Avaliação do reparo tecidual em defeito ósseo por microtomografia tridimensional por raio X / Evaluation of tissue repair after bone fracture using 3D X-ray microtomography

A fratura é uma descontinuidade óssea que pode ser produzida cirurgicamente ou causada por um impacto que excede a resistência mecânica do osso dando início a uma sequência de eventos sistêmicos e específicos de resposta do tecido. Exames radiológicos são comumente realizados na clínica e em experimentos com animais para o monitoramento do reparo ósseo dando informações sobre o alinhamento dos fragmentos e da evolução de reparo. Outras técnicas de monitoramento qualitativas e quantitativas podem ser utilizadas em experimento animal (histologia e ensaios mecânicos) e em experimento animal e clínico (tomografia computadorizada por raio X, ressonância magnética, ultra-sonografia). A microtomografia 3D por raio X é uma nova técnica de monitoramento para uso em experimento animal e com grande potencialidade. A quantificação do reparo ósseo com novas metodologias tem larga aplicação em investigações sobre técnicas invasivas e não-invasivas de tratamento de fraturas utilizando-se experimento animal. O objetivo dessa investigação foi a utilização da microtomografia 3D por raio X policromático (\'mü\'CT) para a avaliação do reparo ósseo em defeito ósseo na tíbia direita de rato macho da raça Wistar com peso aproximado de 280 g. O defeito foi produzido por uma broca odontológica com alta rotação. Foram estabelecidos quatro grupos experimentais caracterizados pela utilização ou não utilização do tratamento do defeito por ultra-som pulsado de baixa intensidade (LIPUS, 30 mW/\'CM POT.2\') e pela duração do experimento. No grupo 1 o tratamento por ultra-som teve a duração de 14 dias, 5 sessões de tratamento por semana. No grupo 2 não houve tratamento por ultra-som e a duração foi de 14 dias. No grupo 3 o tratamento por ultra-som teve a duração de 21 dias, 5 sessões de tratamento por semana. No grupo 4 não houve tratamento por ultra-som e a duração foi de 21 dias. Nos grupos 1 a 4 foram utilizados 10 animais para a avaliação por CT. O defeito ósseo da tíbia direita nos animais dos grupos 1 e 3 foi tratado com ultra-som de baixa intensidade. A avaliação por CT foi realizada através dos softwares NRecon, Data Viewer , CT-Analyzer e CT-Vol fornecidos pelo fabricante do microtomógrafo (Skyscan, Bélgica). Não foi observada diferença estatísticamente significante na quantificação do reparo ósseo dos defeitos dos grupos 1 e 2 e dos grupos 3 e 4. / Fracture is a bone discontinuity that can be surgically produced or caused by an impact that exceeds the mechanical strength of bone by initiating a series of systemic events and specific tissue response. Radiological tests are commonly performed in clinical and animal experiments for monitoring the bone healing by providing information about the alignment of the fragments and the evolution of repair. Other techniques for monitoring quality and quantity can be used in experimental animals (histology and mechanical tests) and in animal experiments and clinical studies (computed tomography X-ray, MRI, ultrasound). The 3D microtomography by X-ray is a new monitoring technique for use in animal experiment and with great potential. The quantification of bone repair with new methods has wide application in research on invasive and noninvasive treatment of fractures using animal experiment. The goal of this research was to use the 3D microtomography by non monochromatic X-ray (\'mü\'CT) to evaluate the bone healing in bone defect in the tibia of Wistar male rat weighing approximately 280 g. The defect was produced by a dental drill with high speed. It was established four experimental groups characterized by the use or non use of low intensity pulsed ultrasound (LIPUS, 30 mW/\'CM POT.2\') for the bone defect treatment as well as the duration of the experiment. In group 1 the LIPUS treatment last 14 days, 5 treatment sessions per week. In group 2 the LIPUS treatment was not used and the duration was 14 days. In group 3 the LIPUS treatment last 21 days, 5 treatment sessions per week. In group 4 the LIPUS treatment was not used and the duration was 21 days. In groups 1 to 4, 10 animals were used for evaluation by CT. The evaluation was conducted by CT through software NRecon, Data Viewer, CT-Vol and CT-Analyzer supplied by the microtomography manufacturer (SkyScan, Belgium). No significant statistical differences were found between the results of groups 1 and 2 as well as the results of groups 3 and 4.

