Stratégies chromatographiques en phase liquide et supercritique pour l'analyse de candidats médicaments / Liquid and supercritical chromatographic strategies for analysis of drug candidates

Le profilage d’impuretés de candidats médicaments est une préoccupation majeure des industries pharmaceutiques. L’identification et la quantification des impuretés doivent être strictement contrôlées pour assurer l’efficacité et la toxicité limitée du principe actif. Il est donc nécessaire de disposer de méthodes analytiques performantes afin de s’assurer que l’ensemble des impuretés est identifié. L’HPLC phase inverse sur phase C18 reste aujourd’hui la méthode de choix pour cette tâche. Cependant, il arrive que cette méthode échoue, notamment lorsque le principe actif n’est pas suffisamment retenu sur la colonne ou que les impuretés ne sont pas parfaitement séparées du composé principal. Il est alors essentiel de pouvoir se tourner vers des méthodes analytiques alternatives et complémentaires.Ce travail de recherche traite du développement et de l’évaluation de méthodes analytiques alternatives à l’HPLC phase inverse sur phase C18 pour le profilage d’impuretés de principes actifs pharmaceutiques. L’HPLC phase inverse sur d’autres phases stationnaires, l’HPLC mixed-mode ainsi que la SFC ont été explorées et leurs performances chromatographiques comparées. La comparaison et l’étude des différentes méthodes ont permis de proposer une stratégie d’analyse du candidat médicament. / Impurity profiling of drug candidates is a significant concern of pharmaceutical industries. The identification and quantification of impurities must be strictly controlled to ensure the efficacy and limited toxicity of the active ingredient. It is therefore necessary to have efficient analytical methods to ensure that all impurities are identified. Today, reversed-phase HPLC with C18 column remains the method of choice for this task. However, this method sometimes fails, particularly when the active pharmaceutical ingredient is not sufficiently retained on the column or when the impurities are not resolved from the main compound. It is therefore essential to turn to alternative and complementary analytical methods.This work deals with the development and evaluation of alternative analytical methods to reversed-phase HPLC on C18 phase for impurity profiling of pharmaceuticals. Reversed-phase HPLC on other stationary phases, mixed-mode HPLC as well as SFC were explored and their chromatographic performances compared. The comparison and the study of the different methods allowed proposing a strategy of analysis of the drug candidate.

Les microplastiques et leurs additifs dans les produits de la pêche : développements méthodologiques et prévalence / Microplastics and their additives in seafood products : methodological developments and prevalence

L'augmentation de la production de matière plastique depuis les années 1950 combinée à une mauvaise gestion des déchets plastiques ainsi qu'aux mauvais comportements des citoyens conduit à la contamination des écosystèmes terrestres comme marins. L'ingestion par des organismes marins de particules plastiques mesurant moins de 5 mm, appelées microplastiques, a été décrite. L'objectif de ces travaux de thèse était d'évaluer le danger "microplastique" pour le consommateur de produits de la pêche. Afin de répondre à cet objectif, la sélection et la caractérisation d'une méthode permettant de digérer les tissus des produits de la pêche a été. En parallèle, une méthode de Pyrolyse-GC/MS a été optimisée afin d'identifier les microplastiques les plus petits possibles. Ses limites d'applications ont été calculées puis la méthode a été appliquée sur des microplastiques provenant de différents produits de la pêche. Ces deux méthodes ont été appliquées dans la recherche de microplastiques dans deux espèces de bivalves et trois espèces de poissons. Pour les bivalves, la contamination aux microplastiques était comprise entre 0.15 et 0.74 MP/g de chair humide alors que pour les poissons la contamination était assez faible : 0,01 MP/g de tractus gastro-intestinal et 0,001 MP/g de filet. Pour finir, une étude exploratoire de la lixiviation d'un additif plastique en utilisant un modèle de digestion in vitro a été entreprise. Globalement, ces travaux de thèse permettent de proposer des voies d'harmonisation des méthodes pour l'étude des microplastiques dans les produits de la pêche pour, in fine, évaluer le danger pour le consommateur. / Increase in plastic production since the 1950's in combination with plastic waste mismanagement lead to the presence of plastic particle in terrestrial and marine environment. Plastic particles measuring less than 5 mm, called microplastics, are ingested by a wide range of organisms. The goal of this PhD thesis was to evaluate the microplastic hazard for seafood consumers. firstly, method for digesting seafood tissue without degrading plastic polymer was selected and applications limits were determined for digestion of fish digestive tracts. Concurrently, a Pyrolysis-GC/MS method was optimized, limit of detection were determined and this method was applied on environmental samples. Both methods were used to study microplastics contamination in bivalve and fish species used for human consumption. Bivalve contamination was between 0.15 and 0.74 MP/g of tissue wet weight whereas contaminations in fish were low. Indeed, digestive tracts were contaminated by in mean 0.01 MP/g of tissue wet weight and muscles were contaminated by in mean 0.001 MP/g of tissue wet weight. Lately, leaching of a plastic additive was studied using an in vitro enzymatic model of digestion. However, such phenomenon was not highlighted. Overall, these results will help to standardize the study of microplastics in seafood products.

