Příprava derivátů cyklodextrinů použitelných pro modifikace pevných povrchů / Preparation of cyclodextrin derivatives usable for modifications of solid surfaces

This master thesis deals with preparation of cyclodextrin (CD) derivatives that are suitable for bonding on solid surfaces such as dopamine polymer and gold. The nucleophilic attack of 6I -O-p-toluenesulfonyl--CD (6-tosyl-β-CD) by dithiols created derivatives with substituents bound to CD skeleton by one of the sulphur atom while keeping a free terminal sulphanyl group. Despite initial problems caused by oxidation of the thiols to disulfides, CDs were modified by alkyl or oligoethylene glycol spacers prepared in 60 - 88% yields. The same reaction was used for preparation of derivatives of β-CD with amino group at the end of the tether that was linked to CD by nitrogen atom. For these purposes were used corresponding diamines and reactions proceeded smoothly reaching up to 92% isolated yield. Next, also the chemoselectivity of the reaction with an ambident spacer bearing both amino and sulphanyl functional group was studied and by 2D NMR experiments was proved that substituent was bound to CD through sulphur atom. It was demonstrated on the case of preparation of CD derivative with dimethylene linker that was prepared in 84% yield. Příprava derivátů γ-cyklodextrinu použitelných pro tvorbu samoskladných struktur / Preparation of γ-cyclodextrin derivatives usable for construction of self-assembled structures

This thesis is focused on the preparation of monosubstituted derivatives of -cyclodextrin ( -CD) which can form supramolecular polymers. These polymers can be utilized in capillary electrophoresis. Set of monosubstituted 2I -O-, 3I -O- a 6I -O-(3-(naphthalen-2-yl)prop-2-en-1-yl) derivatives of -CD was prepared (naphthylallyl derivatives). Reaction was performed by cross metathesis of O-allyl derivatives of -CD with a 2-vinylnaphthalene in yields 16 - 25 %. Hoveyda-Grubbs 2nd generation catalyst was used. Alkylation of -CD with 2-(3-bromoprop-1-enyl)naphthalene followed by peracetylation of remaining hydroxyl groups and separation of isomers resulted in the per-O-acetyl-3I -O- a per-O- acetyl-6I -O-(3-(naphthalen-2-yl)prop-2-en-1-yl)- -CD in yields 5 and 1 %. A set of dimers of -CD 1,4-bis( -CD-2I -O-yl)-but-2-ene, 1,4-bis( -CD-3I -O-yl)-but-2-ene and 1,4-bis( - CD-6I -O-yl)-but-2-ene was prepared, too. Reaction was performed by cross metathesis of O-allyl derivatives of -CD in yields 1 - 8 % using again Hoveyda-Grubbs 2nd generation catalyst. The dimers of naphthalene guests (1,2-bis(naphthalen-2-yl)ethene and 1,2-bis(naphthalen-2-yl)ethane) for preparation of complexes with CD dimers were also prepared. Mechanistic Study of Pollutant Degradation

