Orientador: Daniela Carvalho dos Santos / Resumo: Devido ao uso indiscriminado de inseticidas de largo espectro na produção agrícola e a vulnerabilidade dos inimigos naturais a este tipo de exposição, efeitos colaterais podem afetar a função biológica destes inimigos, tornando sua utilização no controle biológico impraticáveis. A busca por pesticidas mais seletivos sobre esses organismos benéficos é necessária, e entre as alternativas, enfatizamos o aumento do uso dos biopesticidas. Um dos biopesticidas amplamente utilizados é o óleo de nim (Azadirachta indica), e sua segurança e compatibilidade com os inimigos naturais têm sido mais esclarecidos através de estudos ecotoxicológicos que verificam os efeitos da exposição indireta através da ingestão de presas contaminadas. Assim, este estudo analisou a resposta das células epiteliais do mesêntero de adultos do crisopídeo Ceraeochrysa claveri quando expostos por ingestão de presa contaminada com óleo de nim durante a fase larval e a ocorrência de efeitos letais e subletais em seu desenvolvimento. Larvas de C. claveri foram alimentadas com ovos de Diatraea saccharalis tratados com óleo de nim nas concentrações de 0,5%, 1% e 2% durante todo período larval. Fêmeas adultas obtidas a partir destes tratamentos foram utilizadas em análises morfológicas e ultraestruturais da resposta celular do mesêntero. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Due to the indiscriminate use of broad-spectrum insecticides in agricultural production and the vulnerability of natural enemies to this type of pesticide exposure, side effects can affect the biological function of these enemies, making their use for biological control unfeasible. A search for more selective pesticides on these beneficial organisms is necessary, and among the alternatives, we emphasize the increased use of biopesticides. One of the biopesticides widely used is the neem oil (Azadirachta indica), and its safety and compatibility with natural enemies have been more clarified through ecotoxicological studies which check the effects of indirect exposure by intake of poisoned preys. Thus, this study analyzed the response of midgut epithelial cells of adult of lacewing Ceraeochrysa claveri when exposed by intake of contaminated prey with neem oil during the larval stage and the occurrence of lethal and sublethal effects on its development. Larvae of C. claveri were fed on eggs of Diatraea saccharalis treated with neem oil at a concentration of 0.5%, 1% and 2% throughout the larval stage. Aspectos gerais da criação de Ceraeochrysa cincta (Schneider, 1851) (Neuroptera : Chrysopidae) em laboratório /

Orientador: Sergio Antonio De Bortoli / Resumo: No controle biológico, além dos microrganismos benéficos, devem ser também destacados parasitoides e predadores, sendo um bom exemplo deste último grupo, os crisopídeos (Neuroptera: Chrysopidae). O controle biológico aplicado, com a utilização de insetos predadores, como é o caso dos crisopídeos, está na dependência de alguns fatores, destacando-se dentre eles o conhecimento da biologia do inseto e a necessidade do domínio de uma técnica de criação eficiente e econômica em laboratório, que permita sua produção massal para subsequentes liberações em áreas de exploração agrícola. Já é de conhecimento que larvas de alguns crisopídeos são mantidas em laboratório, com boa eficiência, alimentadas com fontes de alimento obtidas de outras criações de insetos também mantidas em laboratório, dentre os quais podem ser destacados ovos de alguns lepidópteros. Ovos de Corcyra cephalonica (Stainton, 1866) (Lepidoptera: Pyralidae) são benéficos ao desenvolvimento de crisopídeos, produzindo adultos com alta viabilidade, porém há poucos relatos que estabelecem relação entre a espécie com Ceraeochrysa cincta (Schneider, 1851). A good example of this latter group is the crisopids (Neuroptera: Chrysopidae). The biological control applied, with the use of predatory insects, as in the case of the crisopids, is dependent on some factors, especially the knowledge of insect biology and the need to master an efficient and economical breeding technique in the laboratory, allowing its mass production for subsequent releases in farm areas. It is known that larvae of some chrysopids are maintained, with good efficiency, with food sources obtained from other insect creations also kept in the laboratory, among which lepidopteran eggs can be highlighted. Diversidade e análise faunística de crisopídeos (Neuroptera: Chrysopidae) em fragmento de floresta estacional semidecidual em Três Pontas, Minas Gerais /

