Perfil tripanocida de lignanas dibenzilbutirolactônicas: avaliação das propriedades terapêuticas in vivo nas fases aguda e crônica da doença de Chagas experimental / Trypanocidal profile of dibenzylbutyrolactone lignans: evaluation of therapeutic properties in vivo, in acute and chronic experimental Chagas\' disease

A doença de Chagas, transmitida pelo protozoário Trypanosoma cruzi, afeta mais de 18 milhões de pessoas na América Latina e mesmo tendo sido descrita há 100 anos por Carlos Chagas, esta ainda representa um importante problema de saúde pública, sendo encontrados casos em 18 países em desenvolvimento na América do Sul e Central. No Brasil, o benzonidazol, (ROCHAGAN), é a única droga de atividade tripanocida disponível no mercado mesmo apresentando vários efeitos colaterais e limitada eficácia na fase crônica da infecção. Em vista da necessidade de novas substâncias com atividade biológica sobre T.cruzi, o interesse pela pesquisa vem crescendo com o intuito de se obter compostos capazes de atuarem sobre o parasita, desprovidos dos graves efeitos colaterais. Estudos desenvolvidos em nosso laboratório envolvendo lignanas demonstraram uma importante atividade destes compostos sobre as formas tripomastigotas sanguíneas. Assim este estudo tem como objetivo avaliar a atividade terapêutica de derivados de lignana dibenzilbutirolactônicas em sistema in vivo, em fase aguda e crônica da infecção por T. cruzi. Como critérios de estudo avaliamos o comportamento parasitêmico dos animais na fase aguda, quantificamos a enzima -galactosidase nos tecidos do fígado, baço e coração além da análise morfométrica dos tecidos. Com relação à parasitemia, dos animais na fase aguda, as substâncias cubebina e hinoquina, apresentaram significante redução parasitêmica comparado ao controle negativo e melhor atividade no tratamento pela via oral. Na análise por caracterização de -galactosidase, as substâncias apresentaram melhores resultados comparados ao controle positivo (Benzonidazol) e também melhor eficácia da resposta para o tratamento pela via oral. Nos resultados apresentados pela cariometria os grupos de tratamento com as substâncias cubebina e hinoquinina apresentaram valores próximos ao controle não infectado, mostrando que ocorreu uma resposta celular positiva comparado ao controle infectado. / Chagas disease, transmitted by the protozoan Trypanosoma cruzi, affects over 18 million people in Latin America. Estudos in vitro sobre a atividade anti-helmíntica de lignanas extraídas da piper cubeba e in sílico com as proteínas β-tubulina e fosfoetanolamina metil transferase do haemonchus contortus /

Orientador: Rosangela da Silva Laurentiz / Resumo: O estudo sobre o uso de plantas e metabolitos secundários tem sido uma das alternativas mais estudadas nos últimos anos como forma de controlar a parasitose em ovinos provocada principalmente pelo nematoide Haemonchus contortus. A Piper cubeba é uma pimenta asiática que apresenta uma série de propriedades biológicas, incluindo atividade antiparasitária, sendo essa atividade, muitas vezes atribuida às lignanas cubebina, hinoquinina e diidrocubebina, que estão presentes na planta em maior concentração. Estudos in vitro com o extrato da Piper cubeba e com as lignanas cubebina, dihidrocubebina e hinoquinina demonstraram que esses compostos apresentam importante ação contra os vários estágios do desenvolvimento de vida do nematoide Haemonchus contortus, sendo a hinoquinina a mais eficaz (CE50 = 68 μg / mL contra ovos, CE50 < 35 μg / mL contra o estágio L1 e CE50 = 0.34 μg / mL contra o estágio L3). Devido à relevância desses resultados foram então realizados estudos de docking molecular com essas lignanas com o objetivo de avaliar a ação desses compostos com as proteínas fosfoetanolamina metil transferase (FMT) e a proteína tubulina do H. contortus usando o albendazol como fármaco de referência. O estudo de docking consensual utilizou os softwares Maestro 10.1, GOLD 5.3 e Autodock 4.2. