With the development of writing and the advent of new technical devices, it creates new ways to store and transport information. The memory is no longer restricted to the limits of the here and now of the subject and undergoes transformations. In this scenario, the media start to play an important role in publicizing and construction of embodiments of memory. This study aims to analyze the conformation of the memory of political groups during the process of receiving audiovisual fiction. For this purpose, a corpus of four chapters of the soap opera Love and Revolution was used as a cognitive device for remembering. The television series, broadcast by SBT between April 2011 and January 2012, went back to the beginning and development of the military dictatorship in Brazil, in the cities of Rio de Janeiro and S?o Paulo. Three militants of various affiliations Communists who acted against the regime in Rio Grande do Norte and neighboring states, were participants in this study. Using the method of oral history, the research was divided into two stages: in-depth interviews, which dealt with the history of life of employees with the militancy in the Communist parties and other social movements, and the assistance of a drive dramatic soap opera Love and Revolution. Comparing these two phases of the study, we analyze the flow of mediations that crossed memories of militancy and media framework; shifts the narrative of remembrance during reception, and the opposition between memory and represented the experiences of the receivers / Inst?ncia constru?da subjetiva e socialmente, a mem?ria n?o ? um fen?meno natural, mas um espa?o de disputa entre v?rias organiza??es sociais pelo controle e legitima??o de um passado.

To understand this phenomenon of meaning production in the cordel media, we used hermeneutics as a method and applied the general theory of interpretation in six chosen leaflets. We worked with a constructivist perspective that grounds the discussion of everyday reality and fiction, concepts that are raised around the essence of the character that is real, but it is also part of the creative activities of poets, how both are interrelated and constitute the understanding that individuals have the real. From the analyzes, we realize that each poet presents the fields of significance of Mr. Lunga in a different way, based on subjectivity, intention and mediations between each of them and the discourses they produce. Each cordelist contributes in his own way of significance for the construction of the imaginary Mr. Lunga. The speech of the cordelists contains a number of elements that aim to legitimize as truth the actions described. In this confrontation, our goal in this work is to understand the construction of the fields of signification, where these discourses are located, the production of meaning around a character who is not in a finite field, but transits through many of them, making the boundaries between reality and fiction dynamic / Neste trabalho pretendemos, a partir do caso de Seu Lunga, um personagem da cultura nordestina, cujas hist?rias circularam oralmente at? virarem versos de cordel, compreender o folheto, produto cultural t?o significativo, como m?dia, com caracter?sticas de linguagem espec?ficas, que atuam como formas de constru??o e transmiss?o de realidades.

The present study aims to develop a model of analysis of female protagonism in audiovisual narratives. The model intends to identify the presence of proactive female protagonism and its performance within the narrative. Based on the concept of Proactive Feminine Protagonism, the female protagonist is an active participant in the plot, playing or not a leadership role, represented in an equivalent way to the male protagonists, and exerting influence and relevance in the events of the narrative. The chosen cut was the universe of science fiction, in which the dystopic genre is evidenced in the audiovisual productions. The films Hunger Games (2012) are used as analysis objects; Catching Fire (2013); The Hope: Part 1 (2014); The hope: the end (2015). To analyze the narrative arc of these dystopias, we use the scheme adapted by Vogler (2015) from Joseph Campbell's Hero's Journey. In the analysis of the protagonist Katniss (Hunger Games); contextualizes the formation of identity based on Castells (2013) and Butler (2016). Using the content analysis method, it investigates symbolic phenomena through coding and categorization. The procedure adopted is the case study, A through Narrative Studies (in the media) and the feminist studies to identify structures, elements and similarities in the analyzed audiovisual pieces. As a data collection technique, from the perspective of feminist studies, it presents the compilation of data collected through interviews, aiming to identify perceptions of different publics about the empirical object (Voracious Games), generating 1119 interviews. The results point to divergences about the presence of female protagonism in the audiovisual perceived by men and women, as well as the main positive and negative characteristics perceived by the public. The divergence between the masculine and feminine looks on the mediated woman evidences the imbalance in the representations of gender of the audiovisual.

