Morphological differentiation of eggs and comparative efficacy of oviposition and gravid traps for Aedes vectors at different habitats

Mosquito surveillance is an integral part of understanding mosquito-borne disease, including and not limited to the La Crosse virus. The primary container-inhabiting Aedeni vectors include Aedes triseritatus, Aedes albopictus, and Aedes japonicus. To better understand the efficacy of gravid traps and oviposition traps as surveillance methods for these mosquitoes, field studies were conducted in three different habitat types. Ae. triseriatus, Ae. albopictus, Ae. japonicus, and Aedes aegypti eggs were described with the aid of color images and SEM. All eggs were broadly cigar shaped with Ae. triseriatus and Ae. japonicus eggs being dull or matte black while Ae. albopictus and Ae. aegypti eggs were shiny jet black. Ae. triseriatus eggs were larger, lighter in color, and have a rougher appearance when compared to Ae. japonicus. Ae. albopictus and Ae. aegypti can be differentiated by the distinct presence of a micropylar collars in Ae. aegypti. Ovitrap and gravid trap efficacy for the surveillance of Ae. triseriatus, Ae. albopictus, and Ae. japonicus were measured in three different habitats on two different sites. Both sites contained the same 2ha habitats with varying degrees of forest canopy disturbance. Ae. triseriatus was the most abundant mosquito on all sites. Ovitraps and gravid traps were efficient in collecting Ae. triseriatus, while ovitraps were not efficient in collecting Ae. japonicus and Ae. albopictus. / Master of Science in Life Sciences

Characterizing the Molecular Changes of Austrofundulus limnaeus As It Develops Towards and Enters Diapause II

Austrofundulus limnaeus is a species of annual killifish which inhabits ephemeral ponds in South America. The species is able to survive seasonally desiccating ponds due to their ability to produce robust embryos. The embryos of this species are capable of entering a developmental arrest, termed diapause II, which precedes the onset of drought. While in this arrested state embryos exhibit the greatest tolerance to anoxia of any characterized vertebrate at 25ºC. Furthermore, when raised at 30ºC, embryos escape the entrance to diapause II and go on to develop directly. Currently, little is known about the molecular mechanisms which induce and maintain this developmentally arrested state. In this study I have developed methods to analyze changes in histone modifications in the context of diapause II. Histone modifications were chosen due to their extreme conservation and well characterized role as modulators of gene expression in other systems. Results utilizing adapted immunobased assays show significant changes in the global amount of H3S10P, H3K27me and H3K4me, as the embryos progress from early embryogenesis through the exit of diapause. Additionally, it is revealed that there exists a degree of phenotypic plasticity with regards to the entrance into diapause II which is modulated by the environment (severe hypoxia 0.1% O2). This work builds a foundation for future histone modification studies and contributes the development of several tools to the field. This study contributes to a greater molecular understanding of the cue(s) which influence the remarkable phenomenon of obligate developmental arrest in a vertebrate embryo.

Insulin-like Growth Factor Pathway Described in <i>Austrofundulus limnaeus</i> Diapause and Escape Embryos

Development in the annual killifish Austrofundulus limnaeus can follow two distinct developmental trajectories. Typical development includes the entrance of embryos into a state of metabolic and developmental arrest termed diapause. Alternately, embryos can escape diapause and develop directly without pause. These two trajectories are characterized by differences in the rate and timing of developmental, morphological, and physiological traits. Insulin and Insulin-like growth factor (IGF) signaling (IIS) is known to regulate entrance into diapause in a variety of invertebrates. In this thesis I explore the possible role of IGFs in the regulation of development and diapause in embryos of A. limnaeus. Here I report stage-specific expression of IGF-I and II proteins and their associated mRNA transcripts. Patterns of IGF-I protein expression are consistent with IGF signaling playing a major role in supporting the escape trajectory. In addition, treatment of embryos with a potent inhibitor of the IGF-I receptor (IGF1R) mimics the diapause developmental pattern even under conditions that should favor direct development. Evaluation of mRNA gene expression patterns in the two developmental trajectories suggests a role for IGF-I signaling through the RAS-MAPK-ERK pathway, which may be promoting the escape phenotype. Additionally, IGF-I activity may be enhanced in escape trajectory embryos though upregulation of IGF binding protein 2 (IGFBP-2) mRNA. These data suggest a major role for IGF signaling in the promotion of the escape trajectory, and thus we predict that specific mechanisms are in place in diapause-bound embryos that block IGF signaling and thus promote entrance into diapause. The data presented here suggest that blocking IGF signaling is critical for induction of diapause, but also suggests that other signaling pathways are likely also at play. Other pathways such at the TGF-beta signaling molecules and SMAD pathway, may also be involved in the direct regulation of the diapause phenotype, as has been shown for other animal models of developmental arrest.

Regulation of Diapause Entry and Termination in the Swede Midge, Contarinia nasturtii (Diptera: Cecidomyiidae)

This thesis is an investigation of several aspects of diapause in the swede midge, Contarinia nasturtii (Kieffer). After developing methodology for induction and quantification of diapause entry in the laboratory, heritability of diapause entry was assessed for sibling larvae reared under diapause-inducing conditions. The diapause-quantification technique was efficient, but diapause frequencies were highly variable across studies employing similar diapause-inducing conditions. The diapause entry trait may be weakly heritable. A field study was conducted on diapause entry and emergence patterns. Diapause frequency was inversely correlated with photoperiod and absolute maximum air temperature. Photoperiod did not influence emergence timing. Two emergence phenotypes were observed before mid-July and a third minor emergence phenotype may exist in mid-to-late August. Approximately one third of emergence in the field was attributed to larvae in diapause for two or more years. Isolation and identification of genes expressed during diapause in horn fly, Haematobia irritans (L.)

There is a discrepancy in the current literature concerning the stage of development in which horn flies arrest during pupal diapause. A study was therefore conducted to describe the morphologies of horn fly pupae and its central nervous system (CNS) throughout nondiapause pupal development and diapause. Morphologies of diapausing pupae and CNS indicated that developmental arrest occured early in pupal development during the interval between head eversion and pupal-adult apolysis. Morphological descriptions are necessary for defining compariable tissues between nondiapausing insects and diapausing insects. These tissues can then be used for molecular differential analysis to determine genes specific to either diapause or nondiapause. One such differential analysis technique, subtractive hybridization, was used to isolate putative diapause up-regulated genes from the horn fly. Seven different cDNAs were closed and partially sequenced. Comparisons of the cDNA sequences with known DNA and protein sequences indicated homology with transferrin, cytochrome oxidase I, Kunitz family serine protease inhibitor, tyrosine hydroxylase (TH), and carboxylensterase. Two cDNAs did not have homology to entries in DNA and protein databases. Northern blot analyses were used to study expression of each gene by probing total RNA extracted from whole pupae throughout nondiapause pupal development and diapause. Expression of TH was also determined in total RNA extracted from CNS tissue of nondiapausing and diapausing pupae. Cytochrome oxidase was equally expressed in nondiapause and diapause destined pupae, and therefore not considered to be a diapause up-regualted gene. Expression patterns differed slightly for each of the remaining clones; however, expression tended to be highest in diapause destined pupae during pupation compared to nondiapausing pupae. These genes and their products are involved in many aspects of insect phsiology including metamorphosis, melanization and sclerotization of the puparium and cellular defense.
