Mutational effects on myosin force generation and the mechanism of tropomyosin assembly on actin

The cyclical interaction between the force-generating protein myosin and actin is the mechanism responsible for muscle contraction among all muscle types. Cardiac muscle contraction is tightly controlled to ensure that blood pumps effectively and efficiently from the heart to peripheral organs. Mutations in various cardiac proteins can lead to cardiac dysfunction and a number of cardiomyopathies. The first part of this dissertation studies two disease-linked mutations in the regulatory light chain of the cardiac myosin molecule, D166V and K104E, and assesses the kinetic and mechanochemical effects of the mutations via the in vitro motility assay. The data show that D166V mutant myosin force generation is reduced compared to wild type, and exogenous phosphorylation of the mutant light chain rescues force generation. In contrast, the K104E mutation showed no deficit in force production but exhibited increased calcium sensitivity of activation. These results are consistent with contractile defects associated with cardiomyopathies caused by various mutation-induced changes to protein function and mechanism of interaction. The second part uncovers the actin-binding mechanism of one of the chief muscle regulatory proteins tropomyosin. In cardiac and skeletal muscle, tropomyosin and troponin modulate muscle contraction. Tropomyosin binds along the length of actin filaments and blocks myosin-binding sites. Following an excitatory stimulus, calcium binds troponin and causes tropomyosin to shift its position on actin, allowing myosin to bind. The precise mechanism of how tropomyosin monomers with low actin affinity bind to form a stably bound, high affinity chain is unknown. By directly observing fluorescently labeled tropomyosin binding to actin filaments, it was shown that tropomyosin molecules bind randomly along the actin filament. Subsequent monomer binding, and formation of tropomyosin end-to-end bonds, increases the probability of sustained chain growth by decreasing the probability of detachment prior to additional monomer binding. Tropomyosin molecules added to the growing chain at approximately 100 monomers/(μM*s). Different tropomyosin isoforms segregate to distinct functional and structural regions of cells. The last chapter presents data that show spatial segregation of two different tropomyosin isoforms on actin filaments. This suggests that tropomyosin sorting in cells is, at least partly, an intrinsic property of the binding mechanism.

A influência da densidade do filtro de cigarros nos teores da fumaça

Em meio ao declínio do consumo de cigarros devido ao aumento das campanhas restritivas e a crescente consciência pública dos riscos associados à saúde dos fumantes, as indústrias de cigarros se deparam com o constante desafio de desenvolver novas tecnologias de fabricação e materiais que minimizem os efeitos da fumaça de cigarros sobre a saúde dos consumidores. O sabor e a composição da fumaça ingerida pelo fumante durante o ato de fumar dependem fundamentalmente da combinação do design do cigarro e do blend de fumo utilizado. O interesse na realização desta pesquisa foi avaliar a influência da densidade do filtro de cigarros na redução dos teores dos principais constituintes da fumaça. Para a realização do estudo, protótipos de cigarro foram fabricados em escala industrial, utilizando o mesmo blend de fumo e demais parâmetros de design do cigarro, apenas variando a especificação do denier do acetato de celulose utilizado na fabricação do filtro. O desempenho dos filtros como material filtrante da fumaça foi avaliado em termos da queda de pressão e das propriedades da fibra do cabo de acetato. E, utilizando métodos padronizados, os protótipos de cigarro foram fumados em máquina de fumar e a composição dos teores de alcatrão, nicotina e monóxido de carbono foi determinada para a avaliação da capacidade de retenção dos filtros de cigarro fabricados com diferentes densidades. Os resultados mostram que a utilização de cabos de acetato com menor denier por filamento nos filtros de cigarro apresenta grande desempenho na redução dos constituintes da fase particulada da fumaça, como o alcatrão e a nicotina, porém não apresenta efeito na redução do monóxido de carbono, substância volátil presente na fase vapor da fumaça. No entanto, cabos de acetato com menor espessura dos filamentos somente podem ser utilizados em filtros de cigarro se combinados com outros parâmetros de design de cigarro a fim de não apresentar prejuízo na aceitação do produto junto aos consumidores, devido ao aumento no esforço para a realização da tragada. / In the middle of the cigarette consumption decline caused by the increase of restrictive advertising campaigns and smokers’ health risk public awareness, tobacco industries now encounter the permanent challenge of developing materials and new manufacturing technologies to minimize cigarette smoke effects in regards to consumers’ health. The taste and formulation of the smoke absorbed in the smoking activity crucially depend on the combination between cigarette design and tobacco blend. The aim in this research was to evaluate the influence of cigarette filter density to reduce the level of smoke main constituents. In order to conduct the study, cigarette prototypes were manufactured at industrial proportion, using the same tobacco blend and respective cigarette design parameters, but only varying the denier specification of the cellulose acetate filter tow used to produce the filter. The performance of filters as smoke-filtering materials were evaluated in terms of pressure drop and filter tow fiber properties. And, according standardized methods, cigarette prototypes were smoked in smoking machines, and the composition of tar, nicotine and carbon monoxide levels was determined to evaluate the retention capacity of the cigarette filters manufactured with different densities. Results show that the use of filter tows with low denier per filament in cigarette filters increases smoke particulate phase components removal efficiency, such as tar and nicotine, but it does not present effects to reduce carbon monoxide, volatile substance that exists in the smoke vapor phase. However, filter tows with of filaments can only be used in cigarette filters if combined with other design parameters in order to avoid damage in the product acceptance by consumers due to the increase of effort in the cigarette puff.