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The study on the use of plants and special metabolites has been one of the most studied alternatives in recent years as a way to control parasitosis in sheep caused mainly by the nematode Haemonchus contortus. Piper cubeba is an Asian pepper that has a series of biological properties, including antiparasitic activity, and this activity is often attributed to cubebin, hinokinin and dihydrocubebin lignans, which are present in the plant in greater concentration. In vitro studies with the Piper cubeba extract and with the lignans cubebin, dihydrocubebina and hinokinin demonstrated that these compounds present important action against the several stages of the development of life of the Haemonchus contortus, being the hinokinin the most effective (EC50 = 68 μg / mL against eggs, EC50 <35 μg / mL against stage L1 and EC50 = 0.34 μg / mL against stage L3). Due to the relevance of these results, molecular docking studies were then carried out with these lignans in order to evaluate the action of these compounds with the phosphoethanolamine methyl transferase (FMT) proteins and the H. contortus tubulin protein using albendazole as a reference drug. Síntese e caracterização de nanopartículas poliméricas para veiculação de lignanas bioativas /

Orientador: Rosangela da Silva de Laurentiz / Resumo: Os produtos naturais são uma fonte inesgotável de compostos químicos com as mais variadas propriedades biológicas, entretanto, muitos deles têm baixa biodisponibilidade e/ou são degradados pelas condições fisiológicas, o que os torna de pouco interesse para uso como potenciais fármacos. Dentre os vários produtos naturais bioativos as lignanas apresentam um significativo potencial citotóxico contra várias linhagens de células tumorais, como a (-)-hinoquinina (HNK), (-)-cubebina (CB) e (-)-6,6’-dinitrohinoquinina (DN), determinado em outros trabalhos. Entretanto, essa lignanas possuem caráter hidrofóbico e podem ser degradadas em condições fisiológicas dependendo do pH. Desta forma, o presente trabalho teve por objetivo estudar a incorporação dessas lignanas em nanopartículas poliméricas (NNPs), como forma de melhorar a biodisponibilidade, perdas por degradação e os efeitos da incorporação sobre a atividade citotóxica desses compostos. Neste estudo a HNK e CB foram extraídas da Piper cubeba e a DN foi sintetizada a partir da reação de nitração da HNK. Essa lignanas foram submetidas a ensaios de citotoxicidade contra as linhagens tumorais Hep-2 e Si-Ha. Among the several bioactive natural products the lignans present a significant cytotoxic potential against several tumor cell lines, as (-)-hinokinin (HNK), (-)-cubebin (CB) and (-)-6,6’-dinitrohinokinin (DN), determined in previous works. However, these lignans have a hydrophobic character and can be degraded in physiological conditions depending on pH. Nos ensaios de citotoxicidade todas as lignanas foram ativas, com destaque para a inibição de 98% da HNK sobre a linhagem Hep-2. As três lignanas foram incorporadas em NNPs de PLGA utilizando o método de nanoemulsão-evaporação com agitação mecânica e gotejamento, entretanto a estabilidade dessas NNPs e as porcentagens de incorporação indicaram que apenas as NNPs-PLGA-HNK apresentaram os parâmetros necessários para a utilização nos ensaios de citotoxicidade. Portanto, outros estudos foram realizados com as três lignanas visando a melhora nos métodos de obtenção e estabilidade com o uso de sonicador e diferente composição do PLGA (50:50) e PLGA-PEG. As análises físicas mostraram que as nanopartículas de PLGA (50:50) obtidas através da metodologia por sonicação foram mais estáveis com menores tamanhos e maior potencial zeta em valores absolutos, entretanto, a DN e a CB não foram encapsuladas de forma eficiente. Desta forma, somente a HNK prosseguiu no estudo. O uso do PLGA-PEG alterou de forma significante os parâmetros físicos e químicos das NNPs-PLGA-PEG-HNK em relação as de PLGA-HKN. Os testes de liberação mostraram melhores resultados com as NNPs-PLGA-HNK, principalmente em pH 7,4. O tempo de liberação total da HNK em todas as NNPs avaliadas foi em torno de 30 minutos o que sugere que as nanopartículas obtidas são nanocápsulas. A comparação dos resultados dos ensaios de citotoxicidade mostraram que o CI50 das NNPs de PLGA-HNK foi de 29,8 µM sobre as células tumorais Si-Ha, enquanto a HNK livre apresentou CI50 = 110 µM. Para as outras células tumorais mais agressivas as NNPs de PLGA(50:50)-HNK foram menos eficazes do que a HNK livre, com cerca de 50% de perda da atividade. In the cytotoxicity assays all lignans were active, with emphasis on inhibition of 98% da (-)-hinokinin about the Hep-2 tumoral line. The three lignans were incorporated into PLGA (65:35) NNPs, however the stability of these PNPs and the percentages of incorporation indicated that only the PLGA-HNK PNPs had the parameters required for use in the cytotoxicity assays. Therefore, other studies were performed with the three lignans aiming to improve the methods of obtaining