O objetivo deste estudo foi avaliar a influência de dois aditivos, propionaldehyde (propanal aldeído) e 2,3-butanedione (diquetona diacetil), nas propriedades de comonômeros, copolímeros e resinas compostas à base de Bis-GMA, quando diluído com TEGDMA, Bis-GMA propoxilado (CH3Bis-GMA) e Bis-GMA propoxilado fluorinado (CF3Bis-GMA). Comonômeros, copolímeros e compósitos experimentais foram preparados combinando Bis-GMA com TEGDMA, CH3Bis-GMA, CF3Bis-GMA e, aldeído (24 e/ou 32 mol%) e diquetona (24 e/ou 32 mol%). No caso dos compósitos, partículas híbridas silanizadas (bário aluminosilicato; 60 peso%) foram adicionadas aos comonômeros. Para a fotopolimerização, adicionou-se canforoquinona (0,2 peso%) e N,N-dimetil-p-toluidina (0,2 peso%). Os comonômeros e copolímeros experimentais tiveram as seguintes propriedades avaliadas: viscosidade, contração de polimerização, grau de conversão, ângulo de contato (em esmalte, dentina e vidro), temperatura de transição vítrea (DSC e equação de Fox), microdureza, alteração de rugosidade superficial e desgaste por abrasão. Os compósitos experimentais, tiveram avaliadas as seguintes propriedades: resistência flexural, módulo de elasticidade, tração diametral, módulo de resiliência, microdureza, alteração de rugosidade superficial e desgaste por abrasão. Os sistemas Bis-GMA/TEGDMA e Bis-GMA/CH3Bis-GMA tiveram suas propriedades significantemente alteradas pela incorporação de aditivos (propanal ou diquetona). Os aditivos (propanal/diquetona) não influenciaram de forma significante as propriedades do sistema Bis-GMA/CF3Bis-GMA. Bis-GMA/TEGDMA and Bis- GMA/CH3Bis-GMA systems had properties significantly influenced by incorporation of additives (either propanal or diketone). Additives (propanal/diketone) did not significantly influence on Bis-GMA/CF3Bis-GMA system properties. The effect of additives on the evaluated material properties is mainly attributed to their ability in improving some resin systems degree of conversion

Bis-GMA (2,2-bis[4(2-hydroxy-3-metacryloxypropoxy)phenil]propane) is the base monomer more frequently used in restorative composite resins. However, this monomer presents some disadvantages, such as high viscosity and two aromatic rings in its structure that can promote allergic reactions to the humans. In this work, the main purpose was to synthesize new monomers from glycidyl methacrylate to use in dental restorative materials. Structural characterization of the monomers was carried out through FTIR and NMR 1H, and eight composites were produced from the new monomers, by addition of silane-treated alumino silicate particles (inorganic filler) and a photocuring system (camphorquinone and ethyl 4-dimethylaminebenzoate). The composites were analyzed by environmental scanning electronic microscopy and the water sorption and solubility, compressive strength and elastic modulus were determined. A commercial composite resin [Z100 (3M)] was used to comparison effect. The new composites presented general characteristics similar to the commercial ones; however, they didn t present the properties expected. This behavior was attributed to the lower degree of monomer reaction and to the granulometry and size distribution of the mineral filler in the polymeric matrix / Atualmente, as resinas compostas constituem-se no material restaurador direto mais utilizado nas interven??es da cl?nica odontol?gica, dada a sua versatilidade e excel?ncia est?tica. O mon?mero mais freq?entemente usado na matriz polim?rica dessas resinas ? o Bis-GMA (2,2-bis[4(2-hidr?xi-3-metacriloiloxipropoxi)fenil]propano). No entanto, esse mon?mero apresenta algumas desvantagens, tais como elevada viscosidade e a presen?a de dois an?is arom?ticos em sua estrutura que podem causar rea??es al?rgicas ao organismo humano. Dual Bypass Gas Metal Arc Welding Process and Control

GMAW (Gas Metal Arc Welding) is one of the most important arc welding processes being adopted in modern manufacturing industry due to its advantages in productivity, energy efficiency and automation. By monitoring and improving some of the important properties of GMAW such as production rate, metal transfer and base metal heat input, researchers could bring the process efficiency and stability to a new level. In recent years, some innovative modifications of GMAW such as Twins, Tandem and laser-MIG hybrid welding have been adopted into many industrial applications for better productivity. In this dissertation, a novel GMAW called DB-GMAW (Dual Bypass Gas Metal Arc Welding) using two GTAW torches and one GMAW torch to construct a welding system, is proposed and developed. In DB-GMAW, two GTAW torches perform the bypass system which decouples the total welding current into base metal current and bypass current after the melt down of filler wire. Compared to conventional GMAW, DB-GMAW has many advantages in droplet formation, base metal heat input and penetration achievement due to its unique characteristics in welding arc and current flow. In the first place of the research, experimental system of DB-GMAW is constructed. Then, sufficient experiments under different parameters are performed to provide us a good understanding of the behaviors and characteristics of this novel GMAW process. Observation about metal transfer formation and base metal heat input is studied to verify its theoretical analysis. Full penetration of work piece via DB-GMAW is achieved based on a series of parameter testing experiments. Moreover, image processing techniques are applied to DB-GMAW to monitor the welding process and construct a feedback system for control. Considering the importance of maintaining stable full penetration during many welding applications, a nonlinear model of DB-GMAW full penetration is developed in this dissertation. To do that, we use machine vision techniques to monitor the welding profile of the work piece. A control algorithm based on the nonlinear model using adaptive control technique is also designed. The achievement of this dissertation provides a fundamental knowledge of a novel welding process: DB-GMAW, and a good guidance for further studies about DBGMAW.

