Development of a Double Antibiotic Electrospun scaffold for Root Canal Disinfection

Indiana University-Purdue University Indianapolis (IUPUI) / Objective: This study synthesized electrospun polymer-based scaffolds containing ciprofloxacin (CIP) and doxycycline (DOX), as a scaffold mimic of Double Antibiotic Paste (DAP) and determined, in vitro, its mechanical properties, chemical composition, and antimicrobial effectiveness against multiple endodontic bacterium. Methods: Polydioxanone sutures (PDS) were dissolved in 1,1,1,3,3,3-hexafluoro-2-propanol (HFP), mixed with CIP/DOX (i.e., 50%), and electrospun under optimized conditions into fibrous scaffolds. Tensile testing was used to evaluate the mechanical properties. Antimicrobial efficacy was determined over time using aliquots collected at 1, 4, 7, 14 day and agar diffusion assays. Two-Way Anova. Significance P < 0.05. Results: Tensile strength (MPa) of the CIP/DOX scaffold did not show significant difference from the control (pure PDS). Elongation at break (%) did show a significant difference between CIP/DOX scaffolds and the control group. Young’s modulus of elasticity (MPa) showed a significant difference between CIP/DOX scaffolds and the control. CIP-containing scaffolds did not inhibit Gram-negative (F. nucleatum and P. gingivalis) bacteria as effectively when compared to Gram-positive bacteria (E. faecalis and S. gordina). DOX-containing scaffolds showed less inhibition against Gram-positive (E. faecalis and S. gordina) bacteria then Gram-negative bacteria (F. nucleatum and P. gingivalis). In combination, CIP/DOX scaffolds showed significant inhibition against G(-) and G(+) bacteria. Conclusion: Electrospun double antibiotic scaffold demonstrated increased antimicrobial efficacy proving the potential for future clinical use to disinfect the RCS in permanent immature necrotic teeth to aid in regenerative treatment and or in persistent infections.

Topical antibiotic treatment of infected dental pulps of monkeys

Indiana University-Purdue University Indianapolis (IUPUI) / A modified double-blind method of investigation was used in which the pulps of 52 monkey teeth were surgically exposed and left open to the oral environment for a period of 24 hours. One-half of the exposed pulps were treated with an antibiotic preparation and one-half with a pure starch control. The antibiotic compound consisted of erythromycin estolate 10 percent, streptomycin sulfate 10 percent, and starch q. s. as the vehicle. The teeth were extracted at 30 and 90 day intervals after treatment and histologically evaluated. Inflammation of a varying degree was observed in all of the teeth treated with either the antibiotic preparation or the starch control. However, those teeth treated with the antibiotic capping material exhibited much less inflammation than did the great majority of teeth treated with the starch control, in which abscess formation and necrosis were frequently observed. The pulps of those teeth treated with the antibiotic capping material demonstrated a decidedly more favorable reaction than did those pulps treated with the starch capping material. Calcific repair at the exposure site was not observed to be complete in any instance. The histologic findings for the antibiotic treated teeth were encouraging and warrant additional investigations of longer duration.

Comparison of giant vacuoles found in the inner wall of Schlemm’s canal in human eyes between high and physiologic pressure