Environmental pollution has been a serious concern worldwide. Many degradation methods have been developed to clean sites contaminated with pollutants. More knowledge and better understanding in this field will help to protect our environment. The goal of the research in this thesis is to gain a better understanding of the mechanism of organic pollutant degradation in Fenton reactions and sonochemical reactions. Fenton degradation uses hydroxyl radical to oxidize organic compounds. The radical is produced by catalytic decomposition of hydrogen peroxide with Fe(II). Further research has found that addition of cyclodextrins can enhance degradation efficiency of hydrophobic organic pollutants. To study the mechanism of the enhancement, pollutant-cyclodextrin-Fe(II) aqueous systems were studied by fluorescence and NMR techniques. The results indicated the formation of pollutant/carboxymethyl-â-cyclodextrin/Fe(II) ternary complexes in the solution. With the ternary complex, the catalyst Fe(II) becomes closer to the pollutant, therefore leading to more efficient hydroxyl radical attack on the pollutant. Additional studies showed that hydropropyl-â- cyclodextrin, â-cyclodextrin and á-cyclodextrin bound pollutant well, but bound Fe(II) poorly. Sulfated-â-cyclodextrin did not bind well with pollutant although it bound Fe(II) well. Sonochemical degradation is another important pollutant treatment method in practice. It was found that phenol sonolysis can be enhanced by volatile hydrogen atom scavengers such as carbon tetrachloride and perfluorohexane. The non-volatile hydrogen atom scavenger iodate did not enhance phenol degradation. The first order rate constant for aqueous phenol degradation increased by about 2.2-2.8 times in the presence of 150 ìM carbon tetrachloride. In the presence of less than 1.5 ìM perfluorohexane the first order rate constant increased by about 2.3 times. Hydroquinone was the major observed reaction intermediate both in the presence and absence of hydrogen atom scavengers. Hydroquinone yields were substantially higher in the presence of hydrogen atom scavengers, suggesting that hydroxyl radical pathways for phenol degradation were enhanced by the hydrogen atom scavengers. The additives investigated in this study have potential to improve pollutant degradation efficiency. Other fields may also benefit from the information gained in this study. For example the improvement could be achieved in synthetic processes that rely on hydroxyl radical as a key intermediate.

Complexação de guaiacol com ß-ciclodextrina para tratamento de alveolite seca: avaliação in vitro e in vivo / ß-cyclodextrin complexation of guaiacol for the treatment of dry socket: in vitro and in vivo evaluation

A alveolite seca (AS) é uma das complicações pós-operatórias mais comuns e sintomáticas na odontologia, porém, até o momento não há um protocolo de tratamento definido. O composto fenólico guaiacol (Gu) é um dos materiais utilizados para revestimento intra-alveolar devido às suas propriedades analgésicas, antioxidantes e antimicrobianas. Contudo, sua desvantagem é a dificuldade de manipulação decorrente da sua baixa estabilidade, alta volatilidade e sensibilidade à oxidação. Para melhorar suas propriedades e aumentar sua aplicabilidade clínica, um complexo de inclusão de Gu com ß-ciclodextrina (ßcd) foi desenvolvido. A formação do complexo supramolecular de Gu:ßcd foi caracterizada mediante a ressonância magnética nuclear (RMN), nos experimentos de 1H e 2D ROESY. A atividade antibacteriana do Gu e Gu:ßcd frente a Escherichia coli, Staphylococcus aureus, Streptococcus mitis, Streptococcus mutans, Streptococcus sanguis e Aggregatibacter actinomycetemcomitans foi analisada pelo método da microdiluição e sua citotoxicidade em osteoblastos de calvária de rato, foi estudado com o ensaio do MTT. O processo de reparo alveolar induzido pelo Gu:ßcd foi avaliado histologicamente após tratamento de alveolite seca em molares inferiores de ratos. A RMN mostrou correlações espaciais entre os hidrogênios internos (H3 e H5) da ßcd e os hidrogênios aromáticos, H(a) e H(b) do Gu, confirmando a formação do complexo. A complexação do Gu na ßcd potencializou seu efeito antibacteriano e reduziu sua citotoxicidade em osteoblastos. O estudo in vivo evidenciou a ocorrência de ossificação no ápice alveolar dos ratos tratados com Gu:ßcd, no 7o dia. No 14o dia, as trabéculas ósseas ocuparam também o terço médio do alvéolo e no 21o dia, todo o alvéolo se encontrava preenchido por osso neoformado. Estes resultados foram similares ao controle negativo e superiores ao controle positivo (Alvogyl®). Os benefícios obtidos pela inclusão do Gu na ßcd foram demonstrados pela melhora das propriedades biológicas do Gu in vitro e o adequado reparo alveolar in vivo. / Dry socket is one of the most common and symptomatic complications in dentistry, however, there is still not a settled treatment for this condition. The phenolic compound guaiacol (Gu) is one of several alveolar dressings used in dry socket because it has analgesic, antioxidant and antimicrobial properties. Nevertheless, its disadvantage is the difficulty of manipulation due to its low stability, high volatility and sensitivity to oxidation. To improve its properties and increase its clinical applicability, an inclusion complex of Gu with ß-cyclodextrin (ßcd) was developed. The Gu:ßcd supramolecular complex was characterized by Nuclear Magnetic Ressonance (NMR), in the 1H and 2D ROESY experiments. The antibacterial activity of Gu and Gu:ßcd over Escherichia coli, Staphylococcus aureus, Streptococcus mitis, Streptococcus mutans, Streptococcus sanguis and Aggregatibacter actinomycetemcomitans was analyzed using the microdilution method and its cytotoxicity in rat calvaria-derived osteoblast was evaluated with the MTT assay. The alveolus repair process induced by Gu:ßcd was histologically studied after the treatment of dry socket in rat mandibular molars. The NMR showed spatial correlations between internal hydrogens (H3 and H5) of ßcd and aromatic hydrogens, H(a) and H(b), of Gu confirming the inclusion complex formation. Gu:ßcd complex potentiated Gu antibacterial effect and reduced its cytotoxicity in osteoblasts. The in vivo study revealed that ossification occurred in the alveolar apex of rats treated with Gu:ßcd, by day 7. In the 14th day, the trabecular bone occupied the apical and middle thirds of the socket and on the 21st day, the entire alveolus was filled by newly formed bone. These results were similar to the negative control and superior to the positive control (AlvogylTM). Benefits gained from inclusion of Gu in cyclodextrin have been particularly demonstrated by the improvement in Gu biological properties in vitro and the appropriate alveolus repair in vivo.

