Les ventes des fromages AOP subissent un affaiblissement. L’utilisation de ces fromages sous forme d’ingrédient représente une alternative intéressante à la dégustation et permettrait de maintenir une activité durable aux producteurs de ces fromages. L’objectif de ce travail est d’explorer les potentialités de 4 catégories de fromages à pâte persillée AOP du Massif-Central à être utilisés comme ingrédient à chaud pour répondre à des propriétés telles que « fondant, nappant, filant, gratinant». Les fonctionnalités recherchées pour ce type d’usage ont été prospectées à l’aide de méthodes physico-chimiques et instrumentales classiques mais également à l’aide de méthodes spectrales et sensorielles afin d’enrichir les observations de la macro et microstructure fromagère et de la perception du consommateur. Dans un premier temps, les caractéristiques physicochimiques, rhéologiques et sensorielles à froid et certaines propriétés à chaud ont été décrites pour les 4 catégories de fromages. Ces premiers résultats ont permis de cataloguer les fromages selon des caractéristiques essentiellement liées à l’organisation matricielle des composants tels que les protéines, les matières grasses, les minéraux. L’analyse sensorielle a permis de décrire précisément certaines propriétés à froid qui ne sont pas mises en évidence par d’autres analyses. Les pâtes persillées ont une grande hétérogénéité de pâte due à la présence de« veines » de moisissures. La seconde partie est une étude sur la capacité de la SFS en tant que méthode d’étude de la micro structure, à prédire la diversité de composition de ces fromages. Malgré la forte hétérogénéité des fromages, la SFS permet une « empreinte » identifiable des fromages et prédit certains mais pas la totalité des paramètres physicochimiques. L’évolution de la macro et microstructure de ces fromages au cours du chauffage et du refroidissement a ensuite été étudiée par SFS et par test de compression dynamique. Ces méthodes décrivent bien la fonte de la matière grasse et de la matrice fromagère. Elles sont corrélées entre elles et démontrent une relation entre la structure moléculaire et les propriétés rhéologiques de ces fromages. Une étude comparative des propriétés sensorielles des fromages a été menée à froid et après chauffage. Les attributs décrivent des caractéristiques de texture différentes entre les 2 conditions. Cette démarche a permis de bien différencier les 4 catégories de fromages et de mettre en évidence certains attributs de qualité ou de défaut pour une utilisation à chaud. En conclusion, ces 4 catégories de fromages à pâte persillée ont des potentialités à être utilisées comme ingrédient pour usage à chaud. L’analyse sensorielle a caractérisé les perceptions des consommateurs vis à vis de ces fromages chauffés et ainsi aider à préciser leurs fonctionnalités. Les analyses rhéologiques et spectrales ont apporté des explications liées à ce traitement au niveau de la macro et microstructure de la matrice fromagère. / Sales of PDO cheeses undergo a decline. The use of these cheeses in the form of ingredient represents an interesting alternative to the tasting and would allow to maintain a sustainable business activity to the producers of these cheeses. The objective of this work is to investigate the potentialities of four categories of Blue cheese PDO of the Massif Central as an ingredient to meet their properties during heating such as "melting, stretchability, browning". The functional properties desired for this type of cheese have been investigated by physico-chemical methods and instrumental classics and also by spectral method and sensory evaluations to enrich the observations of the macro and microstructure of the cheese and the consumer perception. First, the physicochemical, rheological, sensory characteristics and certain properties during heating have been reported for four categories of Blue cheese. These initial results allowed us to characterize the cheese studied according to their characteristics essentially which were related to the organization matrix constitutes such as proteins, fats, minerals. The sensory analysis allowed to describe exactly certain properties which are not revealed by theother analyses. Blue-veined cheeses have a great heterogeneity due to the presence of "veins" of mold. The second part is a study on the ability of the SFS method to study the micro structure and to predict the diversity of composition of these cheeses. Despite the high heterogeneity of cheeses, SFS allows an identifiable 'fingerprint' of cheese and predicts some but not the totality of the physicochemical parameters. The evolution of the macro-and microstructure of these cheeses during heating and cooling was then studied by SFS and dynamic shear test. These methods describe well the melting temperature of the fat and the cheese matrix. They are correlated between them and demonstrate a relationship between the molecular structure and the rheological properties of these cheeses. A comparative study of the sensory textural properties of cheeses was carried out in the non- and heated forms. The attributes describe different characteristics of texture between the two conditions. This approach allowed to differentiate well the four categories of cheeses and to highlight on certain attributes of quality or defects to be used in cooking application. In conclusion, these four categories of Blue-veined cheeses have the potential to be used as an ingredient in cooking application. The sensory analysis characterized the perceptions of the consumers in both non- and heated forms and thus helps to specify their features. The rheological and spectral analyses have provided explanations related to this treatment at the level of the macro and microstructure of the cheese matrix.