A captura ativa foi realizada com rede entomológica confeccionada com tecido voil e armadilhas contendo melaço de cana-de-açúcar a 10% como fonte atrativa. As coletas foram realizadas semanalmente através de captura ativa no período de 13 de setembro de 2008 a 05 de setembro de 2009, e duas coletas com armadilhas atrativas em cada estação do ano. As coletas com rede entomológica foram iniciadas as 7:00 h e finalizadas as 17:00 h. Após esse horário, nas datas determinadas, foram distribuídas as armadilhas atrativas, que eram recolhidas no outro dia até as 7:00 h. Os insetos capturados foram montados em alfinetes entomológicos, etiquetados, triados e identificados. Plantas que tiveram alta concentração de crisopídeos foram coletadas, preparadas exsicatas e encaminhadas a especialista para identificação. Foram capturados 1.467 crisopídeos, distribuídos em 33 espécies, pertencentes as tribos Chrysopini e Leucochrysini. A espécie de maior ocorrência foi Ceraeochrysa tucumana (Navás, 1919), tendo a presença relatada em todas as amostragens. Não foi encontrado diferença entre os horários de coleta para os gêneros de crisopídeos. Nas coletas noturnas, foram capturados seis indivíduos, pertencentes as espécies Leucochrysa (Nodita) vignisi Freitas & Penny, 2001; Ceraeochrysa achillea Freitas e Penny, 2009; Chrysopodes spinella (Adams e Penny, 1987) e C. tucumana. Foi possível verificar associação entre espécies de crisopídeos com plantas do gênero Casearia / Abstract: The objective of this work was to study the diversity and faunistic analysis of lacewings species in Semideciduous Forest located in Três Pontas, southern Minas Gerais State, Brazil. Active capture was performed with an sweep net made of cheesecloth and traps with molasses cane sugar to 10% as attractive. Samples were weekly with active collected in period from 13/IX/2008 to 05/IX/2009 and two collections with attractive traps in each season. Sampling insect nets were opened at 7:00 pm and finished 17:00 h am. After this time, on certain dates, were distributed attractive traps, which were collected the next day until 7:00 pm. The insects were mounted on entomological pins, labeled, sorted and identified. Plants that have high concentrations of lacewings were collected, prepared and dried specimens sent to a specialist for identification. 1,467 lacewings were captured, distributed in 33 species belonging to the tribes Chrysopini and Leucochrysini. The species most frequent was Ceraeochrysa tucumana (Navás, 1919) and reported presence in all the samples. No difference was found between sampling times for the genus of Chrysopidae. In nocturnal collections were captured six individuals of the species Leucochrysa (Nodita) vignisi Freitas & Penny, 2001; Ceraeochrysa achillea Freitas and Penny, 2009; Chrysopodes Spinella (Adams and Penny, 1987) and C. tucumana. Estudos de variabilidade genética em populações de Chrysoperla externa Hagen, 1861 (Neuroptera: Chrysopidae) do estado de São Paulo, Brasil /

A espécie Chrysoperla externa encontra-se tanto em ambientes de vegetação nativa quanto em áreas de cultivo agrícola e possui várias características que a tornam um potencial agente de controle biológico. Entretanto, a capacidade de uma espécie se adaptar à ambientes exóticos, como agroecossistemas, está relacionada à sua variabilidade genética. Assim, os objetivos desse trabalho foram investigar e comparar a variabilidade genética de C. externa em populações estabelecidas no estado de São Paulo, em áreas de vegetação nativa e agroecossistemas, bem como analisar os efeitos da deriva genética e do gargalo populacional sobre essas populações. Foram utilizadas 13 populações coletadas em agroecossistemas, três em áreas nativas e uma linhagem mantida em laboratório sob forte endogamia. Para todas as populações, foram utilizados marcadores moleculares ISSR e para as populações coletadas em agroecossistemas, empregou-se também o gene mitocondrial COI. Altos níveis de variabilidade genética foram observados em todas as populações, o que configura uma característica da espécie. A correlação entre as distâncias genética e geográfica foi baixa, havendo grande compartilhamento de haplótipos entre todas as populações de agroecossistemas. Para essas populações, o gene mitocondrial COI indicou ausência de estrutura genética, enquanto para o marcador ISSR esta pôde ser observada, indicando que as populações estabelecidas em agroecossistema formavam uma única unidade populacional que vem se diferenciando. The species Chrysoperla externa is found in native vegetation and agricultural crops and has several features that make it a potential biological control agent. However, the ability of a species to adapt to exotic environments, such as agroecosystems, is related to the genetic variability. The aims of this study were to investigate and compare the genetic variability of C. externa in established populations in the state of São Paulo, in areas of native vegetation and agroecosystems, as well as to analyze the effects of genetic drift and bottleneck on these populations. We used 13 populations collected in agroecosystems, three in native areas and one lineage maintained in the laboratory under strong inbreeding. ISSR molecular markers were used for all populations, but for the populations collected in agroecosystems we used also the mitochondrial COI gene. High levels of genetic variability were observed in all populations, which is one characteristic of the specie. The correlation between genetic and geographic distances was low and a large number of haplotypes was shared among all populations of agroecosystems. For populations of agroecosystems, the mitochondrial COI gene showed no genetic structure, whereas the ISSR marker demonstrated genetic structure, indicating that the established populations in agroecosystems formed a single population unit, which has been under differentiation. Eficiência de predação de Chrysoperla externa (Hagen, 1861) (Neuroptera: Chrysopidae) sobre o tripes do amendoim Enneothrips flavens (Moulton, 1941) (Thysanoptera: Thripidae) /

