Growth and yield responses of cowpeas (Vigna unguiculata L.) to water stress and defoliation.

Cowpea (Vigna unguiculata L.) is an important legume, especially in the hot, dry tropics and subtropics of sub-Saharan Africa. It has been widely reported to be drought tolerant. Cowpea is a highly nutritious, multi-purpose crop, used as a leafy vegetable and grain legume with potential to contribute to food security in marginal areas. However, the crop is still classified as a neglected underutilised species; legume research focus has been mainly devoted to established legumes such as common bean and soybeans. There is a need to collect empirical information on cowpea which could be used to advise farmers on management strategies. This study evaluated cowpea responses to water stress under controlled and field conditions. Initially, two cowpea varieties (Brown and White birch) were evaluated for seed quality using the standard germination that was laid out in a completely randomised design and each variety was replicated for times. Electrolyte conductivity test was also performed under laboratory conditions. Thereafter, a pot trial was conducted to evaluate cowpea response to water stress imposed at different growth stages under varying growth temperatures. The pot trial comprised three factors: temperature [High (33/27ºC), Optimum (27/21ºC) and Low (21/15ºC)], water regimes (no stress, terminal stress, intermittent stress – vegetative and intermittent stress - flowering) and cowpea varieties. Lastly, a field trial was conducted to evaluate cowpea production as well as the effect of sequential leaf harvesting on yield under irrigated and rainfed conditions. The field trial was laid out as a split-plot design, with water regime (irrigation vs. rainfed) as main factors, cowpea varieties as sub-factor and sequential harvesting (no harvest, harvested once and harvested twice), replicated three times. All treatments were arranged in a randomised complete block design. Results of the initial study showed that germination capacity and vigour of cowpea varieties were significantly different (P < 0.001). White birch had higher electrolyte leakage than Brown birch. Pot trial results showed that cowpea growth (leaf area, leaf number and plant height) was vigorous in the high temperature regime compared with optimum and low temperature regimes. Chlorophyll content index was higher under high temperature relative to optimum and low temperature regimes, respectively. Under low and optimum temperature regimes, cowpea growth was stunted; cowpea failed to flower and form yield. Whereas, under high temperature regime, cowpea growth was vigorous hence flowered and formed yield. Vegetative growth was more sensitive to water stress than flowering stage. Terminal stress and stress imposed during flowering resulted in increased proline accumulation relative to no stress and stress imposed during vegetative growth. Harvest index was lower when water stress was imposed during vegetative relative to flowering stage. Field trial results showed that cowpea growth was sensitive to water stress. Plant height, leaf number, chlorophyll content index and stomatal conductance were lower under rainfed relative to irrigated conditions. Sequential harvesting of leaves had no significant effect on cowpea yield. It is concluded that tropical temperature conditions are most suitable for cowpea production; the controlled environment study showed best crop performance under 33/27ºC. In the context of varieties used for the present study, vegetative growth was the most sensitive stage to water stress. Cowpea performed better under rainfed relative to irrigated conditions with respect to yield formation. Low temperature was found to be more limiting to cowpea growth, development and productivity compared with water stress. Whereas, under high temperature conditions, water stress was more limiting to plant growth and productivity. Methyl jasmonate and salicylic acid enhance chilling tolerance in lemon (citrus limon) fruit.