This diploma thesis researches coffee authenticity problematice, mainly focusing on the authenticity of geographic origin. In the theoretical part of this work, botanical classification is described as well as production technology and processes. The work also includes chemical composition of coffee, describing the major components and changes during production phases. It describes major production areas of the world, in terms of general description and brief history. Problematics with coffee fraud and its identification are also described. Theoretical part also includes general geological description of 17 studied coffee growing regions. Experimental part is devoted to trace amount analysis of selected elements and volatile compounds. The element analysis was conducted using mass spectrometry or optical emission spectrometry, volatile compounds were determined using gas chromatography combined with mass spectrometry detection. Results were statistically described and analyzed, resulting in several discrimination models based on geographic origin.

<p>Steroidna jedinjenja spadaju u grupu supstanci sa širokim spektrom biološkog delovanja i predstavljaju dobru polaznu osnovu za sintezu mnogih derivata sa željenim biološkim potencijalom. Organskim sintezama se došlo do velikog broja steroidnih derivata, od kojih su neki pokazali značajnu biološku aktivnost, kao što je citotoksičnost prema različitim ćelijskim linijama kancera. Karakterizacija novosintetisanih jedinjenja može se izvesti eksperimentalnim i računarskim (in silico) metodama. U ovoj doktorskoj disertaciji predstavljeno je eksperimentalno određivanje lipofilnosti 17α-pikolil i 17(E)-pikoliniliden androstanskih derivata primenom visokopritisne tečne hromatografije na obrnutim fazama, a potom hemometrijska analiza hromatografskog ponašanja (hromatografske lipofilnosti) QSRR pristupom. Hemijska struktura analiziranih derivata opisana je numerički, pomoću izračunatih molekulskih deskriptora. U drugom delu doktorske disertacija predstavljena je QSAR analiza citotoksične aktivnosti 17α-pikolil i 17(E)-pikoliniliden androstanskih derivata prema ćelijama androgen-receptor negativnog kancera prostate (AR-neg. PC-3). Odabir najkvalitetnijih QSRR i QSAR modela obavljen je na osnovu izračunatih statističkih parametara, a njihovo rangiranje izvedeno je primenom metode sume razlika rangova (SRD). This doctoral thesis was conducted in the framework of a co-supervised PhD between the Department of Agriculture, Forestry and Food Sciences of the University of Turin and the Unit of Pharmacognosy, Bioanalysis and Drug Discovery of the Faculty of Pharmacy at the Université libre de Bruxelles. The present manuscript was conceived as a thesis of articles and is composed of 9 different scientific publications. The general introduction of the work was issued from a published narrative review, while the result and discussion part is composed by eight chapters based on different scientific articles issued from the PhD project. The cocoa bean shell (CBS) is the external tegument that covers the cocoa bean, and is one of the major by-products in cocoa industry. It is normally discarded or underutilized, which could result in economical and environmental issues. However, CBS represents a notable source of polyphenols and methylxanthines (theobromine and caffeine) which can give it different biofunctionalities such as antioxidant and antidiabetic properties, among others. It also contains high amounts of dietary fiber (about 50% w/w), minerals, vitamins, and proteins. CBS has low-fat content, and interesting cocoa-aroma compounds. All this could make CBS useful as a food ingredient, and source of biofunctional compounds. The first part of the experimental work of this thesis is devoted to the chemical characterization of CBS and the establishment of its polyphenolic and volatile organic compound (aroma) profiles. The utilization of such profiles, determined by both complete characterization methods and screening methods, was also proposed for authentication purposes of CBS depending on its geographical origin and variety. In a second step, the utilization of CBS as a low-cost food ingredient for functional food production was envisaged. CBS-based beverages and biscuits were proposed as model foods. The influence of the CBS addition to the model foods was evaluated from both technological and nutritional points of view. Changes on the physicochemical characteristics of the model foods were assessed as well as their content in compounds of interest and potential biofunctionalities. Moreover, these studies served also to evaluate the effect of the different food matrices on the bioaccessibility and intestinal permeability of the bioactive compounds contained in CBS. In the third and last part of this work, a different utility was given to the study of the cocoa by-product. The antimicrobial potential of CBS was assessed against different bacterial and fungal strains and a metabolomic strategy was applied in order to individualize the putative active compounds against the Streptococcus mutans proliferation. Improvements on Instrumentation to Explore the Multidimensionality of Luminescence SpectroscopyMoore, Anthony 01 January 2015 (has links)