Criopreservação do sêmen ovino com incorporação de colesterol por ciclodextrina / Criopreservation of ram semen with colesterol loaded cyclodextrin

A preocupação com a qualidade do sêmen ovino congelado tem sido motivo de muitas pesquisas, principalmente pela dificuldade da transposição cervical durante a inseminação artificial. Contudo, a inseminação intra-cervical é frequentemente usada em ovinos e resulta em redução da fertilidade com o uso do sêmen congelado. Neste sentido, esse estudo foi dividido em dois experimentos. No 1o experimento foi verificado o potencial da ciclodextrina pré-carregada com colesterol como aditivo ao diluidor na proteção da cinética espermática, integridade das membranas plasmática e acrossomal, função mitocondrial, capacitação espermática e produção de espécies reativas ao oxigênio (ROS) em espermatozoides criopreservados ovinos. Cinco ejaculados de seis carneiros (n = 30) foram divididos em três tratamentos: apenas diluidor (CON); diluidor + colesterol incorporado a ciclodextrina (CLC + CHO); e diluidor + ciclodextrina pura (CLCP). Após a diluição (50x106 espermatozóides/mL), o sêmen foi envasado em palhetas, identificado e criopreservado utilizando um sistema automatizado. Duas palhetas da mesma partida de cada tratamento foram descongeladas (a 37°C durante 30 segundos) e analisadas quanto motilidade (CASA), morfologia dos espermatozoides (DIC), integridade do plasma (PI-H342) e acrossomal (FITC-PSA) membranas, potencial mitocondrial (JC-1), produção de radicais livres (CellRox), peroxidação lipídica (BODIPY) e fluidez da membrana celular (merocianina 540). As comparações entre os grupos foram analisadas pelo PROC MIXED do SAS e o efeito de grupo foi detectado pelo teste de Tukey (p<0,05) ou pela estatística não paramétrica de ordem (Kruskal-Wallis), quando era necessário. No 2º experimento os mesmos ejaculados foram divididos por congelabilidade baseados na motilidade total pós-descongelação (MTPD) e na porcentagem de redução na motilidade total (%RMT) quando comparados o sêmen in natura e o sêmen pós-descongelação (somente diluidor), sendo divididos nos grupos alta (MTPD>60% e %RMT<40%), intermediária (60%>MTPD>40% e 60%>RMT>40%) e baixa (MTPD<40% e %RMT>60%) congelabilidade. Foram analisados os tratamentos CON e CLC+CHO dentro de cada animal e de cada grupo quanto a motilidade (CASA), a integridade das membranas plasmática (PI-H342) e acrossomal (FITC-PSA), potencial mitocondrial (JC-1), peroxidação lipídica (BODIPY) e fluidez da membrana celular (merocianina 540). As comparações foram realizadas por análise de variância (ANOVA) em arranjo fatorial 3X3 (Tratamento CLC+CHO, CON e in natura X grupos alta, intermediária e baixa congelabilidade), as comparações entre os grupos foram analisadas pelo PROC MIXED do SAS e o efeito de grupo foi detectado pelo teste de Tukey (p <0,05) ou pela estatística não paramétrica de ordem (Kruskal-Wallis), quando era necessário. No 1o experimento o tratamento CLC+CHO se mostrou mais eficaz em preservar os parâmetros de motilidade, integridade de membranas e potencial mitocondrial quando comparado aos demais tratamentos. O grupo CLCP mostrou queda nos parâmetros de motilidade, integridade de membrana, potencial mitocondrial, mostrando, com isso, uma queda na preservação espermática. No 2o experimento observou-se que o tratamento CLC+CHO mostrou maior grau de preservação para motilidade total e progressiva, células rápidas, preservação de membranas plasmática e acrossomal e potencial mitocondrial quando comparado ao CON. Esse efeito teve maior significância nos grupos de baixa e intermediária congelabilidade, não sendo tão expressivo no grupo de alta. Pode-se concluir com esse estudo que o tratamento com ciclodextrina acrescida de colesterol contribui