Aplicação da análise inversa no projeto de fornos com aquecedores de filamento

No projeto de um forno, normalmente busca-se uma configuração para a posição dos aquecedores que resulte em aquecimento uniforme na superfície inferior, a superfície de projeto. Neste trabalho, o fluxo de calor e a temperatura são prescritos na superfície de projeto e os elementos aquecedores devem ser projetados em um conjunto de quatro filamentos, ou um filamento em cada quarto simétrico do forno, a fim de satisfazer às condições prescritas. O forno é modelado como uma cavidade tridimensional preenchida com meio transparente; assume-se que as paredes sejam cinzas e difusas e que o único mecanismo de transferência de calor presente é a radiação térmica. Esse problema, convencionalmente, é solucionado através de um procedimento de tentativa-e-erro; neste trabalho, a solução é obtida através da análise inversa, uma técnica mais versátil e eficaz de projeto, embora exija tratamentos matemáticos especiais. O problema inverso é resolvido nesta pesquisa de forma implícita, como um problema de otimização. A solução é obtida através do método da Otimização Extrema Generalizada (GEO), um método de otimização estocástico e global, utilizado para encontrar as posições para os aquecedores respeitando a condição de formar um filamento. A metodologia é aplicada para se obter a configuração geométrica e posição do filamento aquecedor, uma abordagem inédita na literatura, conduzindo a resultados com desvio máximo inferior a 2%. Por fim, nesta pesquisa, realiza-se um estudo acerca do fator de forma dos elementos aquecedores que indica a aplicabilidade da solução obtida para filamentos com diferentes espessuras. / In the design of a oven is desired to obtain the positions for the heaters that results in a uniform heating to the bottom surface, the design surface. The heat flux and temperature are prescribed in the design surface and the heating elements are positioned in a single filament so as to satisfy prescribed conditions. The oven is modeled as a three-dimensional cavity filled with transparent medium, it is assumed that the walls are gray and diffuse and the dominant heat transfer mechanism is thermal radiation. This problem, conventionally, is solved through a trial and error procedure, in this work, the solution is obtained by inverse analysis. The inverse problem is solved implicitly, as an optimization problem. The solution is obtained by the method of optimization extreme (GEO) a stochastic global optimization method used to find the locations for the heaters respecting the condition of build a filament. The methodology leads to satisfactory results, with maximum error less than 2%. Finally, this research, realized a study of the view factor of the elements heaters that indicates the applicability of the solution into filaments with different thicknesses.

Drosophila non-muscle myosin II bipolar filament formation: Importance of charged residues and specific domains for self-assembly

Search the library catalog for the location and call number. / Non-muscle myosin II generates contractile forces for processes such as cytokinesis, motility, and polarity. Contractility requires assembly of myosin molecules into bipolar mini-filaments through electrostatic interactions between coiled-coil tail domains of the heavy chains. Analyses of myosin II from various organisms have revealed "assembly domains" within the C-terminal portion of the tail domain that mediate filament formation. However, it has been unclear precisely how assembly domains interact with one another, or otherwise contribute to tail-tail interactions, to form the bipolar mini-filament structure. To understand tail domain interactions, we first identified a 90-residue region (1849-1940) of the Drosophila non-muscle myosin II tail domain that was necessary and sufficient for filament formation, using salt-dependent solubility and a novel fluorescence energy transfer assay. We identified residues within this "assembly domain" that were critical for filament assembly by analyzing the effect of point mutations. We found that single point mutations in specific positively charged regions completely disrupt filament assembly. Surprisingly, none of the negatively charged regions within the assembly domain are required for assembly. Most of the mutations in positively charged residues that disrupted filament assembly clustered within a 15-residue segment (1880-1894) that appears to form a critical interaction surface. Using this information, along with known geometrical constraints and electrostatic calculations, we constructed a structural model of the bipolar mini-filament. This model features one favored anti-parallel tail overlap and multiple slightly less stable alternative overlaps. The ability of the positive segment to interact with multiple negative regions explains the lack of required negatively charged residues in the assembly domain. To our knowledge, this structural model of the non- muscle myosin II bipolar filament is consistent with all physical observations and provides a framework for understanding the detailed mechanism by which this fundamental cellular structure is generated. Comportamento mecânico de cilindros de carbono/epóxi processados por enrolamento filamentar para semieixos de transmissão

