Avaliação in vitro dos efeitos da radiação ionizante em tecido ósseo bovino por espectroscopia ATR-FTIR e análise dinâmico-mecânica / In vitro evaluation of ionizing radiation effects in boné tissue by FTIR spectroscopy and dynamic mechanical analysis

A radiação ionizante de fontes de radiação gama ou geradores de raios-x é frequentemente utilizada na ciência médica, como em exames de radiodiagnóstico, radioterapia e esterilização de aloenxertos. A radiação ionizante é capaz de quebrar cadeias polipeptidicas e provocar a libertação de radicais livres, pela radiólise de moléculas de água. A radiação ionizante interage também com o material orgânico a nível molecular, podendo alterar as suas propriedades mecânicas. No caso específico do tecido ósseo, estudos reportam que a radiação ionizante induz alterações nas moléculas de colágeno e reduzem a densidade de ligações cruzadas intermoleculares. O objetivo deste estudo foi verificar as alterações promovidas por diferentes doses de radiação ionizante no tecido ósseo utilizando a técnica de Espectroscopia Transformada de Fourier com Reflexão Total Atenuada (ATR-FTIR), e também a análise dinâmico-mecânica. Amostras de osso bovino foram irradiadas usando irradiador de Cobalto-60, com cinco doses diferentes: 0,01 kGy, 0,1 kGy, 1 kGy, 15 kGy e 75 kGy. Para estudar os efeitos da radiação ionizante sobre a estrutura química do osso foram avaliadas a relação material orgânico por material inorgânico, a relação de sub-bandas de amida I e o índice de cristalinidade. As alterações mecânicas foram determinadas por meio do módulo de elasticidade e do valor do amortecimento. Para verificar se as mudanças químicas e as características mecânicas de osso possuem alguma relação, realizou-se um estudo sobre a correlação entre as análises feitas com os dados espectroscópicos e as análises mecânicas. Foi possível avaliar os efeitos de diferentes doses de radiação ionizante no tecido ósseo. Com a análise por espectroscopia ATR-FTIR, foi possível observar as modificações dos componentes orgânicos e na organização cristais de hidroxiapatita. Também foram observadas alterações no módulo elástico e na tangente de delta (dissipação de energia mecânica). Microtração de sistemas adesivos autocondicionantes em dentina desmineralizada de dentes decíduos após preparo cavitário com laser de Er: YAG: avaliação in vitro / Microtensile bond strength of selfetch adhesive systems in demineralised dentin of primary teeth after Er: YAG laser irradiation: in vitro evaluation

O objetivo deste estudo foi avaliar a resistência à microtração de adesivos autocondicionantes em dentina desmineralizada de dentes decíduos após a irradiação com laser de Er: YAG. Oitenta e dois molares decíduos tiveram a porção oclusal seccionada transversalmente e os espécimes foram desgastados com lixas de carbeto de silício de granulações 360 e 600 até expor a dentina. Os 60 espécimes utilizados no teste de microtração foram preenchidos com resina composta fotopolimerizável na região correspondente à câmara pulpar, isolados com esmalte cosmético mantendo a superfície dentinária exposta e, em seguida imersos em solução desmineralizante por seis dias em estufa a 37° C para indução de lesão de cárie artificial em dentina. Após a desmineralização, os espécimes foram preparados com laser de Er: YAG (n=30) com os parâmetros de 260 mJ, 3 Hz, 47 J/cm2, 0,81W e com broca de aço esférica em baixa rotação (n=30). Sobre as superfícies preparadas foram aplicados três sistemas adesivos autocondicionantes Clearfil SE Bond (Kuraray); Adper™ Easy One (3M ESPE) e Adper™ SE Plus (3M ESPE) e, em seguida foram confeccionados platôs de resina composta Z 250. Após termociclagem (500 ciclos), os corpos de prova foram seccionados produzindo palitos de aproximadamente 1 mm2 de área transversal, que foram submetidos ao teste de microtração e os valores obtidos foram analisados estatisticamente através de ANOVA e teste de Duncan (5%). A análise qualitativa dos tipos de fraturas foi realizada por meio de lupa estereoscópica (aumento de 40X) e da interface adesiva por meio de MEV. Para a análise química da dentina através de espectroscopia no infravermelho por transformada de Fourier (FTIR), foram utilizados discos de dentina após o preparo superficial com a broca (n=5) e com o laser (n=5), e comparadas com as análises de tecido dentinário sadio e desmineralizado obtidas dos mesmos espécimes. Os resultados foram submetidos ao teste estatístico t−Student (5%). O teste de microtração demonstrou que houve diferença significativa entre os adesivos, sendo que o grupo controle (Clearfil e broca) obteve o melhor desempenho comparativamente a todos os grupos avaliados. O preparo com laser de Er: YAG afetou adversamente a adesão para os sistemas adesivos Clearfil SE Bond e Adper™ Easy One em comparação com os grupos preparados com broca. Nos grupos irradiados com laser, o sistema adesivo Clearfil SE Bond foi o que proporcionou o maior valor médio de microtração. Os resultados da FTIR demonstraram que, com a irradiação da dentina desmineralizada, houve um decréscimo na quantidade de matéria orgânica e de carbonato em relação ao grupo da dentina sadia. No substrato preparado com broca, observouse aumento da matéria orgânica e redução de carbonato. Concluise que, em relação aos sistemas adesivos, o Clearfil SE Bond teve o melhor desempenho no teste de resistência à microtração em dentina de dentes decíduos. After thermocycling (500 cycles), specimens were sectioned to produce multiple sticks of approximately 1 mm2 crosssectional area, which were subjected to microtensile test and the data were analyzed with ANOVA and Duncan test at a significance level of 0.05. The qualitative analysis of the types of fractures was performed using a stereomicroscope (X40 magnification) and the morphological analyses of adhesive interfaces were performed by scanning electron microscopy (SEM). Chemical analysis of the dentin by Fourier transform infrared spectroscopy (FTIR) were performed in dentin discs after surface preparation with the steel drill (n = 5) and laser (n = 5) and compared with the values previously obtained from the analysis of sound and demineralized dentin tissue in the same specimens. The results were analyzed by Student\'s t−test at a significance level of 0.05. Regarding the microtensile bond strength there was a significant difference between the adhesive systems, and the control group (Clearfil and steel drill) had the best performance compared to all groups. Er: YAG laser irradiation adversely affected bond strength to the Clearfil SE Bond and Adper™ Easy One compared with the groups prepared with steel drill. Coordination Chemistry of Monocarboxylate and Aminocarboxylate Complexes at the Water/Goethite Interface