para uma melhor preservação dos parâmetros espermáticos do sêmen ovino, principalmente em animais apresentando baixa congelabilidade, contudo não apresenta diferença em ejaculados de alta congelabilidade. / Frozen ram semen has been the subject of many researches, mainly due to the difficulty of cervical transposition during artificial insemination. However, intra-cervical insemination in sheep is often used, resulting in reduced fertility with the use of frozen semen. To this end, this study was divided into two experiments. In the first experiment was verified the potential of cyclodextrin loaded with cholesterol as an additive to the extender in the protection of sperm kinetics, integrity of plasmatic and acrosomal membranes, mitochondrial function, sperm capacitation and production of reactive oxygen species (ROS) in cryopreserved sheep sperm. Five ejaculates from six rams (n = 30) were divided into three treatments: only extender (CON); extender + cyclodextrin loaded with cholesterol (CLC + CHO); and extender + pure cyclodextrin (CLCP). After dilution (50x106 spermatozoa/mL), semen was stored in straws, identified and cryopreserved using an automated system. Two straws from each ejaculate and treatment were thawed (at 37° C for 30 seconds) and analyzed for motility (CASA), morphology of spermatozoa (DIC), plasmatic (PI-H342) and acrosomal (FITC-PSA) membrane integrity, mitochondrial potential (JC-1), production of free radicals (CellRox), lipid peroxidation (BODIPY) and cell membrane permeability (Merocyanine 540). Comparisons between groups were analyzed using PROC MIXED of SAS and the effect of group was detected by Tukey (p <0.05) or by the order of nonparametric statistics (Kruskal-Wallis) test, when necessary. In the 2nd experiment the same ejaculates were divided by freezability based on the total post-thaw motility (TPTM) and the percentage of reduction in total motility (%RTM) when compared fresh and post-thaw semen (only extender), and divided into the groups: high (TPTM ≥ 60% and %RTM ≤ 40%), intermediate (60%> TPTM > 40% and 60%> %RTM > 40%) and low (TPTM ≤ 40% and %RTM ≥ 60%) freezability. CON and CLC + CHO treatments were analyzed within each animal and each group for motility (CASA), plasmatic (PI-H342) and acrosomal (FITC-PSA) membrane integrity, mitochondrial potential (JC-1), lipid peroxidation (BODIPY) and cell membrane permeability (Merocyanine 540). Comparisons were performed by analysis of variance (ANOVA) with a factorial arrangement 3x3 (groups CLC+CHO, CON and fresh X groups high, medium and low freezability), comparisons between groups were analyzed using PROC MIXED of SAS and the group effect was detected by the Tukey test (p < 0.05) or no statistical order parametric (Kruskal-Wallis), when necessary. In the first experimente, CLC + CHO treatment was more effective in preserving the parameters of motility, membrane integrity, and mitochondrial potential compared to other treatments. The CLCP group showed a fall in the parameters of motility, membrane integrity, mitochondrial potential, showing thereby a decrease in sperm preservation. In the 2nd experiment, it was observed that the treatment CLC + CHO showed greater preservation for total and progressive motility, rapid cell preservation, plasmatic and acrosomal membranes and mitochondrial potential when compared to CON. This effect was most significant in the groups of low and intermediate freezability, not being as significant in the group of high freezability. We can conclud from this study that treatment with cyclodextrin loaded with cholesterol contributes to better preservation of sperm parameters in ram semen, especially in animals displaying low freezability, however there where no diference in the high freezability group.

