Redução de sulfato em biorreator operado em batelada e batelada alimentada seqüenciais contendo biomassa granulada com agitação mecânica e Draft-Tube / Sulfate reduction in bioreactor in sequencing batch and fed-batch containing granulated biomass with mechanical stirring and draft-tube

O presente projeto avaliou um reator anaeróbio operado em batelada e batelada alimentada seqüenciais (ASBR), em ciclos de 8 horas, utilizando biomassa granulada e agitação mecânica em um draft-tube, alimentado com água residuária sintética (500 mgDQO/L), contendo sulfato em diferentes relações DQO/[\'SO IND.4\' POT.2-\']. Em todos os ensaios o reator apresentou uma operação estável, produzindo alcalinidade e com concentração de ácidos voláteis totais em níveis adequados. Para os tempos de alimentação de 10 min, 3 h e 6 h, respectivamente, as eficiências de remoção de sulfato foram de 30, 72 e 72% nas operações nas quais o reator foi alimentado com uma relação DQO/[\'SO IND.4\'POT.2-\'] de 1,34. Nos ensaios nos quais o reator foi alimentado na relação DQO/[\'SO IND.4\'POT. 2-\'] de 0,67, as eficiências para a redução de sulfato foram de 25, 58 e 55%, respectivamente. Na operação com relação DQO/[\'SO IND.4\'POT.2-\'] de 0,34, as eficiências para redução de sulfato foram de 23, 37 e 27%, respectivamente. Desta maneira, pode-se concluir que as operações em batelada alimentada favoreceram a remoção de sulfato, enquanto foi observado que nas operações em batelada a remoção de matéria orgânica atingiu melhores eficiências. / This present work evaluate an anaerobic sequencing batch reactor (ASBR), fed in batch and fed-batch, and cycles of 8 hours, using granulated biomass and mechanical stirring in a draft-tube, fed with synthetic wastewater (500 mgCOD/L), enriched with sulfate in some COD/[\'SO IND.4\'POT.2-\'] relations. In all operations the reactor showed a stable operation, producing alkalinity and maintaining the volatile acids in adequate levels. Considering the fed periods of 10 min, 3 h and 6 h, respectively, the removal efficiencies of the sulfate was 30, 72 e 72%, in the operations when the reactor was fed with a COD/[\'SO IND.4\'POT.2-\'] relation of 1,34. In the essays when the reactor was fed in COD/[\'SO IND.4\'POT.2-\'] relation of 0,67, the efficiencies of the sulfate reduction was 25, 58 e 55%, respectively. When the reactor was operated with COD/[\'SO IND.4\'POT.2-\'] relation of 0,34, the efficiencies of sulfate reduction 23, 37 e 27%, respectively. Thus, is possible to conclude that the operations in fed-batch increased the efficiency of sulfate removal, at what time was observed that in batch operations the organic matter removal attained improved efficiencies.

Influência da estratégia de alimentação no desempenho do reator anaeróbio em batelada seqüencial contendo biomassa imobilizada / Influence of feeding strategy on the performance of anaerobic sequencing batch reactor containing immobilized biomass

Submeteu-se um reator anaeróbio operado em batelada seqüencial a diferentes tempos de enchimento, com o objetivo de verificar-se o desempenho do sistema. O reator, construído em acrílico e com capacidade de 6,3 L, tratou por ciclo um volume de 2,5 L de esgoto sintético com concentração de aproximadamente 500 mgDQO/L. O sistema foi operado à temperatura de 30 graus mais ou menos 1, com duração de 8 h para cada ciclo (tC) e agitação de 500 rpm implementada mecanicamente. Cubos de espuma de poliuretano foram usados como suporte para imobilização da biomassa. No início de cada ciclo, o reator foi carregado com 60 % do volume do esgoto; o restante foi preenchido durante tempos de enchimento (tF) que caracterizaram as diferentes estratégias de alimentação. Os resultados obtidos demonstram que para razões tF/tC <= 0,5 o sistema alcança médias de remoção de matéria orgânica acima de 75 % e 70% para amostras filtradas e não filtradas, respectivamente. Nos ensaios em que tF/tC > 0,5, registrou-se perda na eficiência e formação de polímeros extracelulares, apesar da estabilidade observada. O estudo contribuiu para o melhor entendimento do sistema e definição de técnicas de operação em futuras aplicações. / An anaerobic sequencing batch reactor was operated at different fill times with the objective to asses system performance. The reactor, made of plexiglas and with a capacity of 6.3 L, treated per cycle 2.5 L of synthetic wastewater with a concentration of nearly 500 mgCOD/L. The system was operated at a temperature of 30oC, with 8-h cycles (tC) and mechanical agitation rate of 500 rpm. Cubic particles of polyurethane foam were used as support material for anaerobic biomass immobilization. At the start of each cycle, the reactor was fed with 60 % of the wastewater volume, the remainder was filled at different feeding times (tF) that characterized the different feeding strategies. The results obtained showed that for a ratio of tF/tC <= 0.5 the system attained average filtered and non-filtered substrate removal efficiency of 75 % and 70 %, respectively. Ratios of tF/tC > 0.5, resulted in loss in efficiency and formation of extra cellular polymers, in spite of system stability. The study contributed to a better understanding of the system and definition of operational techniques for future applications.

