Peroxisome proliferator-activated receptor (PPARy) is a transcription factor that regulates adipogenic genes and preadipocyte factor-1 (pref-1) is a transmembrane protein that regulates preadipocyte differnetiation. The role of PPARy was investigated using reverse transcription polymerase chain reaction (RT-PCR) by measuring PPARy mRNA levels in bovine adipose depots and longissimus dorsi muscle. No significant differences in PPARy mRNA levels were observed between 10 Charolais and 10 Holstein cattle for either tissue. Differences were observed between depots within these breeds. Correlations were performed among PPARy, carcass characteristics, and adipogenic genes. Pref-1 antibodies were used to immunolocate preadipocytes in bovine muscle tissue to the perimycium, near fat cells and blood vessels. The preadipocytes may exist in muscle tissue for short periods of time or may arise from a source external to the muscle. A computer image analysis program was developed for the quanitifaction and characterization of intramuscular fat in whole muscle tissues. / xvi, 154 leaves : ill. (some col.) ; 28 cm.

Four experiments were conducted to determine the reproductive, metabolic, growth and IGF-I response to ultrasound selection for increased breast muscle thickness (MT) in Pekin ducks. Selection for body weight had a negative ( P < 0.05) effect on the reproductive performance while the selection for greater breast muscle thickness resulted in egg production, fertility and hatchability similar to those of the control line (C). Males from the MT line reached market weight at 6 wk but the breast muscle thickness improved from 6 to 7 wk. Compared to females from the C line, MT females had higher (P < 0.05) carcass yield, plasma glucose, and body protein; lower fat and plasma uric acid; and no difference (P > 0.05) in plasma triglycerides and total cholesterol. Males responded more efficiently to increasing dietary protein than females, and males selected for greater MT can be slaughtered at an earlier age when a high protein feeding program (HP) was followed. Males receiving HP had greater (P < 0.05) pectoralis muscles yield, longer keel bone, and lower breast skin and total skin fat yields. Female dissection data show similar (P > 0.05) effects of high and medium protein programs. Increasing dietary protein reduced (P < 0.05) carcass fat and increased (P < 0.05) crude protein (CP). In vivo breast muscle measurement correlated positively (P < 0.05) with body weight, pectoralis yield, and keel bone length. Birds with higher pectoralis yield tended to have more CP and less fat in their carcasses. A low correlation (r = 0.19) was found for the caliper measurement of breast skin plus fat thickness and carcass fat. We determined the metabolic differences between lean and fat male and female ducks, associated metabolic parameters and body composition, and assessed the difference between two feeding solutions for the determination of metabolic endogenous losses. The effects of dietary fat on the reproductive performance of broiler breeder chickens have been studied. Individually caged broiler breeders were fed diets with or without added fat during 19-wk reproductive period. In Experiment 1, only the females were subjected to dietary treatments: control (no added fat), 3% animal-vegetable fat blend (A-V fat, 3% corn oil (CO)), while the males received a non-added fat diet. In Experiment 2, only the males were fed diets containing 0 or 3% of either A-V fat or CO while the females were provided a non-added-fat diet. / In Experiment 1, added dietary fat produced significantly higher fertility and early embryonic livability. In Experiment 2, inclusion of 3% CO in the males' diets significantly increased late embryo livability and total embryo survival resulting in a significantly higher hatchability. There were no differences in semen characteristics among dietary treatments. These data indicate that the contribution of the male is not restricted to the fertilization of eggs but is also related to embryonic livability. The addition of fat significantly affected the fatty acid composition of the phospholipid fraction of the spermatozoa cells.

Authenticity and quality of muscle foods : assessing consumer trust and fraud detection approaches