Poly (ethylene glycol)/ β - cyclodextrin crystalline inclusion complexes

The slightly water-soluble β-CD and its crystalline inclusion complexes with different molecular weights of poly( ethylene glycol), abbreviated as PEG, were prepared and characterized. The results show that the β -CD forms a crystalline inclusion complex with PEG (MW 600) and PEG (MW 1500) in the solid state. In the solution state, 2D NMR spectroscopy, shows that the PEG is present in the cavity of the P-CD. Coherent cross peaks were observed in both 2D NMR NOESY and ROESY showing correlation between the inner and outer protons of β -CD with the repeating unit of PEG. Scanning electron microscopy (SEM) shows the formation water soluble crystalline complexes between the β -CD with the amorphous PEG (MW 600) and PEG (MW 1500). Crystal formation was supported by wide angle X-Ray studies, W AXD. W AXD patterns for the β-CD/PEG crystalline complexes show new peaks indicative of formation of structures different from the crystalline β-CD.

Avaliação de complexos de inclusão na inibição do aço carbono em meio ácido / Evaluation of inclusion complexes in the inhibition of carbon steel in acid medium

Fundação Carlos Chagas Filho de Amparo a Pesquisa do Estado do Rio de Janeiro / A indústria de petróleo e gás apresenta sérios problemas relacionados à corrosão. Nas petroquímicas e nas instalações de refino de petróleo, as falhas em materiais estão relacionadas com a corrosão, além disso, os processos de corrosão de metais são problemas no mundo, causando ônus em processos industriais e gerando situações de risco como a corrosão de pilares metálicos em pontes ou em fuselagens de aviões. Dentre os meios corrosivos, um muito comum é o meio ácido, que será o meio estudado neste trabalho. Com todos os problemas citados, se faz necessário o estudo de inibidores de corrosão com alta eficiência, estabilidade e que, preferencialmente, não agridam o meio ambiente. Alguns inibidores de corrosão apresentam solubilidade limitada em água, sendo necessária a utilização de alguns solventes não tão amigáveis ao meio ambiente. Então, propôs-se formar um complexo de inclusão para tornar possível a solubilização de inibidores convencionais em solução aquosa, como algumas tioureias, que apresentam solubilidade limitada em água. Essa completa solubilização do inibidor de corrosão orgânico em meio aquoso é possível com a utilização de um aditivo (hospedeiro) capaz de encapsular tais moléculas (convidados) via interações não covalentes, de modo a alcançar o máximo desempenho de inibição. Para a formação do complexo de inclusão foi usado como molécula hospedeira, a α e a β hidroxipropilciclodextrina e como molécula convidada a dibenziltioureia. A intenção foi testar a melhora da ação inibidora da corrosão do aço carbono em meio de HCl 1mol.L-1 com ensaios de perda de massa, de impedância, polarização e microscopia eletrônica de varredura (MEV), além de evidenciar a formação destes complexos de inclusão através da espectroscopia de absorção vibracional no infravermelho, espectroscopia de RMN de 1H, espectroscopia de absorção no Ultra-violeta e análise térmica diferencial (DTA) / The oil and gas industry presents serious corrosion problems. In the petrochemical and oil refining plants, the flaws in materials is related to corrosion, in addition, metal etching processes are problems in the world, causing burdens on manufacturing processes and resulting hazards such as corrosion of metal pillars on bridges or aircraft bodies. Among the corrosive media, a very common is the acidic, which will be the medium studied in this work. With all these problems, it is necessary to study corrosion inhibitors with high efficiency, stability and, preferably, that is environmentally benign.. Some corrosion inhibitors have limited solubility in water, necessitating the use of organic solvents Then, it was proposed the use of an inclusion complex for making possible to dissolve conventional inhibitors in aqueous solution, as some thiourea, which have such limited solubility in water. The complete dissolution of organic corrosion inhibitor in aqueous media would be possible with the use of an additive (host) capable of encapsulating such molecules (guests) via non-covalent interactions in order to reach the maximum inhibition performance. To form the inclusion complex it was used as the host, the α and β hydroxypropyl-cyclodextrin and as the guest to dibenziltioureia. The intention was to test the improvement of the inhibitory performance of carbon steel corrosion in HCl medium 1molL-1 with mass loss test, impedance, polarization and scanning electron microscopy (SEM), and evidence the formation of these inclusion complexes by absorption vibrational infrared spectroscopy, 1 H NMR spectroscopy, spectroscopy of absorption UV and differential thermal analysis (DTA)