Efeitos da dieta hiperlipídica na estrutura e consolidação de tíbias de ratas em crescimento / Effects of high-fat diet on structure and bone healing in tibias of growing rats

O acúmulo de gordura corporal é considerado um problema de saúde pública com diversas consequências negativas, inclusive relacionado à redução da qualidade do osso, que afeta cada vez mais indivíduos jovens. São crescentes as investigações que buscam esclarecer a relação entre massa gorda e massa óssea. Portanto, o objetivo deste estudo foi investigar o efeito do consumo excessivo de lipídios na estrutura e no processo de consolidação óssea em tíbias de ratas em crescimento. Foram utilizadas cinquenta e duas ratas Wistar com 21 dias de vida, distribuídas em quatro grupos (n=13), sendo: animais submetidos à dieta padrão (DP), animais submetidos à dieta hiperlipídica (DH), animais submetidos à dieta padrão e osteotomia (DPO) e animais submetidos à dieta hiperlipídica e osteotomia (DHO). Cada um deles com período experimental de 5 semanas. Os animais dos grupos DP e DH foram mantidos sem intervenção, enquanto os animais dos grupos DPO e DHO foram submetidos à osteotomia da tíbia esquerda na terceira semana experimental. Durante o experimento a massa corporal e o consumo de ração dos animais foram avaliados. Um dia precedente à eutanásia foi realizado o teste de tolerância à insulina em 5 animais de cada grupo. Após a eutanásia a tíbia esquerda de cada rata foi dissecada, limpa das partes moles e submetidas a análises macroscópica, densitometria, ensaio mecânico, histomorfometria, estereologia do osso neoformado e imunohistoquímica. Os dados foram analisados por teste estatístico em modelo linear geral com ajuste para múltiplas comparações de Bonferroni para comparação entre as variáveis, e teste T-Student e Mann-Whitney para comparação entre dois grupos. Ao final do experimento os animais alimentados com dieta hiperlipídica apresentaram massa corporal similar aos animais alimentados com dieta padrão (p=0,115), no entanto tiveram consumo reduzido de ração (p<0,001) e menor sensibilidade à insulina (p=0,018). A variável cirurgia afetou o consumo de ração sendo que animais operados consumiram menos ração do que animais não operados (p=0,017). Os animais alimentados com dieta hiperlipídica tiveram tíbias mais compridas (p=0,041), menor força máxima (p<0,001) e rigidez relativa (p=0,003) quando comparados aos animais alimentados com dieta padrão, embora a densidade mineral óssea (DMO) tenha sido similar (p=0,958). Os animais operados tiveram menor força máxima (p<0,001), rigidez (p=0,020) e menor quantidade de colágeno proximal (p=0,029) quando comparados aos não operados. O volume de osso neoformado das tíbias submetidas à osteotomia tendeu a ser menor no grupo DHO ao grupo DPO (p=0,054). A avaliação imunohistoquímica não revelou diferença entre os grupos DHO e DPO (p<0,05). Concluímos que a dieta hiperlipídica prejudicou a resistência mecânica das tíbias e causou uma redução do volume de osso neoformado, prejudicando dessa forma a estrutura e consolidação óssea das tíbias de ratas em crescimento / The body fat accumulation have been considered a public health issue with several negative consequences, including reduction of bone quality, that affects young people increasingly. The studies that seek to clarify the relationship between fat mass and bone mass have been increasing. Therefore the aim of this study was to investigate the effect of high consumption of lipids on bone structure and bone healing in growing rats. Fifty-two female Wistar rats 21 days old were used, divided into four groups (n=13), as follows: animals treated with standard diet (SD), animals treated with high-fat diet (HFD), animals treated with standard diet and osteotomy (SDO) and animals treated with high-fat diet and osteotomy (HFDO). Experimental period was of 5 weeks for every groups. The SD and HFD animals were maintained without intervention, while SDO and HFDO groups were submitted to osteotomy in left tibia at third experimental week. Animal body mass and feed intake were assessed during experimental period. One day before euthanasia insulin tolerance test was performed in 5 animal of each group. After euthanasia, the left tibia of each rat was dissected and cleaned of soft tissues and submitted to macroscopy analysis, densitometry, mechanical analysis, histomorphometry, stereology of newly formed bone and immunohistochemistry. Data were analyzed by general linear model with adjustment to multiple comparisons of Bonferroni for comparisons between variables and TStudent e Mann-Whitney test for comparisons between two groups. At end of the experiment the high-fat diet fed animals had body mass similar to standard diet fed animals (p=0.115), however they had lower feed intake (p<0.001) and lower insulin sensibility (p=0.018). Surgery affected feed intake, operated animals had lower feed intake than non-operated animals (p=0.017). High-fat diet fed animals had longer tibias (p=0,041), lower maximal load (p<0.001) and lower stiffness (p=0,003) than standard diet fed animals, although bone mineral density (BMD) had been similar (p=0,958). Operated animals had lower maximal load (p<0.001), stiffness (p=0.020) and lower collagen quantity (p=0.029) than non-operated animals. Newly formed bone of tibias submitted to osteotomy trended to be lower in HFDO group than SDO group (p=0.054). Immunohistochemistry presented no difference between HFDO e SDO group (p<0.05). We concluded high-fat diet affected bone in order to harm tibia strength and caused a decrease in the newly formed bone volume, impairing structure and bone healing

