Desenvolveu-se um método espectrofotométrico para determinação de íons Cu(II), Fe(II) e Fe(III) com o reagente cromogênico di-2-piridil cetona benzoilhidrazona (DPKBH), em condições estacionárias e em fluxo. Fez-se a caracterização estrutural e estequiométrica do complexo de Cu(II) com DPKBH usando-se técnicas espectroscópicas de infravermelho e massas, além de análise térmica e elementar. Estudou-se, ainda, a estequiometria dos complexos de Fe(II) e Fe(III) com DPKBH utilizando espectrometria de massas com ionização por electrospray. Desenvolveu-se, preliminarmente, um método espectrofotométrico para a determinação de íons Cu(II) com DPKBH e aplicou-se em amostras de aguardente. Posteriormente, adaptou-se para análise por injeção em fluxo, utilizando-se injetor comutador manual. Em seguida, desenvolveu-se em condições estacionárias um método espectrofotométrico para determinação de Fe(II) e Fe(III) e Cu(II) em uma mesma amostra, com o uso de agentes mascarantes. Fez-se também a adaptação do método para análise em fluxo empregando multicomutação e amostragem binária. Finalmente, determinou-se a concentração de íons Cu(II), Fe(II) e Fe(III) em amostras sintéticas e Cu(II) e ferro total em amostras de sedimento coletadas no reservatório de Guarapiranga. Os resultados obtidos foram comparados com o método de referência de ICP-OES, apresentando concordância para um nível de confiança de 95% da média. / A spectrophotometric method was developed to the determination of Cu(II), Fe(II) and Fe(III) with the chromogenic reagent di-2pyridyl ketone benzoylhydrazone (DPKBH) in stationary conditions as a flow injection process. The structural characterization and the stoichiometry of Cu(II)/DPKBH complex were achieved using infrared spectrometry, mass spectrometry, thermal and elementar analysis. Toe stoichiometry of the Fe(II) and Fe(III) complexes with DPKBH was studied by electrospray ionization mass spectrometry. The spectrophotometric method for the determination of Cu(II) with DPKBH was developed in stationary conditions and, after this, it was adapted to flow injection analysis, using a manual commutator. Subsequently, a spectrophotometric method was developed to determine Fe(II), Fe(III) and Cu(II) in the same sample, in stationary conditions, using masking reagents. This method was also adapted to flow injection analysis, using multicommutation and binary sampling. Finally, Cu(II), Fe(II) and Fe(III) were determined in sediments from Guarapiranga reservoir. The obtained results were compared with the ICP-OES standard methods, showing a good agreement into a 95% confidence level (t-test).

L'économie circulaire appliquée à un système socio-écologique halio-alimentaire localisé : caractérisation, évaluation, opportunités et défis / Circular economy in a small-scale fishery-dependent socio-ecological system : characteristics, evaluation, opportunities and challenges

L'économie circulaire (EC) émerge depuis les années 2000 dans le débat public, en opposition à l'économie linéaire, modèle dominant aux limites économiques et écologiques reconnues. Cette thèse constitue une première confrontation opérationnelle de l'EC à un système socio-écologique dépendant de la pêche, le Pays de la Cornouaille en Bretagne. Le système est délimité dans sa façade maritime avec la pêche côtière, seule considérée comme production locale.L'approche est systémique, multidimensionnelle et dynamique. L'analyse comporte trois volets : le premier sur l'amont du circuit en mer, le deuxième sur l'aval du système à terre et le dernier sur le système complet. Premièrement, le coût caché des rejets est visualisé via une analyse comptable de flux de matière ("Material Flow Cost Accounting" MFCA) adaptée à la pêche, proposé comme outil pertinent pour améliorer la performance économique et environnementale des pêcheurs. Deuxièmement, l’analyse de flux révèle une forte dépendance des activités à terre vis-à-vis des importations et exportations de bioressources halieutiques malgré un apport significatif de la pêche côtière à l'économie locale. Troisièmement, des scenarii sur le sous-système Sardine, du "business as usual" a l'EC, servent à proposer une trajectoire alternative de développement basée sur un "panier" de biens et services territorialises et la construction d'une valeur territoire à partir de ressources halieutiques locales. Pour atteindre cette EC durable, il s'agira d'optimiser l'utilisation des ressources locales et diminuer les externalités négatives, en plus d'aller vers une gouvernance partagée, adaptée à l'échelle du système socio-écologique considéré. / The years 2000 have favored the emergence of "Circular Economy" (CE) in the public debate, as opposed to the "linear economy", the dominant model that led to acknowledged economie and ecological limits. Our work is a first attempt to address the CE to a fishery-dependent socio-ecological System, the Cornouaille region in Brittany. The approach is systemic, multidimensional and dynamic. It implies the identification of marine boundaries, in which only Coastal fisheries are considered as a local production. The analysis is conducted along three main axes: an analysis of the upstream part at sea (fishing), an analysis looking at the downstream value chain, on land, and a third looking at the whole System. First, a Material Flow Cost Accounting (MFCA) analysis adapted to fisheries is proposed as a relevant tool to improve economy and environmental performance of fishers, providing evidence of hidden costs of fish discards.Second, the fish bioresource flow analysis indicates a strong dependency on importation and exportation flows to maintain land-based activities depending on sea products, albeit evidence of a significant contribution of Coastal fisheries to the local economy. Third, the exploration of the sardine sub-system through scenarii, ranging between from "business as usual" to those lined with CE, shows an alternative development trajectory, based upon a potential "basket of goods and services", a "territorial value", specifically linked to local fisheries. To achieve such a sustainable CE, we would need to not only aim at a more efficient use of local marine resources and decrease negative externalities, but also advocate for a shared governance, suited to the scale of the considered localised socio-ecological System.

