Structural Basis of Guest-Host Interaction in the Gastrointestinal Delivery of Lipophilic Bioactive Compounds using Protein-based Vehicles

Bioactive compounds, such as curcumin, lutein, coenzyme Q10, β-carotene, cholecalciferol, astaxanthin, and β-sitosterol, have antioxidant and anti-inflammatory properties that promote health, but their low solubility, fast metabolism, and degradation have made it difficult to fully harness their potential. Encapsulation techniques, such as nano and microencapsulation using food-based biopolymers, have been employed to address these challenges. However, research efforts in protein-based delivery have mainly focused on encapsulation without considering structural, physicochemical, and matrix compatibility, which is tedious, unsustainable, and not cost-effective. Hence, this thesis reports the structural basis of guest-host interaction in the gastrointestinal delivery of lipophilic bioactive compounds using protein-based vehicles. This research employed fluorescence quenching techniques to estimate the influence of protein modification, fractionation and ionic strength on the nature and strength of interactions between protein and bioactive compounds. Morphological examination was carried out with transmission electron microscopy, confocal and widefield fluorescence microscopy whereas the sizes of the nano and micro-complexes was measured with dynamic light scattering techniques. Thermal stability was measured with differential scanning calorimetry and functional group characterization done with Fourier Transform infrared spectroscopy. Encapsulation efficiency was estimated by UV-Visible spectroscopy whereas in vitro bioactive compound release study was carried out in simulated salivary, gastric and intestinal fluids. Cytotoxicity assessment was estimated by calcein leakage assay. The study showed that protein modification affects the strength of protein-curcumin interaction and encapsulation efficiency. Pea protein succinylation increased electrostatic interaction with chitosan but decreased protein-curcumin interaction. Pea glutelin, albumin and globulin fractions showed different binding strengths with curcumin and the protein hydrophobicity and encapsulation efficiency correlated positively with the binding strength. The study also investigated the impact of bioactive compound lipophilicity and physiological ionic strength on the interaction between protein and bioactive compound. Lipophilicity influenced the strength of protein-bioactive compound interaction, while ionic strength changed the mode of interaction from static to static-dynamic quenching. The morphology of the nano and micro complexes formed with protein varied depending on the nature of encapsulated bioactive compound. Finally, bio-nano interaction involving giant unilamellar vesicles and curcumin-loaded pea protein of various surface functionalities as model biomembrane and nanoparticles respectively, was investigated. The result showed that while the protein/chitosan shell stabilizes bioactive compounds from degradation, the bioactive compound modulates their interaction with biomembrane. Overall, this work has demonstrated that for a rational design of protein-based nano/micro-encapsulation system, it is essential to consider the influence of the structural and physicochemical properties of proteins and bioactive compounds, stabilizing intermolecular forces, ionic strength of the environment, lipophilicity of the bioactive compounds, mechanism of release and modulation of cytotoxicity by bioactive compound. For instance, in high ionic strength solution, the stoichiometric ratio between protein carrier and bioactive compounds influences the stability of the complex. Balancing the intermolecular forces in the shell and core of bilayer complexes is essential for the stability of nanocomplexes and the presence of bioactive compound stabilizes the macromolecular carrier to minimal biomembrane disruption.

Design of optical characteristics of ceria nanoparticles for applications including gas sensing and up-conversion