O semieixo automotivo é um componente usualmente metálico pelo qual o torque é transmitido do diferencial para as rodas do veículo. Diferentemente dos semieixos convencionais, os fabricados em materiais compósitos possuem algumas variáveis de projeto que devem ser levadas em consideração, como a orientação das fibras, a sequência de empilhamento, a espessura e o número de camadas. Neste trabalho, cilindros compósitos para aplicação em semieixos de transmissão foram processados por enrolamento filamentar com o objetivo de substituir peças em aço convencional por compósito laminado em fibra de carbono com matriz epóxi. Para tal, os parâmetros da aplicação do semieixo em compósito, como frequência natural, torque critico de flambagem e capacidade de transmissão de torque foram considerados. Foi realizado um ensaio preliminar de torção e foram selecionados três laminados: [±22/±45], [±89/±45] e [±45]2. Cilindros de carbono/epóxi foram fabricados pelo processo de enrolamento filamentar e ensaios de compressão radial e longitudinal dos cilindros foram realizados. Análises mecânicas foram realizadas por meio de abordagens analíticas e numéricas, sendo que a última foi baseada no método dos elementos finitos. Os resultados analíticos foram comparados com os numéricos e experimentais, apresentando uma boa correlação entre eles. O cilindro com o laminado [±45]2 apresentou o melhor desempenho de resistência sob torção. Para torque critico de flambagem, assim como em compressão radial e longitudinal, o laminado [±45]2 apresentou um desempenho satisfatório para a aplicação, apesar de não ser o melhor desempenho dentre os laminados. Conclui-se que o laminado [±45]2 é o que apresenta melhor desempenho, em geral e que a redução de massa do semieixo desenvolvido em compósito utilizando este laminado, em relação ao de aço é de 47%. / The automotive half shaft is usually a metallic-based component in which the torque is transmitted from the differential to the wheels. Unlike conventional half shafts, in composite half shafts there are several parameters to be improved, namely fiber orientation, stacking sequence, thickness and number of layers. In this study, composite cylinders for application in half shafts have been processed by filament winding in order to replace conventional steel parts by laminated composites with carbon fiber reinforced epoxy composites. For that, it must satisfy several parameters of the composite half shaft, such as critical speed, critical buckling torque and load carrying ability. A preliminary torsional test has been performed and three laminates were selected: [±22/±45], [±89/±45] and [±45]2. Carbon/epoxy cylinders were manufactured by filament winding process and radial and longitudinal compression tests were performed. Mechanical analysis has been accomplished by using analytical and numerical approaches, since the last one is based on the finite element method. Analytical results were compared with the numerical and experimental ones, presenting a good correlation between them. The cylinder [±45]2 has the best performance under torsional loading. For critical buckling torque, as well as radial and longitudinal compression, the laminate [±45]2 presented a satisfactory performance for the application, but not the best performance within the laminates. The main conclusion is that the laminate [± 45]2 has the best performance and the weight reduction of the composite half shaft developed using this laminate is of 47%, when compared to a steel-based shaft.

Primeiro relato de isolamento e identificação taxonômica de fungos filamentosos em infecções endodônticas / First report of isolation and taxonomic identification of filament fungus in endodontic infections