South African ‘Eureka’ lemon fruit must be exposed to chilling temperatures (± 0.6°C) as a mandatory quarantine treatment against insect pests for all its overseas markets. Chilling lemon fruit at such temperatures may develop chilling injury (CI) symptoms on the flavedo. This negative effect on fruit quality reduces fruit marketability. This study evaluated postharvest factors influencing physiological, biochemical and ultra-structural mechanisms involved in alleviating CI in lemon fruit. It was hypothesised that treatment with methyl jasmonate (MJ) and salicylic acid (SA) may enhance chilling tolerance in lemon fruit by maintaining cellular integrity and inducing synthesis of enzymatic and non-enzymatic antioxidants. Furthermore, fruit susceptibility to CI was associated with the source of fruit. Lemon fruit were harvested from three locations representative of moderate subtropical, warm temperate and cool subtropical environments. Harvested fruit were treated either with 10 μM MJ, 2 mM SA or 10 μM MJ plus 2 mM SA, stored either at -0.5, 2 or 4.5°C for 0, 7, 14, 21, or 28 days and afterwards transferred to 23°C for a week as shelf-life simulation. Thereafter, fruit were evaluated for alterations in physiological, biochemical and ultra-structural features involved in the manifestation of CI symptoms. Chilling damage was more severe in untreated lemon fruit than in treated lemon fruit. Storing lemon fruit at 4.5°C accelerated the manifestation of CI symptoms more so than at 2°C while storage at -0.5°C delayed the manifestation of CI symptoms. Lemon fruit of moderate subtropical origin were more chilling-tolerant than lemon fruit of warm temperate and cool subtropical origin. Treatment with 10 μM MJ plus 2 mM SA significantly (P < 0.05) improved chilling tolerance in lemon fruit. This treatment effectively maintained membrane integrity, thereby retarding electrolyte leakage and membrane lipid peroxidation as well as mass loss and respiration rate. Treatment with 10 μM MJ plus 2 mM SA was also effective in enhancing the antioxidant concentrations of vitamin E and carotenoids. The production of these antioxidants could have been part of a defence system against chilling damage, reducing CI and maintaining fruit quality. Treatment with 10 μM MJ plus 2 mM SA enhanced the concentration of compounds involved in chilling resistance, such as proline, soluble sugars, ascorbic acid and total phenolics as well as the enzyme phenylalanine ammonia-lyase (PAL). The enhancement of the defence mechanisms may have played a role in enhancing chilling tolerance in lemon fruit. The treatment also inhibited certain enzymes involved in tissue browning, such as peroxidase (POD) which might have contributed to delaying manifestation of symptoms. Polyphenol oxidase (PPO) was found to not be a good biochemical marker of the occurrence of CI. Treatment with 10 μM MJ plus 2 mM SA appeared to be able to enhance chilling tolerance in lemon fruit by maintaining the ultra-structure of the cuticle, cell wall integrity, cell membrane of parenchyma cells of the flavedo. This treatment also preserved the mineral nutrients of the flavedo (carbon, oxygen, phosphorus, potassium, calcium, magnesium, sulphur, sodium, silicon and aluminium) during cold storage. This could have played a role in protecting the fruit against chilling stress and maintaining fruit quality. Treatment with 10 μM MJ plus 2 mM SA reduced ROS production, while the activity of enzymatic antioxidants such as catalyse (CAT), ascorbate peroxidase (APX) and glutathione reductase (GR), and accumulation of essential proteins was enhanced. This increase in activity of enzymatic antioxidants and the presence of stress-responsive proteins in the lemon flavedo could have been directly involved in enhancing chilling tolerance. The CI symptoms were accompanied by an increase in membrane permeability, membrane lipid peroxidation as well as phospholipase D (PLD) and lipoxygenase (LOX) activity; however, treatment with 10 μM MJ plus 2 mM SA effectively reduced the membrane permeability, membrane lipid peroxidation, and PLD and LOX activity induced by the cold treatment. This could have contributed to the efficacy of 10 μM MJ plus 2 mM SA in inhibiting the manifestation of CI symptoms. Treatment with 10 μM MJ plus 2 mM SA enhanced flavedo total antioxidant capacity measured by ferric reducing ability of plasma; 2,2-diphenyl-1-picrylhydrazyl; 2,2-azinobis (3-ethyl-benzothiazoline-6-sulfonic acid) and the oxygen radical absorption capacity assays. The enhancement of antioxidant capacity in lemon flavedo could have contributed to the fruit’s chilling tolerance. Therefore, the effect of 10 μM MJ plus 2 mM SA treatment, enhancing chilling tolerance, may be attributed to its ability to enhance enzymatic and non-enzymatic antioxidants; activate essential proteins and mitigate the effect of ROS accumulation. With the use of 10 μM MJ plus 2 mM SA treatments, the South African citrus industry will be able to meet the quarantine temperature requirements for exportation of lemon fruit whilst reducing economic losses, depending on the preharvest conditions experienced by the fruit in each shipment. / Thesis (Ph.D.)-University of KwaZulu-Natal, Pietermaritzburg, 2013.

