An investigation into the effects of dystrophin on the lateral mobility of muscle membrane components

Dystrophin is the product of the Duchenne Muscular Dystrophy gene locus, whose absence results in progressive skeletal muscle breakdown. Despite considerable work on the localisation of dystrophin and its associated complex, its role in muscle function remains unclear. In the light of the structural and mechanical instability of the dystrophic membrane, the idea was tested that dystrophin might impart membrane integrity and strength by anchoring membrane proteins and/or delineating the surface into specialised subcellular functional domains. Specifically, because dystrophin shows high sequence, structural and spatial similarities to the cytoskeletal protein spectrin; and because spectrin is proven to sterically restrict protein lateral diffusion through a subplasmalemmal network; the capacity of dystrophin to act as a 'molecular fence' to membrane diffusion was studied by comparing lateral mobility of membrane glycoproteins by fluorescence photobleach recovery in mdx and normal tissue. Secondly, as dystrophin has been proven to interact directly with proteins of the dystrophin associated glycoprotein complex in vivo, experiments addressed whether specific binding and immobilisation of the complex by dystrophin at the membrane was essential for function. Finally, given the homology of dystrophin and spectrin, the presence of dystrophin at the neuromuscular junction, and the importance of spectrins in immobilisation of voltage gated sodium channels in the nervous system, the role of dystrophin in regulating voltage gated sodium channel distribution at the neuromuscular junction was investigated. The results show that membrane glycoproteins were immobile in the presence and absence of dystrophin, suggesting dystrophin is not an essential molecular fence component. Alternatively, viability may have been the major influence on protein and lipid diffusion in these fibres and suggestions are made as to how this may be recognised and overcome for subsequent investigation. Three novel exon specific anti-dystrophin peptide antibodies were generated during the work that will be useful for studies into Duchenne muscular dystrophy in general, and dystrophin revertant fibres in particular.

Architecture has often been described as a reflection of society and its current human condition. The converse opinion states that architecture continues to play a major role in constructing that very same society, long after it is built. These statements are most evident where architecture has been intentionally used as a tool of dominance and control. Within a new democratic South African society, the invisible walls of Apartheid have been replaced by physical fences used to ensure ‘the public’ of their safety and security. This has resulted in the fence fetish. The fence fetish is a common phenomenon around the world, as global architecture trends promote autonomy, object buildings and the urban bourgeois. Habitat preferences of the eastern fence lizard, Sceloporus undulatus, in southwestern Virginia

Habitat preference of the eastern fence lizard, Sceloporus undulatus, was investigated in southwestern Virginia. Habitat features were measured at 158 lizard-centered plots and at paired random plots. Landscape-level variables, southerly aspect and mixed forest type, distinguished lizard-centered from random sites. Hatchlings were associated with relatively high temperature at perch height (23 °C), relatively high amounts (per 1- m2) of coarse woody debris (15%) and bare ground (15%), and relatively low amount of litter (34%). Adults and juveniles were associated with a relatively high number of rocks (22 per 0.01 hectare) and amount of coarse woody debris (9% per 1- m2). Habitat preferences were modeled with a Geographic Information System (GIS) using landscape-level variables and with logistic regression and Akaike's Information Criterion using site-level variables. The best-fitting site-level model for adults/juveniles included % CWD. The best-fitting model for hatchlings included % CWD and number of rocks, and the second best-fitting model also included % litter. Landscape (both classes) and site-level models (adult/juveniles only) were tested at 15 GIS-predicted "suitable" study areas and at 15 GIS-predicted "unsuitable" areas. Site-level models for hatchlings were tested with independent data collected at two study areas. Sixteen lizards were found at "suitable" areas and one at an "unsuitable" area; the GIS-based model was a good predictor of lizard presence at the landscape level. The best-fitting site-level models for adults/juveniles and hatchlings were poor predictors of lizard presence while the second best-fitting hatchling model was a good predictor of hatchling presence. / Master of Science

Modeling of an Underground Mine Backfill Barricade

In this thesis finite element analyses were performed to investigate the behavior of fill fences installed in underground mines to retain Cemented Paste Backfill (CPB) pressure. For this purpose, two fill fences installed and tested in the Cayeli mine in Turkey were modeled using a 2-D nonlinear finite element analysis program, Augustus-2, and a 3-D nonlinear finite element analysis program VecTor4, and the results were compared with measured field data. Different models were employed representing the material properties, boundary conditions, reinforcement ratio, and geometric properties, and it was found that boundary conditions (stiffness of surrounding rocks) has the highest influence on the pressure capacity of the fence among the other factors. The accuracy of the Augustus-2 program was investigated by modeling and comparing the analytical response with test results of 12 axially restrained beams tested by Su et al. (2009).