TARGETED DELIVERY OF DASATINIB FOR ACCELERATED BONE FRACTURE REPAIR

<p>Approximately 6.3 million bone fractures occur annually in the USA, resulting in considerable morbidity, deterioration in quality of life, loss of productivity and wages, and sometimes death (e.g. hip fractures). Although anabolic and antiresorptive agents have been introduced for treatment of osteoporosis, no systemically-administered drug has been developed to accelerate the fracture healing process. To address this need, we have undertaken to target a bone anabolic agent selectively to fracture surfaces in order to concentrate the drug’s healing power directly on the fracture site. We report here that conjugation of dasatinib to a bone fracture-homing oligopeptide via a releasable linker reduces fractured femur healing times in mice by ~60% without causing overt off-target toxicity or remodeling of nontraumatized bones. Thus, achievement of healthy bone density, normal bone volume, and healthy bone mechanical properties at the fracture site is realized after only 3-4 weeks in dasatinib-targeted mice, but requires ~8 weeks in PBS-treated controls. Moreover, optimizations have been implemented to the dosing regimen and releasing mechanisms of this targeted-dasatinib therapy, which has enabled us to cut the total doses by half, reduce the risk of premature release in circulation, and still improve upon the therapeutic efficacy. These efforts might reduce the burden associated with frequent doses on patients with broken bones and lower potential toxicity brought by drug degradation in the blood stream. In addition to dasatinib, a few other small molecules have also been targeted to fracture surfaces and identified as prospective therapeutic agents for the acceleration of fracture repair. In conclusion, in this dissertation, we have successfully targeted dasatinib to bone fracture surfaces, which can significantly accelerate the healing process at dasatinib concentrations that are known to be safe in oncological applications. A modular synthetic method has also been developed to allow for easy conversion of a bone-anabolic warhead into a fracture-targeted version for improved fracture repair.</p><p></p>

Stimulation of bone healing in new fractures of the tibial shaft using interferential currents

The aims . of this research were twofold, firstly to find out if interferential currents could reduce the healing time for fractures of the tibia and thereby prevent nonunion and secondly to develop a model which could predict nonunion, given the subject characteristics such as race, mechanism of injury, severity of fracture etc. Subjects, males only between the ages of 12 and 86, who had sustained fractures of the tibiae were entered into this double blind clinical trial on admission to the orthopaedic wards at Groote Schuur Hospital (between January 1989 and October 1991). According to strict inclusion and exclusion criteria, a final sample of 227 cases (208 subjects) were entered by block randomisation into three groups; an experimental group (n=41), placebo group (n=35) and control group (n= 151). lnterferential currents were applied to the experimental group via suction electrodes for, 30 minutes per day for 10 days, using a beat frequency of 10 - 25 Hz and a swing mode of 6 ϟ 6. The placebo group had the suction electrodes applied which produce a rhythmical massage effect. Subjects commented on pain relief which resulted in the addition of the control group as a check on the possible effect of suction, the control group received no intervention. The data were analysed firstly, by using the ANOV A with continuous covariates which resulted in a finding of no significant difference in the time taken to union for the three groups. The second statistical analysis using the same data set, were logistic regression models demonstrating risk factors for nonunion within 24, 32 and 40 weeks. These models were then validated, showing sensitivity and specificity for a variety of possible cutoffs. The conclusions reached about the validity of these models were that they could not be used to predict, accurately enough, those cases where surgical intervention would be necessary; however, for low cost non-invasive intervention they may have value.