Stable Galerkin Finite Element Formulation for the Simulation of Electromagnetic Flowmeter

Sethupathy, S January 2016 (has links) (PDF)
Electromagnetic flow meters are simple, rugged, non-invasive flow measuring instruments, which are extensively employed in many applications. In particular, they are ideally suited for the flow rate measurement of liquid metals, which serve as coolants in fast breeder reactors. In such applications, theoretical evaluation of the sensitivity turns out to be the best possible choice. Invariably, an evaluation of the associated electromagnetic fields forms the first step. However, due to the complexity of the problem, only numerical field computational approach would be feasible. In the pertinent literature, couple of e orts could be found which employ the well-known Galerkin Finite Element Method (GFEM) for the required task. However, GFEM is known to suffer from the numerical stability problem even at moderate flow rates. This problem is quite common in fluid dynamics area and several stabilization schemes have been suggested as a remedial measure. Among such schemes, the Streamline Upwinding Petrov Galerkin (SU/PG) method is a simple and widely employed approach. The same has been adopted in some of the moving conductor literatures for obtaining a stable solution. Nevertheless, in fluid dynamics literature, it has been shown that the SU/PG solution can suffer from distortion/peaking at the boundary. The remedial measures proposed are nonlinear in nature and hence are computationally demanding. Also, even the SU/PG scheme by itself requires significant additional computation for quadratic and higher order elements. Further, the value of stabilization parameter is not accurately known for 2D and 3D problems. The present work is basically an attempt to address the above problem for flow meter and other rectilinearly moving conductor problems. More specifically, but for the requirement of (graded) structured mesh along the flow direction, it basically aims to address a more general class of problems not just limited to the flow meter. Following the classical approach employed in fluid dynamics literature, first the problem is studied in its 1D form. It was observed that a relatively better performance of GFEM over FDM scheme is basically due to the difference in their Right Hand Side (RHS) terms, which represents the applied magnetic field. Taking clue from this, it was envisaged that a better insight to the numerical problem can be obtained by using the control system theory's transfer function approach. An application of FDM or GFEM to the 1D form of the governing equation, leads to flalge-braic equations with space variable in discrete form. Hence, a Z-transform based approach is employed to relate the applied magnetic field to the vector potential of the resulting reaction magnetic field. It is then shown that the presence of a pole at Z = -1 is basically responsible for the oscillations in the numerical solution. It is then proposed that by using the control systems pole-zero cancellation principle, stability can be brought into the numerical solution. This requires suitable modification of RHS terms in the discretised equations and accordingly, two novel schemes have been proposed which works within the framework of GFEM. In author's considered opinion, the use of Z-transform for analysing the stability of the numerical schemes and the idea of employing pole-zero cancellation to bring in stability, are first of its kind. In the first of the proposed schemes, the pole-zero cancellation is achieved by simply restating the input magnetic field in terms its vector potential. Solving the difference equations given by the application of FDM or GFEM to 1D version of the governing equation, it is analytically shown that the proposed scheme is absolutely stable at high flow rates. However, at midrange of flow rates there is a small error, which is analytically quantified. Then the scheme is applied to the original flow meter problem which has only axially varying applied field and the stability is demonstrated for an extensive range of flow rates. Note that the discretisation along the flow direction was restricted in the above exercise to graded regular mesh, which can readily be realised for problems involving rectilinearly moving conductors. In order to cater for more general cases in which the applied field varies in both axial and transverse directions, a second scheme is developed. Here the RHS term representing the input magnetic field is considered in a generic weighted average form. The required weights are evaluated by imposing apart from the need for an essential zero yielding term, the flux preservation and other symmetry conditions. The stability of this scheme is proven analytically for both 1D and 2D version of the problem using respectively, the 1D and 2D Z-transform based approaches. The analytical inferences are adequately validated with numerical exercises. Also, the small error present for the midrange of flow rates is analytically quantified. Then the second scheme is applied to the actual flow meter with a general magnetic field pro le. The proposed scheme is shown to be very stable and accurate even at very high flow rates. As before, the discretisation was restricted to graded regular mesh along the flow direction. By solving for the standard TEAM No. 9 benchmark problem, applicability of the second scheme for other rectilinearly moving conductor problem has been adequately demonstrated. Even though the problems considered in this work readily permits the use of a graded regular mesh along the flow direction, for the sake of completeness, discretisation with arbitrary quadrilateral and triangular mesh is also considered. The performance of the proposed schemes for such cases even though found to deteriorate, is still shown to be considerably better than the GFEM. In summary, this work has successfully proposed two novel, computationally effcient and stable GFEM schemes for the simulation of electromagnetic flow meters and other rectilin early moving conductor problems.