This thesis investigates the impact of doping on the optical and structural characteristics of cerium oxide (ceria) nanoparticles synthesized using chemical precipitation. The dopants selected are samarium and neodymium, which have positive association energy with oxygen vacancies in the ceria host, and negative association lanthanides, holmium and erbium, as well as two metal dopants, aluminum and iron. Characteristics measured are absorption and fluorescence spectra and the diameter and lattice parameter of ceria. Analysis of the characteristics indicates qualitatively that the dopant controls the O-vacancy concentration and the ratio of the two cerium ionization states: Ce+3 and Ce+4. A novel conclusion is proposed that the negative association lanthanide dopants can act as O-vacancies scavengers in ceria while the O-vacancy concentration increases in ceria doped with positive association lanthanide elements. Doped ceria nanoparticles are evaluated in two applications: dissolved oxygen (DO) sensing and up-conversion. In the first application, ceria doped with either Sm or Nd and ceria doped with aluminum have a strong correlation between the fluorescence quenching with the DO concentration in the aqueous solution in which the ceria nanoparticles are suspended. Stern-Volmer constants (KSV) of doped ceria are found to strongly depend upon the O-vacancy concentration and are larger than some of the fluorescent molecular probes currently used to measure DO. The KSV measured between 25-50oC is found to be significantly less temperature dependent as compared to the constants of commercially-available DO molecular probes. In the second application, up-conversion, ceria nanoparticles doped with erbium and an additional lanthanide, either Sm or Nd, are exposed to IR radiation at 780 nm. Visible emission is only observed after the nanoparticles are calcinated at high temperature, greatly diminishing the concentration of O-vacancies. It is concluded that O-vacancies do not play a dominant role in up-conversion, unlike that drawn for down-conversion, where the fluorescence intensity is strongly correlated with the O-vacancy concentration. Correlations between annealing temperatures, dopant, and dopant concentrations with the power dependence of up-conversion on the pump and the origin of the intensities of the visible emission are presented. These studies show the promise of doped ceria nanoparticles. / Ph. D.