Considerando-se que o tabagismo afeta a maioria dos sistemas do corpo humano, que os estudos concentram-se mais sobre os efeitos deletérios do tabaco em doenças cujos órgãos vitais são afetados e que seu impacto na formação e na consolidação óssea ainda não está estabelecido, a proposta neste estudo foi avaliar os efeitos da exposição à fumaça de cigarro no mecanismo de consolidação e remodelamento da matriz óssea e relacioná-los ao processo inflamatório desencadeado por fratura em modelo desenvolvido em camundongos. Para isso, camundongos C57BL6 machos foram distribuídos em quatro grupos: C (n=29) expostos ao ar ambiente; F (n=23) expostos ao ar ambiente e submetidos à osteotomia da tíbia direita; CS (n=29) expostos à fumaça de cigarro; FCS (n=23) expostos à fumaça de cigarro e submetidos à osteotomia da tíbia direita. Nossos dados indicaram diminuição do volume trabecular, da mineralização, MS/BS% (p < 0,05), e da formação óssea, MAR (p=0,0004), bem como aumento de osteoclastos, Oc.S/BS% (p=0,0361), e de reabsorção óssea, ES/BS% (p=0,0114), no grupo CS, quando comparado ao grupo C. Identificamos uma elevação significativa de IL-6 nos grupos CS e FCS (p < 0,001) na comparação com os grupos C e F (p < 0,05). Observamos diminuição de VEGF nos grupos CS e FCS, quando comparados com os grupos C e F (p=0,01). O índice de IGF demonstrou-se diminuído nos grupos CS e FCS, quando comparados com os grupos C e S (p < 0,05). A matriz fibrilar apresentou um intenso remodelamento, caracterizado pelo aumento da expressão de Colágeno V na avaliação por imuno-histoquímica nos grupos CS, F e FCS, comparados com C (p < 0,05), e da expressão dos genes COL5A1 (p < 0,0001) e COL5A2 nos grupos F e FCS (p < 0,0001). Identificamos também diminuição significativa de Colágeno I nos grupos CS, F e FCS, comparados com C (p < 0,001), e da expressão dos genes COL1A1 no grupo CS (p=0,008) e COL1A2 nos grupos CS, F e FCS (p=0,005). Nosso estudo mostra que a exposição à fumaça de cigarro altera a composição das fibras de colágeno pertencentes à matriz fibrilar óssea, retarda a mineralização e modifica a interação entre os diferentes tipos de colágeno, importantes no desempenho funcional do osso / Introduction: The impact of cigarette smoke on bone metabolism is not yet fully understood. This study aimed to verify the effects of cigarette smoke exposure in bone healing in an experimental tibial fracture model, evaluating the mineralization process, bone cellular differentiation/function and collagen types deposition. Materials and Methods: C57BL/6 male mice were assigned into four groups: C(n=29): exposure to room air; F(n=23): exposure to room air and right tibia osteotomy; CS(n=29): exposure to cigarette smoke; FCS(n=23): exposure to cigarette smoke and right tibia osteotomy. Results: Histomorphometry of Bone Mineral Matrix revealed that cigarette smoke exposure significantly reduced thickness of bone trabeculae, associated with a decrease in mineralizing surface and in rate of mineral apposition, which was reflected in lower bone formation rate and consequently in a longer time for mineralization. Both resorption surface and osteoclastic surface were higher in the CS group, evidencing the increase of the resorptive action of cigarette smoke. Histomorphometry of Bone Fibrillar Matrix: Type I collagen demonstrated a decrease in CS and FCS compared to C (p < 0.01); Type V collagen demonstrated an increase in CS, FC and FSC compared to C (p < 0.0001). The cytokines expression evaluation demonstrated that only CS exposure has already induced a VEGF and IGF decrease with a concomitant increase in IL-6 and these changes were intensified under fracture conditions. COL1A1 gene expression was reduced in the CS and FCS groups, similar result was observed in COL1A2 evaluation. COL5A1 gene expression, showed an increase in CS and Fracture groups while the COL5A2 gene expression increase was detected only in Fracture groups. Conclusion: Cigarette smoke exposure alters bone matrix composition and worsens bone mineralization, leading to bone fragility by increasing collagen V synthesis and deposition and impairing collagen I fibril forming and assembling