and stability from sonicator and different composition in PLGA: (50:50) and PLGA-PEG. The physical analyzes showed that PLGA (50:50) nanoparticles obtained by sonication were more stable with smaller sizes and higher zeta potential in absolute values. However, even in this method, DN e CB were not efficiently encapsulated. In this way, only HNK continued the study. The use of PLGA-PEG altered the physical and chemical parameters of PLGA-PEG-HNK NNPs obtained when compared to PLGA-HNK NNPs. Release tests showed better results with NNPs-PLGA-HNK, especially at pH 7.4. The total release time of HNK in all NNPs evaluated was around 30 minutes, which suggests that the nanoparticles obtained are nanocapsules. Comparison of cytotoxicity assay results showed that PLGA-HNK PNPs IC50 was 29.8 μM on the Si-Ha tumor cells, while the free HNK showed IC50 = 110 μM. For the other more aggressive tumor cells PLGA (50:50)-HNK PNPs were less effective than free HNK with about 50% loss of activity. These results indicate the effectiveness of PLGA(65:35)-HNK NPPs on cells of the Si-Ha line, and that the encapsulation of HNK in PLGA NNPs may be an alternative to decrease decomposition and increase bioavailability for conducting biological assays in vivo. / PSDE/88881.134923/2016-01

A isoforma 1 esta relacionada com a citoproteção gástrica, agregação plaquetária e função renal e a isoforma 2 relacionada a processos inflamatórios. Estes inibidores seletivos apesar de não apresentarem os efeitos colaterais (ulceras e gastrites) dos anti-inflamatórios não esteroidais (AINEs) clássicos por inibirem apenas a COX-2, apresentam grave risco cardiovascular, o que motivou à retirada do rofecoxibe do mercado. Porém, por ser um eficiente inibidor seletivo da COX-2 a estrutura do rofecoxibe tornou-se referência no estudo de novas substâncias capazes de inibir seletivamente a COX-2. Dentre as ferramentas utilizadas na busca destas novas estruturas está a modelagem molecular através de programas como o GOLD 5.1, que foi utilizado neste trabalho. O uso do GOLD 5.1 possibilitou o estudo do comportamento das estruturas avaliadas em ligação com as ciclooxigenases. O objetivo foi obtenção de estruturas com comportamento semelhante ao rofecoxibe (em relação às COXs) como potenciais candidatos ao desenvolvimento de novos inibidores seletivos para a COX-2. O estudo foi realizado com 480 estruturas modeladas a partir de lignanas naturais como a hinoquinina, cubebina, deoxipodofilotoxina e podofilotoxina, que apresentam atividade anti-inflamatória in vivo ou in vitro, além de semelhanças estruturais com o rofecoxibe. A deoxipodofilotoxina por apresentar seletividade para a COX-2 em ensaio in vitro também foi utilizada como estrutura de referência além do rofecoxibe. Os resultados observados a partir da simulação molecular permitiram concluir que embora tanto o rofecoxibe como a deoxipodofilotoxina (3) inibam seletivamente a COX-2 in vitro, o fazem de modo diferente. Em relação a COX-2 as duas estruturas ocupam a mesma região do sítio ativo, mas o rofecoxibe apresenta interações mais fortes com o bolso hidrofílico desta isoforma (condição necessária para a inibição seletiva para os coxibes). Já para a COX-1 enquanto o rofecoxibe ocupa a porção superior do canal hidrofóbico (sítio ativo) como os demais AINEs, a deoxipodofilotoxina ocupa uma região vizinha. Pelos resultados obtidos é possível sugerir que tanto a maior flexibilidade das estruturas como a presença do anel lactônico, são importantes para um comportamento análogo ao rofecoxibe ou à deoxipodofilotoxina. Com relação à interação com o bolso hidrofílico da COX-2, os resultados sugerem que a presença de grupos aceptores de prótons menos volumosos nas posições C3 e C4, C3’ e C4’ ou C4 levam a resultados melhores que grupos aceptores de maior volume. A presença de grupos doadores de prótons apesar de permitirem forte interação com o bolso hidrofílico da COX-2 leva a resultados globais insatisfatórios, pois formam interações fortes com o resíduo Arg120 do sítio ativo da COX-1, interação considerada importante para a inibição não seletiva. Resultado semelhante à deoxipodofilotoxina foi observado apenas para a estrutura 17. As estruturas 37, 188, 266, 267, 348 e a hinoquinina (4) apresentam resultados semelhantes ao rofecoxibe, para as duas isoformas. Deste modo permite-se sugerir a partir dos resultados obtidos neste estudo que a hinoquinina (4) e as estruturas 17, 37, 188, 266, 267 e 348 apresentam-se como possíveis protótipos