Efeito das soluções conservadoras renais e da hipotermia sobre a atividade fagocitaria e liberação de superoxido de macrofagos peritonais de comundongos, em cultura

The phagocytes were incubated in RPMI-1640 supplemented with 10% fetal bovine serum at 37°C (Control), in the same solution at 8ºC (Cold Control) and in three preserved solutions, Collins, Euro-Collins and University of wisconsin (UW)at 8ºC, during 1 or 24 hours. In phagocytic actívíty protocol, the results showed that in 1 hour of cold preservation only the Euro-Collins group was different of Control and other experimental groups. Macrophages preserved in Euro-Collins solution during 1 hour showed the smallest phagocytic index. The preservation by 24 hours promoted drop of phagocytlc activity in experimental groups. All groups presented statistical differences when compared with Control. The phagocytes preserved in Collins by 24 hours exposed the biggest phagocytic index when compared with Cold Control, Euro-Collins and UW solution. In other protocol, free radical production, the results showed that Euro-Collins group produced little superoxide when compared with Control, Collins and UW group. By the other hand, during 24 hours of preservation, macrophages preserved in Euro-Collins solution released superoxide like Control and UW groups. The Collins group showed little release of superoxide when compared with Control, Euro-Collins and UW solution. These results led us to say that: the time and temperature of preservation, as well as, the lonic composition of preserved solution affected the phagocytic activity and the productlon of superoxide; Collins and Euro-Collins group always were different, so the magnesium sulfate should be important in the functional activity of these cells; the preserved solutions didn't avoid the release of free radicals; and the substances of UW utilized to eliminate free radicals were not efficient in our experimental conditions / Mestrado / Fisiologia / Mestre em Ciências Biológicas
