Etude de résolutions catalysées par des lipases sous irradiation micro-onde / Study of resolutions catalyzed by lipases under microwave irradiation

La demande en composés chiraux est en plein essor ces dernières années. Pour accéder à leur synthèse, la biocatalyse, couplée à l’irradiation pourrait être une méthode innovante. Il existe en effet de nombreux cas dans la littérature où l’utilisation de micro-onde semble avoir un effet activateur sur l’efficacité enzymatique.Cependant, l’effet de l’irradiation micro-onde est mal compris et controversé. Le but de cette thèse était d’étudier l’impact de l’irradiation micro-onde sur des lipases, immobilisées ou non, en étudiant différentes réactions modèles, allant de la résolution d’alcools secondaires linéaires simples à la résolution de polyols complexes, et alcools polyfonctionalisés, par comparaison entre chauffage sous irradiation micro-onde (en conditions drastiques ou non) au chauffage classique. L’étude de l’irradiation micro-onde sur la stabilité enzymatique et sur paramètres intrinsèques de l’enzyme après modification des paramètres réactionnels a permis de mettre en évidence un rôle indéniable de l’irradiation micro-onde sur l’efficacité des réactions enzymatiques. Il a été possible d’une part de diminuer de façon importante les temps réactionnels, comparé au chauffage traditionnel,et d’autre part de contrôler efficacement l’énantio préférence et la sélectivité de la lipase pour l’obtention de molécules d’intérêt. Par des procédés innovants, l’impact de la puissance d’irradiation a été montré comme hautement dépendant du modèle réactionnel étudié. En optimisant les conditions réactionnelles pour obtenir les meilleures sélectivités et activités enzymatique sous irradiation micro-onde, la synthèse de a-hydroxyamides chiraux et de polyols parfaitement résolus a pu être entreprise de façon rapide, propre, tout en respectant les principes de chimie verte. / Chiral molecules demand is booming in recent years. Biocatalyse under microwave irradiation is found to be an attractive way to synthesise these molecules. Indeed, in the literature, some cases show an activation effect on the enzymatic efficiency by microwave irradiation, compared to classical heating. However, the effect of microwave irradiation is not well understood and controversial. The aim of his PhD was to study the impact of the microwave irradiation on some lipases, immobilized or not. Different model reactions were studied from secondary linear alcohol resolution, to the resolution of more complex polyols or polyfunctionalized secondary alcohols comparing the microwave heating (in drastic conditions or not) to the classical heating. Studying the enzymatic stability and intrinsic parameters after modifying the reaction parameters, we observed a clearly microwave effect on the efficiency of the enzymatic resolutions. Using microwave heating, it could be possible to decrease, in an important way, the reaction time, compared to the classical heating and to control the énantiopréférence of the lipase to efficiently obtain chiral product. Trough innovative processes, the impact of the irradiation has been studied and it depends on the model reaction.Optimizing the conditions to obtain the best enzyme selectivities and activities under microwave irradiation, we could synthesise chiral a-hydroxyamides and polyols, in a rapid, clean way, respecting the principles of green chemistry.