<p>This thesis is a summary of five papers with focus on adsorption processes of various monocarboxylates and aminocarboxylates at the water/goethite interface. Interaction of organic acids at the water/mineral interfaces are of importance in biogeochemical processes, since such processes have potential to alter mobility and bioavailability of the acids and metal ions.</p><p>In order to determine the coordination chemistry of acetate, benzoate, cyclohexanecarboxylate, sarcosine, MIDA (methyliminediacetic acid), EDDA (ethylenediamine-N,N’-diacetic acid) and EDTA (ethylenediamine-N,N’-tetraacetic acid) upon adsorption to the goethite (alpha-FeOOH) surface, a combination of quantitative measurements with attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) was utilized.</p><p>Over the pH range studied here (pH 3- 9) all ligands, except for sarcosine, have been found to form surface complexes with goethite. In general, theses were characterized as outer sphere surface complexes i.e. with no direct interaction with surface Fe(III) metal ions. Furthermore, two types of different outer-sphere complexes were identified, the solvent-surface hydration-separated ion pair, and hydration-shared ion pair. For the monocarboxylate surface complexes distinction between these two could be made. At high pH values the solvent-surface hydration-separated ion pair was the predominating complex, while at low pH the surface complex is stabilized through the formation of strong hydrogen bonds with the goethite surface. However, it was not possible to clearly separate between the two outer-sphere complexes for coordination of the aminocarboxylates with the surface of goethite. Additionally, EDDA also formed an inner-sphere surface complex at high pH values. The EDDA molecule was suggested to coordinate to the surface by forming a five membered ring with an iron at the goethite surface, through the amine and carboxylate groups.</p><p>Contrary to the other ligands studied, EDTA significantly induced dissolution of goethite. Some of the dissolved iron, in the form of the highly stable FeEDTA- solution complex, was indicated to re-adsorb to the mineral surface as a ternary complex. Similar ternary surface complexes were also found in the Ga(III)EDTA/goethite system, and quantitative and spectroscopic studies on adsorption of Ga(III) in presence and absence of EDTA showed that EDTA considerably effects speciation of gallium at goethite surface.</p><p>The collective results in this thesis show that the affinity of these ligands for the surface of goethite is primarily governed by their chemical composition and structure, and especially important are the types, numbers and relative position of functional groups within the molecular structure.</p>