Phenotypic plasticity in thermal tolerance : life history strategy of an invasive freshwater fish

Background: Temperature has a fundamental effect on organisms because it alters the speed of biochemical reactions and thus metabolism. This influence scales up to have ecosystem wide effects as the life history strategies of individual species differ in response to temperature. With the prospect of increasing global temperatures ecosystem functions could be interrupted. In order to predict the consequences of changing environmental conditions it is important to first establish how fitness related traits are affected by changing thermal conditions. Aims: The aim of this thesis was to develop a detailed understanding of the thermal niche of an invasive, tropical freshwater fish species. Methods: Using ecologically realistic conditions this thesis investigates the effect of environmental variation within and between generations on behavioural, growth, physiological, and reproductive characteristics of the Trinidadian guppy (Poecilia reticulata). Results: The results provide an insight into the fundamental thermal niche of a widely used model fish species as well as detailed measures of how thermal change alters phenotypic characteristics. Guppies are demonstrated to have a broad thermal tolerance and be phenotypically responsive to changing environmental conditions. The results also suggest that environmental characteristics of the guppy's habitat make an important contribution to the differences observed between populations of guppies in Trinidad. Conclusions: Water temperature in the guppy's natural environment varies widely over a daily cycle and I suggest that this is partly responsible for the guppy becoming phenotypically plastic and thermally tolerant. Furthermore, phenotypic flexibility is an important characteristic that will enable guppies to withstand some climate warming and continue to expand their invasive range poleward. Using experimental conditions which resemble those in the natural environment is important for developing accurate model parameters. These are necessary for predicting the ecosystem effects of environmental variation and for adaptive mitigation or pre-emptive management of range extensions by invasive species.

O objetivo geral desse trabalho foi o de avaliar o efeito de diferentes manejos de cortinas e sistemas de resfriamento na redução da temperatura do ar interna de ambientes protegidos. Esse trabalho foi realizado em ambientes protegidos com solo nu na área experimental do Departamento de Engenharia Rural, da Escola Superior de Agricultura "Luiz de Queiroz", da Universidade de São Paulo em Piracicaba, SP. Os ambientes protegidos foram instalados no sentido leste-oeste, com dimensões de 6,4 m de largura por 17,5 m de comprimento. A avaliação da distribuição espacial da temperatura do ar (em graus centígrados) no interior do ambiente protegido foi avaliada através da medição com termopares (cobre-constantã) instalados formando malhas, com espaçamento horizontal entre eles de 3,0m e nas alturas de 0,5m, 1,0m, 2,0m, 3,0m e 4,0m em relação ao solo, constituindo um conjunto de 78 pontos de amostragem. Os valores de temperatura do ar foram armazenados em um sistema de aquisição de dados. A nebulização em ambientes protegidos mostrou-se eficaz no processo de redução de temperaturas do ar durante o seu acionamento. A combinação do manejo de cortinas e nebulização foi o sistema mais eficiente na redução de temperatura do ar e pode reduzir a temperatura do ar no ambiente protegido a níveis muito próximos à temperatura do ar externa e em alguns casos a temperaturas abaixo da mesma. O efeito na redução de temperatura média do ar no ambiente protegido causado pela abertura da janela zenital foi maior com a menor abertura das cortinas laterais devido ao processo convectivo ou "efeito termo-sifão". A distribuição da temperatura média do ar no ambiente protegido com as cortinas laterais abertas a 3,0m de altura praticamente não foi modificada pela abertura da cortina zenital, principalmente quando a velocidade de vento externo era superior a 2,0m/s. A análise da distribuição da temperatura do ar por meio de isotermas permitiu uma visão global instantânea do processo de redução de temperatura do ar. Através das isotermas observou-se que a distribuição de temperatura do ar no ambiente protegido não é homogênea e a escolha do local ideal para instalação de sensores para medidas dos elementos meteorológicos é um aspecto importante a ser considerado. Nesse estudo, o sensor que correspondeu a medida de temperatura média do ar no ambiente protegido com as cortinas laterais fechadas estava localizado no centro do ambiente protegido, a 1,0m de altura em relação ao nível do solo. / The objective of this study was to evaluate different managements, such as fog and handling curtains, to