Identification Studies of Bacillus Spores Using Fluorescence Spectroscopy

Fluorescence spectroscopy was examined as a potential technique for identifying aerosol particles like bacterial spores. This technique was used for laboratory measurements on some common biological agent simulants. We have measured the intrinsic steady-state fluorescence emission spectra as a function of the excitation wavelength for several bacterial spores (washed and unwashed) in dry and aqueous suspensions at room temperature using excitation wavelengths from 200 to 600 nm. These measurements were compared to those of common, naturally occurring biological components like fungal spores and pollen and non spore samples like ovalbumin. The spectra of samples were combined into fluorescence profiles or fluorescence fingerprints. Different substrates were used for collection and detection of spores. Each bacterium produces a unique in vitro fluorescence profile when measured in dried and aqueous suspension and exhibits a strong maximum in its fluorescence emission spectrum near 330-340 nm. The fluorescence profiles were reproducible. The complexity of microorganisms made the interpretation of their spectral signature a difficult task. Principal components analysis (PCA) and cluster analysis were done as a data reduction technique for detection and identification from different backgrounds. PCA illustrates that linear combination of detected fluorescence intensities, which are present in different ratios in each of samples studied, can be used to discriminate biological agent simulants from other biological samples. The hydration effects, washing effects and the role of tryptophan on spore fluorescence were also investigated. The emission spectra of the dried spores showed a maximum near 330 nm, suggesting a hydrophobic environment for its tryptophan residues. The aqueous solution of tryptophan showed fluorescence shifted to 360 nm and in ethanol solution the maximum was shifted to 340 nm, suggesting a rather more polar average location of the tryptophan. To find the limit of detection we measured the quantum efficiency (QE) of a few samples. We concluded that spectroscopy techniques coupled with effective interpretation models are applicable to biological simulants agents. Biogeochemical interactions of natural organic matter with arsenic in groundwater

Over 100 million people are at health risk due to arsenic contaminated groundwater, especially in the Bengal Basin in south-east Asia. Dissolved organic matter (DOM), geology and geomicrobiology are important factors affecting arsenic mobility. This study focuses on interactions of different aspects of natural organic matter in arsenic-contaminated environments. A literature review specifically includes past studies done on fundamentals of arsenic geology, geomicrobiology, DOM characterization and relevant analytical methods and tools. Based on background information already collected, this research is focused on specific research questions and corresponding hypotheses. The overarching goal of this investigation is to better understand the mechanisms by which DOM influences arsenic mobilization. The specific goals of this research are: 1) to evaluate role of oxidized humic quinones in reductive dissolution of Fe-As minerals and subsequent arsenic mobilization via electron shuttling, 2) to quantify the rate of microbially mediated reductive dissolution in the presence of oxidized humic quinones, 3) to evaluate DOM-Fe-As ternary complex formation and its influence on arsenic mobility and 4) to characterize DOM in the arsenic-contaminated aquifers of West Bengal, India and evaluate its role in arsenic mobilization using groundwater flow and contaminant transport modeling approach. Results of this study revealed that oxidized quinone like moieties (such as fulvic acids) serve as an electron shuttle and enhance the reductive dissolution process under reducing conditions, hence mobilize the arsenic in groundwater. Another key result from this study suggested that arsenic binds with non-aromatic portion of the humic-like DOM under reducing conditions and increases its solution concentration. A field study conducted in West Bengal, India revealed that the mechanisms studied in the laboratory exists in reducing aquifer. A groundwater flow and reactive transport model was created to explain multiple interactions of DOM and arsenic spatial scales. Broader impacts of this study include significant addition to scientific knowledge about subsurface biogeochemistry and the role of DOM in biogeochemical reactions in the subsurface.