A proposta deste trabalho foi demonstrar in vivo, por meio de técnicas específicas de isolamento, a presença de fungos filamentosos nos canais radiculares de dentes portadores de necrose pulpar e lesão periapical, relacionar sua presença a pacientes com resposta imunológica comprometida e realizar o estudo taxonômico dos isolados. Foram realizadas culturas de 60 canais radiculares com polpa necrosada e lesão periapical. Os pacientes responderam a um questionário de saúde e assinaram permissão para realização da pesquisa. Após isolamento absoluto e assepsia do campo operatório, as amostras foram coletadas utilizando-se 3 pontas de papel absorvente estéreis inseridas no canal radicular uma a uma, durante 1 minuto. Em campo isolado por duas lamparinas, o material coletado foi inoculado em um tubo de ensaio contendo meio de Saboraud Agar acrescido de Cloranfenicol. Próximo ao campo isolado pelas duas lamparinas, foi colocada uma placa de Petri aberta contendo o mesmo meio de cultura contido no tubo de ensaio, com o intuito de verificar a acuidade do isolamento do campo (controle negativo). Ao mesmo tempo foi efetuado um controle positivo, através de uma placa de Petri que permaneceu aberta durante a coleta, fora do campo isolado. Os tubos e placas de Petri foram mantidos em temperatura ambiente por um período de sete a quatorze dias observando o crescimento micelial. Aqueles que apresentaram crescimento foram semeados em meios específicos para microcultivo CYA Agar, Extrato de Malte Agar, ou Agar Batata. Com o auxílio de microscopia óptica, as colônias foram identificadas em gênero e espécie. Das 60 amostras coletadas, 17 apresentaram cultura positiva para fungos filamentosos, perfazendo um total de 28,3%. O gênero Aspergillus foi isolado, in situ, de 7 amostras (11,6%), sendo que a espécie Aspergillus ustus foi isolada de 3 pacientes (5%). O Aspergillus granulosus, Aspergillus niger e o Aspergillus sydowii foram isoladas de 3 pacientes respectivamente. A espécie Emericella quadriluniata, forma sexuada de Aspergillus, foi isolada de 1 paciente. O gênero Penicillium foi isolado, in situ, de 4 amostras (6,6%), sendo as espécies isoladas: Penicillium implicatum, Penicilium micsynvisk, Penicillium lividum e Penicillium citrionigrum. O gênero Fusarium foi isolado, in situ, de 2 amostras (3,3%), sendo as espécies identificadas como Fusarium moniliforme, Fusarium melanochorum, respectivamente. As espécies Aureobasidium pullulans, Exophiala jeancelmei, Eurotium amstelodame e Cladosporium sphaerospermum foram isoladas in situ de 4 amostras. Todos as amostras nas quais foi isolado fungo filamentoso pertenciam a pacientes com comprometimento da resposta imunológica. Nas placas de Petri empregadas no raio das duas lamparinas, não houve crescimento micelial. Entretanto nas placas controle positivo abertas para verificar a contaminação do meio ambiente houve crescimento de fungos ambientais que não eram compatíveis com os isolados dos canais radiculares, além de não apresentarem culturas puras. Pôde-se concluir que canais radiculares, com necrose pulpar e lesão periapical, podem apresentar cultura positiva para fungos filamentosos e que este resultado, neste estudo, apresentou relação direta com a deficiência da resposta imunológica do hospedeiro. / The purpose of the present study was to evaluate in vivo, with specific techniques that isolates the presence of filament fungus in the root canal of teeth with necrotic pulp and perirradicular disease, see the relationship between the presence of these fungus and patients with immunological deficiency and carry out a taxonomic study of the fungus isolated. It was fulfilled the culture of 60 root canals of teeth with necrotic pulp and perirradicular disease. The patients answered a health questionnaire and signed a term to allow the research. After the absolute isolation and cleaning the operating field the samples were collected using three sterile paper points inserted into the root canal during 1 minute. In an isolated field by two lamps, the collected materials were inoculated in a tube with Saboraud Agar with Cloranfenicol. Nearby the isolated field with lamps it was placed an opened Petri plaque with the same culture material from the tubes with the purpose to verify the perceptiveness of the isolated field (negative control). At the same time it was carried out the positive control group with an opened Petri plaque that was maintained far from the isolated fields. The tubes and the Petri plaque were maintained in atmosphere temperature during seven to fourteen days to observe micelial growth. Those that showed growth were spread on specific environment for microcultivation CYA Agar, Extract of Agar Malt or Agar potato. With the help of an optic microscope the colonies were identified in genre and species. Among the 60 samples collected, 17 showed positive culture for filament fungus, making up a total of 28,3%. The genre Aspergillus was isolated, in situ, of 7 samples (11,6%), and the species Aspergillus ustus was isolated from 3 pacients (5%). The Aspergillus granulosus, Aspergillus niger e o Aspergillus sydowii were isolated from 3 pacients respectivaly. The species Emericella quadriluniata, sexual form of Aspergillus, was isolated from 1 pacient. The genre Penicillium was isolated, in situ, from 4 samples (6,6%), where the species isolated were Penicillium implicatum, Penicilium micsynvisk, Penicillium lividum and Penicillium citrionigrum. The genre Fusarium was isolated, in situ, from 2 samples (3,3%), where the species identified were Fusarium moniliforme, Fusarium melanochorum, respectively. The species Aureobasidium pullulans, Exophiala jeancelmei, Eurotium amstelodame and Cladosporium sphaerospermum were isolated in situ from 4 samples. All the samples where it was isolated the filament fungus belonged to pacients with the imunolgical response compromised. On the Petri plaques employed at the range of the two lamp, it was not seen micelial growth. Thereby, on the positive control plaques, opened to check the contamination of the environment showed the growth of ambiental fungus that were not compatible with the fungus isolated from the root canal, and did not shor pure culture. It was concluded that root canal with necrotic pulp and periapical disease, can show positive culture for filament fungus and these results were directly related to the deficiency of the immunological response of the hostess.
