Para tanto, ser?o utilizadas algumas concep??es te?ricas sobre mem?ria individual e mem?ria coletiva e as reflex?es de Henri Bergson (1999) e de Le Goff (1984), em contraponto com as contribui??es de Maurice Halbwachs (2006) e outros autores, no que se refere ao assunto. / In this research, we intend to deliver a possible reading of the narrative Oiteiro: mem?rias de uma Sinh?-Mo?a (1958), by the writer Magdalena Antunes, placing it within the context of the Brazilian Literature of Rio Grande do Norte. The author?s personal and social relations described in her book as well as the outcomes within an autobiographical, memorialistic and fictional approach will be under inquiry here. Our intent is to reach an understanding of the autobiographical aspects in Antunes? work that reveal the ?writing of the self? and its probable fictional traits, by means of the tensions identified in the narrative. For this reason, we rely on contributions by Lejeune (2008); Amorim (2007; 2012); Walty (1985) and Iser (2002), among others. Aditionally, Antunes? work is also constituted of a memorialistic nature, which emerges in the book through the reinterpretation of memories based on the author?s childhood and adolescence. For a theoretical description of those moments, we will utilize some conceptions on individual and collective memory and the thoughts of Henri Bergson (1999) and Le Goff (1984), as opposed to the contributions brought by Maurice Halbwachs (2006) and other authors.

Cinco destas cepas de S. aureus (ATCC 33591, AM 13, AM 18, AM 20, AM 21) apresentaram resistência a todos os agentes antimicrobianos avaliados e desta forma foram selecionadas para o estudo do sinergismo entre o ácido úsnico e os agentes antimicrobianos através do método do tabuleiro de xadrez (checkerboard method). Os critérios utilizados para avaliar a atividade sinérgica foram definidos pelo Índice da Concentração Inibitória Fracionada (FICI). Todas as cepas de S. aureus foram suscetíveis ao ácido úsnico, determinado pelo método de microdiluição. O FICI variou de 0,25 – 1,0, sugerindo uma interação sinérgica frente as cepas de S. aureus MRSA. A associação do ácido úsnico com o ciprofloxacino apresentou efeito sinérgico sobre todas as cepas S. aureus MRSA. A oxacilina apresentou sinergismo em associação com ácido úsnico sobre as cepas de S. aureus ATCC 33591, AM 13, AM18 e AM24 e exibiu os menores valores FICI. A associação do ácido úsnico com a gentamicina foi sinérgica sobre as cepas AM18, AM21 e AM24. A associação do ácido úsnico com a penicilina apresentou-se indiferente para todas as cepas exceto para S. aureus AM13. Este estudo demonstrou que o ácido úsnico, quando associado à antimicrobianos fluoroquinolônicos, betalactâmicos e aminoglicosídeos pode agir sinergicamente, inibindo cepas de S. aureus MRSA.

Living organisms employ different kinds of mechanisms, to regulate the functions of genes or their products, which may help in maintaining homeostasis inside the cell or may help in fighting hostile environment in the case of pathogenic organisms. These mechanisms act at the transcriptional, post-transcriptional, translational, and post-translational levels. In order to understand the physiology of an organism, it is essential to obtain an in-depth knowledge of such mechanisms, in which several proteins participate in interlinked pathways. In this regard, the present study focuses on two such proteins: (i). the newly identified Fic (Filamentation induced by cAMP) protein; and (ii). NDK (Nucleoside Diphosphate Kinase), which had been studied for decades. Fic protein and NDK share several common features: (i). both use nucleoside triphosphate (NTPs) or nucleoside diphosphate (NDPs) or their derivatives as one of their substrates; (ii). they have been found to be involved in diverse cellular pathways, involving different types of substrates that form the second substrate of these proteins; (iii). both are ubiquitously present in all the living organisms - from bacteria to humans to plants. However, there is very little information on these proteins from mycobacterial systems, which include some major human pathogens, Mycobacterium tuberculosis and Mycobacterium leprae, which are the causative agents of Tuberculosis and Leprosy, respectively. In view of these reasons, in the present study, the structural and/or functional features of the Fic and NDK proteins from Mycobacterium tuberculosis, were analysed, as it might be of medical significance for effectively combating the pathogen. The Chapter 1 of the thesis contains the Introduction to the research work and Chapter 2 is on the overall Materials and Methods. The remaining chapters pertain to the data obtained on the structural and/or functional features of the Fic and NDK proteins from Mycobacterium tuberculosis. Chapter 3. Cloning, Expression and Purification of Mycobacterium tuberculosis Fic The role of FIC (Filamentation induced by cAMP) domain containing proteins in the regulation of many vital pathways, mostly