This dissertation presents experimental and instrumentation developments that take full advantage of the multidimensional nature of line narrowing spectroscopy at liquid nitrogen (77 K) and liquid helium (4.2 K) temperatures. The inconvenience of sample freezing procedures is eliminated with the aid of cryogenic fiber optic probes. Rapid collection of multidimensional data formats such as wavelength time matrices, excitation emission matrices, time-resolved excitation emission matrices and time resolved excitation emission cubes is made possible with the combination of a pulsed tunable dye laser, a spectrograph and an intensifier-charged coupled device. These data formats provide unique opportunities for processing vibrational luminescence data with second order multivariate calibration algorithms. The use of cryogenic fiber optic probes is extended to commercial instrumentation. An attractive feature of spectrofluorimeters with excitation and emission monochromators is the possibility to record synchronous spectra. The advantages of this approach, which include narrowing of spectral bandwidth and simplification of emission spectra, were demonstrated with the direct analysis of highly toxic dibenzopyrene isomers. The same is true for the collection of steady-state fluorescence excitation-emission matrices. These approaches provide a general solution to unpredictable spectral interference, a ubiquitous problem for the analysis of organic pollutants in environmental samples of unknown composition. Since commercial spectrofluorimeters are readily available in most academic institutions, industrial settings and research institutes, the developments presented here should facilitate the widespread application of line-narrowing spectroscopic techniques to the direct determination, no chromatographic separation, of highly toxic compounds in complex environmental matrixes of unknown composition.

Chemometric Applications To A Complex Classification Problem: Forensic Fire Debris AnalysisWaddell, Erin 01 January 2013 (has links)
Fire debris analysis currently relies on visual pattern recognition of the total ion chromatograms, extracted ion profiles, and target compound chromatograms to identify the presence of an ignitable liquid. This procedure is described in the ASTM International E1618-10 standard method. For large data sets, this methodology can be time consuming and is a subjective method, the accuracy of which is dependent upon the skill and experience of the analyst. This research aimed to develop an automated classification method for large data sets and investigated the use of the total ion spectrum (TIS). The TIS is calculated by taking an average mass spectrum across the entire chromatographic range and has been shown to contain sufficient information content for the identification of ignitable liquids. The TIS of ignitable liquids and substrates were compiled into model data sets. Substrates are defined as common building materials and household furnishings that are typically found at the scene of a fire and are, therefore, present in fire debris samples. Fire debris samples were also used which were obtained from laboratory-scale and large-scale burns. An automated classification method was developed using computational software that was written in-house. Within this method, a multi-step classification scheme was used to detect ignitable liquid residues in fire debris samples and assign these to the classes defined in ASTM E1618-10. Classifications were made using linear discriminant analysis, quadratic discriminant analysis (QDA), and soft independent modeling of class analogy (SIMCA). The model data sets iv were tested by cross-validation and used to classify fire debris samples. Correct classification rates were calculated for each data set. Classifier performance metrics were also calculated for the first step of the classification scheme which included false positive rates, true positive rates, and the precision of the method. The first step, which determines a sample to be positive or negative for ignitable liquid residue, is arguably the most important in the forensic application. Overall, the highest correct classification rates were achieved using QDA for the first step of the scheme and SIMCA for the remaining steps. In the first step of the classification scheme, correct classification rates of 95.3% and 89.2% were obtained using QDA to classify the crossvalidation test set and fire debris samples, respectively. For this step, the cross-validation test set resulted in a true positive rate of 96.2%, a false positive rate of 9.3%, and a precision of 98.2%. The fire debris data set had a true positive rate of 82.9%, a false positive rate of 1.3%, and a precision of 99.0%. Correct classifications rates of 100% were achieved for both data sets in the majority of the remaining steps which used SIMCA for classification. The lowest correct classification rate, 69.2%, was obtained for the fire debris samples in one of the final steps in the classification scheme. In this research, the first statistically valid error rates for fire debris analysis have been developed through cross-validation of large data sets. The fire debris analyst can use the automated method as a tool for detecting and classifying ignitable liquid residues in fire debris samples. The error rates reduce the subjectivity associated with the current methods and provide a level of confidence in sample classification that does not currently exist in forensic fire debris analysis.