Zoning of the commercial poultry industry in Ontario as a method of avian influenza mitigation

L’Organisation mondiale de la santé animale (OIE) est l’institution internationale responsable de la mise en place des mesures sanitaires associées aux échanges commerciaux d’animaux vivants. Le zonage est une méthode de contrôle recommandée par l’OIE pour certaines maladies infectieuses, dont l’influenza aviaire. Les éclosions d’influenza aviaire été extrêmement coûteuses pour l’industrie avicole partout dans le monde. Afin d’évaluer la possibilité d’user de cette approche en Ontario, les données sur les sites de production avicole ont été fournies par les fédérations d’éleveurs de volailles ce cette province. L’information portant sur les industries associées à la production avicole, soit les meuneries, les abattoirs, les couvoirs, et les usines de classification d’œufs, a été obtenue par l’entremise de plusieurs sources, dont des représentants de l’industrie avicole. Des diagrammes de flux a été crée afin de comprendre les interactions entre les sites de production et les industries associées à ceux-ci. Ces industries constituaient les éléments de bas nécessaires au zonage. Cette analyse a permis de créer une base de données portant sur intrants et extrants de production pour chaque site d’élevage avicole, ainsi que pour les sites de production des industries associées à l’aviculture. À l’aide du logiciel ArcGIS, cette information a été fusionnée à des données géospatiales de Statistique Canada de l’Ontario et du Québec. La base de données résultante a permis de réaliser les essais de zonage. Soixante-douze essais ont été réalisés. Quatre ont été retenus car celles minimisaient de façon similaire les pertes de production de l’industrie. Ces essais montrent que la méthode utilisée pour l’étude du zonage peut démontrer les déficits et les surplus de production de l’industrie avicole commerciale en Ontario. Ceux-ci pourront servir de point de départ lors des discussions des intervenants de l’industrie avicole, étant donné que la coopération et la communication sont essentielles au succès du zonage. / The World Organisation for Animal Health (OIE) is the international reference body for international trade standards for live animals. Zoning is a method of controlling certain infectious diseases, including avian influenza, recommended by the OIE for use when appropriate. Avian influenza outbreaks have been extremely costly to the poultry industry throughout the world. In order to assess whether this approach was possible in Ontario, data on poultry industry production sites were provided by the Ontario poultry marketing boards. Zone borders were formed based on two criteria. The first criterion was the supply of essential products and services such that within-zone commercial poultry production could be maintained. The second was the contiguity of the zone’s territory. Four associated industries were identified which provide essential products and services: feed mills, abattoirs, hatcheries, and egg grading stations. A product flow analysis was completed to understand the direction of product movements between the poultry production sites and the sites of the four associated industries. This analysis was used to create a database of input requirements and output production capacity from each type of poultry production site. Using ArcGIS, this information was merged with geospatial data from Statistics Canada on Ontario and Quebec to create the database used for zoning scenarios. Seventy-two scenarios were completed; of these, four were chosen which minimized production loss over the whole industry. These scenarios demonstrate that the method used for the zoning study can identify the production deficits and surpluses of the commercial poultry industry in Ontario. These scenarios can serve as a starting point for discussion among industry stakeholders, as cooperation and communication are essential to the success of zoning.