Foram investigadas através da técnica de absorção eletrônica Uv-Vis e emissão de fluorescência a interação entre uma série de porfirinas e zinco-porfirinas com o polímero de eumelanina sintética obtida pela auto-oxidação da L-DOPA (dihidroxi-fenilalanina). Ocorre a formação de dois complexos na interação entre as porfirinas catiônicas e a eumelanina. Um complexo mais fraco em concentrações mais baixas e intermediárias de eumelanina, e um complexo mais forte em concentrações maiores. Estes complexos podem ser observados pelas variações obtidas nos espectros de absorção em função da concentração da melanina. A eumelanina suprime eficientemente a fluorescência das porfirinas catiônicas, em um processo de supressão por esfera de ação. Este processo ocorre particularmente para os complexos fracos entre a porfirina e a eumelanina, podendo ser inferido pela natureza de polieletrólito da eumelanina que deve apresentar uma ampla superfície de contato carregada negativamente, facilitando a orientação das porfirinas em uma disposição planar em relação ao polímero. A determinação das constantes de supressão estáticas aparentes (KEA) mostra que os substituintes alquílicos influem consideravelmente na formação destes complexos com a eumelanina. As diferentes interações são devidas aos fatores estruturais tais como dimensão da porfirina, polarização dos substituintes e formação de interações de natureza hidrofóbica. As zinco-porfirinas são de forma geral menos suprimidas que as bases livres. Este comportamento pode ser uma evidencia da aproximação planar em relação ao polímero da melanina, uma vez que zinco porfirinas apresentam moléculas de água ligadas axialmente, logo estas moléculas coordenadas axialmente impediriam uma aproximação mais efetiva. Foi realizado um estudo da formação de filmes baseados na técnica de auto-montagem eletrostática entre as porfirinas e a melanina utilizando como substrato o vidro. Observa-se que a adsorção das porfirinas em vidro é diferenciada sendo a TBzPyP a base livre que apresenta a maior adsorção. Pelos espectros de emissão dos filmes de porfirina em vidro foi possível verificar que alguns derivados interagem com uma participação mais efetiva dos piridínios que outros em que a interação é menos direcionada. Nos filmes porfirina melanina também é observada a supressão de fluorescência da porfirina. A diferença na magnitude de supressão nos filmes sugere que estes derivados possam ser utilizados em sistemas de diagnóstico relacionados à detecção de tumores melanóticos. Em um estudo paralelo, foram investigadas as propriedades de fotoisomerização de grupos azobenzênicos coordenados axialmente a uma ftalocianina de Silício(IV). Este sistema apresenta fotoisomerização E-Z e isomerização térmica e sensibilizada para o processo Z-E. As conversões E-Z e Z-E seguem uma cinética de primeira ordem, sendo o processo térmico Z-E cerca de dez vezes mais lento que o fotoquímico E-Z. A intensidade de fluorescência da ftalocianina é dependente do estado de isomerização dos grupos axiais, sendo que a forma E apresenta maior emissão de fluorescência que a forma Z. Portanto este sistema se constitui em um dispositivo molecular do tipo “on-off", onde a intensidade de fluorescência pode ser controlada por um estímulo externo, no caso o comprimento de onda de excitação para a reação E-Z direta ou Z-E sensibilizada. / In this work we have investigated the interaction between a series of cationic porphyrins and zinc-porphyrins and the synthetic eumelanin polymer obtained by the auto-oxidation of L-DOPA (dihidroxy-phenylalanine) by means of UV-Vis and fluorescence emission techniques. The corresponding Zinc-porphyrins are less efficiently quenched by eumelanin, probably due the fact this molecules are axially coordinated by a water molecule, precluding a more close approximation between zinc-porphyrin and the eumelanin polymer. Electrostatically self-assembled films of porphyrins and eumelanin on glass have been studied. Different porphyrin adsorption on glass were observed, and TBzPyP derivative shown the highest adsorption in the investigated porphyrin series. Fluorescence emission suggests different interaction properties of the adsorbed pophyrin on glass, with specific pyridinium participation for some porphyrin derivatives. Fluorescence quenching of the porphyrins is observed in porphyrin – melanin films. The differences observed in the quenching for each derivative suggest a possible application of these compounds for diagnostic procedures related to melanotic tumors. In a complementary study, have been investigated the photoisomerization properties of a Si(IV)-phthalocyanine axially coordinated by azobenzene groups. Photochemical E-Z and thermal and sensitized Z-E isomerization processes were observed. Both E-Z photochemical and Z-E thermal follow first-order kinetics. Z-E process as expected is considerably slower than E-Z. The pthalocyanine emission is dependent on the isomerization state of the apendent azobenze groups, and the E form is more emissive than the Z form. This system can be considered a molecular device since the emission properties of the phthalocyanine moiety can be modulated by the isomerization state of the axially coordinated azoarenes, creating an on-off (E-Z states) fluorescence signal, and differently from other reported systems the recovery of the initial state does not depend only on the rate of the thermal reaction, since it can be controlled by a sensitized mechanism. This system seems promising taking into account the stability of the axially coordinated moieties, the greater versatility in the achievement of the desired isomerization state and the broad spectral region considering both direct and sensitized excitation.

Optical methods play an important role in process analytical technologies (PAT). Four examples of optical process and quality sensing (OPQS) are presented, which are based on three important experimental techniques: near-infrared absorption, luminescence quenching, and a novel method, photon density wave (PDW) spectroscopy. These are used to evaluate four process and quality parameters related to beer brewing and polyurethane (PU) foaming processes: the ethanol content and the oxygen (O2) content in beer, the biomass in a bioreactor, and the cellular structures of PU foam produced in a pilot production plant.

Nové metody pro rychlou detekci biologického materiálu na čipu / New technique on a chip for rapid detection of biological materials