Authenticity issues and fraudulent practices regarding animal products are affecting consumer confidence. Verifying the description, composition, processing or origin of foods can be challenging. To explore British and Kurdish consumers’ perceptions of kebab meat products, focus groups and questionnaire surveys were applied. About 40% of participants in the UK tend to purchase fewer processed meats after the European horsemeat scandal. Issues raised by participants indicated their concerns about the declaration of species, meat content, and other ingredients incorporated in kebab and other meat products. Lack of consumer trust has been linked to authenticity issues. Reactions towards the addition of fat-replacing inulin were positive by more than half of respondents. A further study aimed to investigate the effect of commercial inulin (CI) and Jerusalem artichoke (JA) tubers as fat replacers on the eating quality and overall acceptability of kebabs. Inulin flour prepared from JA by a simple protocol presented advantages with about 10% higher cooking yield and overall acceptability when compared with CI. Levels of inulin as low as 0.5% were detected in meat products using enzymatic assay, which could be relevant to detect additives and enforce labelling requirements. The authenticity (origin and species) was investigated in fish samples from commercial markets in Erbil, Kurdistan Region of Iraq (KRI). The declared fish species was checked using DNA barcoding with Cytochrome b region. A 10 % rate of mislabelling occurred only for wild common carp (Cyprinus carpio), with 9 out of 12 discovered to be the related species goldfish (Carassius auratus), which was deemed to be accidental rather than deliberate fraud. Such occurrences were from street markets and fishmongers, while none were from supermarkets. Wild and farmed common carp samples were not discriminated by DNA barcoding. Further fingerprinting using compositional profile and nearinfrared spectroscopy (NIRS) together with chemometric analysis aimed to predict composition and discriminate between wild and farmed common carp and species identity. NIRS-predictions of composition and some macrominerals of fish have a strong correlation with the references. NIRS with chemometric analysis is promising, but were not satisfactorily accurate for micro-minerals. Even with no clear solution from principal component analysis (PCA), NIRS-PCA may contribute to discriminating sample groups, but not for authentication when used alone. Having reliable techniques for authentication of food of animal origin may discourage deliberate replacement in retail, wholesale and international trade, and may contribute to reductions in food mislabelling, therefore protecting consumers from fraudulent practices.

Em cada linha de processamento, foram coletadas amostras de dois tipos de esteiras (modular e lisa) higienizadas com água e não higienizadas, em quatro tempos distintos (T0, T1, T2 e T3). As amostras foram submetidas à contagem de mesófilos, enterobactérias e pesquisa de L. monocytogenes. Culturas de L. monocytogenes foram submetidas à sorotipagem e a PFGE. Foram realizadas dez repetições do experimento em cada linha de processamento. Com base nos resultados foi possível observar que o efeito da utilização da aspersão com água foi dependente de diversos fatores, tais como a linha de processamento, tipo de corte, design do equipamento e o tipo de esteira. A higienização por aspersão não foi o fator primordial para garantir a eliminação de L. monocytogenes, uma vez que o patógeno foi isolado independente da linha de processamento e tipo de esteira, além disso, os pulsotipos encontrados evidenciaram a disseminação do patógeno nas linhas de processamento avaliadas. Deste modo, a decisão de retirar a água do processo de higienização de esteiras condutoras deve ser precedida de um estudo criterioso para garantir que a sua ausência não venha influenciar na inocuidade dos produtos de origem animal / This study aimed to evaluate the effect of the use of water spray on cleaning of conveyor belts in four broiler lines processing in Brazilian slaughterhouses. Surface samples were collected of two types of conveyor belt (modular and smooth) which passed through two processes (with and without water spray), in four times (T0, T1, T2 and T3). The samples were subjected to mesophilic and enterobacteria count, and research of L. monocytogenes. Cultures of L. monocytogenes were serotyped and submitted to PFGE. Ten replicates of the experiment were performed on each processing line. The results demonstrated that the effect of the use of spraying with water was dependent on several factors, such as the processing line, product type, equipment design and the type of conveyor belt. The cleaning spray is not the primary factor in ensuring the elimination of L. monocytogenes, because the pathogen was isolated independent of the type of conveyor belt and processing line, moreover, PFGE demonstrated the dissemination in the processing lines. Thus, the decision to remove the water from the cleaning of conveyor belts must be preceded by a study to ensure that the absence of this procedure does not influence the safety of products of animal origin