Characterization of carbon nanotubes grown by chemical vapour deposition

Carbon nanotubes (CNTs), discovered by Ijima in 1991, are one of the allotropes of carbon, and can be described as cylinders of graphene sheet capped by hemifullerenes. CNTs have excellent electrical, mechanical, thermal and optical properties and very small size. Due to their unique properties and small size, CNTs have a great potential for use in electronics, medical applications, field emission devices (displays,scanning and electronprobes/microscopes) and reinforced composites. CNTs can be grown by different methods from a number of carbon sources such as graphite, CO,C2H4, CH4 and camphor. Under certain conditions, a metallic catalyst is used to initiate the growth. The three main methods used to grow CNTs are: Arc-discharge, laser ablation (LA) and chemical vapour deposition (CVD). In the present work CNTs were grown from a mixture of camphor (C10H16O) and ferrocene (C10H10Fe) using Chemical Vapour Deposition (CVD) and argon was used as a carrier gas. The iron particles from ferrocene acted as catalysts for growth. The substrates used for the growth of CNTs were crystalline Si and SiO2 (Quartz) placed in a quartz tube in a horizontal furnace. Several parameters have been found to affect the CNT growth process. The effects of three parameters: growth temperature, carrier gas (Ar) flow rate and catalyst concentration were investigated in the present work in order to optimize the growth conditions with a simple and economical CVD setup. The samples were characterized using electron microscopy (EM), thermogravimetirc analysis (TGA), Raman and FTIR spectroscopy techniques. It was found that the quality and yield of the CNTs were best at 800°C growth temperature, 80sccm flow rate and 4% catalyst concentration.

Molecular Investigation Of Ptz-induced Epileptic Activities In Rat Brain Cell Membranes And The Effects Of Vigabatrin

The epilepsies are a heterogenous group of symptom complexes, whose common features is the recurrence of seizures. There is no certain therapy for epilepsy. In order to promote new advances for the prevention of epilepsy the molecular mechanism of epileptic activities should be clarified. In the present study the goal is to obtain information for molecular mechanism of epilepsy. To achieve this, molecular alterations from pentylenetetrazol (PTZ)-induced epileptic activities on rat brain tissue and cell membranes were investigated by Fourier Transform Infrared (FTIR) spectroscopy, Fourier Transform Infrared Microscopy and Atomic Force Microscopy (AFM). Moreover, the therapeutic role of an antiepileptic agent vigabatrin (VGB) on epileptic rat brain membranes were examined at molecular level. For better understanding of the action mechanism of PTZ and an antiepileptic drug VGB in cell membranes we firstly studied at model level using multilamellar liposomes (MLVs). We investigated PTZ-DPPC MLVs interactions in terms of lipid phase behavior, order and dynamics and nature of hydrogen bonding around its polar part, using Fourier Transform Infrared (FTIR) spectroscopy, Differential Scanning Calorimetry (DSC), Electronspin Resonans Spectroscopy (ESR) and Steady State Fluorescence Spectroscopy. According to our data, PTZ has no ability to interact with membrane lipids. On the other hand, the results of VGB-DPPC interactions showed that VGB strongly interact with the head group and/or the region near the head of membrane phospholipids. The molecular investigation of PTZ-induced epileptic activities revealed that PTZ-induced seizures cause a decrease in the lipid and protein content, membrane fluidity and glycogen level. They stimulate alterations in membrane packing and the secondary structure of proteins as well as lipid peroxidation. In addition, our results show the transcription of early genes following high dose PTZ administration. All these molecular alterations variatins are only resulted from the consequences of epileptic activities not from convulsant agent PTZ itself. The important finding is that, VGB restored some of the alterations by PTZ-induced epileptic activities on brain cell membrane. For instance, it restored membrane fluidity, lipid peroxidation, phospholipid degradation and changes in membrane organization. However, it was found that VGB has no significant effects on the changes in protein secondary structure.

Study Of Bone Characteristics And Muscle Quality In Metabolic Disorders

Although the effects of diabetes on bone mineral content has been studied, little is known about the structural alterations in collagen, maturation of apatite crystals and carbonate content in diabetic bone. The first part of this study aimed to investigate the mineral and organic properties of cortical, trabecular and growth plate regions of rat femur tissues in type I diabetes using FTIR microspectroscopy and Vickers microhardness test. A decrease in mineral content (degree of mineralization), decrease in microhardness, increase in carbonate content, increase in size and maturation of hydroxyapetite crystals, which are the implications of increased osteoporosis, were observed in diabetic bone. In addition, a decreased carbonate substitution into bone apatite and an increase in labile type carbonate was observed in diabetic bone. There was a decrease in the level of crosslinking of collagen in cortical and trabecular regions of diabetic femurs, implying a decrease in bone collagen quality that may contribute to bone fragility. Recent evidence implies that intramyocellular lipid accumulation is directly correlated with insulin resistance, a key parameter in the generation of obesity. The second part of this study is mainly focused on the determination of the structural and compositional characterization of macromolecules of longissimus dorsi and quadriceps muscles of Berlin fat mouse inbred (BFMI) lines using ATR-FTIR spectroscopy and FTIR microspectroscopic imaging, together with the quantification of fiber specific distribution of lipids in these muscles by the use of confocal microscopy. The study groups included 10 weeks old standard breeding diet fed (juvenile) and 20 weeks old high fat diet fed control and BFMI lines. The results revealed the loss of unsaturation in lipids, increased triglyceride content, increased amount of lipids having shorter chain length, increased lipid peroxidation and fiber specific accumulation of lipids in type IIa and intermediate fibers in skeletal muscles of both 10 weeks old and 20 weeks old BFMI lines, emphasizing their obese phenotype. However, the alterations were more prominent in skeletal muscles of 20 weeks old high fat diet fed BFMI lines, displaying a more severe obesity phenotype. The results of the characterization revealed that BFMI860 and BFMI861 lines are convenient models for the study of spontaneous obesity and studies to enlighten the genetic basis of obesity.