V dnešním světě je neustále se zvyšující poptávka po spolehlivých automatizovaných mechanismech pro detekci a lokalizaci osob pro různé účely -- od analýzy pohybu návštěvníků v muzeích přes ovládání chytrých domovů až po hlídání nebezpečných oblastí, jimiž jsou například nástupiště vlakových stanic. Představujeme metodu detekce a lokalizace osob s pomocí nízkonákladových termálních kamer FLIR Lepton 3.5 a malých počítačů Raspberry Pi 3B+. Tento projekt, navazující na předchozí bakalářský projekt "Detekce lidí v místnosti za použití nízkonákladové termální kamery", nově podporuje modelování komplexních scén s polygonálními okraji a více termálními kamerami. V této práci představujeme vylepšenou knihovnu řízení a snímání pro kameru Lepton 3.5, novou techniku detekce lidí používající nejmodernější YOLO (You Only Look Once) detektor objektů v reálném čase, založený na hlubokých neuronových sítích, dále novou automaticky konfigurovatelnou termální jednotku, chráněnou schránkou z 3D tiskárny pro bezpečnou manipulaci, a v neposlední řadě také podrobný návod instalace detekčního systému do nového prostředí a další podpůrné nástroje a vylepšení. Výsledky nového systému demonstrujeme příkladem analýzy pohybu osob v Národním muzeu v Praze.

My Master's thesis deals with the problems of flow compression in network devices. Its outcome should alleviate memory consumption of the flows and simplify the processing of network traffic. As an introduction I provide a description of protocols serving for data storage and manipulation, followed by discussion about possibilities of compression methods that are employed nowadays. In the following part there is an in-depth analysis of source data that shows the structure and composition of the data and brings up useful observations, which are later used in the testing  of existing compression methods, as well as about their potential and utilization in flow compression. Later on, I venture into the field of lossy compression and basing on the test results a new approach is described, created by means of flow clustering and their subsequent lossy compression. The conclusion contains an evaluation of the possibilities of the method and the final summary of the thesis along with various suggestions for further development of the research.

Určování stresu z řečového signálu / Stress recognition from speech signal

Předložená disertační práce se zabývá vývojem algoritmů pro detekci stresu z řečového signálu. Inovativnost této práce se vyznačuje dvěma typy analýzy řečového signálu, a to za použití samohláskových polygonů a analýzy hlasivkových pulsů. Obě tyto základní analýzy mohou sloužit k detekci stresu v řečovém signálu, což bylo dokázáno sérií provedených experimentů. Nejlepších výsledků bylo dosaženo pomocí tzv. Closing-To-Opening phase ratio příznaku v Top-To-Bottom kritériu v kombinaci s vhodným klasifikátorem. Detekce stresu založená na této analýze může být definována jako jazykově i fonémově nezávislá, což bylo rovněž dokázáno získanými výsledky, které dosahují v některých případech až 95% úspěšnosti. Všechny experimenty byly provedeny na vytvořené české databázi obsahující reálný stres, a některé experimenty byly také provedeny pro anglickou stresovou databázi SUSAS.

<p> </p> <p>Learning about ‘behavior’ has always been at the heart of my research endeavors. While my undergraduate work in evolution and ecology exposed me to the science behind why a behavior exists, in my graduate work, I intended to explore how to use something’s behavior to widen its applicability. In this thesis, <em>I will present three works that utilize some of the fundamental</em></p> <p><em>behaviors (i.e., properties) of elastin-like polypeptides (ELP) to improve existing protein purification methods or explore their applicability in bladder cancer imaging and immunotherapy. </em></p> <p>Bladder cancer has high recurrence rates (60-70 % annually) that necessitate multiple follow-up therapies making it one of the costliest cancers per patient. In this work, we have attempted to address two leading causes of the recurrence. First is a low sensitivity (62-84 %) and variable specificity (43-95 %) of white light cystoscopy used to diagnose and remove tumors. We aimed to address the heart of this problem, i.e., the non-specific mode of detection using white light. Only the trained eyes can discern abnormal from normal-appearing tissues even then, leaving up to 45% of tumors unresected to colonize and spread. <em>We developed and characterized near infrared dye-peptide-ligand conjugates (NIR-ELP-ligand) that undergo receptor-mediated binding and internalization to human bladder cancer cells in vitro and tissues ex vivo.</em> By using a molecular target-based probe in combination with NIR imaging, we can aid in improving the detection limit via selective binding to the tumor and reduction in background autofluorescence.</p> <p>Bacillus-Calmette Guérin (BCG) instillation in the bladder is the gold-standard</p> <p>immunotherapy used after surgical removal of bladder tumors. This was approved as a response to the inefficiency of surgery alone in improving cancer status. It has succeeded by reducing the recurrence rate to 30-50 %. But it comes with the complications of putting a live mycobacterium</p> <p>in the human body and giving a patient a urinary tract infection right after surgical tumor resection. <em>Thus, we aimed to deliver nucleic acid as immunotherapeutic cargo in a selective manner to elicit robust anti-tumor immune responses while minimizing the side effects due to its carrier.</em> Towards</p> <p>this goal, we have developed a highly modular and adaptable ELP-ligand fusion protein-based nucleic acid delivery carrier targeted toward bladder cancer. Before developing targeted peptide-based cancer imaging and nucleic acid delivery modalities, we addressed the Achilles heel of peptide-based approaches. The peptide and protein industry suffers</p> <p>through complex, time-consuming, inconsistent, and low-yielding purification methods. <em>We have developed a scalable, facile, and reproducible protein purification method that delivers ELP and ELP fusion proteins free of host cell proteins and nucleic acids and has low lipopolysaccharide</em></p> <p><em>content in just 3 h starting from a bacterial pellet. </em>Thus, for a coherent narrative, the thesis is structured as follows:</p> <p>1. <b>Sustainability Analysis of Critical Materials in Electric Vehicles with Emphasis on Circular Economy Principles</b>

