As avaliações foram realizadas na CEAGESP e nos laboratórios do Departamento de Agroindústria, Alimentos e Nutrição da Escola Superior de AgriculturaLuiz de Queiroz ESALQ/USP. Foram realizadas coletas quinzenais, em três atacadistas, com as amostras recolhidas de forma aleatória, padronizadas em frutos tipo 10 (caixa contendo 10 frutos), compostas de dez frutos por atacadista/origem. As amostras foram coletadas nos lotes oferecidos para comercialização pelos atacadistas. Foram avaliadas as seguintes características físicas e físico-químicas: peso do fruto inteiro, peso da coroa, avaliação visual da coloração da casca, densidade, coloração da polpa, pH, teor de sólidos solúveis totais, acidez titulável e ratio. Para a análise estatística foi utilizado o Programa SAS. Pelos resultados observou-se que dentre as regiões produtoras de abacaxi houve destaque em relação à regularidade de oferta para os três atacadistas o pólo de Canápolis ( Smooth Cayenne ) e Sapé e Miracema do Tocantins ( Pérola ). As diferentes regiões produtoras de abacaxi da cultivar Pérola apresentaram uma grande variação em relação ao peso médio dos frutos comercializados, indicando diferenças na tecnologia de produção adotada, com destaque para Itaberaba e Frutal. Exploring the genetic mechanisms underlying superficial scald development and prevention in apple fruit during postharvest storage.

Apple is one of the most valuable fruit consumed worldwide due to its excellent quality features and high degree of acceptation by consumers. However, apples are susceptible to various postharvest disorders, leading to significant economic losses. To slow down food decay and provide constant fresh food to the market, postharvest strategies including decreasing the temperature can slows down the metabolism of fruits, but in parallel can also trigger the occurrence of typical chilling injury related disorders. Among all the physiological disorders, superficial scald is one of the most challenging, causing discoloration and skin browning triggering therefore consumers unacceptance. The development of superficial scald in apple fruit is a complex process influenced by various factors, including genetic background, storage conditions and preharvest management practices. Recent research partially disclosed the molecular mechanisms underlying the development of superficial scald in apple fruit. It has been shown that this disorder is associated with the accumulation of reactive oxygen species (ROS) and the oxidation of alfa-farnesene, a sesquiterpene largely considered as a major actor in the onset of this disorder. Various postharvest treatments have been used to control the development of superficial scald in apple fruit, such as including the application of the ethylene competitor 1-methylcyclopropene (1-MCP) or controlled atmosphere storage. This thesis aimed to identify and characterize the genetic factors involved in symptoms development and to disclose the regulation of this phenomenon. To this end, a comparative multidisciplinary study integrating transcriptomics and metabolomics analysis have been performed in a wide range of storage conditions and across different genetic background. The mechanism of action and effectiveness of two preventing strategies, such as the exogenous application of 1-MCP and storage at low oxygen concentration were assessed in two apple cultivars, ‘Granny Smith’ and ‘Ladina’. The metabolite and transcriptomic profiles revealed that 1-MCP treatment actively stimulated the production of unsaturated fatty acids such as oleic acid (C18:1), linoleic acid (C18:2) and linolenic acid (C18:3), thereby successfully preventing the development of superficial scald in ‘Granny Smith’. ‘Ladina’, on the other hand, was less responsive to these strategies and was characterized by a higher accumulation of very long chain fatty acids (VLCFAs), together with an increased level of fatty acid saturation. Additionally, the integration of the transcriptomic survey and VOCs assessment underlined that storage at low oxygen concentration stimulated the expression of genes involved in anaerobic respiration, such as malate, alcohol dehydrogenase, and pyruvate decarboxylase, leading to the accumulation of ethanol and acetaldehyde in both cultivars. The expression of three members of the VII subgroups of ERF genes was also observed in ‘Granny Smith’, which coordinate the acclimation process to hypoxia in plants. The effect of controlled atmosphere in the prevention of superficial scald was furthermore investigated in fruit of ‘Granny Smith’ stored for different time and employing both a static and dynamic controlled atmosphere approach. The transcriptome analysis found that the duration of storage had an important effect on the coordination of gene expression, and the expression patterns of DEGs identified a distinct networks and types of transcriptomic hubs. Samples that developed superficial scald were characterized by higher concentration of chlorogenic acid and a time-specific increase in the expression of PAL and PPO genes. Prevention of superficial scald was related to a peculiar realigning episodes that involved the accumulation of specific antioxidant metabolites, very long chain fatty acids, and the expression of genes that coordinate the hypoxia acclimation process, such as RAP2-like and PCO. A complementary research was conducted to investigate the connection between the application of 1-MCP, low oxygen, and superficial scald. The study highlighted the possible application of genes involved in key fruit ripening pathways, including ethylene-related pathways and sugar/fermentation metabolism, as potential biomarkers for molecular characterization of the major changes that occur during fruit storage. The specific expression of polyphenol oxidase, along with sorbitol-6-phosphate dehydrogenase, were also used to investigate the onset of superficial scald in apples and correlated with the accumulation of the sugar alcohol sorbitol, which plays a protective role against chilling injuries. Assessing the transcriptional signature of these biomarkers can contribute to the development of new tools for a more informed understanding of the physiological progression of postharvest ripening in apples. Overall, these studies provide insights into the genetic and metabolic mechanisms underlying the development and prevention of superficial scald in apple cultivars during storage. The results obtained here paved the way for the identification of a core set of genes to be used as functional marker suitable to better comprehend this phenomenon and to select new accession characterized by a superior fruit quality and postharvest performance.