TARGETED AND NON-TARGETED METABOLITE ANALYSIS FOR DISEASE RISK ASSESSMENT: MEASURING BIOMARKERS OF SMOKE EXPOSURE AND HABITUAL DIETWellington, Nadine L January 2019 (has links)
Exposomics applies metabolomics methods and technologies to the comprehensive
analysis of all low molecular weight molecules (< 1.5 kDa) in complex biological
samples to characterize the interaction between cellular metabolism and exogenous
lifestyle exposures that determine health and quality of life. To fully access the
diverse classes of biological molecules related to an individual’s metabolic profile,
metabolomics frequently requires the use of complementary analytical platforms,
and employs targeted and untargeted molecular profiling strategies to identify
biomarkers that are clinically relevant to an individual’s health status. Chapter 2
describes a quinoline-based boronic acid biosensor for N-acetylneuraminic acid that
undergoes a striking binding enhancement under strongly acidic conditions. For the
first time, this work allows for direct analysis of acidic sugars with high selectivity
when using UV absorbance or fluorescence detection based on formation of a
highly stable boronate ester complex with metabolites containing an α-hydroxycarboxylate moiety. Chapter 3 describes a targeted analysis of 24 different
organic contaminants using GC-MS that can serve as biomarkers of recent smoke
exposure following search-and-rescue training exercises by firefighters located at
three different sites across the province of Ontario. Importantly, skin and possible
respiratory uptake of various polycyclic aromatic hydrocarbons, methoxyphenols,
and resin acids was confirmed by peak excretion of several wood smoke biomarkers
in urine within 6 h following acute exposure. Chapter 4 applied a cross-platform
metabolomics strategy based on CE-MS and GC-MS in order to identify and
validate dietary biomarkers in matching plasma and urine samples collected from
healthy participants in the pilot Diet and Gene Interaction Study (DIGEST). For the
first time, we demonstrate that a panel of metabolites can serve as reliable
biomarkers following contrasting Prudent and Western diets over 2 weeks of food
provisions, which correlated well with self-reported diet records. This work paves the way for the development of objective biomarkers for accurate assessment of
wood smoke exposures, as well as complex dietary patterns as required for new
advances in occupational health and nutritional epidemiology. / Dissertation / Doctor of Philosophy (PhD) / Exposomics is an emerging multidisciplinary science aimed at deciphering the
complex interactions that impact human health and gene expression, such as
lifestyle choices (i.e., habitual diet) and lifelong environmental exposures. There is

cardiometabolic disorders, including heart disease, type 2 diabetes and cancer. The
research in this thesis focuses on developing new analytical methods for identifying
and quantifying metabolites that may allow for better assessments of human health,
and has contributed to the development of novel biosensors for the targeted analysis
of N-acetylneuraminic (sialic) acid and related acidic sugars, as well as high
resolution methods for broad spectrum analysis of biotransformed organic
contaminants from smoke exposure by GC-MS, and plasma and urinary metabolites
that differentiate contrasting Prudent and Western diets and correlate well with self-reported
diet records.