The dissertation work focuses on the whole route of material development starting from the investigations of properties of the initial (raw) HBPs to their applications. Each research step is given in a separate chapter to enhance attention to various aspects of the aim of the work. Thus, every chapter is started with an introduction. After that, the methods applied and experimental procedure are described. Next part tries to give the comprehensive description of the results obtained. At the end of the chapter, the main points are summarized. The Chapter 1 gives the theoretical description of the main experimental techniques used in this work. In Chapter 2 the chemical (chemical composition, purity, typical structure elements) and physical (glass transition temperature, the temperature of the maximum decomposition, the thermal stability at the high temperatures, molar mass, polydispersity and possible aggregation in solution) properties examined by different techniques of polymer analysis are described. The Chapter 3 is divided into three separate parts: In Chapter 3.1 the description of the formation and modification of inter- and intramolecular hydrogen bonds of hydroxyl terminated HBP is presented to reveal the information of hydroxyl groups re- and/or association due to the high temperatures applied. In Chapter 3.2 the nature of the solid-liquid interface of HBP thin films have been studied by different surface sensitive techniques with respect to further protein adsorption investigations. In Chapter 3.3 the strategy for the fabrication of surface attached carboxyl terminated HBP using &amp;quot;grafting to&amp;quot; technique is developed. The Chapter 4 consists of two parts: The first (theoretic) part outlines the basic principles of protein chemistry, factors influencing on the protein molecule stability in aqueous medium, the mechanism of protein adsorption and forces involved in the adsorption process. In the second part the combination of different in situ techniques was applied to obtain a comprehensive description of complex adsorption processes of protein molecules on different polymer surfaces.

Differenzierung von humanen Plattenepithelkarzinomen mittels IR-mikrospektroskopischem Imaging

Die Dissertation befasste sich mit der Entwicklung einer neuen diagnostischen Methode für in-situ-Gewebeuntersuchungen. Der Ansatzpunkt war die Untersuchung von pathologischen Veränderungen im Gewebe, die sich biochemisch in den Zellen widerspiegeln und deshalb mit schwingungsspektroskopischen Methoden, wie der IR-Spektroskopie, erfassbar sind. Das Ziel der Arbeit war die IR-spektroskopische Charakterisierung und Klassifizierung von benignen, präkanzerösen und malignen Geweben mittels chemometrischer Algorithmen auf der Basis multivariater Informationen der IR-Spektren. Um komplexe spektrale Veränderungen zu charakterisieren und die Ergebnisse statistisch abzusichern, ist für jeden Gewebetyp eine Vielzahl an Spektren erforderlich. Daher wurde zur Spektrenakkumulation das IR-mikrospektroskopische Imaging mittels Focal Plane Array Detektor (FPA) genutzt. Die Herausforderung liegt in der Datenanalyse. Der große Datenumfang macht die Anwendung multivariater Algorithmen notwendig. Angewendet wurden Clusteralgorithmen zur Spektrendifferenzierung und die SIMCA (Soft Independent Modelling of Class Analogies) zur Spektrenklassifizierung. Die Validierung der Ergebnisse erfolgt über die histologische Untersuchung der nach der spektroskopischen Messung gefärbten Gewebedünnschnitte. Die genaue Vorgehensweise bei der Auswertung wird in dieser Arbeit anhand humaner Gewebeproben dargestellt. Die untersuchten Plattenepithelkarzinome und Adenokarzinome gehören zu den epithelialen Tumoren, die oralen bzw. zervikalen Ursprungs sind. Die Übertragbarkeit der spektralen Modelle wurde mit Gewebeproben mehrerer Patienten innerhalb einer und zwischen verschiedenen Tumorarten untersucht. Das ist ein erster Schritt zum Einsatz spektroskopischer Methoden in der medizinischen Forschung und Diagnostik.
