Exploring ruthenium dye synthesis and TiO2-dye-I-/I3- electron transfer reactions in a dye-sensitised solar cellChadwick, Nina January 2013 (has links)
Octahedral, six co-ordinate ruthenium complexes containing acid substituted polypyridyl ligands have proved particularly successful as dyes for Dye-Sensitised Solar Cells (DSSCs); thus there have been hundreds, if not thousands of these types of complexes synthesised and studied. Ruthenium dyes are now incorporated into commercial DSSCs; yet there is limited understanding of the interactions between the dye and the liquid I-/I3- electrolyte, which can both facilitate and hinder the generation of current and voltage within the cell. Monodentate –NCS ligands incorporated into ruthenium dyes appear to interact strongly with I- and I2 within the electrolyte. The analogous –NCO containing dye has therefore been synthesised to study the effect of the chalcogen atom on these interactions. Substituting –NCS for -NCO resulted in a significant change in the spectroscopic and electronic properties of the molecular dye, with destabilisation of the HOMO of the dye causing a red shift in dye absorption. Possibly due to this change in properties, the nature of the chalcogen atom was shown to have a significant impact on the performance of the dye in a DSSC. The effect of the nature of substituents on ancillary ligands of Ru(H2-dcbpy)(4,4`-Y2- bpy)(NCS)2 dyes on recombination across the TiO2 – electrolyte interface within the cell has been proven significant due to changes in the strength of binding of I2 to the substituent groups. However, few substituent groups have been investigated; therefore a series of halogenated dyes, where Y = Cl, Br, were synthesised. The effect of the nature of the halogen on dye recombination was significant, although the trends observed were not consistent with the reported data for iodine binding. Similar trends were observed for the analogous series, Ru(H2-dcbpy)(5,5’-Y2- bpy)(NCS)2 where Y = F, Cl, Br. By comparison of this series of dyes with the 4,4` dyes, it was discovered that the position of the substituent had a significant effect on the rate of recombination within the solar cell, as well as the electrochemical and spectroscopic properties of the dyes themselves. Such isomeric effects have not been previously reported. In the synthesis of these dyes, and in attempting the synthesis of five other ruthenium dyes, many barriers to efficient dye synthesis were discovered. Therefore, an investigation into the synthesis of ruthenium dyes has been conducted. By analysis of the breakdown products formed a number of avoidable side reactions, including decarboxylation and ruthenium catalysed nucleophilic substitution of the bpy ligands, were shown to occur. Problems associated with the high lability of the ruthenium centre at high temperatures have also been explored, and the use of UV/vis monitoring to aid optimisation of the reaction conditions was implemented. Thus, the development of two novel synthetic procedures allowed the synthesis of the dyes investigated during the course of this thesis.

Thesis (MScEng) -- Stellenbosch University , 2001. / ENGLISH ABSTRACT: The electrochemical analysis of selected metal ions with the newly biodegradable
chelating agents, lminodisuccinic acid (IDS) and Polyaspartic acid (PASP) was
performed by cyclic voltammetry using a thin mercury film glassy carbon
microelectrode. IDS and PASP are of interest from an environmental perspective. They
were introduced as alternatives to chelants and dispersants.
The thermodynamic stability constants of these selected metal complexes were
determined by cyclic voltammetry (CV). A comparison with similar data obtained using
potentiometric methods shows a good correlation between the two methods. This
reaffirms that CV is a useful and facile means for evaluating metal-ligand complex
thermodynamic data. Furthermore, our results were used to validate the predicted models
obtained using an Equilibrium Speciation model JESS program.
Capillary Electrophoresis (CE) was used to separate metal-IDS and metal-PASP
complexes at different pH's. The separation of these chelates was achieved within 7
minutes. The obtained results were compared with speciation and a reasonable agreement
was observed, although the separation ofmetal-PASP showed poor broad peaks. / AFRIKAANSE OPSOMMING: Die elektrochemiese analise van geselekteerde metaal-ione met nuwe biodegredeerbare