Data representation for fluorescence guided stereotactic brain tumor biopsies : Development and evaluation of a visual and auditory user interface

Background and Objective In stereotactic brain tumor biopsies, the combination of real-time fluorescence spectroscopy with the detection of microvascular perfusion using laser Doppler flowmetry provides an improved localization of the brain tumor while decreasing the risk of intra-cranial hemorrhage. The surgeon using the measurement probe is required to view signal values on a screen or usually, when her or his visual focus is directed at the patient, the verbal feedback of a biomedical engineer who is monitoring the measurement signals is needed. In this process possible important information can be overlooked and time is lost. The aim of the thesis was the development a visual and auditory user interface (UI) for use in stereotactic brain tumor biopsies. Materials and Methods The system translates the fluorescence intensity of protoporphyrin IX (PpIX) into sound and visual indicators that are easy and fast to recognize and transmits warning signals in case of signal error or the detection of microvascular perfusion. The increasing and de-creasing fluorescence values at tumor margins were reproduced to improve the precision of de-tecting varying fluorescence intensities when entering tumor tissue with color gradient models. The algorithm produced five signal values when specific fluorescence intensities were measured and compared at different wavelengths.For the development of the UI, a user-centered design was implemented. The user-, operating room- and safety requirements were gathered by communicating with the biomedical engineers and neurosurgeons who had experience in working with fluorescence guided brain tumor biop-sies. The requirements were considered when designing the UI’s features in LabVIEW and the auditory feedback was generated using OSC (Open Sound Control). The user interface intended to deliver measurement data to the user that triggered a high response accuracy by being easy to understand while inducing high user acceptance. The user interaction and function response accuracy of the visual and auditory interface were evaluated in statistical tests where operating room situations were mimicked. The user acceptance of the UI was evaluated. Results Signals for no, low (increasing and decreasing) and high fluorescence indicators, as well as two warning indicators for a blocked signal and vessel occurrence were represented visually and auditorially by the user interface. An intensity/time graph and intensity/wavelength graph, along with the option of recording measurement files and opening saved files allowed the inspec-tion of detailed measurement values. The user study exhibited auditory response accuracy of 95 ± 3% in the intuition test and 91±16% in a memory test. The testing of the response accuracy of the individual signal values displayed accurate responses in 84% to 100% of times a signal was played back. The user acceptance rating of the auditory and visual interface showed no negative results. Conclusion A UI was developed to visually and auditorially represent measurement values to a neurosurgeon performing a stereotactic brain tumor biopsy procedure and biomedical engineers monitoring the measurement signals. The visual display was successful in representing data in a way that was easy to understand. The auditory interface showed high response accuracies for the individual tones representing measurement values. The majority of the test subjects per-ceived the signals to be intuitive, easy to understand and easy to remember. The auditory and visual UI showed high user acceptance ratings, indicating that the user interface was useful and satisfactory in its application.

Propriedades estruturais de membranas modelo em interação com o composto anti-Leishmania miltefosina / Structural properties of model membranes in interaction with the leishmanicidal compound miltefosine

A leishmaniose é uma doença tropical negligenciada causada por diferentes espécies do gênero Leishmania que atinge grande parte da população mais pobre do mundo e sua manifestação visceral, que é fatal se não tratada, tem se alastrado atingindo grandes cidades, aumentando o número de pessoas com risco de infecção. Dentre os medicamentos em uso, está o análogo lipídico sintético hexadecilfosfocolina (miltefosina), administrado oralmente, que age nas membranas celulares do parasita e pode induzir apoptose, mas com modo de ação não totalmente esclarecido. O primeiro local de interação desse fármaco é a membrana celular do parasita, sendo importante o conhecimento da sua forma de interação. Neste trabalho examinamos propriedades de diversos modelos de membrana com diferentes composições, levando em consideração o conhecimento existente sobre a composição da membrana plasmática da Leishmania. Assim, as membranas modelo foram vesículas unilamelares grandes e gigantes (LUVs e GUVs), de fosfolipídios puros, de misturas binárias com fosfolipídios e colesterol e ainda misturas ternárias com ceramida, um esterol presente nas membranas de Leishmania. A interação com a miltefosina foi estudada em diferentes intervalos de concentração do fármaco. Como técnicas principais utilizamos