This study investigated the morphologic differences of the giant vacuoles (GVs) in the inner wall endothelium of Schlemm’s canal (SC) in human eyes perfused at either 30 mmHg or 7 mmHg (physiologic pressure in enucleated eyes) using serial block-face scanning electron microscopy (SBF-SEM) paired with three-dimensional reconstruction software. Two normal human eyes were perfused at 30 mmHg with fluorescent tracers to mark regions of active and inactive flow, followed by perfusion-fixation. Tissue wedges (n = 6) of trabecular meshwork including SC from high-, low-, and non-flow areas of each eye (determined by tracer distribution) were dissected and processed for SBF-SEM. Four types of GVs were identified: Type I GVs which lack both a basal opening and an Ipore; Type II GVs which have a basal opening but lack an I-pore; Type III GVs which have an I-pore but lack a basal opening; and Type IV GVs which possess both a basal opening and an I-pore. Types and spans of GVs were collected from the SBF-SEM images, and volumes of GVs from a random subset were measured using 3D reconstruction. Results were compared with findings from an earlier study conducted with two eyes perfused at 7 mmHg and prepared in the same manner (Soares, 2022). In total, 19,047 SBF-SEM images were analyzed between 7 mmHg (n = 9586) and 30 mmHg (n = 9461) using Reconstruct. Statistical analysis comparing data between the two pressures was performed using R. There were more GVs found at 30 mmHg (n = 1541) when compared with 7 mmHg (n = 1312), and there were more Type IV GVs at 30 mmHg when compared with 7 mmHg. Type IV GVs occurred most frequently in high-flow areas at both pressures. GVs with I-pores were greater in size (both span and volume) than GVs without I-pores in all flow areas at both pressures. Type IV GVs were larger than Type II GVs which were larger than Type I GVs at both pressures. The span of GVs without I-pores was significantly greater at 7 mmHg. However, there was no significant difference between the volumes of GVs with or without I-pores between the two pressures. The result that GVs with I-pores were larger in size than GVs without I-pores at all conditions appears to support the theory that GV size is an important contributing factor to I-pore formation. The differences in span but not volume of GVs without I-pores between two pressures suggest that GVs at high pressure may be more convex in shape and may protrude further into SC, a situation which could contribute to thinning of the cellular membrane of GVs. Finally, the result that more Type IV GVs were found in high-flow areas at both pressures implies that the changing percentage of Type IV GVs likely plays a role in regulating segmental flow. / 2025-02-26T00:00:00Z

The Historic Canal System in Bangkok, Thailand: Guidelines for Reestablishing Public Space Functions

This thesis proposes guidelines for reestablishing the historic canal system on Rattanakosin Island, Bangkok as a public space system and a connector of key public spaces. The study examines the historic value and cultural symbolism of the canals through evolutionary morphological analysis, establishing that the canals are primary structural elements in the city, since they have retained the integrity of their physical form over time, and have come to hold cultural meaning for the Thai people. The canals have also accommodated different functions over time, in response to a changing urban context. There is potential for them to accept new functions as recreational spaces, connectors of key public spaces, and as tourist destinations. Typological analysis of structural characteristics of the canals yields seven canal types that have potential to accommodate public space functions. The study proposes guidelines for the seven canal types that will enhance these potentials and ensure the preservation of the canals' physical form. / Master of Landscape Architecture