through the transfer of NMPs from NTPs to specific target proteins (NMPylylation), in microorganisms, higher eukaryotes, and plants is emerging. In order to understand the biological role of FIC domain containing proteins in mycobacteria, the gene for the FIC domain containing protein of the human pathogen, Mycobacterium tuberculosis, MtuFic, was cloned, overexpressed, purified to homogeneity, and biochemically characterised. Neither the His-tagged nor the GST-tagged MtuFic protein, overexpressed in Escherichia coli, nor expression of Mtufic in Mycobacterium smegmatis, yielded the protein in the soluble fraction. However, the maltose binding protein (MBP) tagged MtuFic (MBP-MtuFic) could be obtained partly in the soluble fraction. Denatured-refolded protein was used for the antibody generation in mice and rabbit. The cellular localisation and secretion of MtuFic were characterised using the antibody. Chapter 4. Biochemical Characterisation of Mycobacterium tuberculosis Fic Sequence alignment with several FIC motif containing proteins, complemented with homology modeling on the FIC motif containing protein, VbhT of Bartonella schoenbuchensis as the template, showed conservation and interaction of residues constituting the FIC domain. MtuFic, possesses the critical His144 residue, in the characteristic FIC Motif, HPFREGNGRSTR (HPFxxGNGRxxR), spanning 144th to 155th residue. Site-specific mutagenesis of the His144, or Glu148, or Asn150 of the FIC motif, or of Arg87 residue that constitutes the FIC domain, or complete deletion of the FIC motif, abolished the NTP to NMP conversion activity. The activity of MtuFic was consistent with the biochemical activities hitherto reported for a variety of bacterial FIC domain containing proteins. Studies were also carried out on NMPylylation in the presence of eukaryotic proteins and eukaryotic and mycobacterial cell lysates. Although formation of NMPs from NTPs mediated by MBP-MtuFic could be detected, we could not identify any protein as the target substrate either in the human macrophage (THP1) cells or in the M. tuberculosis cells. VopSΔ30 (kind gift from Dr. Kim Orth), along with human G proteins as targets, were used as the positive controls. Various possibilities for the inability to detect a protein target substrate are discussed. Chapter 5. Transcriptional Analysis of Mycobacterium tuberculosis fic Gene (Mtufic) In parallel, in order to understand the transcriptional regulation of Mtufic, primer extension analysis was carried out. The Transcription Start Site (TSS; +1 site) of Mtufic were mapped under different growth/stress conditions, which tubercle bacilli encounter in human host. Mtufic got expressed mainly through two transcripts, T1 and T2, arising from two different transcription start sites (TSS). Putative promoter regions were cloned in a promoter probe vector, which expresses a GFP protein of very high intensity, in order to qualitatively detect the activity of the promoters. The half-life of the gfp mRNA was determined to be 4 min and therefore justifiably quantitated the Mtufic promoter activity by determining the gfp mRNA levels. The levels of Mtufic mRNA were two-fold higher under nutrient-depleted stationary phase of growth, as compared to the levels at mid-log phase. The activity of P1 and P2, as quantitated real-time using the short half-life gfpm2+ mRNA levels in Mycobacterium smegmatis transformants, showed that the activity of P2 was upregulated two-fold under nutrient-depleted stationary phase of growth, while that of P1 remained unaltered while of P1 and P2 were low under hypoxia. Co-transcription of Mtufic, with the immediate upstream gene, Rv3642c, of unknown function, was observed. Taken together, the data strongly indicated that the expression of Mtufic gets altered under nutrient-depleted and hypoxic conditions, which are the stress conditions experienced by tubercle bacilli in granuloma in tuberculosis patients. Chapter 6. Functional Characterisation of Mycobacterial FtsZ-NDK Interaction During the past few decades, our laboratory has been carrying out extensive molecular and functional studies on the cytokinetic protein, FtsZ, of different mycobacterial species, and of a variety of other mycobacterial proteins that are believed to be interacting with the cell division machinery. In this regard, in parallel to the work on MtuFic, we carried out work on the identification and characterisation of the proteins that interact with mycobacterial FtsZ. In this context, we found for the first time that the nucleoside diphosphate kinase (NDK), which can generate NTPs from ATP/GTP and NDPs, interacts with FtsZ and that the interaction was conserved across several mycobacterial species. Therefore, the FtsZ-NDK interaction was extensively characterised in vitro, using the recombinant, purified FtsZ and NDK proteins from different mycobacterial species. This novel finding on the interaction of NDK with FtsZ adds another role to NDK, namely in bacterial cell division.

