not enough to characterize these species as vectors of H. canis mainly because many ticks were used in experimental infections and few were infected with the parasite. It is likely that R. sanguineus has little epidemiological importance in the transmission of H. canis in Brazil, since this research and previous researchers demonstrated that R. sanguineus was unable to experimentally get infected with the protozoan

Ação da luz ultravioleta e da riboflavina na inativação da Leishmania infantum chagasi em sangue canino conservado em bolsas para transfusão

O presente trabalho visa caracterizar as possíveis alterações hematológicas e bioquímicas, ocorridas durante o período de 21 dias de armazenamento das amostras de sangue canino colhidas em bolsas plásticas para transfusão, investigando ainda a eficiência do tratamento destas bolsas com luz ultravioleta e riboflavina na inativação de Leishmania infantum chagasi. Para isto, realizou-se hemograma, teste de fragilidade osmótica, determinação da concentração sérica de sódio e potássio, testes sorológicos e moleculares, comparando-se um grupo de bolsas de sangue colhidas de cães parasitados com L. infantum chagasi, com um grupo de bolsas de sangue obtidas de cães hígidos. Após as análises laboratoriais in vitro, o sangue canino parasitado foi submetido ao tratamento com riboflavina e luz UV por 30 e 45 minutos e inoculado em hamsters. O sangue de cães com leishmaniose armazenado em bolsas para transfusão demonstrou: anemia, hiperproteinemia, trombocitopenia, hipercalemia, diminuição do pH, 100% das RIFIs positivas (título 1:640) e PCR convencional positiva em 20% dos animais, demonstrando a importância do hemograma, exames bioquímicos e dos testes laboratoriais de diagnóstico da leishmaniose na seleção dos doadores. O sangue parasitado não perdeu a capacidade de produzir a infecção após o período de armazenamento. Os hamsters inoculados com sangue tratado com riboflavina e luz UV por 30 e 45 minutos apresentaram PCR positiva, apesar de não apresentarem sinais e sintomas clínicos da enfermidade. Na qPCR pode-se identificar que a associação da riboflavina com a luz UV reduziu o número de leishmanias, diminuindo a carga parasitária, porém não eliminou completamente os parasitas / Providing safety during the transfusion of blood products is of major importance in transfusion medicine; this requires a proper blood to minimize the occurrence of pathogen transmission. The present work aims to characterize the possible hematological and biochemical changes that occurr during the 21 days of storage of canine blood samples collected in plastic bags for transfusion and to investigate the treatment efficiency of these bags with ultraviolet light and riboflavin in inactivating Leishmania infantum chagasi. Total blood count, osmotic fragility test, determination of serum sodium and potassium, serological and molecular tests were performed comparing a group of bags of blood taken from dogs infected with L. infantum chagasi, with a group of bags of blood obtained from healthy dogs. After this, the parasitized canine blood was subjected to the treatment with riboflavin and UV light for 30 to 45 minutes and inoculated in hamsters. The blood of dogs with leishmaniasis stored in bags for transfusion showed anemia, hyperproteinemia, thrombocytopenia, hyperkalemia, decreased pH, 100 % of RIFIs positive (1:640 title) and conventional PCR positive in 20% of animals, demonstrating the importance of blood count, biochemical tests and laboratory tests for the diagnosis of leishmaniasis in blood donor selection. The parasitized blood has not lost the ability to produce infection after the storage period. Hamsters inoculated with blood treated with riboflavin and UV light for 30 and 45 minutes were PCR positive, although not presenting clinical signs and symptoms of the disease. The qPCR can identify that the combination of riboflavin with UV light reduced the number of Leishmania, reducing the parasitic load, but did not completely eliminate parasites