As comparações foram realizadas por análise de variância (ANOVA) em arranjo fatorial 3X3 (Tratamento CLC+CHO, CON e in natura X grupos alta, intermediária e baixa congelabilidade), as comparações entre os grupos foram analisadas pelo PROC MIXED do SAS e o efeito de grupo foi detectado pelo teste de Tukey (p <0,05) ou pela estatística não paramétrica de ordem (Kruskal-Wallis), quando era necessário. No 1o experimento o tratamento CLC+CHO se mostrou mais eficaz em preservar os parâmetros de motilidade, integridade de membranas e potencial mitocondrial quando comparado aos demais tratamentos. O grupo CLCP mostrou queda nos parâmetros de motilidade, integridade de membrana, potencial mitocondrial, mostrando, com isso, uma queda na preservação espermática. No 2o experimento observou-se que o tratamento CLC+CHO mostrou maior grau de preservação para motilidade total e progressiva, células rápidas, preservação de membranas plasmática e acrossomal e potencial mitocondrial quando comparado ao CON. Esse efeito teve maior significância nos grupos de baixa e intermediária congelabilidade, não sendo tão expressivo no grupo de alta. Pode-se concluir com esse estudo que o tratamento com ciclodextrina acrescida de colesterol contribui para uma melhor preservação dos parâmetros espermáticos do sêmen ovino, principalmente em animais apresentando baixa congelabilidade, contudo não apresenta diferença em ejaculados de alta congelabilidade. / Frozen ram semen has been the subject of many researches, mainly due to the difficulty of cervical transposition during artificial insemination. However, intra-cervical insemination in sheep is often used, resulting in reduced fertility with the use of frozen semen. To this end, this study was divided into two experiments. In the first experiment was verified the potential of cyclodextrin loaded with cholesterol as an additive to the extender in the protection of sperm kinetics, integrity of plasmatic and acrosomal membranes, mitochondrial function, sperm capacitation and production of reactive oxygen species (ROS) in cryopreserved sheep sperm. Five ejaculates from six rams (n = 30) were divided into three treatments: only extender (CON); extender + cyclodextrin loaded with cholesterol (CLC + CHO); and extender + pure cyclodextrin (CLCP). After dilution (50x106 spermatozoa/mL), semen was stored in straws, identified and cryopreserved using an automated system. Two straws from each ejaculate and treatment were thawed (at 37° C for 30 seconds) and analyzed for motility (CASA), morphology of spermatozoa (DIC), plasmatic (PI-H342) and acrosomal (FITC-PSA) membrane integrity, mitochondrial potential (JC-1), production of free radicals (CellRox), lipid peroxidation (BODIPY) and cell membrane permeability (Merocyanine 540). Comparisons between groups were analyzed using PROC MIXED of SAS and the effect of group was detected by Tukey (p <0.05) or by the order of nonparametric statistics (Kruskal-Wallis) test, when necessary. In the 2nd experiment the same ejaculates were divided by freezability based on the total post-thaw motility (TPTM) and the percentage of reduction in total motility (%RTM) when compared fresh and post-thaw semen (only extender), and divided into the groups: high (TPTM ≥ 60% and %RTM ≤ 40%), intermediate (60%> TPTM > 40% and 60%> %RTM > 40%) and low (TPTM ≤ 40% and %RTM ≥ 60%) freezability. CON and CLC + CHO treatments were analyzed within each animal and each group for motility (CASA), plasmatic (PI-H342) and acrosomal (FITC-PSA) membrane integrity, mitochondrial potential (JC-1), lipid peroxidation (BODIPY) and cell membrane permeability (Merocyanine 540). Comparisons were performed by analysis of variance (ANOVA) with a factorial arrangement 3x3 (groups CLC+CHO, CON and fresh X groups high, medium and low freezability), comparisons between groups were analyzed using PROC MIXED of SAS and the group effect was detected by the Tukey test (p < 0.05) or no statistical order parametric (Kruskal-Wallis), when necessary. In the first experimente, CLC + CHO treatment was more effective in preserving the parameters of motility, membrane integrity, and mitochondrial potential compared to other treatments. The CLCP group showed a fall in the parameters of motility, membrane integrity, mitochondrial potential, showing thereby a decrease in sperm preservation. In the 2nd experiment, it was observed that the treatment CLC + CHO showed greater preservation for total and progressive motility, rapid cell preservation, plasmatic and acrosomal membranes and mitochondrial potential when compared to CON. This effect was most significant in the groups of low and intermediate freezability, not being as significant in the group of high freezability. We can conclud from this study that treatment with cyclodextrin loaded with cholesterol contributes to better preservation of sperm parameters in ram semen, especially in animals displaying low freezability, however there where no diference in the high freezability group.