Comparação biomecânica e da redução óssea do Clamp and Rod Internal Fixation e placa de reconstrução bloqueada em fraturas de acetábulo / Comparação biomecânica e da redução óssea de Clamp and Rod Internal Fixation (CRIF) e placa de reconstrução bloqueada em fraturas de acetábulo

Este estudo objetivou comparar, biomecanicamente, por meio dos ensaios de flexão destrutível e ciclagem, bem como a qualidade da redução óssea, após avaliação do molde da superfície articular de osteotomias centrais do acetábulo de cão fixadas com Clamp and Rod Internal Fixation (CRIF) 2,7 mm ou placa de reconstrução bloqueada 2,7 mm. Os implantes foram divididos em dois grupos, denominados grupo CRIF (GC) e grupo Placa (GP). Foram utilizadas nove pelves de cadáveres de cão com peso corporal entre 15 e 35 kg, posteriormente as pelves foram subdivididas em 18 hemipelves e distribuídas aleatoriamente para testar os dois implantes, um contralateral ao outro em uma mesma pelve. Para realização dos testes, foi simulado uma fratura central no acetábulo, por meio de osteotomia linear, realizada com auxílio de serra oscilatória e em seguida, fixadas segundo os padrões AO/SIF, sendo utilizados três parafusos craniais e dois caudais a osteotomia. Após comparação biomecânica de resistência máxima à flexão o GP (22,38&#177;8,44 N.m) demonstrou superioridade estatística em relação ao GC (15,60&#177;5,76 N.m), (p=0,02408). Na comparação do molde da superfície articular após 30 ciclos o GP (0,27 &#177; 0,24mm) demonstrou valores menores para lacuna da linha articular que o GC (0,49 &#177; 0,29mm), sendo o GP estatisticamente superior, (p=0,04784). Em conclusão, a aplicação da placa de reconstrução bloqueada é simples e deve ser considerada para redução de fraturas do acetábulo que necessitem de reconstrução anatômica. / The aim of this study was to compare the accuracy of reduction and biomechanical characteristics of acetabular osteotomies repaired with 2.7-mm Clamp and Rod Internal Fixation (CRIF) constructs or 2.7-mm locking reconstruction plates in cyclic and load to failure flexion tests. Nine dog cadavers with body weight between 15 and 35 kg were included in this study. All 18 hemipelves were randomly assigned into two groups named after the implants tested, group CRIF (GC) and group Plate (GP). All pelves were used to test both groups with each hemipelvis belonging to a different implant group. Acetabular osteotomies were created with an oscillatory saw and then fixed according to the AO/ASIF standards, using either 5-hole, 2.7-mm locking reconstruction plates with three locked screws cranial and two caudal to the osteotomy line, or CRIF constructs with 5 clamps, three located cranial and two caudal to the osteotomy line. Load to failure at GP (22.38 &#177; 8.44N.m) was significantly higher than GC (15.60 &#177; 5.76N.m), (p = 0.02408). Imprinted cast of acetabular reduction gap after 30 cycles was statistically smaller at GP (0.27 &#177; 0.24mm) than GC (0.49 &#177; 0.29mm) (p = 0.04784). We concluded that the appliance of locking reconstruction plates is simple and should be considered for acetabular fractures, that require perfect anatomical reconstruction.

Avaliação do efeito de centrifugado osteogênico de medula óssea na consolidação de fratura: estudo experimental em coelhos / Effect of centrifuged osteogenic bone-marrow aspirate on bone fracture healing: an experimental study in rabbits