Os ecossistemas marinhos do Estado de São Paulo sofrem intensa pressão antrópica. Considerando que os estudos tróficos de peixes são essenciais para o manejo desses ecossistemas e para a gestão pesqueira, foi construído um banco de dados alimentares de peixes para sintetizar o conhecimento já obtido na área. Foram inseridos, individualmente ou agrupados, dados primários (não-publicados) e secundários (publicados), representando 142 espécies. Com base nesse banco, foi comparada a dieta dos peixes nos sistemas de plataforma continental do litoral Norte, plataforma continental do litoral Centro, canal de São Sebastião, baía de Santos e complexo estuarino-lagunar de Cananéia-Iguape. Foi verificado que a dieta variou entre os sistemas, provavelmente devido à variação na disponibilidade alimentar. Na plataforma Norte, foi analisada a variação sazonal (outono/inverno e primavera/verão) da dieta e verificou-se que a diferença interespecífica sobrepujou a diferença intraespecífica temporal. A dieta da corvina (Micropogonias furnieri), espécie abundante e de grande importância comercial, foi analisada em detalhe e concluiu-se que ela possui estratégia alimentar mista, consumindo majoritariamente alguns itens (poliquetas e camarões) e esporadicamente outros. Foram elaboradas considerações sobre a publicação de dados científicos brutos e sobre a metodologia aplicada aos estudos de conteúdo estomacal. / The marine ecosystems of São Paulo State are under intense anthropic pressure. Considering that trophic studies of fishes are essential to the management of ecosystems and fisheries, a fish stomach content database was built in order to summarize the available data in the region. Primary (unpublished) and secondary (published) data were inserted, representing 142 species. Based on this database, diet compositions of fishes were compared among 5 marine systems of São Paulo State (North continental shelf, Central continental shelf, São Sebastião channel, Santos bay and Cananéia lagoon estuarine system). It was observed that diet composition varied among systems, probably associated to the food resource availability. Seasonal comparison (autumn/winter and spring/summer) performed in the North continental shelf showed that the interspecific variation was larger than the intraspecific temporal variation. Diet of Micropogonias furnieri, an abundant and commercially important species, was analyzed in detail and it was concluded that it has a mixed feeding strategy, consuming mostly few items (polychaetes and shrimps) and sporadically many others. Some considerations on scientific raw data publication and on methodology of stomach content studies were made.

La bioconversion des biomasses lignocellulosiques est actuellement un grand défi pour le développement de technologies de bio-raffinage. Le manque de connaissances des mécanismes de liquéfaction et de saccharification est l’un des principaux facteurs qui pénalisent le développement des procédés de bio-raffinage. Ce travail est centré sur le développement d’analyses physiques et biochimiques in-situ (viscosimétrie, focus beam reflectance measurement) et ex-situ (rhéometrie, granulométrie laser, morphogranulométrie, sédimentation…) pour améliorer la compréhension des mécanismes de déstructuration desfibres lignocellulosiques et caractériser les cinétiques de libération de carbone fermentescible. Des substrats modèles (cellulose microcristalline, papier Whatman) et industriels (pâte à papier, bagasse de canne à sucre) ont été utilisés avec différentes conditions d'hydrolyse (1% à 30%w/v, 0.1 à 0.5mL enzyme/ g cellulose). Les résultats obtenus ont permis:- de proposer et de valider les mesures in-situ de la viscosité de la suspension et de la distribution des longueurs de corde des particules, ainsi que sa conversion en distribution de diamètre.- de montrer l'impact de la nature et de la concentration de substrat et des ratios enzyme/substrat sur les évolutions des paramètres physico-biochimiques lors de l'hydrolyse. Ces effets ont été quantifiés sur les limitations de transfert.- d'établir un modèle phénoménologique de comportement rhéologique des suspensions initiales- de montrer que les cinétiques physico et bio-chimiques sont des cinétiques du second ordre- de montrer que, pour des hydrolyses à haute teneur en matière sèche, on peut réduire considérablement la limitation des transferts liée aux hautes concentrations et contrôler la cinétique de production de glucose par une stratégie d’ajouts cumulés desubstrat. / In the context of biofuels and chemicals production of petroleum substitutes from renewable carbon, bioconversion of lignocellulose biomasses is currently a major challenge. The limited knowledge of liquefaction and saccharification mechanisms stands as the main factor which penalizes bio-refinery progress. The present work is centred on the development of in-situ(viscosimetry, focus beam reflectance measurement) and ex-situ (rheometry, diffraction light scattered, morphometry, decantation…) physical and biochemical analysis to expand our understanding of the destructuration mechanisms of lignocellulose fibres and to characterise the release kinetics of fermentable cellulosic carbon. Model (microcrystalline cellulose,Whatman paper) and industrial (paper-pulp, sugarcane bagasse) lignocellulose matrices under a large range of hydrolysis conditions (1% up to 30%w/v and 0.1 up to 0.5mL enzyme/g cellulose) were studied during 24h hydrolysis experiments (pertinent period to appreciate transfer limitations). Our scientific results allow:- to propose and validate the in-situ measurements of the suspension viscosity and chord length distribution together with its conversion into particle size distribution.- to demonstrate the impact of the substrate nature and concentration and of the enzymatic ratios on the evolution of physical- and biochemical parameters during hydrolysis. Their impacts on transfer phenomena were quantified.- to establish phenomenological models for rheological behaviour of initial suspensions.- to describe all physical (viscosity, particle size) and biochemical (substrate and product) kinetics by second order reaction models.- to demonstrate that, for high dry matter concentration hydrolysis, a cumulative feeding substrate strategy allows considerably reducing the transfer limitations linked to high concentrations and to control the glucose production kinetics.