Molecular Dynamics Simulations of Biomimetic Carbohydrate Materials

The present thesis honors contemporary molecular dynamics simulation methodologies which provide powerful means to predict data, interpret observations and widen our understanding of the dynamics, structures and interactions of carbohydrate systems. With this as starting point my thesis work embarked on several cutting edge problems summarized as follows. In my first work the thermal response in crystal cellulose Iβ was studied with special emphasis on the temperature dependence of the crystal unit cell parameters and the organization of the hydrogen bonding network. The favorable comparison with available experimental data, like the phase transition temperature, the X-ray diffraction crystal structures of cellulose Iβ at room and high temperatures, and temperature dependent IR spectra supported our conclusions on the good performance of the GLYCAM06 force field for the description of cellulose crystals, and that a cautious parameterization of the non-bonded interaction terms in a force field is critical for the correct prediction of the thermal response in cellulose crystals. The adsorption properties of xyloglucans on the cellulose Iβ surface were investigated in my second paper. In our simulations, the interaction energies between xyloglucan and cellulose in water were found to be considerably lower than those in vacuo. The van der Waals interactions played a prevailing role over the electrostatic interactions in the adsorption. Though the variation in one side chain did not have much influence on the interaction energy and the binding affinity, it did affect the structural properties of the adsorbed xyloglucans. The interaction of the tetradecasaccharide XXXGXXXG in complex with the hybrid aspen xyloglucan endo-transglycosylase PttXET16-34 was studied in the third paper. The effect of the charge state of the “nucleophile helper” residue Asp87 on the PttXET16-34 active site structure was emphasized. The results indicate that the catalysis is optimal when the catalytic nucleophile is deprotonated, while the “helper” residue and general acid/base residue are both protonated. In my forth paper, the working mechanism for a redox-responsive bistable [2]rotaxane based on an α-cyclodextrin ring was investigated. The umbrella sampling technique was employed to calculate the free energy profiles for the shuttling motion of the α-cyclodextrin ring between two recognition sites on the dumbbell of the rotaxane. The calculated free energy profiles verified the binding preferences observed experimentally. The driving force for the shuttling movement of the α-cyclodextrin ring was revealed by the analysis of the free energy components. / QC 20110513

Assessment of the Antiprotozoal Activity of some Tubulin Inhibitors Following Cyclodextrin Complexation.

