Technologinių veiksnių įtaka chloramfenikolio kiekio kitimui pieno produktuose / The influence of chloramphenicol on the milk technological properties

Tiriamojo darbo tikslas - ištirti pagrindinių technologinių veiksnių (gaminant įvairius pieno produktus) įtaką chloramfenikolio suardymui ar jo sumažinimui. Lietuvoje daug dėmesio skiriama antibiotiko chloramfenikolio likučių aptikimui superkamame piene. Kadangi tai vienas iš stipriausių, plataus veikimo spektro, antibiotikų, todėl labai svarbu, kad per pieno produktus šis antibiotikas nepatektų į žmogaus organizmą. Vykdant šį darbą buvo ištirta ar chloramfenikolio koncentracija gali pasikeisti (sumažėti) veikiant įvairiems technologiniams veiksniams, kurie vykdomi gaminant pieno produktus. Paaiškėjo, kad pasterizacijos temperatūra nuo 63 iki 90°C neturėjo įtakos chloramfenikolio kiekiui. Jo koncentracija, prieš ir po temperatūrinio apdorojimo, nepakito. Išlaikymas pieno mėginių su chloramfenikoliu esant 92°C temperatūrai iki 15 min taip pat neturėjo įtakos jo koncentracijai. Pieno laikymas, atšaldžius, esant 8°C temperatūrai ir laikant iki 50val. neturėjo įtakos chloramfenikolio kiekiui. Struktūriniai pieno koloidinės sistemos pokyčiai, pieno mėginius užšaldant (-20°C) ir atšildant (+ 10°C), kurie dalinai gali vykti valgomųjų ledų gamybos metu, bei laikant ar transportuojant pieno mėginius prieš tyrimus, taip pat nepakeitė chloramfenikolio koncentracijos. Rauginant su raugais (Yomix 433, Yomix 860, XT 312) ir su atskiromis pieno rūgšties bakterijomis (Str. lactis, Str. thermophylus, L. acidophylus) pieno mėginius su chloramfenikoliu, nustatyta, kad jo koncentracija... [toliau žr. visą tekstą] / Great attention is paid to the detection of the leftover antibiotic chloramphenicol in the bought up milk in Lithuania. This antibiotic is amongst the strongest ones and is of a wide activity range. Therefore, it is of the utmost importance to prevent its access into the human organism through dairy products consumption. The concentration of the chloramphenicol was analyzed during the study. The analysis focused on whether the concentration can change, i.e. decrease, under the influence of various technological factors, which are set in action during the production of the dairy products. The results showed thai the pasteurization temperature (from 63°C to 90°C) had no impact on the amount of chloramphenicol as its concentration remained the same both prior and alter the temperature processing. Keeping the milk samples with chloramphenicol in the temperature of 92°C up to 15 minutes had no influence either. Furthermore, storing the cooled milk in the temperature of 8°C up to 50 hours did not have any effect upon the amount of chloramphenicol as well. Monitoramento e operação de cultivos de células animais em sistemas de perfusão. / Monitoring and operation of animal cell cultures in perfusion systems.

Os processos biotecnológicos com células animais tem despertado interesse crescente devido à sua capacidade de sintetizar moléculas complexas de altíssimo valor agregado, mesmo exigindo condições especiais de cultivo. Torna-se portanto fundamental o correto monitoramento de tais processos, para se manter seus parâmetros dentro de limites desejados. O objetivo deste trabalho foi efetuar cultivos de células animais em processo de perfusão, aos quais se acoplavam sistemas de monitoramento visando melhor controle, de modo a contribuir para o aumento da produtividade. Realizaram-se ensaios com células BHK e hibridoma em biorreator de 12L em alta densidade celular (20E6 cel/mL), para produção de fator de coagulação FVIII e anticorpo anti-TNF. O processo de perfusão foi analisado, buscando-se melhoria em seu controle através da correta estimativa da concentração celular. O sistema de transferência de oxigênio através de membranas de silicone foi estudado, tendo-se elaborado uma simplificação para a estimativa da velocidade de consumo de oxigênio, baseando-a na pressão média da membrana de silicone, e tendo-se determinado a influência da concentração de oxigênio no crescimento e na produção e seu valor limitante (entre 5 e 10%). Por fim, estudou-se o efeito da osmolalidade do meio no processo, tanto com relação ao crescimento e consumo de substratos, como na produtividade. Observou-se o aumento do tamanho da célula com o aumento da osmolalidade (até 470mmol/kg) e como este fato influencia o cálculo da velocidade específica de produção. Tentou-se sem sucesso fazer um monitoramento on-line deste aumento de tamanho, com o uso da sonda Lasentec FBRM. No entanto esta se mostrou capaz de estimar a concentração celular. O trabalho demonstra a importância de se efetuar um adequado monitoramento do processo para garantir a correta operação, em especial em cultivos com células animais em perfusão. / Animal cell culture technology has become apparent, due to its capacity to synthesize highly valuable complex molecules, even requiring special attention. In this scenario, adequate process monitoring becomes essential to keep its parameters well controlled. The aim of this work is to perform perfusion cultures with animal cells, and couple them to monitoring systems, in order to improve the efficiency of the control and consequently productivity. Runs were realized with BHK and hibridoma cells in a 12-L bioreactor at high cell density (20E6 cel/mL), to produce clotting factor FVIII and antibody anti-TNF. The perfusion process was analyzed, to enhance its control through reliable cell density estimate. Oxygen transfer system through silicone tubing was studied, resulting in a simple way to estimate oxygen uptake rate and establishing the influence of oxygen partial pressure in the growth and production. At last, medium osmolality effect was investigated related to growth, substrate consumption, and productivity. Cell enlargement was observed at higher osmolalities (till 470mmol/kg) and its results over the specific production rate were analyzed. The cell size change was measured off-line. Lasentec FBRM probe could not follow the size change, but showed capable to estimate cell density This work makes evident the importance in having adequate monitoring systems, in order to assure correct process operation, and specially cell culture perfusion processes.