a espectroscopia de fluorescência, estática e resolvida no tempo, a espectroscopia de correlação de fluorescência, microscopia de fluorescência e confocal e imagens por tempo de vida de fluorescência. Observou-se que a interação entre o fármaco e as membranas lipídicas ocorre de diferentes formas, dependendo i) da razão molar entre o fármaco e os lipídios; ii) da concentração real do fármaco, se abaixo ou acima da concentração micelar crítica (CMC); iii) da composição do modelo de membrana e da fase lipídica da bicamada. Em concentrações abaixo da CMC, a miltefosina tem efeito de fluidificação das bicamadas, principalmente quando elas se encontram em na fase gel, mas esse efeito é pouco pronunciado na presença de colesterol, pois esse composto protege a bicamada do efeito do fármaco. Em vesículas de misturas ternárias de fosfolipídio, colesterol e ceramida em alta concentração, não há separação de fases, e a presença de 10 mol% de miltefosina promove a formação de domínios de ceramida; nas vesículas em que a ceramida está em concentração molar mais baixa, formando domínios, a separação de fases fica menos evidente com o acréscimo de miltefosina. Em razões de concentração miltefosina/lipídio elevadas, mas ainda abaixo da CMC, observa-se diminuição no tamanho das vesículas, por formação de agregados de fármaco/lipídio com porções da bicamada. Em concentrações acima da CMC, ocorrem efeitos drásticos com solubilização de partes cada vez maiores da bicamada da membrana, e esses efeitos ocorrem em tempos menores quanto maior a concentração de miltefosina. Portanto, de maneira geral, o colesterol protege a bicamada do efeito da miltefosina, mas o fármaco tem efeito pronunciado em modelos de membrana de misturas ternárias contendo ceramida. Os efeitos variam com a concentração da miltefosina, com aumento da fluidez da bicamada em baixas razões fármaco/lipídios, solubilização de pequenas porções da bicamada e diminuição do tamanho das vesículas em razões maiores, mas ainda abaixo da CMC, e acima da CMC, formação de agregados do fármaco com porções dos lipídios da bicamada e fragmentação da membrana. / Leishmaniasis is a complex of diseases part of the neglected tropical diseases caused by several species of the genus Leishmania. It reaches a large part of the poorest people in the world and its visceral form, which is fatal if left untreated, has been spread around big cities, increasing the number of people at risk of infection. Among the used drugs for the treatment, there is the synthetic lipid analogue hexadecylphosphocholine (miltefosine), orally administrated, which acts in the cell membranes and can induce apoptosis like death, but its mechanism of action is not totally clear. The first interactions site of this drug is the cell membrane, and it is important to know its mechanism of interaction. In this work we explore properties of several membrane models with different compositions, taking into account the existent knowledge about the composition of the Leishmania plasma membrane. Therefore, the model membranes were giant and large unilamellar vesicles (GUVs and LUVs), formed from pure phospholipids, binary mixtures of phospholipids and cholesterol and ternary mixtures with ceramide, a sterol present in the Leishmania membranes. The interaction with miltefosine was studied in different intervals of drug concentration. The main techniques used were the steady-state and time-resolved fluorescence spectroscopy, fluorescence correlation spectroscopy, confocal and fluorescence microscopy and fluorescence lifetime imaging. The interaction depends on i) the molar ratio of drug and lipids; ii) the real concentration of the drug, if it is below or above the critical micelle concentration (CMC); iii) the composition of the model membrane and the lipid phase of the bilayer. In concentration below the CMC, miltefosine has an effect of bilayer fluidization, mainly when it is in a more ordered phase, but this effect is less pronounced in cholesterol presence, because this compound protects the bilayers from the drug effect. In vesicles from ternary mixtures of phospholipid, cholesterol and ceramide in high concentration, there is no phase separation, and the presence of 10 mol% of miltefosine promotes ceramide domains formation; in vesicles in which ceramide is in low concentration, forming domains, the phase separation is less evident with miltefosine addition. In high concentration ratio miltefosine/lipids, but below CMC, it is observed a decrease in vesicles size with drug/lipids aggregates formation from portion of the bilayer. In concentrations above the CMC, drastic effects occur, with solubilization of bigger portions of the membrane bilayer, and the effects occur in lower times for higher drug concentration. Therefore, generally, cholesterol protects bilayer from the effect of miltefosine, but the drug has a pronounced effect in model membranes of ternary mixtures containing ceramide. The effects vary with miltefosine concentration, increasing the bilayer fluidity in lower drug/lipid ratio, solubilization of small portions of the bilayer and decrease of vesicles size in higher ratios, but still below CMC, and above CMC, formation of aggregates of the drug with portions of bilayer lipids, and membrane fragmentation.