INTRODUÇÃO: O objetivo deste estudo foi de avaliar a eficácia de um centrifugado osteogênico de medula óssea para estimular a consolidação de osteotomias da fíbula em coelhos. MÉTODOS: Este estudo experimental envolveu a utilização de dez coelhos machos adultos da raça Nova Zelândia albino. Realizou-se uma osteotomia transversa médio-diafisária da fíbula direita, seguida da adição local de uma esponja de colágeno absorvível embebida em um centrifugado osteogênico, obtido pela centrifugação de aspirado de medula óssea do osso ilíaco ipsilateral. A fíbula esquerda foi utilizada como controle, sendo feita a mesma osteotomia, porém neste caso adicionando-se somente a esponja de colágeno absorvível. O centrifugado de medula óssea elaborado em laboratório foi submetido à contagem do número de células nucleadas e a teste de viabilidade celular antes de ser administrada no local das osteotomias. Após quatro semanas os animais foram sacrificados para estudo dos calos ósseos formados. Os critérios de avaliação foram a mensuração da densidade mineral utilizando-se a densitometria óssea com DEXA, do volume do calo com tomografia computadorizada multi-slice e dos tecidos formados por meio de histomorfometria. RESULTADOS: O método utilizado para a centrifugação dos aspirados de medula óssea resultou em uma concentração média de células nucleadas três vezes maior que o número destas células nos aspirados originais, sem destruição celular significativa. A utilização deste centrifugado osteogênico resultou em um aumento médio na densidade mineral óssea dos calos de 40,3% e da quantidade relativa de tecido ósseo de 9,4%, sem aumento significativo nas quantidades relativas de cartilagem ou fibrose. Não houve aumento significativo no volume dos calos ósseos. CONCLUSÃO: A administração de centrifugado osteogênico de medula óssea utilizado neste estudo favoreceu a consolidação óssea de osteotomias experimentais em coelhos, observando-se uma melhora qualitativa do calo ósseo. / INTRODUTION: The purpose of this study was to evaluate the efficacy of a centrifuged osteogenic bone-marrow aspirate to stimulate rabbit fibular osteotomies healing. METHOD: Ten white New Zeeland rabbits were used. A transverse middle-diaphysis fibular osteotomy was performed at the right fibula, where a collagen absorbable sponge embedded in the osteogenic centrifuged bone marrow aspirate was inserted. The left fibula was used as the control group, where the collagen absorbable sponge was inserted without the osteogenic centrifuged aspirate. The centrifuged bone-marrow aspirate was arranged at the laboratory and submitted to nuclear cell count and cell viability test. The rabbits were killed at four weeks after surgery to evaluate bone callus formation. The results analysis was performed with DEXA bone densitometry to evaluate callus mineral mass, multislice computer tomography to evaluate callus volume and histomorphometry to evaluate the relative rate of tissue formation. RESULTS: The bone-marrow centrifugation technique increased the number of nucleated cells by three compared with the number of that cells in the original bone-marrow aspirates, without significant nucleated cell dead. The apply of centrifuged osteogenic bone-marrow aspirate resulted in an increased callus bone mineral mass by 40,3%, and increased relative rate of bone tissue formation by 9,4%, without increase the relative rate of cartilage or fibrous tissue. There was not increased callus volume. CONCLUSION: This study shows that the centrifuged osteogenic bone-marrow aspirate was able to improve the healing of experimental fibular osteotomies in rabbits by qualitative improve of bone callus.

Einfluss des Immunsystems auf die Frakturheilung in der Maus

Noch immer gibt es trotz neuester medizinischer Operationstechniken Patienten, bei denen eine Fraktur zu einer verzögerten Heilung führt oder gar nicht heilt. Indizien häufen sich, dass das Immunsystem, seine Zellen und Zytokine eine wichtige Rolle im Prozess der Heilung einer Fraktur spielen könnten. In dieser Arbeit wurde der Einfluss des adaptiven Immunsystems auf die Fraktur-heilung untersucht. Dazu wurde der Heilungsverlauf in Wildtyp Mäusen mit RAG1-/- Mäusen, Mäusen ohne reife B- und T-Lymphozyten, verglichen. Histologische, immunhistologische sowie Mikroarray Analysen der verschiedenen Tierstämme lassen darauf schließen, dass T-Lymphozyten einerseits der Schlüssel für ein gutes Heilungsergebnis sind, andererseits ihr Fehlen maßgeblich zu einer Veränderung der Heilung führt. Diese resultierte darin, dass die Knorpelphase verkürzt war und eine ungeordnete Bildung von Geflechtknochen einsetzte. / Despite of advanced methods in trauma surgery some patients suffer from a delayed healing or a non-union one. There is more and more evidence that the immune system with its cells and mediators could play an important role in such healing situations. Hence this thesis investigated the role of the adaptive immune system in the process of fracture healing comparing healing in wildtype mice and RAG1-/- mice. RAG1-/- mice do not have mature B- and T-lymphocytes. Histological, immunohistological and microarry analyses of different mice strains reveal that T lymphocytes are the key cell for a normal healing outcome. Their absence leads to a different healing outcome with a diminished cartilage phase and an irregular distribution of woven bone in the whole callus region.