Study of the Degradation of New Lubricant Oil Formulations with the Design and Demands of Current and Future EnginesGarcía Barberá, Antonio 03 March 2022 (has links)
[ES] El análisis de los aceites lubricantes de motor, englobado dentro de las tareas realizadas en el Oil Condition Monitoring (OCM), resulta ser una herramienta poderosa con la cual se es capaz de extraer información de utilidad. Por este motivo, en esta Tesis se decidió explotar este campo realizando un estudio en profundidad centrado en tres aspectos: mejorar las técnicas y protocolos ya existentes, desarrollar procedimientos propios acorde con las necesidades y requisitos del momento y, para terminar, poner en valor la información obtenida del análisis de los lubricantes mediante el uso de tratamientos estadísticos y quimiométricos. Bajo estas tres premisas, se ha desarrollado toda la tarea de investigación de esta Tesis.
En primer lugar, en relación con el análisis de los aceites lubricantes, se realizó un estudio acerca de la degradación de cuatro formulaciones de aceites lubricantes. En este estudio se analizaron aspectos propios de los aceites como: su viscosidad cinemática, oxidación, nitración, caída de los aditivos antioxidantes, etc. así como parámetros de desgaste de motor (monitorizando la presencia de metales). Como resultado de este estudio fue posible encontrar limitaciones en las técnicas actuales, lo cual condujo a realizar mejoras y/o adaptaciones de estas técnicas para conseguir obtener la información deseada.
El siguiente aspecto tratado en esta Tesis es una consecuencia de la necesidad de, en ciertas situaciones, disponer de información acerca de uno o varios parámetros de una determinada forma y con un grado de precisión elevado. El caso en cuestión analizado en la Tesis está relacionado con la cuantificación de hollín en el aceite lubricante. Respecto a la cuantificación del hollín se requirió encontrar una técnica que fuera capaz de aportar información de una forma rápida y, además, poder detectar pequeños cambios. Esta técnica fue la espectroscopia Ultravioleta-Visible, con la cual se diseñó un protocolo analítico que permitiera trabajar con muestras de aceites lubricante con muy poco uso y, por ende, poca cantidad de hollín disuelta, consiguiendo resultados muy satisfactorios en periodos de tiempo cortos.
Por último, debido a la disponibilidad de una cantidad de datos considerable fruto del trabajo realizado durante todo el tiempo de duración de la Tesis, se decidió emplear toda ella para conseguir extraer la máxima información posible gracias al uso de tratamientos estadísticos y quimiométricos. Este trabajo, nuevo en la línea, se dividió en dos vertientes: la primera focalizada en el conjunto de datos de los aceites ya caracterizados (procedentes del estudio de la degradación de los aceites) y la otra parte en encontrar sinergias entre las técnicas espectroscópicas (FT-IR y NIR) y los ensayos de caracterización de los lubricantes. Como resultado de ello, se obtuvieron relaciones entre parámetros que anteriormente no se tenían presentes, así como modelos predictivos de propiedades de los lubricantes a partir del análisis de sus espectros.
El presente documento de Tesis se encuentra redactado en dos idiomas: en español y en inglés. Principalmente el documento se encuentra redactado en inglés para hacer especial énfasis en aquellas labores de investigación desarrolladas en esta etapa, así como los resultados derivados de los diferentes estudios realzados. Mientras que, la parte confeccionada en español hace referencia a aquellos aspectos más descriptivos del documento de Tesis. / [CA] L'anàlisi dels olis lubrificants de motor, englobat dins de les tasques realitzades en el Oil Condition Monitoring (OCM), resulta ser una eina poderosa amb la qual s'és capaç d'extraure informació d'utilitat. Per aquest motiu, en aquesta Tesi es va decidir explotar aquest camp realitzant un estudi en profunditat centrat en tres aspectes: millorar les tècniques i protocols ja existents, desenvolupar procediments propis d'acord amb les necessitats i requisits del moment i, per a acabar, posar en valor la informació obtinguda de l'anàlisi dels lubrificants mitjançant l'ús de tractaments estadístics i quimiomètrics. Sota aquestes tres premisses, s'ha desenvolupat tota la tasca d'investigació d'aquesta Tesi.
En primer lloc, en relació a l'anàlisi dels olis lubrificants, es va realitzar un estudi sobre la degradació de quatre formulacions d'olis lubrificants. En aquest estudi es van analitzar aspectes propis dels olis com: la seua viscositat cinemàtica, oxidació, nitració, caiguda dels additius antioxidants, etc. així com paràmetres de desgast de motor (monitorant la presència de metalls). Com a resultat d'aquest estudi va ser possible trobar limitacions en les tècniques actuals, la qual cosa va conduir a realitzar millores i/o adaptacions d'aquestes tècniques per a aconseguir obtindre la informació desitjada.