The Suspicious Transaction Reporting Responsibilities of Attorneys in Terms of South African Anti-Money Laundering Legislative Frameworks

Dorey, Frank C. January 2014 (has links)
With the implementation of more and more stringent measures to prevent money laundering, criminals are resorting to the expertise of lawyers for assistance in the formulation of increasingly complex money laundering schemes. This expertise is provided both wittingly and unwittingly. The purpose of this research was to consider whether the South African anti-money laundering legislation places suspicious transaction reporting obligations, which are in line with and meet international directives, conventions and best practice frameworks, on attorneys. The study entails a consideration of the suspicious transaction reporting obligations of lawyers introduced by the Financial Action Task Force, the European Union, the United Kingdom and South Africa and provides an understanding of the concept of money laundering, the money laundering process and the areas in which lawyers are vulnerable to money laundering. The research found that the suspicious transaction reporting responsibilities of attorneys in terms of South African anti-money laundering legislation are not in line with international frameworks and best practice. / Dissertation (MPhil)--University of Pretoria, 2014. / tm2015 / Auditing / MPhil / Unrestricted

<b>Post-translational modifications governing neuro-migration and infection</b>

Sherlene Brown (18087418) 04 March 2024 (has links)
<p dir="ltr">This dissertation delves into two research projects that aim to characterize post-translational modifications in two distinct proteins, each originating from a different species – one from the eukaryotic sea slug Aplysia californica and the other from the bacterial pathogen Bordetella bronchiseptica.</p><p dir="ltr">Aplysia have an unusually large neuron and therefore serve as an excellent model for studying cell signaling regulating neuronal chemotaxis. Cortactin is an actin binding protein that is regulated by post-translational modifications, including acetylation and phosphorylation. Studies have shown that Src2 tyrosine kinase phosphorylates cortactin to regulate lamellipodia protrusion and filopodia formation in Aplysia bag cell neurons. However, these in vivo phenotypes have not been tested mechanistically in vitro. To this end, the goal of my thesis work was to validate in vivo observations. The following work describes the methodology we developed to purify homogenous non-phosphorylated proteins. Our collaborative results show that Src2 phosphorylates cortactin at Y499, although Y505 is the preferred site in vitro.</p><p dir="ltr"> Filamentation induced by cAMP (Fic) proteins constitute a recently characterized family of enzymes that are being recognized to regulate diverse cellular processes in bacteria and metazoans. While Fic proteins predominantly utilize adenosine triphosphate (ATP) to post-translationally modify target proteins via a covalent addition of AMP, two Fic proteins have been reported that utilize uridine triphosphate (UTP) and cytidine diphosphate-choline (CDP-choline) to alter the activity of their target. In this dissertation, we report the discovery of the first guanosine triphosphate (GTP) specific Fic protein – BB0907 (BbFic) from Bordetella bronchiseptica. BbFic displays weak to no binding to ATP; instead has a 10-fold increased preferential usage for GTP. We identify key residues involved in GTP recognition. Additionally, sequence similarity network (SSN) analyses reveal that BbFic represents a distinct clade of Fic proteins, highlighting BbFic as a representative new class of guanylyltransferase. Our discovery adds to the functional diversity of the growing Fic protein family and frames the groundwork for understanding Fic-mediated GMPylation as a novel signaling paradigm. </p><p dir="ltr">Taken together, my thesis work provides novel insights into biological consequences of Fic-mediated GMPylation in bacteria and Src-mediated phosphorylation in filopodia formation.</p><p><br></p>