Global optimization applied to kinetic models of metabolic networks

Recientemente, el uso de técnicas de manipulación genética ha abierto la puerta a la obtención de microorganismos con fenotipos mejorados, lo que a su vez ha llevado a unas mejoras significativas en la síntesis de algunos productos bioquímicos. Sin embargo, la mutación y selección de estos nuevos organismos se ha llevado a cabo, en la mayoría de casos, por ensayo y error. Es de esperar que estos procesos puedan ser mejorados si se usan principios de diseño cuantitativos para guiar la búsqueda hacia el perfil enzimático ideal. Esta tesis está dedicada al desarrollo de un conjunto de herramientas de optimización avanzadas para asesorar en problemas de ingeniería metabólica y otras cuestiones emergentes en biología de sistemas. Concretamente, nos centramos en problemas en qué se modelan las redes metabólicas usando expresiones cinéticas. La utilidad de los algoritmos desarrollados para resolver tales problemas es demostrada por medio de varios casos de estudio. / In recent years, the use of genetic manipulation techniques has opened the door for obtaining microorganisms with enhanced phenotypes, which has in turn led to significant improvements in the synthesis of certain biochemical products. However, mutation and selection of these new organisms has been performed, in most cases, in a trial-and-error basis. It is expected that these processes could be further improved if quantitative design principles were used to guide the search towards the ideal enzymatic profiles. This thesis is devoted to developing a set of advanced global optimization tools to assess metabolic engineering problems and other questions arising in systems biology. In particular, we focus on problems where metabolic networks are modeled making use of kinetic expressions. The usefulness of the algorithms developed to solve such problems is demonstrated by means of several case studies.

Osteoblast Behaviour on Injectable Biomaterials Intended for Augmentation of Vertebral Compression Fractures

Biomaterials used for stabilization of compressed vertebraes due to osteoporosis, have mainly been based on resin materials, like PMMA (polymethyl methacrylate), but have recently expanded to consist of injectable ceramics, such as calcium-aluminate. In this in vitro study human osteoblast-like cells, MG-63, were cultured on three different injectable biomaterials based on: Ca-aluminate, Bis-GMA (bisphenol A-glycidylmethacrylate) and PMMA, to investigate the cellular response elicited by these materials. Cell proliferation was measured by the NucleoCounter® system, cell viability was investigated by LDH (lactate dehydrogenase) analysis, cell differentiation and mineralization was evaluated by mRNA gene expression of the osteoblastic markers: ALP (alkaline phosphatase), OC (osteocalcin) and COLL-I (collagen type I) by qPCR (quantitative polymerase chain reaction) analysis. Two control materials were used: TCP (tissue culture polystyrene, negative control) and PVC (polyvinyl chloride, positive control). The results showed that all the bone cement materials were non-toxic and biocompatible, i.e. they provided good cell viability and proliferation of the MG-63 cells. They are specific for bone cells, since they expressed high values of the osteoblast-specific differentiation markers, and are thus promising as injectable bone cement materials. Among the bone cements, Xeraspine appears to be the most biocompatible material for bone cells. It is followed by Cortoss and then Vertebroplastic.

In these days are probiotic lactic acid bacteria (LAB) very often used in food processing industry, such as milk products, cheese and fermented salami production. As well as the food conservation agent. Except of the industry usage of the LAB there are microbiological aspects. Identification of the bacterial species methods based on the isolation and amplification of DNA are very often used last few years. Diploma thesis deal with the bacterial cell of Lactobacillus species lysis and it´s optimalization. At first it was tested the optimal concentration of lysozyme (3mg/ml, 5mg/ml, 10mg/ml) and the exposure times (3, 5 a 10 hours). Another testing was aimed to the use and suitability of washing powder to the bacterial cell of Lactobacillus species lysis. I tested the Amway washing powder optimal concentration (1%, 2%, 3% a 4%). Four of another comercial washing powders were tested too. All these tests were performed at the pure Lactobacillus bacterial culture. To ensure the results I tested the washing powders at the real food matrix (Acidified milk, yogurt mango, yogurt white). All the methods were evaluated at the amplification method PCR with specific primers for the Lactobacillus genus. The DNA isolation was performed with the paramagnetic microsperes P(HEMA-co-GMA) and the amount of the DNA was quantified spectrophotometrically. The PCR products detection was performed with the agarose gel electrophoresis.