reduce air temperature in a plastic greenhouse. The experiment was conducted in two plastic greenhouses installed in an East-West direction and located at the Department of Rural Engineering, "Escola Superior de Agricultura Luiz de Queiroz", University of São Paulo, Piracicaba, São Paulo State. The plastic greenhouses have a dimension of 6.4m width and 17.5m of length. The air temperature (°C) spatial distribution was measured using 78 thermopar sensors installed in a grid of 3.0m horizontally and five different high, 0.5, 1.0, 2.0, 3.0 and 4.0m. The sensors were connected to a data logger system and the data were stored there. The fog system showed a good efficiency to decrease the greenhouse air temperature. A combination of fog system and handling curtains was the most efficient management to decrease the plastic greenhouse air temperature. Due to the convection process, the opened of zenith curtains showed efficient to decrease the plastic greenhouse temperature only when the side curtains is not totally opened. The air temperature distribution was not modified when the side curtains were opened from 3.0m of the soil and the wind speed outside was higher than 2.0 m/s. Isotherms lines were determined and described the spatial distribution of plastic greenhouse air temperature. Through the isotherms lines were observed that the spatial distribution of plastic greenhouse air temperature was not homogeneous. Due to this fact, the places to install the sensors are an important factor to be considered. In this study, the sensor that showed the average value of greenhouse air temperature was located in the middle of the plastic greenhouse, and 1.0m of high.

Nutritional and environmental effects on triploid Atlantic salmon skeletal deformity, growth and smoltification

The Atlantic salmon (Salmo salar) is an iconic species that dominates the global finfish production sector with increasing market demand. The Scottish industry and government alone aspires for expansion of the sector to 210,000 t by 2020 with 154, 000 t produced in 2013. As such, there are pressures to improve sustainable development in particular to minimise the genetic impact of escapees on wild populations and reduce sea lice infection which are required for the granting of “green licenses” in Norway. The use of triploidy has been tested in the 1980’s with little success owing to suboptimal rearing conditions leading to elevated mortalities, poorer growth and a higher prevalence of deformities, in particular of the skeleton. Collectively: recent success of triploid trout farming, expansion to the salmon production sector and potential resulting pressure on wild stocks through escapee increases have reinstated interest to implement artificially induced triploid Atlantic salmon in commercial production. As diploid Atlantic salmon have undertaken extensive domestication to achieve the high quality production and welfare standards observed to date, triploid conspecifics too require husbandry optimisation to realise potential. In particular, industrialisation requires that higher observations of deformities and inconsistent growth trajectories during seawater ongrowing be resolved through optimisation of rearing regimes and subsequent standardization of husbandry protocols. Triploids possess additional genomic material and increased cell size yet reduced frequency that reflects known differences in physiology and supports that, in effect, triploids should be considered as a new species relative to diploid conspecifics. Therefore, this doctoral thesis aimed to study nutrition and temperature effects on triploid Atlantic salmon traits throughout the production cycle from ‘egg to plate’. Nutrition trials aimed to improve growth potential and mitigate skeletal deformities both in freshwater (FW) and saltwater (SW) whilst attempts were made to define a window of smoltification to ensure optimal ongrowing performance. Finally, impacts of embryonic temperature regimes that are known to impact long term performance and deformity development in triploids, were examined in relation to DNA regulation and yolk composition in an attempt to underpin potential mechanisms for the environmental impact of temperature on developmental phenotype. One of the main restrictions to triploid Atlantic salmon implementation is the increased prevalence and severity of skeletal deformities, particularly after the maring phase. The work performed in this thesis first demonstrated that protein and/or phosphorous (P) supplementation throughout SW ongrowing not only reduced the level of severely deformed (≥ 10 deformed vertebrae observable by x-radiography) individuals by 30 % but also sustained 6.8 % faster growth and improved harvest grade compared to triploids fed a standard grower diet (chapter 2). Comparison of x-radiography and severely deformed individuals