Dissolved carbon dioxide driven repeated batch fermentation

Dissolved carbon dioxide driven repeated batch fermentation has been performed under four glucose concentrations: ~150, ~200, ~250 and ~300 g glucose l-1, with three dissolved carbon dioxide (DCO2) control conditions: without DCO2 control, with DCO2 control at 750 and 1000 mg l-1 levels. No residual glucose was observed under all performed fermentation conditions, and the repeated batch fermentation system could be operated by a computer as self-cycling system. The collected fermentation results presented that, under the same feeding concentration, ethanol concentration in the presence of DCO2 control was significantly lower than that in the absence of DCO2 control; and a higher biomass concentration in the presence of control was observed in this comparison as well. A higher biomass concentration resulted in a shorter fermentation time, which contributed to a higher ethanol production rate. The highest final ethanol concentration was observed as 113.5 g l-1 at 1000 mg DCO2 l-1 control level under ~300 g glucose l-1 condition, where the lowest ethanol production rate of 1.18 g l-1 h-1 was observed. The highest ethanol production rate was 4.57 g l-1 h-1 and its corresponding ethanol concentration was 66.7 g ethanol l-1 at 1000 mg l-1 DCO¬2 control level under ~200 g glucose l-1 condition. For all fermentation conditions, the viabilities of yeast at the end of fermentation were maintained at near 90% where their corresponding final ethanol concentrations were lower than 100 g l-1. As soon as the final ethanol concentration at the end of each cycle was greater than 110 g l-1, its corresponding viability decreased to ~70%. The ethanol conversion efficiency was maintained at ~90% and ~65% in the absence and presence of DCO2 control, respectively. Based on the changing of biomass concentration profiles in the stabilized cycles, two cell growth phases could be identified in the absence of DCO2 control, and only one cell growth phase was noticeable in the presence of DCO2 control cases. Meanwhile, a sudden decline of DCO2 readings at the end of fermentation was constantly observed in both of in the absence and in the presence of DCO2 control cases, which resulted in developing two control algorithms to determine self-cycling time. Comparison of carbon balance analysis between in the absence and in the presence of DCO2 control suggested that the availability of DCO2 control might alter the metabolic flow during fermentation; and the figure of ethanol concentration against fermentation time illustrated that the changing of DCO2 control level did not affect fermentation results, significantly. Moreover, comparisons of ethanol production rate between different processes and different initial glucose concentrations concluded that the ethanol production rate in the presence of DCO2 control was generally higher than that in the absence of DCO2 control under the same glucose concentration; and the ethanol production rate was decreased with the increasing of glucose concentration under the same DCO2 control condition. The experiment results were scaled up to 106 L as a sample analysis in production scale, which suggested that the fermentation with ~200 g glucose l-1 feeding concentration in the absence of DCO2 controlled would provide best profits in the all fermentation conditions.

This research was conducted to determine the effects of M. fimbriolata damage levels in sugarcane on cane quality, yeast stability, fermentative process and ethanol production. Spittlebug attack negatively affected cane quality, Sucrose content, Total Reducing Sugars and juice purity were lower and total, volatile acidity and lactic acid contents and phenolic compounds were higher at higher damage levels. Also, we observed that the clarification process does not remove totally phenolic compounds of juice. Pest attack also affected microorganisms in fermentation process, reducing yeasts cell and bud viabillty. There was an increase in bacterial contamination, resulting in wines with lower alcohol content and fermentation efficiency.

Pesquisas recentes indicam que a praga pode causar perdas de produção e reduzir a qualidade tecnológica da matéria-prima destinada ao processamento industrial. A presente pesquisa tem como objetivo estudar a influência dos danos promovidos pela cigarrinha-das-raízes em cana-de-açúcar sobre a qualidade da matéria-prima destinada ao processamento, estabilidade das leveduras, desempenho fermentativo, e produção de etanol.