A presença de Salmonella em 200 amostras de alimentos crus de origem animal foi investigada empregando-se os dois ensaios imunoenzimáticos rápidos TECRA&#8482; Salmonella VIA e TECRA&#8482; Salmonella UNIQUE (TECRA Diagnostics, Rosewille, NSW, Australia) e o método de cultura convencional empregado rotineiramente no Instituto Adolfo Lutz, São Paulo, SP. Quarenta e cinco amostras (22.5%) foram Salmonella positivas por pelo menos um dos três métodos. O número de amostras positivas de acordo com o método analítico foi 34 (75,6%) para o método de cultura convencional, 29 (64,4%) para TECRA&#8482; Salmonella VIA e 27 (60.0%) para TECRA&#8482; Salmonella UNIQUE. O método de cultura convencional detectou quatro amostras positivas não detectadas por nenhum dos outros dois métodos rápidos. TECRA&#8482; Salmonella UNIQUE detectou sete amostras positivas não detectadas pelos demais métodos. Uma amostra foi positiva apenas pelo método TECRA&#8482; Salmonella VIA. Considerando todos os resultados (positivos e negativos) o teste de qui quadrado de McNemar indicou que as diferenças entre os resultados obtidos pelos métodos rápidos, quando comparados aos obtidos pelo método convencional, não foram estatisticamente significativas (p>0.05). / The presence of Salmonella in 200 raw food samples of animal origin was investigated by means of rapid immunoassays TECRA&#8482; Salmonella VIA and TECRA&#8482; Salmonella UNIQUE (TECRA Diagnostics, Rosewille, NSW, Australia) and the cultural procedure used routinely in Instituto Adolfo Lutz, Sao Paulo, SP. Forty-five samples (22.5%) were Salmonella positive by at least one of the three methods. The number of positive samples according to the analytical method was 34 (75.6%) for the cultural procedure, 29 (64.4%) for TECRA&#8482; Salmonella VIA and 27 (60.0%) for TECRA&#8482; Salmonella UNIQUE. The cultural method detected four positive samples that both rapid methods were unable to detect. TECRA&#8482; Salmonella UNIQUE detected seven positive samples that were not detected by the two other methods. One sample was positive by the TECRA&#8482; Salmonella VIA exclusively. Considering overall results (positive and negative) McNemar\'s chi square tests indicated that the differences between results given by the rapid immunoassays when compared to those of the cultural method were not significant (p>0.05).

A geléia real é um dos principais produtos da colméia que serve como alimento para as larvas em desenvolvimento, assim como para a abelha rainha por toda a sua vida. Tem sido utilizada pelas pessoas como um complemento alimentar devido às suas propriedades revitalizantes. O principal objetivo deste trabalho foi estabelecer as análises físico-químicas e químicas que poderão ser usadas para a caracterização da geléia real e detecção de sua autenticidade ou adulteração, enfatizando-se a determinação do ácido 10-hidroxi-trans-2-decenóico (10-HDA). A proposta do trabalho está justificada pela falta de legislação com relação a padrões de identidade e qualidade da geléia real e derivados. Foram determinados emalostras puras e adulteradas (com clara de ovo, água potável, iogurte natural, pasta de amido e uma mistura de leite condensado com própolis) os teores de umidade, cinzas, Iípides, proteínas, carboidratos, 10-HDA, pH, acidez titulável, reação com lugol e teste de solubilidade em meio alcalino. Como análises complementares foram determinados os teores das vitaminas B6 , A, pró-vitamina A (&#946;-caroteno) e vitamina E nas amostras puras. Considerando-se os resultados obtidos preconizou-se as determinações de umidade, cinzas, lípides, proteínas, carboidratos, 10-HDA e teste de solubilidade em meio alcalino como os principais itens a serem observados. / Royal jelly is one of the most important products from beehive, which is used by growing up larves like food, and also for the queen bee for her entire life. It has being used by people as a complement because of the revitalizing properties. The main objective of this work was to establish the physicalchemical and chemical analysis, which could be used for characterization of the adulteration of royal jelly, mainly the 10-hidroxy-trans-2-decenoic acid (10-HDA). The purpose of this study is justified by the lack of legislation concerning the quality and identity standards of royal jelly and derivatives. There were determined in pure and adulterated samples (with condensed milk, water, yogurt, starch, egg white) the analysis of moisture, ash, lipids, proteins, carbohydrates, 10-HDA, pH, titrate acidity, iodine reaction, solubilíty test in alkaline solution. For complementary tests It was determined the values of vitamin B6 , A, provitamin A, and E in pure samples. Considering the obtained results it was suggested the use of moisture content, ash, lipids, proteins, carbohydrates, 10-HDA and solubility in alkaline solution, as the main items to be observed.