O objetivo deste estudo foi avaliar o pH e a liberação de íons cálcio dos cimentos AH Plus, MTA Fillapex e Sealapex, em diferentes períodos, quando empregados em obturações de canais radiculares, bem como, a qualidade das obturações, após 1 ano de realizadas. O pH e a liberação de íons cálcio foram avaliados por meio de um peagômetro e espectrofotômetro de absorção atômica, respectivamente. A qualidade das obturações foi avaliada por meio de um estereomicroscópio. Cento e vinte e quatro pré-molares inferiores humanos extraídos, unirradiculados, foram divididos em 01 grupo controle e 02 grupos experimentais, de acordo com a padronização do diâmetro do forame apical. Em seguida, os 02 grupos experimentais foram divididos em 4 subgrupos, considerando as técnicas de instrumentação e obturação empregadas. Após o preparo dos canais radiculares cada subgrupo teve o número de dentes divididos por dois, formando novos subgrupos, de acordo com os cimentos obturadores utilizados. Imediatamente após a obturação, a raiz de cada dente foi imersa em 15 mL de água ultrapura para avaliação do pH e liberação de íons cálcio nos seguintes períodos: 1 h, 1, 3, 7, 30, 180 e 365 dias, após a obturação. Após o período de 1 ano, os dentes foram seccionados transversalmente a 2, 4, 6 mm do forame apical para análise da qualidade das obturações, analisando-se a porcentagem de cimento, guta-percha e espaços vazios na massa obturadora. Os cimentos apresentaram pH neutro, porém, aos 180 dias, os valores mostraram um ligeiro aumento quando utilizados em canais com forames de diâmetro 0,25 mm. Quando utilizados em canais com forames de diâmetro 0,45 mm, o pH mostrou-se com valores maiores para todos os períodos sem distinção do cimento. Todos os cimentos liberaram íons cálcio, com menores valores para o AH Plus. Na estereomicroscopia, observaram-se variações nas porcentagens de cimento e guta-percha, considerando-se os diâmetros foraminais e técnicas obturadoras, e porcentagem insignificante de espaços vazios. / The objective of this study was to evaluate the pH and calcium ion release of AH Plus, MTA Fillapex and Sealapex cements at different time periods when used in root canal obturations, as well as the quality of the obturations one year after being performed. The pH and calcium ion release were evaluated by means of a pH meter and an atomic absorption spectrophotometer, respectively. The quality of the obturations was evaluated using a stereomicroscope. One hundred and twenty-four extracted human uniradicular mandibular premolars were divided into 1 control and 2 experimental groups, according to the standardization of the apical foramen diameter. Next, the 2 experimental groups were divided into 4 subgroups, considering the instrumentation techniques and obturations employed. After the preparation of the root canals, each subgroup had the number of teeth divided by two, forming new subgroups, according to the sealers used. Immediately after the obturation, the root of each tooth was immersed in 15 mL of ultrapure water for the evaluation of the pH and calcium ion release in the following time periods: 1 hr, 1, 3, 7, 30, 180 and 365 days after the obturation. After the period of one year, the teeth were transversely sectioned at 2, 4 and 6 mm from the root apex to examine the quality of the obturations, analyzing the percentage of cement, gutta-percha and voids in the obturator mass. The cements presented a neutral pH, which showed slightly increase as of 180 days when used in canals with foramens of 0.25 mm diameters. When used in canals with foramens with diameters of 0.45 mm, the pH presented higher values in all periods for all cements. All cements released calcium ions, with lower values for the AH Plus. In the stereomicroscopy, there were variations in the percentages of cement and gutta-percha, considering the foraminal diameters and obturation techniques and an insignificant percentage of voids.

"Influência do grau de proficiência técnica dos operadores no preparo de canais curvos, quando do emprego de instrumentos rotatórios de níquel-titânio" / Influence of the operator’s proficiency in the preparation of curved canals, using of rotary nickel-titanium instruments