Clonagem, expressão e caracterização de proteinas recombinantes de Xylella fastidiosa / Cloning, expression and characterization of Xylella fastidiosa proteins

A bactéria Xylella fastidiosa (X. fastidiosa) é um patógeno de planta que causa a clorose variegada dos citros, também conhecida como amarelinho. Xylella fastidiosa é uma bactéria limitada ao xilema, sendo transmitida de planta a planta através de insetos vetores. Através da determinação experimental da seqüência primária de algumas proteínas marcadamente expressas pela X. fastidiosa e a comparação destas com seqüências de ácidos nucléicos do genoma identificou entre elas três ORFs codificadoras de proteínas que podem estar relacionadas à patogenicidade desta bactéria no seu hospedeiro. No entanto, a função biológica destas proteínas ainda não foi funcionalmente caracterizada. Para investigar o papel biológico e realizar estudos de estrutura e função, as ORFs correspondentes as proteínas hidroxinitrila liase, corismato sintase e proteína D do sistema de transporte e secreção Tat foram clonadas, seqüenciadas e expressas em Escherichia coli. As proteínas recombinantes expressas foram purificadas por cromatografia de afinidade e suas identidades verificadas por seqüenciamento. As proteínas purificadas foram encontradas como uma única banda em SDS-PAGE. As proteínas recombinantes, pela primeira vez isoladas, foram caracterizadas em termos de estabilidade conformacional e comportamento estrutural frente a mudanças de pH e temperatura utilizando-se as espectroscopias de dicroísmo circular e fluorescência. O sucesso na construção do gene sintético e na clonagem em vetores de expressão de E. coli resultou em quantidade satisfatória de expressão das proteínas recombinantes. Duas delas apresentaram-se na formas solúveis, facilmente purificadas e ativas e permitindo suas caracterizações. / Xylella fastidiosa is the causal agent of citrus variegated chlorosis, also known as \"amarelinho\". Xylella fastidiosa inhabits exclusively the xylem vessels, and is transmitted by sharpshooter vectors. The experimental determination of the primary sequence of some markedly expressed proteins for X. fastidiosa and the comparison with the nucleic acids sequence of its genome aided the identification of three of them as being proteins involved in host pathogenicity. However, the biological role of these proteins should be assigned experimentally. In order to investigate the biological role, function and structure, ORFs corresponding to hydroxynitrile liase, chorismate synthase and type V secretory pathway proteins were cloned, sequenced and expressed in Escherichia coli. The expressed recombinant proteins were purified by immobilized metal affinity chromatography (Ni-NTA resin) and their identities were verified by protein sequencing. The purified proteins were found as a single band on SDS-PAGE. The successful cloning and heterologous expression of recombinant HNL in E. coli resulted in a satisfactory amount of expressed protein. Two of the expressed recombinant proteins were soluble, easily purified, and isolated in an active form. X. fastidiosa recombinant proteins, for the first time isolated, were characterized according to enzyme conformational stability and structural behavior as a function of pH and temperature using circular dichroism and fluorescence spectroscopy.

