Études in-situ dans un microscope électronique en transmission des réactions à l’état solide entre métal et nanofil de Ge / In-situ transmission electron microscopy studies of metal-Ge nanowire solid-state reactions

Le domaine des nanofils semi-conducteurs est en pleine expansion depuis ces dix dernières années grâce à leurs applications dans de nombreux domaines tels que l’électronique ou la conversion d’énergie. Dans cette étude on part d’une base de nanofil de germanium (le canal), on dépose des contacts métalliques qui seront chauffés par effet joule. Une différence de potentiel est alors appliquée au contact d’entrée (la source), le courant électrique est récupéré et mesuré par le contact de sortie (le drain). Une réaction à l’état solide permet aux atomes du métal de diffuser dans le nanofil. La propagation d'une phase métal/semi-conducteur est suivie dans un microscope électronique en transmission (MET) dont la résolution permet une observation à l’échelle atomique au niveau de la source, le drain et le canal. Les dispositifs caractérisés au cours de ce stage ont été élaborés à partir de deux types de membranes, l’une plane et l’autre avec des trous. Chacune d’entre elles sont constituées d’une couche de nitrate de silicium Si3N4 à leurs surfaces présentant l’avantage d’être transparents aux électrons et isolants au courant. / Semiconductor nanowires (NWs) are promising candidates for many device applications ranging from electronics and optoelectronics to energy conversion and spintronics. However, typical NW devices are fabricated using electron beam lithography and therefore source, drain and channel length still depend on the spatial resolution of the lithography. In this work we show fabrication of NW devices in a transmission electron microscope (TEM) where we can obtain atomic resolution on the channel length using in-situ propagation of a metallic phase in the semiconducting NW independent of the lithography resolution. We show results on semiconducting NW devices fabricated on two different electron transparent Si3N4 membranes: a planar membrane and a membrane where devices are suspended over holes. First we show the process of making lithographically defined reliable electrical contacts on individual NWs. Second we show first results on in-situ propagation of a metal-semiconductor phase in Ge NWs by joule heating, while measuring the current through the device. Two different devices are studied: one with platinum metal contacts and one with copper contacts. Different phenomena can occur in CuGe NWs during phase propagation.

The first half of the work describes the way the system should work, classification of construction and operation of the site. The second half describes the calculations of the given parameters on which the system is designed. The calculations deal with - Air exchange and maintaining the required parameters - Designing the muti stage air filtration system - Designing the air flow speed, intake and outlet parameters of the air conditiong distribution system - Desingning the air conditioning units and their possitioning in the utility room

A espectrometria de massas baseada na tecnologia MALDI-TOF (do inglês, matrix-assisted laser desorption ionization-time of flight) (MALDI-TOF MS) tem sido cada vez mais incorporada à rotina de identificações microbiológicas nos laboratórios farmacêuticos de controle de qualidade, principalmente para as atividades do Programa de Monitoramento Ambiental de Salas Limpas. Isso porque o longo tempo necessário para a obtenção dos resultados por meio de métodos convencionais tem incentivado a procura por técnicas que permitam métodos rápidos. O objetivo deste trabalho foi avaliar a adequação da técnica MALDI-TOF MS para a identificação de bactérias isoladas do ambiente de salas limpas utilizadas em algumas etapas da produção de uma vacina viral. Treze espécies bacterianas conhecidas, normalmente isoladas das salas limpas estudadas, e cinco cepas ATCC foram identificadas pela técnica MALDI-TOF MS e por uma técnica bioquímica (BBL Crystal®). O desempenho da técnica MALDI-TOF MS foi superior ao da técnica bioquímica na identificação correta das espécies bacterianas (88,89% e 38,89%, respectivamente) e produziu menos identificações não confiáveis (5,55% e 22,22%, respectivamente). Os resultados evidenciaram que a técnica MALDI-TOF MS pode ser implementada para identificação rotineira de bactérias em um laboratório de controle de qualidade farmacêutico. Entretanto, a dependência de bases de dados exige estudos adicionais de isolados não identificados e, se apropriado, a adição destes a uma base de dados interna. O aperfeiçoamento de métodos de identificação microbiana é muito relevante no contexto de salas limpas, pois permitem ações corretivas e proativas essenciais para garantir a segurança microbiológica do processamento asséptico. / Matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) has been increasingly introduced in routine microbiological identifications of pharmaceutical quality control laboratories, mainly for the activities of the Environmental Monitoring Program of Clean Rooms. The long time needed to obtain the results through conventional methods has stimulated the search for techniques that allow rapid methods, as MALDI-TOF MS. Thus, the objective of this work was to evaluate the suitability of the MALDI-TOF MS technique for the identification of bacteria isolated from the environment of clean rooms used in some stages of the production of a viral vaccine. Thirteen bacterial species commonly isolated from clean rooms studied and five strains ATCC were identified by MALDI-TOF MS technique and by a biochemical technique (BBL Crystal® System). Performance of MALDI-TOF MS was better than biochemical technique for correct species identifications (88.89% and 38.89%, respectively) and produced fewer unreliable identifications (5.55% and 22.22%, respectively). MALDI-TOF MS can be implemented for routine identification of bacteria in a pharmaceutical quality control laboratory. However, as a database-dependent system, maybe some isolated not identified by this technique must be additionally studied and, if appropriate, added to an in-house database.

Development of planar technology for focal planes of future radio to sub-millimetre astronomical instruments

Receiver systems utilising planar technologies are prevalent in telescopes observing at radio to sub-millimetre wavelengths. Receiver components using planar technologies are generally smaller, have reduced mass and are cheaper to manufacture than waveguide-based alternatives. Given that modern-day detectors are capable of reaching the fundamental photon noise limit, increases in the sensitivity of telescopes are frequently attained by increasing the total number of detectors in the receivers. The development of components utilising planar technologies facilitates the demand for large numbers of detectors, whilst minimising the size, mass and manufacturing cost of the receiver. After a review and study of existing concepts in radio to sub-mm telescopes and their receivers, this thesis develops planar components that couple the radiation from the telescope's optics onto the focal plane. Two components are developed; a W- band (75-110 GHz) planar antenna-coupled flat mesh lens designed for the receiver of a Cosmic Microwave Background (CMB) B-mode experiment, and an L-band (1- 2 GHz) horn-coupled planar orthomode transducer designed for the receiver of the FAST telescope. The first developments of a planar antenna-coupled flat mesh lens are presented. The design is driven by the requirement to mitigate beam systematics to prevent pollution of the CMB B-mode signal. In the first instance, a waveguide-coupled mesh lens is characterised. The radiation patterns of the waveguide-coupled mesh lens have -3 dB beam widths between 26 and 19 degrees, beam ellipticity <10%, and cross-polarisation.