O presente estudo avaliou a interferência de operadores com diferentes proficiências no preparo de canais radiculares curvos, simulados em blocos de resina transparente. Os grupos, com 10 indivíduos cada um, foram: Grupo 1 – professores de Endodontia; Grupo 2 - alunos de especialização; Grupo 3 – alunos de odontologia Monitores de Endodontia e o Grupo 4 – alunos de graduação no início do curso de Endodontia. Cada operador instrumentou dois canais com 20 graus de curvatura, o primeiro canal era visualizado para treinamento e o segundo não. O sistema mecanizado com instrumentos rotatórios de níquel-titânio empregado foi o RaCe®. Os parâmetros de avaliação foram: tempo de instrumentação, quantidade de material removido, desvio apical e deformação dos instrumentos. Os resultados encontrados (ANOVA e Teste de Tuckey a= 0,05%), foram: Para o tempo de instrumentação, no primeiro canal, houve diferença estatística entre o grupo 1 e os grupos 3 e 4, o grupo 2 diferiu somente do grupo 4. Já, no segundo canal, não houve diferença estatística entre os grupos. Para a quantidade de material removido do primeiro canal houve diferença estatística entre os graduados (grupos 1 e 2) em relação aos graduandos (grupos 3 e 4), porém, no segundo canal, houve diferença somente entre os grupos 1 e 2. O desvio apical não apresentou diferença estatística entre os grupos em nenhuma das condições experimentais. Após a aplicação do teste do Qui-quadrado (a=0,01%), observou-se que o grupo 1 foi o que produziu as menores deformações nos instrumentos. Conclui-se que: Os desvios apicais promovidos foram semelhantes entre os grupos experimentais. Houve diminuição no tempo de instrumentação do segundo canal em relação ao primeiro, com diferença estatística nos grupos dos monitores e acadêmicos e que não houve diferença estatística entre os grupos, em relação ao tempo de instrumentação do segundo canal. Os grupos dos graduados (professores e alunos de especialização) excisaram maior quantidade de material do primeiro canal, quando comparados aos dos graduandos (monitores e acadêmicos), não havendo diferença estatística entre os grupos em relação à quantidade de material removido do segundo canal, excetuando a comparação entre os professores e os alunos de especialização, onde os primeiros desgastaram significativamente mais que os últimos. Comparados aos demais grupos, os professores produziram menores deformações aos instrumentos. / This study evaluated the interference of operators with different proficiencies in the preparation of curved canals , simulated transparent resin blocks. The groups, with 10 individuals each, were: Group 1 - professors of Endodontics; Group 2 – students of specialization; Group 3 – undergraduate dental students who had concluded the Endodontics course and Group 4 - undergraduate dental students in the beginning of the Endodontics course. Each operator prepared two canals with 20 degrees of bending, the first canal was visualized for training and second no. The rotary instrumentation used was the RaCe®. The parameters of evaluation were: time of instrumentation, amount of removed material, apical transportation and deformation of the instruments. The results (ANOVA and Test of Tuckey = 0.05%): In relation to the instrumentation time, when of the execution of the first canal, it had statistics difference between group 1 and groups 3 and 4, group 2 only differed from group 4. When of the execution of the second canal it did not have difference statistics between the groups. For the amount of material removed of the first canal it had difference statistics between the graduated (groups 1 and 2) relation to the undegraduated (groups 3 and 4). After the second canal instrumentation, only had difference between groups 1 and 2. The apical transportation did not present statistics difference enters the groups in no ne of the experimental conditions. After the application ?2 test (a=0,01%), was observed that group 1 produced lesser deformations in the instruments. Conclusions: The apical transportation was similar between the different groups. It had reduction in the time of instrumentation of the second canal in relation to the first one, with statistics difference in the groups of the monitors and academics. It did not have difference statistics between the groups, in relation to the time of instrumentation of the second canal. The groups of the graduated ones (professors and students of specialization) removed bigger amount of material of the first canal, when compared with the ones of the undergraduate (monitorial and academic). It did not have statistics difference enters the groups in relation to the amount of material removed of the second canal, excepting the comparison between the professors and the students of specialization, where the first ones had consumed significantly more than the last ones. Compared with all groups, the professors had produced minors deformations to the instruments.

Influência do pré-alargamento cervical no desgaste promovido pelos instrumentos rotatórios de níquel-titânio na região apical: Estudo em canais simulados curvos / Influence of cervical preflaring on prepared canal shape in simulated curved root canals prepared with nickel-titanium rotary systems