Imagem de fluorescência aplicada em doenças de citros / Fluorescence image applied on citrus diseases

Nos últimos anos, tem havido um crescente interesse na detecção precoce das doenças que afetam as culturas agrícolas a fim de evitar grandes perdas econômicas devido à contaminação de novas plantas. As principais doenças cítricas, cancro cítrico e greening, são uma séria ameaça à produção de citros em todo o mundo, incluindo regiões do Brasil e dos Estados Unidos. A disseminação rápida das doenças leva à redução do número de pomares cultivados, resultando em danos econômicos aos produtores e às indústrias relacionadas. O desenvolvimento de métodos para o diagnóstico precoce pode resultar em uma importante ferramenta para o controle e gestão dos citros. Algumas deficiências nutricionais como a de ferro e zinco apresentam sintomas visuais semelhantes com o greening, enquanto que o cancro cítrico pode ser confundido com a verrugose ou leprose dos citros, podendo levar ao diagnóstico incorreto. Atualmente, somente testes bioquímicos são capazes de detectar especificamente o cancro cítrico e o greening, e consequentemente diferenciá-los das demais doenças e deficiências de nutrientes. No presente trabalho, a técnica de espectroscopia por imagens de fluorescência em conjunto com os métodos de aprendizado e classificação, SVM (do inglês, Support Vector Machine) e ANN (do inglês, Artificial Neural Network), foram utilizadas a fim de identificar e discriminar as principais doenças que afetam a citricultura nos estados de São Paulo/Brasil e da Flórida/EUA. As amostras em estudo são cancro cítrico, verrugose, greening e deficiência de zinco. O objetivo principal é discriminar as doenças com sintomas visuais semelhantes, no caso, cancro cítrico de verrugose e greening de deficiência de zinco para as amostras do Brasil, e greening de deficiência de zinco para as amostras dos Estados Unidos. Os resultados mostram que é possível utilizar a técnica de espectroscopia por imagens de fluorescência em conjunto com os métodos de classificação na discriminação das doenças que apresentam sintomas visuais semelhantes. Ambos os classificadores apresentaram uma elevada precisão na classificação tanto das amostras do Brasil como dos Estados Unidos, destacando assim eficácia da técnica sob condições diferentes. / In recent years, there has been an increasing interest in early detection of diseases that affect agricultural crops to avoid great economic losses due to contamination of new plants. The main citrus diseases, citrus canker and HLB, are a serious threat to citrus production worldwide, including regions in Brazil and the United States. The rapid spread of the diseases leads to the reduction of cultivated orchards resulting in economic losses to producers and industries. The development of methods for early diagnosis can result in an important tool for the control and management of citrus. Some nutritional deficiencies such as iron and zinc have similar visual symptoms to HLB, while the citrus canker can be confused with citrus scab and citrus leprosies, which may lead to incorrect diagnosis. Currently, only biochemical tests are able to detect, specifically, citrus canker and HLB, and thus distinguish them from other diseases and nutrient deficiencies. In this work, the fluorescence imaging spectroscopy technique with the learning and classification methods, SVM (Support Vector Machine) and ANN (Artificial Neural Network), were used to identify and discriminate the main diseases that affect citrus production in the states of São Paulo/Brazil and Florida/USA. The samples studied are citrus canker, citrus scab, HLB and zinc deficiency. The objective is to discriminate the diseases with similar visual symptoms, such as citrus canker from citrus scab and HLB from zinc deficiency for samples from Brazil and HLB from zinc deficiency for samples from the United States. The results show that it is possible to use the fluorescence imaging spectroscopy technique together with the classification methods for the discrimination of the diseases that have similar visual symptoms. Both classifiers showed high accuracy in the classification of the samples from Brazil and the United States, highlighting the efficiency of the technique under different conditions.