<p dir="ltr"><a href="" target="_blank">The electrification of the transportation sector is pivotal in reducing greenhouse gas emissions and decreasing dependence on fossil fuels. Central to this transition are battery electric vehicles (BEVs) and other clean energy technologies, which rely heavily on critical materials (CMs) such as cobalt, lithium, neodymium, and nickel. </a>These materials are essential for the performance of batteries, advanced electronics, and other components in BEVs. <a href="" target="_blank">However, the limited availability of these CMs poses potential constraints on the widespread adoption of such technologies.</a></p><p dir="ltr">This research delves into the implications of widespread BEV adoption on the demand for CMs in the United States, with a focus on both light-duty vehicles (LDVs) and medium- and heavy-duty vehicles (MHDVs). Various market penetration scenarios were analyzed, revealing that while MHDVs require more CMs per vehicle, the sheer volume of LDV sales drives the overall CM demand, particularly in a scenario with 100% BEV adoption. Key findings highlight that cobalt, graphite, lithium, neodymium, and nickel are critical for BEVs, whereas palladium and rhodium are more crucial for internal combustion engine vehicles (ICEVs). Also explored is the impact of lightweighting on LDVs, revealing that while substituting steel with aluminum increases the total CM quantity per vehicle, it reduces the vehicle's mass, operational energy consumption, and the demand for high-concern battery-related CMs. Additionally, changing the battery cathode chemistry from NMC622 to LFP significantly reduces CM use but increases the demand for strategic materials like copper and phosphorus due to the lower energy density of LFP-based batteries.</p><p dir="ltr">The research also highlights the importance of rare earth permanent magnets (REPMs), <a href="" target="_blank">particularly Neodymium-Iron-Boron (NdFeB) magnets, in clean energy technologies such as electric vehicles and wind turbines.</a> Neodymium, a critical material, faces supply chain risks. To lessen these risks, circular economy strategies have been proposed, including the recovery of needed materials from end-of-life (EoL) products. <a href="" target="_blank">A dynamic material flow analysis (MFA) model was developed to forecast such EoL flows for products containing REPMs and assess the recoverable neodymium from these EoL products. </a>The results indicate that even a modest recycling efficiency of 15% could meet 12% of the Nd demand for EVs by 2050, with reuse meeting up to 70% of the demand.</p><p dir="ltr">With the dynamic MFA model showing that circular economy principles could meet up to 70% of future neodymium demand in 2050, the next step was to investigate the techno-economic feasibility of recycling REPMs. A techno-economic assessment model was developed for establishing a magnet-to-magnet recycling facility for REPMs. Results revealed a net present value (NPV) of $8,867,111 over 20 years, a payback period of 3 years, and an internal rate of return (IRR) of 53%, providing a compelling case for investment in recycling infrastructure. Sensitivity analyses point to the selling price of recycled magnets, feedstock purchase price, facility throughput, and labor costs as the most influential factors on profitability.</p><p dir="ltr"><a href="" target="_blank">Additionally, this research explored the challenges and opportunities in the disassembly and recycling of EoL EV components, particularly traction motors containing REPMs. The complexity of disassembly, driven by varying component sizes and designs, is identified as a significant barrier. By evaluating manual disassembly times and proposing potential automation solutions, the study aims to streamline the disassembly process, thus facilitating more efficient recycling and remanufacturing operations.</a></p><p dir="ltr">The key contributions of this research are summarized as follows:</p><p dir="ltr">· Evaluated the vehicle CM demand of ICEVs and BEVs for LDVs and MHDVs and explored the impact of lightweighting and changing the battery cathode chemistry from NMC622 to LFP on CM demands.</p><p dir="ltr">· Developed a dynamic material flow analysis (MFA) model to forecast end-of-life (EoL) flows of products containing REPMs and assess the recoverable neodymium from these EoL products.</p><p dir="ltr">· Developed a techno-economic assessment (TEA) model to evaluate the viability of a magnet-to-magnet recycling facility.</p><p dir="ltr">· Performed disassembly analysis to assess the ease with which EoL BEV transmissions can be disassembled with a specific focus on the retrieval of traction motors (which house the REPMs) for potential reuse or remanufacturing.</p>