Development of biochemical and physiological indices of maturity of dessert stone fruit in relation to cool storage

The aim of this research was to develop a new harvest maturity index for highly coloured stone fruit that is not affected by seasonal or climatic factors. The judgement of commercial maturity is difficult with some cultivars of stone fruit because they develop intense skin pigmentation several days before they are ripe. Studies of the changes of the physiochemical and physiological parameters associated with ripening confirmed that no single measurement is suitable for the assessment of harvest maturity in the Japanese type plum cultivars(Prunus salicina Lindl). Two distinct patterns of ripening behaviour were identified. The cool storage (0 degrees Centigrade) responses of fruit harvested at three stages of maturity were examined. Total soluble proteins were extracted and separated from fruit during maturation and ripening using 2-dimensional gel electrophoresis. At least three proteins were first detected in fruit at optimum commercial harvest maturity. Since Gulfruby, Beauty and Rubyred (Japanese type plum cultivars) accumulated similar concentrations of aminocyclopropane-1-carboxylic acid(ACC), it was suggested that the suppressed-climacteric phenotype is the result of an impaired ability of the fruit to convert this compound to ethylene / Doctor of Philosophy (PhD) (Ph.D.)

Também foram mensuradas características de produção, entre as quais, peso do cacho, número de frutos, peso médio dos frutos, produtividade, número de pencas; peso, número, comprimento e diâmetro de frutos da 2º penca. Os atributos fisicoquímicos e químicos analisados foram: textura, pH, acidez titulável, sólidos solúveis, açúcares totais, amido e potássio. Foi adotado o delineamento experimental inteiramente casualizado, com 13 tratamentos (genótipos), 5 repetições e duas plantas por parcela. Foi utilizado o teste de Tukey (P 0,05) para a comparação entre médias de genótipos, dentro de cada grupo genômico. Além disso, realizaram-se possíveis correlações entre as características de crescimento e produção. Some growth characteristics were evaluated such as: plant heights, the pseudostem circumference, number of leaves, number of days between planting and flowering, number of days between flowering and harvest and number of days from planting to harvest. These characteristics were measured at the flowering time. The production characteristics such as the bunch weigth, number of fruits, average weigth of the fruits, productivity, number of hands and weight, number, length and diameter of the fruits of the 2nd hand. The physical and chemical characteristics which were analysed were: firmness, pH, titratable acidity, soluble solids, total sugars, starch and potassium. The experimental design was completely randomized, with 13 treatments (genotypes), 5 replications and 2 plants per plot. We used the test of Tukey (P 0.05) for the comparison between average of genotypes, within each group genomic. In addition to that, some possible correlation between growth and production were also established. The results indicated that in the group AAA, ‘Nanicão’ showed good characteristics regarding growth, production and quality of fruits. Desempenho de híbridos de melão rendilhados cultivados em substrato da fibra da casca de coco reutilizada

