This diploma thesis describes the design of a translation stage for focusing objective inside the LIBS interaction chamber. It contains a description of the fundamentals and basic functions of LIBS spectroscopy. Moreover, in the thesis an overview of different translation stages as possible LIBS components is given. Their advantages and disadvantages for use in a vacuum translation stage construction are evaluated. The thesis describes the final solution of a linear translation stage for focusing objective, and his advantages for LIBS method

Využití mikropreparativní izoelektrické fokusace při analýze biologických vzorků / Utilization of micropreparative isoelectric focusing in the analysis of biological samples

The aim of this Master Thesis is the optimization and verification of conditions of separation of proteins from biological materials. For the separation a developed technique called micropreparative isoelectric focusing is being used. The theoretical part describes analytical methods of biological materials with a focus on proteins which were used for the optimization of the developing system in general. This part of the Master Thesis also presents this method itself. Furthermore, basic characteristics of other techniques used to control the efficiency of the micropreparative isoelectric focusing are introduced. The experimental part depicts the individual steps of optimization of the micropreparative isoelectric focusing. The ideal separation procedure was firstly found out on the individual standard proteins, secondly on their mixture and lastly the optimised procedure was applied to a real biological sample. The efficiency of the micropreparative isoelectric focusing was verified by SDS-PAGE and MALDI-TOF mass spectrometry.

The Corona pandemic - a focusing event for insufficient governmental action on climate change mitigation?

This study seeks to examine whether the Corona pandemic has potential to serve as a focusing event for the problem of insufficient governmental action on climate change mitigation. The study is built on the Multiple Streams Framework by John W. Kingdon, with a main focus on the focusing event theory. According to this, focusing events can come in three forms: as crises and disasters, personal experiences of policymakers, and as symbols. Kingdon’s theoretical discussions, alongside my own developments of his work, provides the basis for a set of analytical questions through which the answer to the research question is provided. The analysis reveals that while the pandemic indeed can be considered a crisis or disaster and personal experiences of policymakers, establishing whether these could focus attention to the specific problem of insufficient governmental action on climate change mitigation requires further research, as the perceived cause of the crisis or disaster and personal experience must be established. However, the paper finds that the pandemic indeed has potential to serve as a symbol for the specific insufficient governmental action, for instance by stressing that deforestation increases the risk of zoonotic outbreaks, such as the Corona pandemic.

Studium transferinu jako markeru dědičných poruch glykosylace / Study of transferrin as a marker of congenital disorders of glycosylation

Congenital disorders of glycosylation (CDG) represent a heterogeneous group of mul- tisystemic metabolic disorders which are caused by defects in biosynthetic pathways of glycoproteins. The screening test for N-glycosylation disorders is the analyses of sialylated isoforms of serum transferrin (Tf) by means of isoelectric focusing (IEF). Two distinct pathological IEF patterns of Tf are observed. A type I pattern is cha- racterized by a decrease of tetra- and an increase of di- and asialotransferrin, whereas a type II pattern shows in addition an increase of tri- and monosialotransferrin. The aims of diploma thesis were: 1) to evaluate reference range for spectrum of sialylated forms of Tf separated by IEF and 2) to perform biochemical and molecular analyses in three patients (P1-P3) with clinical suspicion for CDG. Serum and genomic DNA from three patients with clinical suspicion for CDG and family members of P1 were analysed. Sera from 99 healthy volunteers within the age range of 2-42 years served as a control group. Tf was analysed by IEF with direct immunofixation, SDS-PAGE and Western blot using specific antibody against human Tf (Dako). Profiles of Tf were quantified by AlphaEaseFC software (Alpha Innotech). Thesis advisor: Sara Cordes / The format of our quantity representations is a contentious topic of study in the field of numerical cognition with researchers debating whether we use discrete (i.e. number) or continuous (e.g. area, time, volume or density) cues to make quantity judgements. It has been proposed (through the Sense of Magnitude Theory) that continuous quantities are more perceptual in nature and thus do not require the higher order cognitive processes needed to represent abstract number, making it unlikely that number is tracked in the presence of perceptual quantities. In the current dissertation, I examined claims made by the Sense of Magnitude theory by 1) investigating the accuracy with which we represent continuous quantities and the mental processes we may engage in when representing these quantities and by, 2) comparing the relative salience of discrete and continuous quantities and how this may change across development. In Project 1, I investigated the accuracy with which infants make element size discriminations and whether this ability becomes more precise with age. Project 2 examined the precision with which adults track cumulative area and uncover the process by which they do so. Lastly, Project 3 explored the relative salience of number for preschoolers by assessing their “Spontaneous Focusing on Number.” Together, findings from these three projects undermine claims stating that humans at all stages of development are better at, and prefer to, attend to continuous quantities over discrete number. Instead I propose that this dissertation suggests that humans at all stages of development are strongly attuned to number in their environment. Disasters, Smart Growth and Economic Resilience: An Empirical Analysis of Florida Cities

This dissertation examines the relationship between economic resilience, disaster experience, and smart growth policies at the local government level. The study is based upon three research questions that examine spatial distribution of economic resilience in Florida cities, and examines the impact of disaster experience, and smart growth policies adopted by local governments on economic resilience. Based upon the bounce-forward approach (Cowell, 2013; Klein et al. 2003), economic resilience is defined using three dimensions—economic stability, economic equity, and economic diversity. The spatial analysis is conducted by mapping economic resilience scores across 780 Census Designated Places in Florida through standard deviation method of classification, and conducting cluster-outlier analysis. Results suggest difference in economic resilience within coastal and inland communities—with higher scores mostly situated inland. East Central Florida, Tampa Bay, and South Florida were identified as high economic resilience clusters, and Northwest Florida was identified as low resilience cluster. Impact of disaster experience, and smart growth policies on economic resilience was examined based upon logic of focusing events by Birkland (1997, 2010). Data was collected from the U.S. Census, the National Climatic Data Center, and the Energy Sustainable Florida Communities Survey conducted by Florida State University in 2009. Results suggest significant association between disaster experience of communities and their level of economic resilience, highlighting the importance of community learning in building capacity for resilience after disasters. The results also highlight the need of balancing mitigation and adaptation strategies. Mixed-use development policies, green building certification, and energy savings policies were found to have a significant positive impact on economic resilience.