Tato práce navrhuje techniku separace a detekce na čipu pro kvantové tečky (QD, „quantum dots“) konjugované s různými proteiny, za účelem sledování vlivu vazebného činidla na potlačení intenzity uorescence QD způsobené konjugací s proteinem a za účelem provedení multianalytické imunoanalýzy k identifikaci malých množství daného proteinu. Za optimálních podmínek byly biokonjugované QD úspěšně odděleny od těch nezkonjugovaných během 10 minut. Částice a cílové roztoky byly smíchány a detekce na čipu byla provedena za pomoci zařízení vyvinutého v naší laboratoři. Byl použit pouze jeden zdroj excitačního světla v kombinaci s několika filtry pro různé emisní vlnové délky. Fluorescence emitovaná dvěma typy konjugovaných QD mohla být poté zaznamenána současně, protože QD emitovaly světlo na různých vlnových délkách, ačkoliv byly excitovány při stejné vlnové délce. Smícháním dvou typů QD biokonjugovaných se dvěma druhy proteinů a protilátek jsme dokázali detekovat imunokomplexní píky s různými plochami. Plocha pod píkem závisela na koncentraci QD a antigenů, na postupu reakcí protilátka–antigen a ukázalo se, že je lineárně korelována s koncentrací antigenu. Ukázali jsme, že kapilární elektroforéza QD na čipu může být použita jako citlivá technika pro detekci biologických molekul. Hlavními výhodami této metody jsou jednoduchost, malé požadavky na objem vzorku i činidla a také vysoká účinnost separace.

Self-assembly of ionic fluorescent dyes inside polymer nanoparticles : engineering bright fluorescence and switching / Auto-assemblage de colorants ioniques fluorescents à l’intérieur de nanoparticules de polymères : ingénierie de fluorescence efficace et commutable

L’encapsulation dans des nanomatériaux de polymères de colorants ioniques à l’aide de contre-ions hydrophobes volumineux apparaît être une méthode très efficace pour générer des nanoparticules (NPs) fluorescentes ultra-brillantes pour la bioimagerie. Nous avons d’abord étendu cette approche par contre-ions aux colorants cyanine opérant dans la gamme du bleu au proche infra-rouge. A partir de NPs chargés en cyanines, une methode de code-barre multicolore pour le traçage cellulaire à long terme a été développé. Ensuite, le rôle des contre-ions hydrophobes volumineux dans l’auto-assemblage des colorants cationiques à l’intérieur des NPs de polymères a été étudié en testant une large collection d’anions. Nous avons montré qu’une forte hydrophobicité du contre-ion augmente l’encapsulation du colorant, régule son clustering et empêche l’agrégation de nanoparticules, alors qu’une grande taille empêche l’auto-inhibition de fluorescence. Enfin, nous avons introduit les contre-ions à base d’aluminates et de barbiturates, qui sur-performent les tetraphénylborates fluorés. Ce travail procure une base solide au concept d’émission et d’encapsulation augmentées par contre-ions pour la préparation de NPs chargés en colorants fluorescents. / Encapsulation of ionic dyes with help of bulky hydrophobic counterions into polymer nanomaterials emerged as powerful method for generating ultrabright fluorescent nanoparticles (NPs) for bioimaging. Here, this counterion-based approach is extended to cyanine dyes, operating from blue to near-infrared range. Based on cyanine-loaded NPs, a multicolour cell barcoding method for long-term cell tracking is developed. Second, the role of bulky hydrophobic counterion in self-assembly of cationic dyes inside polymeric NPs is studied by testing a large library of anions. We show that high hydrophobicity of a counterion enhances dye encapsulation, prevents particle aggregation and tunes dye clustering, while large size prevents dyes from self-quenching. Third, counterions based on aluminates and barbiturates are shown to outperform fluorinated tetraphenylborates. This work provides a solid basis for counterion-enhanced encapsulation and emission concept in preparation of dye-loaded fluorescent NPs.