A proposta desse estudo in vitro foi avaliar a influência do pré-alargamento cervical no desgaste apical de canais simulados curvos preparados com instrumentos rotatórios de níquel-titânio. Foram confeccionados 30 blocos de resina com canais simulados de 20 graus de curvatura que foram divididos aleatoriamente em três grupos distintos (n=10) de acordo com o tipo de alargamento cervical realizado: Grupo I - sem alargamento cervical; Grupo II - alargamento cervical realizado com brocas CP Drill®; Grupo III - alargamento cervical realizado com brocas LA Axxess®. Previamente ao alargamento cervical os canais foram preenchidos com tinta nanquim em toda sua extensão e realizaram-se as fotografias iniciais com auxílio de máquina digital posicionada em estativa. Para o preparo apical utilizaram-se instrumentos rotatórios K3® na seqüência de 20.02 até 45.02, sendo que, entre o intervalo do uso de cada instrumento o canal foi preenchido, novamente, com tinta nanquim e realizada a fotografia final para cada instrumento. As fotografias pré e pós-operatórias foram sobrepostas e procedeu-se a mensuração do desgaste linear da parede interna e externa do canal simulado no ápice da curvatura e no ápice do canal proporcionado por cada instrumento, com auxílio do software Image Tool. A diferença entre esses valores, de acordo com cada área do canal avaliada, foi submetida à análise estatística. O teste de Regressão Linear permitiu estabelecer uma correlação entre o calibre do instrumento e o desgaste do canal, que pode ser traduzida em uma equação matemática, indicando que é diretamente proporcional. O teste de Kruskal-Wallis (complementado com pós-teste de Dunn) permitiu a comparação do desgaste entre os diferentes grupos experimentais. Por meio desses testes pode-se observar que grupo onde não foi realizado o pré-alargamento cervical apresentou padrões de desgastes diferentes daqueles onde o preparo foi executado. O grupo sem pré-alargamento provocou o desgaste acentuado do lado externo da curvatura, enquanto os grupos preparados com CP Drill® e LA Axxess® mostraram padrões de desgaste semelhantes, denotando a maior centralização do instrumento no canal em ambas as regiões analisadas (ápice da curva e ápice do canal). Pode-se concluir que o pré-alargamento cervical influenciou de forma positiva no preparo da região apical de canais simulados curvos, proporcionando preparos mais centralizados dessa região. / The purpose of this in vitro study was to evaluate the influence of cervical preflaring on prepared canal shape in simulated curved root canals prepared with nickel-titanium rotary systems. Thirty resin blocks with simulated canals with 20º curvature were fabricated and randomly assigned to three groups (n=10) according to the type of cervical preflaring: Group I - no cervical preflaring; Group II - cervical preflaring with CP Drill® burs; Group III - cervical preflaring with LA Axxess® burs. Previously cervical preflaring, the canals were thoroughly filled with India ink and the initial photographs were taken using a digital camera fixed in a static position. Right after, apical preparation was performed using 20.02 to 45.02 K3® rotary instruments. At each change of file, the canals were refilled with India ink and the final photographs were taken for each instrument. The initial and final photographs were superimposed and linear dentin removal produced by each type of rotary instrument was measured with specific software at the internal and external portions of the curvature apex and at the canal apex. The difference between the initial and final values was analyzed statistically, according to each canal region. Statistical analysis by linear regression established a correlation between instrument size and canal transportation, which can be expressed by a mathematical equation. This correlation is directly proportional, which means that the greater instrument, the more accentuated the canal transportation. Additionally, Kruskal- Wallis test (complemented by the Dunn\' post-test) allowed comparing dentin removal among the different experimental groups. These tests revealed that the non-flared group presented different dentin removal patterns, as compared to the groups where cervical preflaring was undertaken. The group without preflaring presented a significantly greater dentin removal on the external side of curvature, while the groups preflared with CP Drill® and LA Axxess® exhibited similar dentin removal patterns, demonstrating a greater centralization of the instrument inside the canal, for both analyzed regions (curvature apex and canal apex). It may be concluded that cervical preflaring influenced positively the apical preparation of simulated curved canals, producing more centralized preparations in this region.

Remoção mecânica de Enterococcus faecalis em canais preparados com Wave One Gold ou One Shape New Generation / Mechanical removal of Enterococcus faecalis in root canals instrumented using Wave One Gold or One Shape New Generation