Développement de méthodes analytiques pour la caractérisaton de lots industriels du nitroxyde SG1 / Development of analytical methods for the characterization of industrial batches of SG1-Nitroxide

Ce travail de thèse s’inscrit dans un contexte industriel. Il consiste en la caractérisation de lots bruts de SG1 : le N-tert-butyl-N- (1-diethylphosphono-2,2-dimethylpropyl)-N-oxyle. La spécificité de cette espèce est son caractère radicalaire stable. La première partie de ce travail consistait à développer une méthode d’analyse, facilement applicable en laboratoire d’analyse de contrôle sur site industriel pour la caractérisation de la pureté en SG1 dans des lots synthétisés. La méthode choisie a été développée par chromatographie liquide haute performance (HPLC). Pour ce faire, un étalon de SG1 a été préparé par purification de SG1 brut industriel en utilisant la technique séparative chromatographique de partage centrifuge (CPC). L’identification des impuretés suspectées majoritaires dans les lots de SG1 bruts a ensuite été réalisée par spectroscopie RMN ou par l’étude des fragments obtenus par spectrométrie de masse en tandem par ionisation electrospray. A l’aide de ces techniques, quinze impuretés majori- taires ont été identifiées. Des méthodes de quantification ont ensuite été développées pour douze de ces impuretés par diverses techniques analytiques chromatographiques : par HPLC-UV, HPLC-MS ou GC-MS ou par des techniques spectroscopiques Infra-Rouge ou encore par conductimétrie. / This PhD study has been carried out within an industrial context. It deals with the characterization of industrial batches of N-tert-butyl-N- (1-diethylphosphono-2,2-dimethylpropyl)-N-oxyle also called SG1. The distinctive feature of this molecule lies in its specific stability which is uncommon for radical species. The first part of this study consists in developping an analytical method, easy to implement in a QC laboratory for the determination of SG1 purity. The chosen analytical method has been developped by High Performances Liquid Chromatography (HPLC). This method requires the use of a standard sample of SG1, which has been obtained after purification of industrial batches of SG1 by the use of an innovative separative technology, the Centrifugal Partition Chromatography (CPC). The identification of the impurities, suspected as the major impurities in the industrial batches of SG1, has been carried out by NMR spectroscopy or by the study of their fragmentation observed by Tandem Mass Spectrometry – Electrospray Ionization. The use of these techniques enabled the identification of fifteen major impurities. Quantification methods by various techniques (HPLC-UV, HPLC-MS, GC-MS, other direct spectroscopic techniques like Infra-Red or other types of techniques like conductimetry) have been developped for twelve impurities.

Multi-Orbital Physics in Materials with Strong Electronic Correlations : Hund's Coupling and Inter-Shell Interactions / Physique multi-orbitalaire dans les matériaux corrélés : Couplage de Hund et interactions inter-couches

Les matériaux corrélés offrent une richesse de nouveaux phénomènes, dont beaucoup ne sont pas encore - ou seulement partiellement - compris. Au centre de cette thèse sont des modèles multi-orbitalaires que j'etudie à travers une palette de méthodes, dont la théorie du champ moyen dynamique. Dans le modèle de Hubbard multi-orbitalaire proche de la transition de Mott, je mets en évidence un régime de mauvais métal induit par le couplage de Hund. Les propriétés de la transition de Mott dans ce système sont analysées. Dans un deuxèime temps, je traite un modèle élargi pour inclure des degrés de liberté des ligands dans les oxydes. Plus spécifiquement, cette thèse étudie les effets des interactions inter-couches entre orbitales corrélés d'un atome de métal de transition et les orbitales p des ligands. Une partie du travail est dédiée au développement de nouvelles méthodes dont une approche de rotateurs esclaves à ce problème. Le dernier chapitre concerne le domaine de la spintronique moléculaire, où j'étudie la physique du "spin-state switching" en fonction de l'hybridation d'un ion de métal de transition avec ses ligands dans les molecules organométalliques du type porphyrine de nickel. / The physics of correlated materials offers a wealth of new phenomena, many of which are not yet - or only partially - understood. In this thesis, we focus on multi-orbital models, which we study using various methods, including dynamical mean-field theory. We show that in the multi-orbital Hubbard model close to the Mott transition, Hund's coupling gives rise to a bad metal regime the properties of which we analyze. Furthermore, we consider a more general class of models that include oxygen ligands. More specifically, we study the effect of inter-shell interactions between correlated metal- and ligand p-orbitals. In this context, we develop and test a new slave-rotor approach to treat such interactions in an effective manner. The final chapter constitutes an excursion to the field of molecular spintronics, where we study the physics of the hybridization-induced spin-state switching in organometallic nickel porphyrin molecules.