between harvest and sea transfer highlighted that protein and P supplementation arrested deformity development whereas prevalence increased in triploids fed a standard grower diet. This implied that severe deformities were of FW origin and strongly suggest requirement for improved nutrition in FW to optimise SW performance. Therefore investigation of higher dietary P inclusion in FW was investigated and results showed significantly reduced number of deformed vertebrae and no severely deformed individuals in those fed 19.7 g total P Kg-1 compared with those fed 13.0 & 16.7 g total P Kg-1 (chapter 3). Most deformities were localised in the central (vertebrae 27 – 31) and caudal (vertebrae 52 – 57) regions for all treatments. However, triploids fed lower dietary P displayed a particular increase in prevalence within the tail region (vertebrae 32- 47) which is consistent with SW ongrowing reports and results from chapter 2, further highlighting FW origin of higher vertebral deformities reported in SW ongrowing in triploids. Higher P supplementation in FW also significantly improved growth in triploid parr compared to diploids and lower supplementation. However, this effect did not transpire in later FW smolt stages where weights were significantly higher in triploids fed lower compared to higher P supplementation. Expression of target genes involved in osteogenesis and bone P homeostasis in vertebrates were then analysed and a ploidy effect of osteogenic genes alp, igf1r and opn as well as a dietary effect on P homeostasis gene fgf23 was apparent in the parr stages but not smolt. In addition, stronger ploidy-diet effects were also observed in parr stages for whole body mineral concentrations. Collectively, growth, gene expression and whole body mineral content results indicate these earlier parr life stages may be more sensitive to P supplementation. This pronounced effect may be a consequence of seasonal accelerated growth associated with this period, where higher temperatures were also observed. The potential for shorter P supplementation windows in commercial production was addressed in chapter 4 with hope to cut economic cost to raw mineral inclusion in feed and also mitigate potential anthropogenic eutrophication on the environment that may be induced by P leached through uneaten feed and faeces. Triploids were fed higher dietary P (17.4 g total P Kg-1) until either early (5 g) or later (20 g) parr stages, or smolt (83 g) and monitored for performance throughout freshwater (FW) development. During later parr development (30 g), x-radiography assessment demonstrated that increased dietary P reduced the number of deformities and severely deformed individuals with no indication that feeding P for shorter windows improved skeletal integrity. Hence, P supplementation may be required throughout FW development for optimal skeletal performance. In addition, no differences in deformities were observed between triploid treatments at smolt. An effect of dietary P supplementation on whole body mineral concentration was observed in the early and later parr stages that was not as pronounced as smolt, which is consistent with results in chapter 3. Together, these results indicate that skeletal assessment during early developmental stages may not reflect smolt performance most likely as a consequence of seasonal effects of improved linear growth in the cooler winter temperatures prior to smolt where reversible deformities observed at parr may also be alleviated. In the same study (chapter 4), the inclusion of the probiotic Pediococcus acidilactici (Bactocell™) was also tested as a means to enhance gut assimilation as suggested in previous studies and therefore reduce the levels of P supplementation. Results clearly indicate superior skeletal performance in parr (30 g) as well as significantly less deformed vertebrae and no severely deformed individuals. However, at smolt (~83g), no effects of the dietary probiotic treatment were observed which may also be attributed to seasonal effects. Overall, nutritional research clearly indicate triploids require higher dietary P for optimal growth and skeletal development, which although is not consistent between life stages, is ultimately required throughout FW for optimal skeletal development at smolt. The use of probiotics offer a promising avenue for reduced P requirement in FW feed and further research should verify results and assess long-term performance. Timing of SW transfer according to correct parr-smolt transformation (PST) is essential for survival and growth performance in ongrowing where feeding and growth rate accelerate post-transfer. So far, SW transfer regimes and in particular the smoltification ‘window’ remains loosely defined in triploid Atlantic salmon and it is crucial that this be addressed to ensure optimal ongrowing survival and performance.