O experimento foi conduzido em casa de vegetação em (FCAV-UNESP) Jaboticabal, Estado de São Paulo, Brasil, de março a julho de 2008. Utilizou-se o delineamento em blocos casualizados, em esquema fatorial 2x3, com quatro repetições. Os tratamentos resultaram da combinação de dois híbridos: ‘Bônus no 2’ e ‘Fantasy’, e três tipos de utilização de substrato (S1 = fibra da casca de coco nova; S2 = fibra da casca de coco utilizada uma vez na cultura do pimentão, e S3 = fibra da casca de coco utilizada por duas vezes: 1a na cultura do pimentão e 2a na cultura do melão). O cultivo foi realizado em vasos plásticos de 13 dm3 e dispostos em fileiras duplas, nos espaçamentos de 0.8 m entre fileiras simples, 0.5 m entre plantas na linha e 1.10 m entre fileiras duplas. Foi utilizada irrigação por gotejamento com solução nutritiva recomendada para a cultura. Após a colheita, avaliaram-se: diâmetro transversal do fruto, diâmetro longitudinal do fruto, índice de formato de fruto, diâmetro transversal do lóculo, diâmetro longitudinal do lóculo, índice de formato do lóculo, diâmetro da inserção do pedúnculo, espessura do mesocarpo, firmeza, rendilhamento da casca, massa do fruto, sólidos solúveis, acidez titulável, pH e produtividade. Foi realizada a diagnose foliar e acaracterização físico-química do substrato. Concluiu-se que, para as características produtivas e qualitativas dos frutos não houve diferenças entre os híbridos de melão rendilhado; a reutilização por até três vezes não afetou o desempenho dos híbridos; a reutilização promoveu, nos substratos, acúmulo de nutrientes, aumento da densidade e volume de água facilmente disponível, redução do espaço de aeração e porosidade total / The present work had as objective to evaluate the performance of net melon hybrids cultivated in reused coconut fiber substrate with fertigation, in protected environment. The experiment was carried out in greenhouse at (FCAVUNESP) Jaboticabal, São Paulo State, Brazil, from March to July 2008. The experimental design was randomized complete block design, in 2x3 factorial scheme, with four replications. The treatments resulted of the combination of two net melon hybrids: ‘Bônus no 2’ and ‘Fantasy’ and three types of substrate (S1 = new coconut fiber; S2 = coconut fiber used once, in the culture of sweet pepper; and, S3 = coconut fiber used twice, 1st in the culture of sweet pepper and 2 nd in the culture of the net melon). The cultivation of net melon was carried out in plastic pots of 13 dm3 containing coconut fiber, were arranged in spacing of 0,8 m between simple rows, 0,5 m between plants in rows and 1,10 m between double rows. It was used drip irrigation with nutrient solution recommended to the culture. After harvest were evaluated: transverse fruit diameter, longitudinal fruit diameter, index of fruit shape, transverse locules diameter, longitudinal locules diameter, index of locules shape, diameter of peduncle insertion, pulp thickness, firmness, skin netting, mass of fruit, soluble solids; titratable acidity, pH, productivity. Leaf diagnosis and physical-chemical characterization of substrate were carried out. Concluded that for the productive and qualitative characteristics of fruits there wasn´t observed differences between net melon hybrids; the reuse of three times didn´t affect the performance of net melon hybrids; the reuse of three times caused in the substrate, accumulation of nutrients, an increasing of bulk density and easily available water content, decreasing of the total porosity and aeration space