Triquentrinas A são produtos naturais marinhos com atividade biológica e alta complexidade estrutural. Estes fatores tornam estes alcaloides e compostos análogos, como os herbindóis, alvos para a síntese total e plataforma para o desenvolvimento de novas metodologias sintéticas. Nesta Dissertação de Mestrado, procurou-se completar a síntese estereosseletiva da (+)-cis-triquentrina A empregando-se intermediários sintéticos protegidos com o grupo benzila que poderão ser usados para a futura prospecção de novos compostos com atividade biológica. A rota proposta baseia-se na obtenção do ácido (S)-3-(1-benzil-4-etil-1H-indol-7-il)butanóico, um intermediário-chave já descrito por Silva e colaboradores no percurso da síntese total da (+)-trans-triquentrina A, e da finalização da síntese de acordo com a abordagem proposta pelo grupo de RajanBabu para conversão do análogo protegido com grupo tosila à (+)-cis-triquentrina A. A resolução enzimática do intermediário-chave com lipase de Pseudomonas cepacia imobilizada em terra diatomácea foi otimizada, resultando em rendimentos de 32% e 99% ee na metade do tempo descrito anteriormente. Na etapa-chave da síntese, o (S)-ácido foi submetido a uma acilação de Friedel-Crafts intramolecular na presença de anidrido trifluoroacético que produziu o produto de ciclização desejado com 40% de rendimento. Na etapa final da síntese, o intermediário sintético protegido com grupo benzila não pode ser convertido à (+)-cis-triquentrina A, empregando-se a metodologia desenvolvida para a redução do composto análogo tosilado, dada a diferença de reatividade imposta pela troca do grupo protetor. As etapas realizadas até o penúltimo intermediário sintético (S)-8, consta com 10 etapas e rendimento global de 1,3%. viii As diferenças eletrônicas e estruturais relacionadas a diferentes grupos protetores poderão ser refletidas em variações na atividade antiproliferativa de indóis relacionados a triquentrinas. Ainda no interesse de preparar moléculas para envio à análises de atividade antiproliferativa preparou-se um composto relacionado à síntese da trans-triquentrina A tendo como etapa-chave uma contração de anel mediada por I(III) em 21% de rendimento, cujo trabalho foi incluído nos anexos. / Trikentrins A are marine natural products with biological activity and high structural complexity. Synthesis of Fmoc-3-(N-ethyl-3-carbazolyl)-L-alanine and Its Incorporation into a Cyclic Peptide

"Ghadiri reported the first synthetic peptide nanotubue in 1993, which has triggered extensive studies on peptide-based nanotubes and their potential application in molecular wires, catalysts and novel drug delivery vehicles. Our concerns focus on chromophore-modified cyclic peptides, which open a new way to design and synthesize novel nanoscale electronic or photonic devices, and are expected to provide the highly efficient electron and energy transfer that such devices require. This research concerned the design and synthesis of chiral a-amino acids with specific chromophores, including N-ethyl-3-carbazolylalanine and 9-anthrylalanine, and an 8-mer linear peptide (H-Aib-Car-Aib-Phe-Aib-Bpa-Aib-Phe-OH) and its corresponding cyclic peptide cyclo(Aib-Car-Aib-Phe-Aib-Bpa-Aib-Phe) that incorporate the N-ethyl-3-carbazolylalanine. This thesis describes the relevant background, synthetic strategies, experiments and results in detail. The carbazole derivatives were found to be very labile to strong acid, which might have caused self-condensation. In order to avoid the formation of acid-derived side-products, the Wittig-Horner reaction was used successfully in preparation of N-protected-3-(N'-ethyl-3-carbazolyl)-DL-alanine methyl ester. Dual enzymatic hydrolyses were developed to produce the chiral amino acids with high enantiomeric excess. ChiroCLEC-BL was used to selectively hydrolyze the N-acetyl-L-amino acid methyl ester, while amanoacylase was adopted to remove the acetyl group from the resulting N-acetyl-L-amino acid. Two model peptides were synthesized, a 4-mer peptide (H-Car-D-Ala-Bpa-D-Ala-OH) via the Boc-strategy, and an 8-mer peptide (H-Ala-D-Ala-Npa-D-MeAla-Ala-D-Ala-Bpa-D-Ala-OH) by the Fmoc-strategy. Eventually, the target linear peptide was synthesized via the Fmoc-strategy and then cyclized in solution."