Caractérisation Spectrale et Temporelle par Quenching de Fluorescence des Interactions Matière Organique-Eléments Métalliques / Spectral and Temporal Characterization of Organic Matter–Metal Elements by Fluorescence Quenching

L’étude du comportement des éléments métalliques est primordiale compte tenu leur effet souvent toxique dansde nombreux écosystèmes. Ces derniers lorsqu’ils interagissent avec la Matiere Organique (MO), peuvent formerdes complexes plus ou moins stables. Ainsi, la MO joue un rôle important dans leur spéciation chimique et leurtransport. Dans ce travail, l’analyse de cette complexation est réalisée par Quenching de Fluorescence (QF).Cette technique permet de modéliser la fixation des sites de complexation à l’aide d’une constante thermodynamiquedéterminée à partir d’un modèle 1 : 1. Le quenching de fluorescence a été mesuré par spectroscopie defluorescence en mode stationnaire et en mode résolue en temps. Les mesures de fluorescence en mode stationnairefournissent des Matrices d’Excitation et d’Émission de Fluorescence (MEEFs). L’extraction des différents composantsde ces MEEFs est effectué par séparation de sources : la décomposition multilinéaire CP/PARAFAC,qui permet de caractériser spectralement les composants. Les mesures par Spectroscopie Laser Résolue en Temps(SLRT) permettent une caractérisation spectrale et temporelle des composants fluorescents. L’étude des lois dedécroissance de la fluorescence induite par impulsion laser nanoseconde en l’occurrence a permis de déterminerle type d’interaction entre la MO et les quencheurs. Pour se faire, un algorithme de déconvolution temporellea été appliqué à chaque décroissance de fluorescence mesurée. L’interprétation des données temporelles a étéaccomplie en utilisant le graphique de Stern–Volmer. Les résultats des interactions du cuivre, de l’europiumet de l’uranium avec les Acides Humiques (AH) et les Acides Fulviques (AF) montrent des décroissances defluorescences importantes et des constantes de stabilité entre 2,04 et 4,52. Le cuivre a permis de valider notremodèle d’étude et l’interaction de l’europium et l’uranium avec les AH et AF étudiés a révélé des constantesde stabilité en général en bonne corrélation avec la littérature. Les résultats de la SLRT ont souvent révélé desdécroissances bi–exponentielles et des temps de vie entre 0,40 et 14 ns et montrent que les interactions étudiéesont principalement engendrer un quenching statique et donc la formation d’un complexe moléculaire à l’étatfondamental. Cette étude a donc permis par caractérisation spectrale et temporelle, de déterminer l’interactionde la matière organique avec les métaux plus ou moins toxiques. / The study of metal elements behavior, considering their impact in various ecosystems, is of paramount importance.The latter, upon contact with Organic Matter (OM) can form weak or strong stable complexes. Therefore,OM plays an important role in their chemical speciation and transport. The analysis of these properties is carriedout by Fluorescence Quenching (QF). This technique allows introspecting ligands–metal interactions andthe 1 : 1 modeling (one ligand site, one metal) gives information about the conditional thermodynamic constant.Fluorescence quenching was measured using steady–state and time–resolved fluorescence spectroscopy.The steady–state measurements provides Excitation and Emission Matrices of fluorescence (EEMs). The extractionof the different components from these matrices is carried out by a multi–mode factor analysis such asCP/PARAFAC, which allows a spectral composition of the samples. Time-Resolved Laser Spectroscopy (TRLS)measurements allow temporal and spectral characterization of the fluorescent components. Indeed, the study ofthe fluorescence decays induced by a nanosecond pulsed laser in this case allowed to measure the interaction betweenthe OM and the quencher. For those purposes, a fluorescence lifetime deconvolution algorithm was appliedto each fluorescence decay. Analysis of the fluorescence lifetime data was accomplished using the Stern–Volmerplot which gave reliable information on the quenching process that takes place. Copper, europium and uraniuminteractions with Humic Acids (HA) and Fulvic Acids (FA) shows significant fluorescence decays and stabilityconstants between 2.04 and 4.52. Copper allowed to calibrate our model study and the interactions of europiumand uranium with the HA and FA studied reported stability constants in agreement with the literature. TRLSresults have often revealed a bi–exponential decays and fluorescence lifetimes between 0.40 and 14 ns and showsthat the studied interactions mainly lead to static quenching and thus the formation of a molecular complex inthe ground state. This study has allowed spectral and temporal characterization to determine organic matterinteraction with toxic metals.