de fármacos que atuem como inibidores seletivos para a COX-2. / The selective inhibitors of the cyclooxygenase-2 (COX-2) as rofecoxib (2) and celecoxib (1), form an important class of anti-inflammatory drugs developed from the discovery of two isoforms of cyclooxygenases (COX-1 and COX-2) in the late 1979. Isoform 1 is related to the gastric cytoprotection, platelet and renal function and isoform 2 related to inflammatory processes. These selective inhibitors although they did not side effects (ulcers and gastritis) of the classic NSAIDs to inhibit only COX-2, have severe cardiovascular risk, which led to the withdrawal of rofecoxib from the market. However, to be an effective selective COX-2 to rofecoxib structure has a reference in the study of new substances capable of selectively inhibiting COX-2. Among the tools used in the search of these new structures is by molecular modeling program such as GOLD 5.1, which was used in this work. Using GOLD 5.1 made it possible to study the behavior of structures evaluated in binding with the cyclooxygenases. With the objective of obtaining structures with similar behavior to rofecoxib (regarding behavior with COX) as potential candidates for the development of new selective inhibitors for COX-2. The study was conducted with 480 structures modeled from natural lignans as hinokinin, cubebin, deoxypodophyllotoxin and podophyllotoxin, which have anti-inflammatory activity in vivo or in vitro as well as structural similarities with rofecoxib. The deoxypodophyllotoxin for presenting selectivity for COX-2 in the in vitro assay was also used as a reference structure beyond rofecoxib. The results observed from the molecular simulation showed that although both rofecoxib (2) as deoxypodophyllotoxin (3) selectively inhibit COX-2 in vitro, they do differently. In relation to COX-2 the two structures occupy the same region of the active site, but rofecoxib has stronger interactions with the hydrophilic pocket of this isoform (a necessary condition for the selective inhibition for coxibs). As for the COX-1 while rofecoxib occupies the upper portion of the hydrophobic channel (active site) like other NSAIDs, the deoxypodophyllotoxin occupies a neighboring region. From the results it is possible to suggest that the greater flexibility of the structures such as the presence of the lactone ring, are important for a similar behavior to rofecoxib or deoxipodofilotoxina. With respect to the interaction with the hydrophilic pocket COX-2, the results suggest that the presence of acceptors groups less bulky protons in posítions C3 and C4, C3 ' and C4' and C4 lead to better results than acceptors groups of larger volume. The presence of proton donors groups despite allowing strong interaction with the hydrophilic pocket COX-2 lead to poor overall results, since they form strong interactions with Arg120 residue of COX-1 active site, considered important interaction for inhibiting non-selective. Results similar to deoxipodofilotoxina was only observed for structure 17. Structures 37, 188, 266, 267, 348 and hinokinin (4) show results similar to rofecoxib for the two isoforSA. Thus it allows suggest from the results obtained in this study hinokinin (4) and structures 17, 37, 188, 266, 267 and 348 are shown as possible prototype drugs that act as selective inhibitors for COX-2.

Estudos de modelagem molecular de lignanas em complexos com ciclooxigenases-1 e 2 /

Orientador: Rosangela da Silva de Laurentiz / Resumo: Os inibidores seletivos da ciclooxigenase-2 (COX-2), como o rofecoxibe (2) e o celecoxibe (1), formam uma importante classe de medicamentos anti-inflamatórios desenvolvidos a partir da descoberta das duas isoformas das ciclooxigenases (COX-1 e COX-2) na década de 1979. A isoforma 1 esta relacionada com a citoproteção gástrica, agregação plaquetária e função renal e a isoforma 2 relacionada a processos inflamatórios. Estes inibidores seletivos apesar de não apresentarem os efeitos colaterais (ulceras e gastrites) dos anti-inflamatórios não esteroidais (AINEs) clássicos por inibirem apenas a COX-2, apresentam grave risco cardiovascular, o que motivou à retirada do rofecoxibe do mercado. Porém, por ser um eficiente inibidor seletivo da COX-2 a estrutura do rofecoxibe tornou-se referência no estudo de novas substâncias capazes de inibir seletivamente a COX-2. Dentre as ferramentas utilizadas na busca destas novas estruturas está a modelagem molecular através de programas como o GOLD 5.1, que foi utilizado neste trabalho.