O preparo do canal radicular visa a remoção de bactérias que podem causar a patologia periapical, e os instrumentos endodônticos estão sendo constantemente aprimorados para tornar o tratamento do canal radicular mais fácil, rápido e seguro. Assim, novos instrumentos precisam ser avaliados quanto ao seu desempenho. O objetivo deste estudo foi avaliar a remoção bacteriana dos sistemas Wave One Gold e One Shape New Generation comparando-os com seus sistemas predecessores. Cinquenta e seis canais disto vestibulares de molares superiores esterilizados por oxido de etileno foram contaminados com Enterococcus faecalis por 21 dias, e então uma amostra bacteriana inicial foi coletada e paqueada em M-enterococcus agar para contagem bacteriana em unidades formadoras de colônias. Os espécimes foram aleatoriamente divididos em quatro grupos de acordo com a instrumentação (n=12): Wave One Gold Primary, One Shape New Generation 25/.06, Wave One Primary e One Shape 25/.06, e os outros 8 canais não contaminados foram o controle de assepsia. Todos os grupos utilizaram água destilada como irrigante. Nova coleta foi feita imediatamente após a instrumentação e aos 7 dias. A redução bacteriana foi calculada em porcentagem, e então feita análise intragrupo pelo teste de Wilcoxon e entre grupos por Kruskal Wallis e teste de Dunn, todos com significância de 5%. Todos os sistemas reduziram significativamente a carga bacteriana do canal radicular tanto na coleta imediata quanto aos 7 dias (p<0,05). Houve aumento do número de bactérias 7 dias após o preparo quando comparado com a coleta imediata (p<0,05). A análise entre grupos mostrou que Wave One Gold e One Shape New Generation promoveram maior redução bacteriana que os sistemas Wave One e One Shape (p<0,05), sem diferença significativa entre Wave One Gold e One Shape New Generation ou entre Wave One e One Shape (p>0,05). Conclui-se que Wave One Gold e One Shape New Generation promoveram maior remoção bacteriana do que seus sistemas predecessores. / The root canal preparation aims to remove the bacteria that can cause periapical pathologies, and endodontic instruments are constantly being improved to make root canal treatment easier, faster and safer. Thus, new instruments need to be evaluated for their performance. The aim of this study was to evaluate the bacterial removal promoted by Wave One Gold and One Shape New Generation systems in comparison to that of their predecessor systems. Forty-six distobuccal root canals of upper molars sterilized with ethylene oxide were infected with Enterococcus faecalis for 21 days, and then root canal initial bacterial sampling was collected and plated on M-enterococcus agar to bacterial count in colony forming unities. The specimens were randomly divided into four groups according to the instrumentation (n=12): Wave One Gold Primary, One Shape New Generation 25/.06, Wave One Primary and One Shape 25/.06, and the other 8 uncontaminated canals were used as asepsis control. All groups used distilled water as irrigant. New sampling was obtained immediately after instrumentation and at 7 days. The bacterial reduction was calculated in percentage, after that intra-group analysis was carried out by Wilcoxon test, and inter-group analysis by Kruskal-Wallis complemented by Dunn\'s test, all at 5% significance. All the systems significantly reduced the bacterial amount in the root canal in both immediate and at 7 days sampling (p<0.05). The bacterial amount increased at 7 days after preparation comparing to immediate sampling (p<0.05). The analysis between groups showed that Wave One Gold and One Shape New Generation promoted greater bacterial reduction than Wave One and One Shape systems (p<0.05). No significant difference was found between Wave One Gold and One Shape New Generation or between Wave One and One Shape (p>0.05). It can be concluded that Wave One Gold and One Shape New Generation promote greater bacterial removal than their predecessor systems.