Effect of tree girdling, harvest time and ripening temperature on "hass" avocado fruit skin colour development during ripening

Thesis (M. Sc. (Horticulture)) -- University of Limpopo, 2018 / ‘Hass’ avocado fruit changes skin colour from green to purple and then black during ripening. However, markets importing South African avocado fruit have been complaining about the ‘Hass’ skin colour not changing to purple/black during ripening. Thus, the study aimed to investigate the effect of tree girdling, harvest time and ripening temperature on ‘Hass’ avocado fruit skin colour development during ripening. The mature ‘Hass’ avocado fruit were harvested from girdled and non-girdled trees during early (April), mid- (May) and late (June) harvest times. Upon arrival, in the laboratory fruit were cold stored at 5.5°C for 28 days. After storage, fruit were ripened at 25, 21 and 16°C for 8, 6 and 4 days, respectively. After withdrawal from clod storage fruit were evaluated for skin colour development, ripening and physiological disorders (chilling injury). Fruit from girdled trees showed high maturity (low moisture content) when compared with fruit from non-girdled trees during early and mid-harvest. With respect to skin colour development, the results indicate that skin eye colour development of fruit from girdled and non-girdled trees minimally increased from emerald green (1) to olive green (3) across all harvest times, ripening temperature and ripening duration. However, late season fruit from non-girdled trees improved to purple (4) when ripened at 21°C when compared with fruit from girdled trees. In terms of objective colour, lightness, hue angle and chroma decreased for fruit from girdled and non-girdled trees, across all harvest times, ripening temperature and ripening duration. Lightness and hue angle of fruit from girdled trees were slightly reduced when compared with fruit from non-girdled trees, throughout all harvest times, ripening temperature and duration. Early and mid-season fruit harvested from girdled trees showed rapid decrease of chroma when compared with fruit from non-girdled trees, throughout ripening temperature and x duration. In terms of softening, fruit from girdled trees showed higher firmness loss and ripening percentage within 6 (16°C) and 4 (21 and 25°C) days when compared with fruit from non-girdled trees during early and mid-harvest, whereas, late harvest fruit from girdled trees reached higher ripening percentage and firmness loss within 4 days throughout ripening temperatures. With respect to cold damage, late harvested fruit from girdled trees showed higher external chilling injury when compared with non-girdled trees, throughout ripening temperature. In general, girdling treatment improved fruit maturity, ripening rate and firmness loss. However, the incidence of variable skin colouring of ‘Hass’ avocado fruit during ripening was also prevalent in early harvested fruit from girdled tree, irrespective of ripening temperature. The mature ‘Hass’ avocado fruit were harvested from girdled and non-girdled trees during early (April), mid- (May) and late (June) harvest times. Upon arrival, in the laboratory fruit were cold stored at 5.5°C for 28 days. After storage, fruit were ripened at 25, 21 and 16°C for 8, 6 and 4 days, respectively. After withdrawal from clod storage fruit were evaluated for skin colour development, ripening and physiological disorders (chilling injury). Fruit from girdled trees showed high maturity (low moisture content) when compared with fruit from non-girdled trees during early and mid-harvest. With respect to skin colour development, the results indicate that skin eye colour development of fruit from girdled and non-girdled trees minimally increased from emerald green (1) to olive green (3) across all harvest times, ripening temperature and ripening duration. However, late season fruit from non-girdled trees improved to purple (4) when ripened at 21°C when compared with fruit from girdled trees. In terms of objective colour, lightness, hue angle and chroma decreased for fruit from girdled and non-girdled trees, across all harvest times, ripening temperature and ripening duration. Lightness and hue angle of fruit from girdled trees were slightly reduced when compared with fruit from non-girdled trees, throughout all harvest times, ripening temperature and duration. Early and mid-season fruit harvested from girdled trees showed rapid decrease of chroma when compared with fruit from non-girdled trees, throughout ripening temperature and x duration. In terms of softening, fruit from girdled trees showed higher firmness loss and ripening percentage within 6 (16°C) and 4 (21 and 25°C) days when compared with fruit from non-girdled trees during early and mid-harvest, whereas, late harvest fruit from girdled trees reached higher ripening percentage and firmness loss within 4 days throughout ripening temperatures. With respect to cold damage, late harvested fruit from girdled trees showed higher external chilling injury when compared with non-girdled trees, throughout ripening temperature. In general, girdling treatment improved fruit maturity, ripening rate and firmness loss. However, the incidence of variable skin colouring of ‘Hass’ avocado fruit during ripening was also prevalent in early harvested fruit from girdled tree, irrespective of ripening temperature.