Orientador: Sérgio de Freitas / Banca: Arlindo Leal Boiça Júnior / Banca: Márcio da Silva Araújo / Resumo: Este estudo teve como objetivo verificar o potencial de predação de E. flavens através da liberação de ovos embrionados e larvas de primeiro ínstar de C. externa em plantas de amendoim cultivadas em casa-de-vegetação. O delineamento experimental utilizado foi o inteiramente casualizado com três tratamentos (plantas que não receberam liberação, plantas que receberam liberação de ovos embrionados e plantas que receberam liberação de larvas de primeiro ínstar) e 15 repetições no primeiro experimento e 20 no segundo. Foram realizadas amostragens antes da liberação e com 4, 9 e 15 dias após. No primeiro experimento e em relação às diferentes amostragens, as plantas que não receberam liberação o número médio de tripes não variou significativamente. As plantas que receberam liberação de ovos e larvas apresentaram aumento significativo do número médio de tripes aos quatro dias. Aos nove dias o número médio de tripes reduziu-se significativamente nas plantas que receberam liberação de ovos e aos 15 dias nas plantas que receberam liberação de larvas. No segundo experimento e em relação às diferentes amostragens, o número médio de tripes nas plantas que não receberam liberação não variou significativamente, como ocorrido no primeiro experimento. As plantas que receberam liberação de ovos apresentaram redução significativa no número médio de tripes aos quatro dias após a liberação. O mesmo ocorreu com as plantas que receberam liberação de larvas. Foi observado no segundo experimento, através da fórmula de Abbott, eficiência de redução da população de tripes de 71 e 74%, quando foram liberados ovos e larvas, respectivamente. Dessa maneira foi possível verificar que C. externa possui potencial de predação sobre o tripes do amendoim E. flavens / Abstract: This study was carried out in order to verify the potential of predation of E. flavens through the release of fertilized eggs and first instar larvae of C. externa peanut plants grown in a greenhouse. The experimental design was completely randomized design with three treatments (plants that did not receive release, plants that received release embryonated eggs and plants that received release of the first larval instar) and 15 repetitions in the first experiment and 20 in the second. Samplings were made before the release and with 4, 9 and 15 days. In the first experiment and for the different samples, plants that were not release the average number of thrips did not vary significantly. The plants that received the release of eggs and larvae showed significant increase in the average number of thrips to four days. At nine days the average number of thrips decreased significantly in plants that received release of eggs and 15 days in plants that received release of larvae. In the second experiment and for the different samples, the average number of thrips on plants that did not receive release did not vary significantly, as occurred in the first experiment. The plants that received the release of eggs showed a significant reduction in the average number of thrips to four days after release. The same occurred with the plants that received release of larvae. Was observed in the second experiment, using the formula of Abbott, efficiency of reducing the population of thrips on 71 and 74%, when they were released eggs and larvae, respectively. Análise dos genes mitocondriais COI e 16S de populações de Chrysoperla externa (Hagen, 1861) (Neuroptera: Chrysopidae) de diferentes localidades geográficas /

Em cada ambiente em que for encontrada poderá sofrer diferentes pressões seletivas do ambiente. Para a verificação das mutações genéticas presentes nas populações de C. externa podem ser usados marcadores moleculares, em especial os genes mitocondriais, que são de fácil manipulação. C. externa é de fácil criação e a mais estudada para multiplicação massal, por isso se faz necessário estudar a resposta das populações quando liberadas em ambientes diferentes, visto que estas podem não sobreviver em áreas biogeográficas distintas, inviabilizando o seu papel como agente de controle de pragas. O objetivo deste trabalho foi caracterizar geneticamente as populações de C. externa nos municípios de Brotas (SP), Jaboticabal (SP), Rifaina (SP), São Carlos (SP), São José dos Campos (SP) e São Sebastião do Paraíso (MG). Para o gene COI foram verificados oito haplótipos, seis mutações e a maior diversidade haplotípica foi encontrada em Brotas e São Sebastião do Paraíso. Para o gene 16S foram observadas quatro mutações, seis haplótipos e a maior diversidade haplotípica