Complexos do íon uranilo (UO22+) apresentam um grande potencial como materiais luminescentes, aplicados em tecnologia laser, sondas luminescentes, células para conversão de energia solar, etc. Neste trabalho avaliou-se a eficiência da transferência de energia no aquo-complexo de UO2(MS)22H2O dopado com Eu3+ (MS = íon metanossulfonato, CH3SO3-) e no aquo-complexo de Eu(MS)36H2O dopado com UO22+ para serem usados como eficientes dispositivos moleculares conversores de luz e/ou em células solares para conversão de energia. Também foram descritos a síntese, caracterização e estudo espectroscópico das matrizes UO2(MS)2.(H2O)n.(x%mol)Eu3+ e Eu(MS)3.(H2O)n.(x%mol) UO22+ (nas quais x= 1, 3, 5 e 10). Os complexos obtidos foram caracterizados por análise elementar CHN e espectrofotometria para determinar a concentração de U6+, espectroscopia de absorção na região do infravermelho, análise térmica e espectroscopia de luminescência. Os dados de infravermelho sugerem que o metanossulfonato atue como um ligante bidentado. O espectro de emissão do complexo UO2(MS)2 dopado com európio exibe bandas fluorescentes características do íon uranilo, atribuídas à transição 3∏u → 1∑g+ da matriz de urânio. Para os complexos de Eu(MS)3 dopados com UO22+, as transições 5Do → 7FJ (J= 0→4) são predominantes do espectro. A intensidade de luminescência do európio associada à sensibilização é registrada no espectro de emissão na faixa de 420-720 nm. A constante de supressão da fluorescência do UO22+ pelo Eu3+ também foi determinada. / Uranyl compounds (UO22+) present a great potential as luminescent materials, for instance, applied in technology laser, luminescent probes, cells for conversion of energy, etc. In this work it is studied the efficiency of energy transfer in the compound Eu3+ doped in UO2(MS)2(H2O)n matrix and UO22+ in Eu(MS)3 (H2O)n for to be used as efficient Light Conversion Molecular Devices (LCMD) and/or in solar cells for energy conversion. It is also described the synthesis, characterization and spectroscopic study of the matrix Eu(MS)3 (H2O)n.(x%mol) UO22+ and UO2(MS)2.(H2O)n.(x%mol) Eu3+ (where x= 1, 3, 5 and 10). The compounds obtained was characterized by elementar analyses for determine the U6+ concentration, infrared spectra, thermal analyses and luminescence spectra. The IR data suggest that the MS ligand acts as bidentate one. The emission spectra of Eu3+ doped in UO2(MS)2.(H2O)2 matrix showed characteristic fluorescence bands of the uranyl ion, attributed to the transition 3∏u → 1∑g+ of the uranium matrix. For the UO22+ in Eu(MS)3(H2O)6 the transitions of 5Do → 7FJ (J= 0→4) are predominant in the spectra. The intense europium luminescence associated with uranyl sensitising is registered in emission spectra of Eu3+ ions in the range of 420-720 nm. The quenching rate constant of UO22+ fluorescence with Eu3+ is also determined.