The Influence of Management Strategies on the Water Productivity in Dairy Farming and Broiler Production

Die Wasserproduktivität in der Tierhaltung ist von vielen Faktoren abhängig. Die Futterproduktion hat den größten Anteil am Wasserbedarf von tierischen Produkten. Weitere Einflussfaktoren sind die Leistung, die Reproduktion und der Gesundheitsstatus der Tiere, das Management und die Haltungsbedingungen. In dieser Arbeit sollte untersucht werden, wie sich diese Faktoren auf die Wasserproduktivität von Milch und Geflügelfleisch in Nord-Ost-Deutschland auswirken. Zehn unterschiedliche Futtermittel wurden hinsichtlich ihres Wasserbedarfes untersucht. Aus diesen Futtermitteln wurden die Rationen für die Tiere erstellt. Die Milchleistung der Kühe wurde zwischen 4.000 und 12.000 kg Milch pro Kuh und Jahr in 2.000 kg Schritten variiert. Für jedes Leistungsniveau wurden zwölf verschiedene Fütterungsstrategien untersucht, welche auf der Erhöhung einzelner Bestandteile der Ration basieren. Der Wasserbedarf von Leitungswasser im Stall wurde mit 38 Wasserzählern ermittelt. Für die Wasserproduktivität des Geflügelfleisches wurden vier verschieden intensive Mastverfahren untersucht. Die Wasserproduktivität steigt mit steigender Milchleistung der Kühe. Das Maximum wird bei 10.000 kg Milch pro Kuh und Jahr und Rationen mit einem hohem Gras- bzw. Maissilageanteil erreicht. Die Kühe, die im automatischen Melksystem gemolken wurden, nahmen mehr Tränkwasser zu sich, als die Kühe im Fischgrätenmelkstand. Dies ist durch die höhere Milchleistung bedingt. Im automatischen Melksystem wurden im Mittel 28,6 Liter Reinigungswasser pro Kuh und Tag benötigt. Für die Reinigung des Fischgrätenmelkstandes wurden 33,8 Liter pro Kuh und Tag genutzt. Die untersuchten Broilermastverfahren zeigten keine Unterschiede hinsichtlich der Wasserproduktivität. Die intensivere Aufzucht und bessere Futterverwertung wurde durch eine niedrigere Wasserproduktivität des Futters kompensiert. Der Anteil des technischen Wassers macht in der Milchkuh- und Broilerhaltung nur einen kleinen Teil am Gesamtwasserbedarf aus. / Livestock production is the main user of water resources in agricultural production. Water is used in animal production for producing feed, watering the animals, and cleaning and disinfecting barns and equipment. The objective of this dissertation was to quantify the effects of management strategies, such as feeding, intensity of production and the replacement process on the water productivity of milk and poultry meat in Germany. Water productivity in milk and broiler production systems was calculated based on the methodology of Prochnow et al. (2012). Own measurements of the drinking and cleaning water demand in milk production were conducted in a dairy cow barn. The study was based on site conditions of North-East Germany with common variations in farm operations. The feed production is the main contributor to water input in dairy and poultry production. The water productivity of milk increased with an increasing milk yield. The most beneficial conditions related to water productivity in dairy farming were found to be with a milk yield of approximately 10,000 kg fat corrected milk and a grass silage and maize silage based feeding. The total technical water use in the barn makes only a minor contribution to water use. Former regression functions of the drinking water intake of the cows were reviewed and a new regression function based on the ambient temperature and the milk yield was developed. In broiler production the intensification of the fattening systems did not increase water productivity. An increase of water productivity in animal production can be achieved with various management strategies with their specific influence on the production process. The feed management should be a focus of the strategies.