mikroelektrode. Vanaf'n omgewingsstandpunt is IDS en PASP van belang. Hulle was
voorgestel as alternatiewe vir chelate en verspreidingsagente.
Die termodinamies stabiliteits konstantes van hierdie uitgesoekte metal komplekse was
bepaal deur middel van sikliese voltametrie (CV). 'n Vergelyking met soortgelyke data,
wat met potensiometriese metodes bepaal was, dui 'n goeie korrelasie aan tussen die twee
metodes bepaal was. Dit bevestig die feit dat CV 'n bruikbare en maklike manier is om
metal ligand komplekse se termodiniese data te evalueer. Die resultate was ook gebruik
om die voorspelde model, wat verkry was deur gebruik te maak van "Equilibrium
Speciation model JESS program, te bevestig.
Kapillere elektroforese (CE) was gebruik om metal-IDS en metal-PASP komplekse by
verskillende pH's te skei. Die skeiding van hierdie chelate was binne sewe minute verkry.
Hierdie resultate was vergelyk met die spesiasie data en 'n aanvaarbare ooreenstemming
was waargeneem, alhoewel die skeiding van die Metaal-PASP swak pieke toon.

Ce travail s’inscrit dans le cadre de la recherche relative au développement de biosenseurs à ADN électrochimiques. Des aspects fondamentaux, ainsi que des aspects d’application de la détection d’hybridation d’ADN sont envisagés.
Dans un premier temps, le comportement interfacial et le processus d’hybridation d’oligonucléotides d’ADN linéaires et ADN hairpin (structure en épingle à cheveux) nonmarqués sont étudiés en formant des monocouches auto-assemblées mixtes de monobrins d’ADN (ssADN) thiolés et d’un hydroxyalcanethiol (4-mercaptobutan-1-ol) par coadsorption spontanée sur des électrodes d’or polycristallin. L’immobilisation de monocouches mixtes ssADN/MCB est caractérisée par voie électrochimique et par spectroscopie des photoélectrons X. Des mesures de chronocoulométrie, en présence de [Ru(NH3)6]3+ (RuHex), permettent de déterminer la quantité d’ADN dans la monocouche mixte formée. Les résultats montrent que l’excès superficiel d’ADN linéaire est plus important que l’excès superficiel d’ADN hairpin sous des conditions de formation identiques.
La réaction de reconnaissance moléculaire d’hybridation est détectée par des mesures d’impédance en présence de [Fe(CN)6]3-/4-. L’hybridation se traduit dans le cas de l’ADN linéaire par une augmentation de la résistance au transfert d’électron Rct tandis que dans le cas de l’ADN hairpin, Rct diminue. Ces différences sont dues au plus faible recouvrement et au changement de conformation des molécules d’ADN hairpin lors de l’hybridation. Des mesures de réflectivité de neutrons nous ont permis de mettre en évidence l’augmentation de l’épaisseur du film d’ADN hairpin et de confirmer le changement conformationel ces sondes lors de la reconnaissance moléculaire.
Dans la seconde partie, nous présentons une nouvelle méthode électrochimique de détection d’hybridation, basée sur les interactions électrostatiques entre le complexe cationique RuHex et les groupements phosphates de l’ADN. Afin d’améliorer la détection des molécules de PNA (peptide nucleic acid) ont été immobilisées comme sondes de reconnaissance moléculaire. Après hybridation des sondes PNA avec le brin complémentaire, RuHex s’adsorbe sur l’ADN hybridé et un signal de réduction de ces complexes redox, enregistré par voltampérométrie alternative, constitue une signature claire de l’hybridation d’ADN à l’interface modifiée. Les interactions RuHex/PNA-ADN ont été étudiées. La constante d’adsorption de RuHex sur l’électrode modifiée PNA/MCB après hybridation est évaluée à 2,9 (±0,3) 105 M-1 en milieu Tris-HCl 0,01M, selon une isotherme de Langmuir.
Les performances analytiques de la méthode de détection (sensibilité, sélectivité et reproductibilité) ont été évaluées et optimisées pour la détection des séquences d’ADN du gène de l’ARNr 23S d’Helicobacter pylori. La méthode de détection électrochimique présentée est assez sélective pour permettre de discriminer les mutations ponctuelles A2143G et A2144C de la séquence de type sauvage. La diminution significative des signaux d’admittance enregistrés en présence des séquences mutées est attribuée à la capacité accrue de discrimination de mutations ponctuelles des molécules PNA.
La réponse de détection est linéaire en fonction du logarithme de la concentration de la cible d’ADN sur plus de quatre ordres de grandeur (10-6 M à 10-10 M). La limite de détection de l’oligonucléotide d’ADN complémentaire de 80 pM est très bonne. La méthode a été appliquée avec succès à la détection de fragments PCR complémentaires de 100 et 400 paires de bases, amplifiés à partir de souches SS1 d’H.pylori.