El següent aspecte tractat en aquesta Tesi és una conseqüència de la necessitat de, en unes certes situacions, disposar d'informació sobre un o diversos paràmetres d'una determinada forma i amb un grau de precisió elevat. El cas en qüestió analitzat en la Tesi està relacionat amb la quantificació de sutge (soot en anglès). Respecte a la quantificació del soot es va requerir trobar una tècnica que fora capaç d'aportar informació d'una forma ràpida i, a més, poder detectar xicotets canvis. Aquesta tècnica va ser l'espectroscòpia Ultravioleta-Visible, amb la qual es va dissenyar un protocol analític que permetera treballar amb mostres d'oli lubrificant amb molt poc ús i, per tant, poca quantitat de soot dissolta, aconseguint resultats molt satisfactoris en períodes de temps curts.
Finalment, a causa de la disponibilitat d'una quantitat de dades considerable fruit del treball realitzat durant tot el temps de duració de la Tesi, es va decidir emprar tota ella per a aconseguir extraure la màxima informació possible gràcies a l'ús de tractaments estadístics i quimiomètrics. Aquest treball, nou en la línia, es va dividir en dos vessants: la primera focalitzada en el conjunt de dades dels olis ja caracteritzats (procedents de l'estudi de la degradació dels olis) i l'altra part a trobar sinergies entre les tècniques espectroscòpiques (FT-IR i NIR) i els assajos de caracterització dels lubrificants. Com a resultat d'això, es van obtindre relacions entre paràmetres que anteriorment no es tenien presents així com models predictius de propietats dels lubrificants a partir de l'anàlisi dels seus espectres.
El present document de Tesi es troba redactat en dos idiomes: en espanyol i en anglès. Principalment el document es troba redactat en anglès per a fer especial èmfasi en aquelles labors d'investigació desenvolupades en aquesta etapa així com els resultats derivats dels diferents estudis realitzats. fa referència a aquells aspectes més descriptius del document de Tesi. / [EN] The analysis of engine lubricating oils, encompassed within the tasks carried out in Oil Condition Monitoring (OCM), is a powerful tool with which it is possible to extract useful information. For this reason, in this Thesis it has been decided to exploit this field by carrying out an in-depth study focusing on three aspects: improving existing techniques and protocols, developing our own procedures in accordance with current needs and requirements and finally, adding value to the information obtained from the analysis of lubricants through the use of statistical and chemometric treatments. All the research work in this Thesis has been carried out under these three premises.
Firstly, in relation to the analysis of lubricating oils, a study was carried out on the degradation of four lubricating oil formulations. This study analysed aspects of the oils, such as their kinematic viscosity, oxidation, nitration, antioxidant additives depletion, etc. as well as engine wear parameters (monitoring the presence of metals). Using this study, it was possible to find limitations in the current techniques, which led to improvements and/or adaptations of these techniques in order to obtain the desired information.
The next aspect dealt with in this Thesis is a consequence of the need, in certain situations, to have information about one or more parameters in a certain form and with a high degree of accuracy. The case in question analysed in the Thesis is related to the quantification of soot. With regard to soot quantification, it was necessary to find a technique that was capable of providing information quickly and, in addition, being able to detect small changes. This technique was Ultraviolet-Visible spectroscopy, with which an analytical protocol was designed to work with samples of lubricating oils with very little use and therefore, with a small amount of dissolved soot, achieving very satisfactory results in short periods of time.
Finally, due to the availability of a considerable amount of information resulting from the work carried out throughout the duration of the Thesis, it was decided to use all of these data to extract as much information as possible, thanks to the use of statistical and chemometric treatments. This work, new in the research line, was divided into two parts: the first focused on the dataset of oils already characterised (from the study of oil degradation) and the other on finding synergies between spectroscopic techniques (FT-IR and NIR) and lubricant characterisation tests. As a result, relationships between previously unknown parameters were obtained, as well as predictive models of lubricant properties based on the analysis of their spectra.
This Thesis document is written in two languages: Spanish and English. The document is mainly written in English in order to place special emphasis on the research work carried out at this stage, as well as the results derived from the different studies carried out.