The IFSP presents itself as a contradictory socio-occupational space: on one hand, serves the interests of capital and on the other, even considered as compensatory policy, it remains an important activity within the education and professionalization for people lower income. Based on the theoretical analysis of empirical data, this study identified objective and subjective contributions in the practice of social assistants, reinforcing the importance of the development and consolidation of the socio-occupational space. The methodology adopted in this research provided better understanding of the limits and possibilities of social assistants professional practice analyzed in the light of empirical categories, such as: the impact of the social assistant practice in PROEJA and PROEJA FIC; professional mediation in PROEJA and PROEJA FIC daily routine, instrumentality, the routine, the relationships with other professionals, among others aspects. In the investigative process, we used primary and secondary sources by conducting semi-structured interviews, group discussion, bibliographical and documental analysis. For the apprehension of the practical dimension of the social assistant, the study prioritized the analysis of strategic actions, procedures and tools used in the planning, organization, implementation and evaluation of actions as well as the identification of principles that guide the design and professional practice in regards to PROEJA and PROEJA FIC. It was verified, despite the limits and contradictions, the existence of progress in the consolidation of professional practice of social assistants and significant socio-occupational space. To the professionals, competes the responsibility of the incorporation of the Social Service premises, seized the objective conditions of social reality, developing a professional praxis oriented and theoretically able to increasingly contribute to the qualification of profession / Esta dissertação tem como objeto o estudo da prática do assistente social no PROEJA e PROEJA FIC no IFSP (Programa Nacional de Integração da Educação Profissional na Modalidade Básica e na Formação Inicial e Continuada no Instituto Federal de Educação, Ciência e Tecnologia de São Paulo). O IFSP apresenta-se como espaço sócio-ocupacional contraditório: de um lado, atende aos interesses do capital; de outro, mesmo considerado como política compensatória, não deixa de ser uma atividade no âmbito da educação e profissionalização importante para a população de menor poder aquisitivo. Com base na análise teórica dos dados empíricos, este estudo identificou contribuições objetivas e subjetivas na prática dos assistentes sociais, reforçando a importância do desenvolvimento e consolidação desse espaço sócio-ocupacional. A metodologia adotada nesta pesquisa propiciou melhor compreensão dos limites e possibilidades da prática profissional do assistente social, analisadas à luz de algumas categorias empíricas, tais como: a relação dos assistentes sociais com os estudantes do PROEJA e PROEJA FIC, a as atribuições e rotinas de trabalho do assistente social, as metodologias e instrumentalidade, as relações interprofissionais, entre outros. No processo investigativo, foram utilizadas fontes primárias e secundárias mediante a realização de entrevistas semiestruturadas, grupo de discussão, análise bibliográfica e documental. Para a apreensão da dimensão prática do assistente social, priorizou-se a análise das ações estratégicas, procedimentos e instrumentos utilizados no planejamento, sistematização, execução e avaliação das ações, bem como a identificação dos princípios que orientam a concepção e prática profissional, no que diz respeito ao PROEJA e PROEJA FIC. Verificou-se, não obstante os limites e contradições presentes, a existência de avanços na consolidação da prática profissional do assistente social e significativo espaço sócio-ocupacional. Aos profissionais compete a incorporação dos pressupostos do Serviço Social, apreendidos nas condições objetivas da realidade social, desenvolvendo uma práxis profissional orientada e teoricamente capaz de contribuir cada vez mais para a qualificação da profissão