Introdução: O transplante renal é amplamente reconhecido como a melhor forma de tratamento para os pacientes que necessitam de terapia de substituição renal. Frequentemente a equipe transplantadora se depara com a difícil questão de determinar se órgãos, provenientes de doadores falecidos e limítrofes ou em parada cardíaca, estão aptos para serem transplantados. É difícil quantificar a intensidade do dano provocado pela isquemia no enxerto a ser utilizado, especialmente se o doador apresentou quadro de instabilidade hemodinâmica com queda significativa da perfusão tecidual e aumento do risco de diminuir a função do enxerto e afetar adversamente sua sobrevida. Desta forma torna-se justificável a utilização da técnica de espectroscopia de fluorescência induzida pelo laser, na tentativa de se avaliar a correlação entre os seus achados e o grau de lesão histológica renal experimental, uma vez que se trata de um método objetivo, não invasivo, rápido e em tempo real que, futuramente, pode ser aplicada nos transplantes renais em humanos. Objetivos: Avaliar a correlação entre os dados da espectroscopia de fluorescência induzida pelo laser e alterações histológicas na isquemia e reperfusão renal em ratos, e se existe diferença significativa na leitura da espectroscopia entre os polos superior, inferior e o terço médio. Materiais e Métodos: Foram utilizados 33 ratos (Rattus norvegicus) machos adultos da linhagem Wistar que, depois de anestesiados, tiveram seus rins esquerdos abordados. Inicialmente os rins foram submetidos à detecção da espectroscopia de fluorescência dos pólos superiores, inferiores e terços médios. As excitações foram geradas por lasers com comprimentos de onda de 408, 442 e 532 nm. Em seguida os pedículos renais esquerdos foram dissecados, isolados e clampeados com auxílio de mini-pinça vascular. Então, os animais foram divididos aleatoriamente em três grupos isquêmicos de 30, 60 e 120 minutos de isquemia quente. Em cada um dos grupos, os rins foram novamente analisados pela espectroscopia de fluorescência, bem como após 5 minutos de reperfusão, utilizando novamente feixes excitatórios com os mesmos comprimentos de onda, nas mesmas regiões renais. Posteriormente os rins esquerdos foram coletados e enviados para estudo histológico. Resultados: O tempo de isquemia mostrou forte influência com a graduação histológica. Com 30 minutos de isquemia, nenhum comprimento de onda (408, 442 e 532 nm) apresentou correlação com a graduação histológica (p = 0,81; p = 0,11; p = 0,21, respectivamente). Com 60 minutos de isquemia, o laser de excitação de 532 nm (na fase de reperfusão) apresentou coeficiente de correlação negativa significativa (r = - 0,61) com a graduação histológica. Na isquemia de 120 minutos, o laser com 442 nm de comprimento de onda (na fase de reperfusão) mostrou o coeficiente de correlação negativa significativa (r = - 0,73) com a graduação histológica. O terço médio renal apresentou média estatística superior à dos polos (p < 0,001) na leitura da espectroscopia de fluorescência. Conclusões: Há correlação entre os dados da espectroscopia de fluorescência induzida pelo laser e as alterações histológicas na isquemia renal em ratos, sendo necessário, durante a investigação, analisar apenas o terço médio renal. / Introduction: Renal transplantation is widely recognized as the best form of treatment for patients who require renal replacement therapy. Often, the transplant team is faced with a difficult question, if organs from deceased marginal donors or non-heart beating donors are able to be transplanted. It is difficult to quantify the intensity of damage caused by ischemia in the graft to be used, especially if the donor had hemodynamic instability with a significant decrease of the tissue perfusion and an increased of the risk of diminishing the graft function which could affect adversely its survival. Thus it is justified to use the technique of laser-induced fluorescence spectroscopy, to assess the correlations between its results and the histological grade in experimental renal injury, since it is an objective, non-invasive, fast and in real-time analysis, which can be applied, in the future, in human kidney transplants. Objectives: To evaluate the correlation between the data of laser-induced fluorescence spectroscopy and histological changes in kidney ischemia and reperfusion in rats, and if there are significant differences of reading between the upper and lower poles and the middle area of such kidneys. Materials and Methods: We used 33 adults male rats (Rattus norvegicus) of Wistar strain, which after anesthetized, had their left kidney addressed. Initially such kidneys were submitted to detection of the fluorescence spectroscopy of the upper pole, lower pole and the middle area. Excitations were generated by lasers having wavelengths of 408, 442 and 532 nm. Then the left renal pedicles were dissected, isolated and clamped. Then the animals were randomized into three ischemic groups of 30, 60 and 120 minutes. In each group, the kidneys were analyzed by fluorescence spectroscopy for the second time, and again after 5 minutes of reperfusion, using excitatory beam with same wavelength, at the poles (upper and lower) and the middle area of the kidneys. Later, the left kidney were collected and sent for histological examination. Results: The ischemia time showed a strong influence on the histological grade. With 30 minutes of ischemia, no wavelength (408, 442 and 532 nm) was correlated with the histological lesions (p = 0.81, p = 0.11, p = 0.21, respectively). With 60 minutes of ischemia, the laser excitation of 532 nm (in the reperfusion phase) showed a significant negative correlation coefficient (r = - 0.61) with the histological grading. In 120 minutes of ischemia, laser with 442 nm wavelength (in the reperfusion phase) showed a significant negative correlation coefficient (r = - 0.73) with the histological grade. The middle area of the kidneys showed a higher average statistically (p< 0,001) than the poles in the reading of fluorescence spectroscopy. Conclusions: There is a strong correlation between the data of laser-induced fluorescence spectroscopy and the histological changes in rats renal ischemia, being necessary, during the investigation, to analyze only the middle area of the kidneys.