Avaliou-se presença de espaços vazios, presença de fendas na interface cimento/dentina, o perímetro de penetração do cimento obturador nos canais e nos istmos.e a ação antimicrobiana intra-dentinária do cimento endodôntico (AH Plus) quando submetido ou não a agitação ultrassônica no momento da obturação. Para a avaliação do preenchimento de istmos foram utilizadas 30 raízes mesiais de primeiros molares inferiores, as quais foram divididas em 2 grupos (n=15). A instrumentação foi realizada com o sistema reciprocante Reciproc (R25) e finalizado com instrumento rotatório Mtwo (35.04). Em seguida, o cimento AH Plus foi espatulado com Rodamina B e foi inserido nos canais com espiral de Lentulo no35; o cimento do grupo 1 foi submetido a agitação ultrassônica enquanto o do grupo 2 não foi agitado, sendo que todos os canais foram obturados por meio da técnica do cone único e armazenados em estufa a 37oC durante 7 dias. As raízes foram seccionadas transversalmente a 2, 4 e 6 mm do ápice radicular para análise em esteromicroscópio e microscopia confocal a laser. Para a avaliação da ação antimicrobiana foram utilizados 30 incisivos de bovinos, os quais foram seccionados em cilindros de 6 mm de espessura. Os canais tiveram seu diâmetro padronizado com uma lima tipo K no 80 e então contaminados com Enterococcus faecalis (ATCC 29212) por 4 dias. Após esse período os espécimes foram divididas em 3 grupos (n=30). No grupo 1 o cimento foi agitado com o ultrassom e obturados com a técnica de cone único; no grupo 2 o cimento não foi agitado e o grupo 3 foi o grupo controle. As raízes foram seccionadas longitudinalmente no sentido vestíbulopalatino e coradas com corante LIVE/DEAD para avaliar a viabilidade bacteriana em microscopia confocal a laser. Para análise da agitação ultrassônica do cimento obturador no preenchimento de canais e istmos, foi utilizado o teste não paramétrico de Mann-Whitney (p <0.05) e para a análise da ação antimicrobiana intradentinária, foi utilizado o teste não paramétrico Kruskal-Waliis e o Teste Dunn (p <0.05). Os resultados indicaram que a agitação ultrassônica do cimento AH Plus favoreceu o preenchimento de istmos e a penetração intratubular. A agitação ultrassônica do cimento AH Plus reduziu significantemente a viabilidade bacteriana. Conclui-se que a agitação ultrassônica do cimento AH Plus favoreceu maior penetração intratubular, menor presença de fendas, menos espaços vazios e maior ação antimicrobiana. / In this study it was assessed the presence voids, gaps in the sealer/dentin interface, the intratubular sealer penetration in canals and isthmuses and intratubular antimicrobial effect of the AH Plus sealer when ultrasonic activation was performed during obturation. 30 mesial roots of mandibular first molars were selected and divided into 2 groups (n=15). Root canal instrumentation was performed with the reciprocating system Reciproc (R25) and finished with the Mtwo (35.04) rotary instrument. Then the AH Plus sealer was mixed with Rhodamine B and was inserted into the root canals with a Lentulo spiral #35. In group 1 the ultrasonic activation of the sealer was performed while in group 2 was not activated. Then, the root canal filling was performed using the single cone technique. After 7 days of storage the roots were sectioned at 2, 4 and 6 mm from the apex and analyzed in the stereomicroscope and by confocal laser scanner microscopy. To the antimicrobial test 30 bovine incisors cut into cylinders with 6 mm in thickness were used. The root canals diameter was standardized with a K file # 80 and then contaminated with Enterococcus faecalis (ATCC 29212) for 4 days. After this period the samples were divided into 3 groups (n = 30). In group 1 the ultrasonic activation of the sealer was perfomed and filled with single cone technique; in group 2 the sealer was not ultrasonic activated and in group 3 the canals were not obturated (control group). The roots were sectioned in the buccal-palatal direction and stained with LIVE / DEAD stain to assess the bacterial viability by confocal laser microscopy. For analysis of ultrasonic activation of the sealer on the filling quality of the canals and isthmuses, a non-parametric Mann-Whitney test (p <0.05) was used. For the analysis of the intradentin antimicrobial action, a Kruskal-Waliis nonparametric and Dunn\'s test was used (p <0.05). The results showed that the ultrasonic agitation of the AH Plus sealer favored the isthmus filling and the intratubular penetration. Also, the ultrasonic activation of AH Plus sealer reduced bacterial viability. It was concluded that the ultrasonic agitation of the sealer favored higher intratubular penetration, lower presence of gaps and voids and higher antimicrobial activity.