El hilo conductor de la historia es su relaci?n con una amiga de infancia, donde sus expectativas y decepciones se mezclan. En los ensayos te?ricos a continuaci?n (a) explicito mi protelo de creaci?n y las influencias literarias m?s importantes durante el per?odo de escrita; (b) investigo los conceptos de imaginaci?n, ficci?n y realidad y su modo de relacionamiento. En el sentido com?n, generalmente entendemos que ficci?n y realidad son conceptos contrarios, siendo el uno el substrato de la negaci?n del otro. ?Pero ser? verdad? Para fundamentar esta reflexi?n, utilizo los te?ricos H. Vaihinger y W. Iser. El primero pr?cticamente empez? las reflexiones acerca del concepto de ficci?n en el sentido de entender la ficci?n como parte de Ia realidad, a saber, muchas de nuestras ideas m?s b?sicas y caras pueden ser comprendidas como ficciones. Iser por su vez, desarrollando las ideas de Vaihinger, piensa como esta polaridad ficci?n vs. realidad deber?a ser comprendida como un juego entre el ficcional, el real y el imaginario, cancelando la polaridad, y por consecuencia, lugares transcendentales desde donde estamos acostumbrados a posicionarnos; (c) intento pensar como el amor, sentimiento subyacente a la relaci?n de los personajes, puede funcionar como un generador de ficciones; Para concluir, (d) hay un ensayo final que intenta retomar las cuestiones esenciales presentes en los ensayos previos. / A parte liter?ria deste trabalho retrata per?odos da vida de um homem da adolesc?ncia ? velhice. O eixo condutor da hist?ria ? a sua rela??o com uma amiga de inf?ncia, onde entram em jogo suas expectativas e decep??es. Nos ensaios te?ricos que se seguem (a) explicito meu processo de cria??o e as influ?ncias liter?rias mais importantes durante o per?odo de escrita; (b) investigo os conceitos de imagina??o, fic??o e realidade e seu modo de relacionamento. No senso comum, geralmente entendemos que fic??o e realidade s?o conceitos contr?rios, sendo um o substrato da nega??o do outro. Mas ser? que isso ? mesmo assim? Para embasar esta reflex?o lan?o m?o dos te?ricos H. Vaihinger e W. Iser. O primeiro praticamente iniciou as reflex?es sobre o conceito de fic??o no sentido de entender a fic??o como fazendo parte da realidade, a saber, muitas de nossas ideias mais b?sicas podem ser entendidas como fic??es. J? Iser, desenvolvendo as ideias de Vaihinger, pensa como esta polaridade fic??o vs. realidade deveria ser compreendida como um jogo entre o ficcional, o real e o imagin?rio, cancelando a polaridade, e como consequ?ncia, lugares transcendentais da onde estamos acostumados a falar; (c) tento pensar como o amor, sentimento subjacente ? rela??o dos personagens, pode funcionar como um gerador de fic??es. In the thesis, the Wilcoxon approach to linear regression problems is combined with the fractal image compression to form a novel Wilcoxon fractal image compression. When the original image is corrupted by noise, we argue that the fractal image compression scheme should be insensitive to those outliers present in the corrupted image. This leads to the new concept of robust fractal image compression. The proposed Wilcoxon fractal image compression is the first attempt toward the design of robust fractal image compression. Four different numerical methods, i.e., steepest decent, line minimization based on quadratic interpolation, line minimization based on cubic interpolation, and least absolute deviation, will be proposed to solve the associated linear Wilcoxon regression problem. From the simulation results, it will be seen that, compared with the traditional fractal image compression, Wilcoxon fractal image compression has very good robustness against outliers caused by salt-and-pepper noise. However, it does not show great improvement of the robustness against outliers caused by Gaussian noise.

The doctoral thesis entitled Por dentro da cidade solid?o e marginalidade em Rubem Fonseca turns itself to his tale, it is considered by many as the most pungent of the author s literature. It is chosen a corpus of varied tales of the author, and still having as foundation the ideas from scholars of literary narratives, among them Antonio Candido (1980; 1987; 2006), Alfredo Bosi (1999; 2006) and still Walter Benjamin, (2000a; 2000b), the thesis discusses the contemporary Brazilian literature, more specifically the fonsequiana fiction, which exposes in an intense way the modern human condition in big cities: our dramas, inconstancies, desires and loneliness. / A fic??o do escritor Rubem Fonseca, desde a sua estreia em 1963, com o livro de contos Os prisioneiros, vem despertando crescente interesse nos seus leitores e na cr?tica especializada.