The purpose of the present study was to evaluate the potential usefulness of tubulin inhibitors when complexed with hydroxypropyl-â-cyclodextrin (ÇPâCD) against a range of protozoan parasites. This approach involved investigations into the complexation of these drugs with ÇPâCD, and subsequent investigations of these drugs and their complexes in regard to cytotoxicity, pharmacokinetics, in vitro efficacy against Giardia, Cryptosporidium and rodent malaria (Plasmodium chabaudi), and their in vivo efficacy against Giardia and malaria. Albendazole (ABZ) is a benzimidazole carbamate with a broad anti-parasite spectrum, while the dinitroanilines trifluralin (TF) and oryzalin (OZ) have recently been found to exhibit activity against certain parasites. All three compounds are microtubule antagonists in either nematodes or weeds and have poor aqueous solubility, with the solubility of ABZ and OZ dependent on pH. Cyclodextrins (CD) have a hydrophobic cavity that allows them to form inclusion complexes with hydrophobic drugs, resulting in increased drug aqueous solubility, and often, improved drug dissolution and bioavailability. Thus the complexation of these drugs with ÇPâCD was investigated. All three compounds exhibited type AL phase solubility diagrams with ÇPâCD complexation, with additional increases in ABZ and OZ solubility achieved through the manipulation of temperature and pH. OZ displayed a stronger interaction with ÇPâCD when ionised over its neutral form. However, insufficient concentrations of the TF/ÇPâCD complex were achieved for drug efficacy studies. The cytotoxicity of the drugs and their complexes was assessed using the assay kit Cytotox 96 with human carcinoma cells. This is a colourimetric assay that measures lactate dehydrogenase release as a consequence of compromised cellular and membrane integrity. Both ABZ and OZ are cytotoxic to rapidly proliferating and differentiating cells but are not cytotoxic to cells in the stationary phase. Complexation did not affect drug cytotoxicity. In pharmacokinetic studies, complexation improved ABZ (and metabolites) bioavailability, but had no significant affect on OZ bioavailability. In vitro drug assessment studies found ABZ to be highly effective against Giardia, and effectiveagainst Cryptosporidium and malaria. OZ on the other hand exhibited no activity against Giardia, but was effective against Cryptosporidium and malaria. Complexation did not improve the antiprotozoal efficacy of either ABZ or OZ. In particular, excess ÇPâCD decreased the antigiardial effects of ABZ, possibly due to competitive complex formation. In addition, complexation did not improve the antiprotozoal effects of ABZ in vivo. However, the cytotoxic effect of the ABZ/ÇPâCD complex was more evident in the treatment of malaria in vivo, resulting in increased anaemia and suppression in weight gain, due to the improved bioavailability of ABZ and metabolites. ÇPâCD alone was found to be cytotoxic at greater than 2.5%, and inhibited Giardia both in vitro and in vivo at greater than 1% and 2% respectively. This was attributed to membrane disruption caused by the dissolution and removal of membrane components. In comparison, malaria grew better in the presence of ÇPâCD in vitro, with no detrimental effect observed at up to 8% ÇPâCD. This was attributed to either the increased solubilization of a necessary media component, or the complexation and removal of an inhibitory compound from the cultivation medium. Therefore ÇPâCD complexation did not improve the antiprotozoal activity of the tubulin antagonists ABZ and OZ. However, the results of the pharmacokinetic studies suggest that anthelmintic activity of ABZ, particularly against systemic infections, may be improved with oral administration of the ABZ/ÇPâCD complex. In addition, the antiparasitic activity of ÇPâCD alone may be promising, especially against intestinal infections. Finally, the improved in vitro cultivation of P. chabaudi in the presence of ÇPâCD presents a promising approach to its potential long term cultivation.