ocorreu no município de São Sebastião do Paraíso. A distância genética encontrada entre as populações de C. externa não foi significativa para os dois genes analisados, evidenciando que provavelmente as populações são geneticamente compatíveis. O estudo da estrutura genética dessas populações de C. externa, de ambos os genes, mostrou que essas populações não apresentam um padrão de distribuição haplotípica. Então, talvez sejam necessários outros estudos com populações desta espécie oriundas de localidades mais distantes geograficamente dos que as utilizadas neste trabalho / Abstract: Chrysoperla externa is a species of green lacewing found in several Brazilian agroecosystems able to feed on various agricultural pests. In each environment that is found may experience different environmental selective pressures. In order to verify genetic mutations in C. externa populations it may be used molecular markers, in particular mitochondrial genes, which are easy handling and extraction. C. externa is easy to create and the most studied for mass multiplication, so it is necessary to study on the response of populations when released in different environments, since they may not survive in different biogeographic areas, derailing its role as a pest controller. The objective of this study was to characterize genetically the populations of C. externa from the cities of Brotas (SP), Jaboticabal (SP), Rifaina (SP), São Carlos (SP), São José dos Campos (SP) and São Sebastião do Paraíso (MG). For the COI gene it was found eight haplotypes, six mutations and greater haplotype diversity in Brotas and São Sebastião do Paraíso. In the 16S there were four mutations, six haplotypes and haplotype diversity was higher in São Sebastião do Paraíso. The genetic distance among populations of C. externa was not significant for the two genes analyzed, showing that the populations are probably genetically compatible. The study of the genetic structure of populations of C. externa, showed for both genes that these populations do not show a pattern of haplotype distribution. Toxicidade e genotoxicidade do biopesticida Azamax™ em ovário e intestino de Ceraeochrysa claveri (Neuroptera:Chrysopidae)

Orientador: Daniela Carvalho dos Santos / Resumo: O biopesticida Azamax™ tem sido utilizado como uma alternativa aos inseticidas sintéticos para o controle de pragas em plantações. Além disso, este produto tem obtido destaque por poder se associar ao uso de predadores naturais no controle biológico previsto pelo Manejo Integrado de Pragas (MIP). Dentre os modelos biológicos, destaca-se a espécie Ceraeochrysa claveri, sobretudo devido sua característica predadora na fase de larva com ampla variedade de presas e alto potencial reprodutivo na fase adulta. Embora a azadiractina, ingrediente ativo do Azamax™, já tenha sido relacionada com a indução de efeitos citotóxicos em insetos, ainda não há dados na literatura sobre os possíveis efeitos da sua ingestão indireta em células germinativas e intestinais de C. claveri. Desta forma, o objetivo deste estudo foi avaliar o efeito tóxico e genotóxico do Azamax™ em células do ovário e do intestino de fêmeas de C. claveri. As larvas foram alimentadas ad libitum, com ovos de Diatraea saccharalis tratados com duas diferentes concentrações do Azamax™ (0,3% e 0,5%) durante todo o seu período larval (n=150 larvas/grupo). As análises morfológicas, ultraestruturais e de genotoxicidade foram realizadas após os indivíduos se desenvolverem até a fase adulta. Os resultados indicaram que a exposição ao Azamax™ somente na fase larval, induziu sinais de toxicidade em todas as fases do desenvolvimento do inseto, como: (a) significante taxa de mortalidade a partir do 3º instar larval; (b) prepupa e pupa... Besides, this product has been highlighted because this association use with natural predators for biological control in the integrated pest management programs (IPM). Among the biological models, is commonly use the specie Ceraeochrysa claveri, above all its predatory characteristic at the larval stage with a wide variety of prey and high reproductive potential in the adult stage. Azadirachtin, active ingredient of Azamax ™, has been related to the induction of cytotoxic effects in insects, although it has not been published in literature about the possible effects of Azamax and its indirect ingestion in germ and intestinal cells of C. claveri. From this, the aim of this study, is to evaluate the toxic and genotoxic effect of Azamax ™ in cells of the ovary and intestine of females of C. claveri. Larvae were fed ad libitum with treated eggs of Diatraea saccharalis at different concentrations of Azamax ™ (0.3% and 0.5%) during all or their larval period (n = 150 larvae/group). Com o propósito de avaliar o efeito do óleo de nim, um extrato vegetal com propriedades bioinseticidas, sobre os crisopídeos, ovos de Diatraea saccharalis foram tratados com óleo de nim em três diferentes concentrações e oferecidos como alimento a larvas de Ceraeochrysa claveri, avaliando deste modo o efeito da ingestão do óleo de nim na fase larval sobre a morfologia do testículo