Complexos do íon uranilo (UO22+) apresentam um grande potencial como materiais luminescentes, aplicados em tecnologia laser, sondas luminescentes, células para conversão de energia solar, etc. Neste trabalho avaliou-se a eficiência da transferência de energia no aquo-complexo de UO2(MS)22H2O dopado com Eu3+ (MS = íon metanossulfonato, CH3SO3-) e no aquo-complexo de Eu(MS)36H2O dopado com UO22+ para serem usados como eficientes dispositivos moleculares conversores de luz e/ou em células solares para conversão de energia. Também foram descritos a síntese, caracterização e estudo espectroscópico das matrizes UO2(MS)2.(H2O)n.(x%mol)Eu3+ e Eu(MS)3.(H2O)n.(x%mol) UO22+ (nas quais x= 1, 3, 5 e 10). Os complexos obtidos foram caracterizados por análise elementar CHN e espectrofotometria para determinar a concentração de U6+, espectroscopia de absorção na região do infravermelho, análise térmica e espectroscopia de luminescência. Os dados de infravermelho sugerem que o metanossulfonato atue como um ligante bidentado. O espectro de emissão do complexo UO2(MS)2 dopado com európio exibe bandas fluorescentes características do íon uranilo, atribuídas à transição 3&prod;u &rarr; 1&sum;g+ da matriz de urânio. Para os complexos de Eu(MS)3 dopados com UO22+, as transições 5Do &rarr; 7FJ (J= 0&rarr;4) são predominantes do espectro. A intensidade de luminescência do európio associada à sensibilização é registrada no espectro de emissão na faixa de 420-720 nm. A constante de supressão da fluorescência do UO22+ pelo Eu3+ também foi determinada. / Uranyl compounds (UO22+) present a great potential as luminescent materials, for instance, applied in technology laser, luminescent probes, cells for conversion of energy, etc. In this work it is studied the efficiency of energy transfer in the compound Eu3+ doped in UO2(MS)2(H2O)n matrix and UO22+ in Eu(MS)3 (H2O)n for to be used as efficient Light Conversion Molecular Devices (LCMD) and/or in solar cells for energy conversion. It is also described the synthesis, characterization and spectroscopic study of the matrix Eu(MS)3 (H2O)n.(x%mol) UO22+ and UO2(MS)2.(H2O)n.(x%mol) Eu3+ (where x= 1, 3, 5 and 10). The compounds obtained was characterized by elementar analyses for determine the U6+ concentration, infrared spectra, thermal analyses and luminescence spectra. The IR data suggest that the MS ligand acts as bidentate one. The emission spectra of Eu3+ doped in UO2(MS)2.(H2O)2 matrix showed characteristic fluorescence bands of the uranyl ion, attributed to the transition 3&prod;u &rarr; 1&sum;g+ of the uranium matrix. For the UO22+ in Eu(MS)3(H2O)6 the transitions of 5Do &rarr; 7FJ (J= 0&rarr;4) are predominant in the spectra. The intense europium luminescence associated with uranyl sensitising is registered in emission spectra of Eu3+ ions in the range of 420-720 nm. The quenching rate constant of UO22+ fluorescence with Eu3+ is also determined.