EXPLORATION OF CIS-1,2-DIAMINOCYCLOHEXANE-BASED CONFORMATIONALLY LOCKED CHIRAL LIGANDS IN ASYMMETRIC SYNTHESIS

Natural products have been demonstrated to be of great significance to the pharmaceutical industry in the development of new drugs and medicine. Unfortunately, synthetic approaches to obtain these natural products often prove increasingly challenging due to the complexity of synthesizing the target drug in the proper stereochemistry. The availability of enantioselective reactions can play a pivotal role in overcoming this challenge, yielding access to optically pure intermediates and products. Chiral ligands based on a trans-1,2-diaminocyclohexane motif are often employed for this purpose and their complexes with transition metals have been demonstrated to act as efficient chiral catalysts in asymmetric reactions. In contrast, studies involving cis-1,2-diaminocyclohexane derivatives as chiral catalysts are strongly underrepresented. We have designed and performed the synthesis of an axially chiral conformationally locked cis-1,2-diamine scaffold, conveniently designed for further derivatization into more complex structures. A key step in this synthesis was the chiral resolution of a racemic intermediate, realized through both chemical and enzymatic means in a comparative study. Utilizing the newly gained optically pure primary diamine scaffold, a library of chemically diverse secondary diamine ligands has been synthesized and characterized through NMR spectroscopy, mass spectrometry, and chiral HPLC. Assignment of the absolute configuration within the cis-1,2-diamine scaffold was realized through single-crystal X-ray crystallography experiments on one of the synthetic intermediates. The synthesized ligands have been evaluated for their potential to function as chiral catalysts in the asymmetric Henry reaction and asymmetric transfer hydrogenation. As a function of both steric and electronic structural variation, a range of catalytic activities and enantioselectivities in the Henry reaction were observed. The ligands proved to be less suitable for asymmetric transfer hydrogenation with only a select number of ligands catalyzing the reaction, and a single example resulting in a decent enantioselectivity. We additionally explored the possibility of incorporating a chemical switch into the scaffold, responsible for switching the axial chirality of the molecule. As a consequence of inverting the axial chirality, the configuration of the potential reaction product in asymmetric synthesis would also be inverted. To this extent, we performed the synthesis of a novel specifically designed crown ether, dicyclohexeno-18-crown-6, furnished with two π-bonds in the cyclohexane rings, allowing for additional modification into more advanced functionalized structures.

Les vinylphosphonates fonctionnalisés représentent une classe importante de briques moléculaires utilisées en synthèse organique et suscitent un grand intérêt au niveau biologique. Ainsi, nous avons étudié différentes voies de synthèse pour accéder à ces composés. Il en ressort une méthode généralisable, faisant intervenir une réaction de substitution organocatalysée, à partir d’un substrat phosphoré original, et permettant d’obtenir de nombreux vinylphosphonates non décrits jusqu’ici. Par la suite, la synthèse de vinylphosphonates azahétérocycliques a été abordée en employant la réaction de métathèse cyclisante à partir de substrats adéquats, eux-mêmes obtenus par le biais de la méthodologie décrite précédemment.Nous avons enfin mis au point la première synthèse totale énantioselective de la molécule UPF 702 (vinylphosphonate cyclique comportant un motif acide aminé), connue pour ses propriétés biologiques notamment comme agoniste de récepteurs du glutamate et présentant ainsi un potentiel thérapeutique contre les maladies du système nerveux central. Deux voies de synthèses ont été imaginées, basées sur des réactions organocatalysées et de métathèse cyclisante. L’introduction de la chiralité a été réalisée via des réactions de désymétrisation ou de dédoublement enzymatiques et l’accès à l’amine par un réarrangement de Curtius. / Functionalized vinylphosphonates constitute an important class of building blocks used in organic synthesis and aroused great interest due to their various biological activities. Thus, we developed several synthetic methodologies to reach these compounds. The most general method entails an organocatalyzed substitution reaction using an original substrate, and allows the synthesis of numerous hitherto unknown vinylphosphonates.Then, the synthesis of azaheterocyclic vinylphosphonates was investigated using the ring closing metathesis from appropriate substrates which are obtained through the above methodology.Finally, we have set up the first enantioselective synthesis of UPF 702 (a cyclic vinylphosphonate including the amino acid moiety), known to exhibit agonist activity towards glutamate receptors, and thus potentially active against central nervous system diseases. Two synthetic approaches were devised, based on the organocatalyzed substitution followed by ring closing metathesis. The enantioselectivity was brought by enzymatic resolution or desymmetrisation, whereas the amino acid was prepared via a Curtius rearrangement.