durante o desenvolvimento de C. claveri, assim como sobre o processo de espermatogênese deste predador. Foram utilizadas a análise histológica, ultraestrutural e imunocitoquímica para a avaliação da toxicidade do óleo de nim sobre as células do testículo de C. claveri; sendo esta avaliação realizada em testículos de larvas de terceiro instar, pupas e adultos. As análises demonstraram que a morfologia dos testículos varia de arredondado em larvas, para alongado em pupas e espiralado em adultos, enquanto sua coloração varia de esbranquiçado a amarelo intenso, respectivamente. Thus, the search for conservation of natural resources is also growing, along with organic agriculture and integrated pest management. In search of insects that can be used in biological control, which is an important tool in organic agriculture and integrated pest management, the lacewings has proved a strong controlling agent, since their larvae are predators of eggs and larvae of many insects pests of different commercial crops. In order to evaluate the effect of neem oil, a plant extract with insecticides properties on lacewings, Diatraea saccharalis eggs were treated with neem oil at three different concentrations and offered as food for the larvae of Ceraeochrysa claveri, thereby evaluating the effect of intake of neem oil on the larval morphology of testis during development of C. claveri, as well as on the process of spermatogenesis of this predator. Histological, ultrastructural, and immunohistochemistry analysis was used to evaluate the toxicity of neem oil on cells of the testis of C. claveri, and this evaluation performed in testes of third instar larvae, pupae and adults. Estudo do padrão de distribuição genético-haplotípico de Chrysoperla externa (Neuroptera: Chrysopidae) em áreas de citros no estado de São Paulo /

Resumo: Os crisopídeos são insetos com grande potencial para uso em programas de controle biológico de pragas agrícolas. Populações de Chrysoperla externa apresentam ampla distribuição geográfica, abrangendo desde o sul dos Estados Unidos até o sul da América do Sul, ocorrendo em diferentes ambientes. Contudo, há relativamente poucos estudos buscando compreender a estrutura genética de agentes de controle biológico, especialmente insetos predadores. Desta forma, os principais objetivos deste trabalho foram caracterizar geneticamente as populações de C. externa por meio de sequências do gene mitocondrial COI e compreender sua estrutura populacional nos municípios amostrados no Estado de São Paulo. Para tanto, indivíduos adultos foram coletados em pomares de citros, e da região torácica foi extraído o DNA total. O gene COI foi amplificado por meio da técnica de PCR e as amostras foram purificadas e sequenciadas. As populações de C. externa analisadas apresentaram elevada diversidade genética, bem distribuída entre os municípios amostrados. Esta homogeneização pode ser decorrência de fluxo gênico, ação antrópica, correntes de ar, proximidade das fazendas com matas nativas e elevado potencial reprodutivo de C. externa. A partir dos resultados obtidos é possível inferir que o agroecossistema, por ser um ambiente homogêneo, esteja contribuindo para a perda de estruturação que havia entre estas populações quando elas viviam em ecossistemas nativos, sendo assim, é fundamental que as populações de C. externa sejam estudas neste ambiente, para que possam ser compreendidas em seu habitat de origem e sem a influência da ação antrópica / Abstract: The green lacewings are insects with great potential for use in programs of biological control of agricultural pests. Populations of Chrysoperla externa are widely distributed geographically, being found from the southern United States until the southern South America, occurring in different environments. However, there are relatively few studies trying to understand the genetic structure of biological control agents, especially predatory insects. Thus, the main objectives of this work were to characterize genetically the populations of C. externa through COI mitochondrial gene sequences and to understand its population structure in sampled municipalities in the State of São Paulo. For this purpose, adult individuals were collected in citrus orchards, and from its torax were extracted the total DNA. The COI gene was amplified by PCR and the samples were purified and sequenced. The populations showed high genetic diversity, well distributed among the municipalities. This homogenization may be due to gene flow, human action, action of winds, proximity of farms to native forests, and high reproductive potential of C. externa. Assim, a busca pela preservação dos recursos naturais também está em crescimento, juntamente com a agricultura orgânica e do manejo integrado de pragas. Na busca por insetos que possam ser utilizados no controle biológico, sendo esta uma importante ferramenta na agricultura orgânica e manejo integrado de pragas, os crisopídeos vem se mostrando um forte agente controlador, uma vez que suas larvas são predadoras de diversos ovos e larvas de insetos pragas de diferentes culturas comerciais.
