Nanogels de polysaccharides pour la délivrance d’insuline

Les nanogels sont de bons candidats pour la délivrance d’actifs. Ces réseaux de polymères réticulés et de taille nanométrique, sont gonflés d’eau. Ils sont donc capables d’encapsuler une protéine à l’intérieur de leurs pores et de la libérer en fonction de l’état de gonflement du réseau. Cet état peut être modulé par la densité de réticulation du réseau ou par l’application d’un stimulus externe tel que le pH, la température ou encore une biomolécule telle que le glucose. Ainsi, les nanogels sensibles au glucose se présentent comme des candidats idéaux pour administrer l’insuline de façon asservie à la glycémie. Afin de satisfaire aux critères de biocompatibilité et de biorésorption des vecteurs, nous avons choisi de développer des nanogels à base de polysaccharide, en particulier à base d’acide hyaluronique (HA). Ceux-ci sont obtenus par réticulation du HA, préalablement modifié par des fonctions réticulables telles que les méthacrylates, dans des nanogouttes d'émulsion eau-dans-huile. Des nanogels de taille et de porosité modulables ont été synthétisés grâce à un bon contrôle 1) de la modification chimique des précurseurs par des fonctions réticulables (taux de méthacrylation), 2) de l’émulsion matricielle (taille, stabilité), 3) des conditions de réticulation par photopolymérisation gouvernant le taux de conversion des méthacrylates. Ce savoir-faire a ensuite été appliqué à la synthèse de nanogels modifiés par des dérivés de l’acide phénylboronique, ligand du glucose, afin d’obtenir des matériaux dont le taux de gonflement varie en fonction de la glycémie. L’intérêt applicatif de ces objets a été évalué vis-à-vis des propriétés d’encapsulation de l’insuline, de dégradabilité enzymatique, et de biocompatibilité. / Nanogels are an attractive class of delivery systems. These soft particles, made of highly swollen polymer network, can physically entrap a drug and release it at a rate depending on its diffusion though the network. Therefore, any change in the swelling degree can trigger the release kinetics. This parameter can be tuned by modifying the density of cross-links in the gel matrix or by changing the environmental conditions such as pH, temperature or analyte such as glucose. Thus, glucose-responsive nanogels are good candidates to be used as self-regulated systems for insulin delivery. To fulfill both biocompatibility and biodegradability criteria, our attention has been focused on the design of new nanogels made of polysaccharides, in particular made of hyaluronic acid (HA), as a main constituent. HA was at first covalently modified with polymerizable methacrylate functions and confined in nanoreactors during photopolymerization using water-in-oil miniemulsions as template. Biodegradable nanogels with a well-defined size and various cross-linking degrees were thus achieved, thanks to a good control of 1) the chemical modification of HA with methacrylates (degree of methacrylation) 2) the emulsion template (size, stability), 3) the photopolymerization conditions which governed the conversion rate of the polymerization. Further modification of the polysaccharide with phenylboronic acid as a glucose-sensitive group yielded nanogels whose swelling behavior could vary as a function of glucose concentration. These systems were further studied as insulin delivery systems. Moreover, their biodegradability, stability and biocompatibility were assessed.

This diploma thesis deals with the optimalization of synthesis of gold nanoparticles and their surface modification allowing their use as contrast agents for in vivo imaging by CT. Gold nanoparticles were prepared by the Turkevich method and characterized by TEM, DLS, MADLS and UV -Vis. Their surface was functionalized with polyethylene glycol containing a thiol group forming a bond with the Au atoms in the surface of gold nanoparticles. The terminal end of the polymer was methylated or containing an aminooxy group forming an orthogonal bond with hyaluronic acid using click-chemistry. The eligibility for in vivo application of the prepared nanoparticles was verified with stability and cytotoxicity tests. The nanoparticles modified by methylated polyethyleneglycol were injected intravenously into a mouse and their application potential as contrast agents were verified by CT.

Hyaluronic acid (HA) is an important substance, which is mostly used in pharmaceutical and cosmetic industry. This substance is commonly found in the human body. HA is one of the factors contributing to virulence of microorganisms. Some bacterial strains produce hyaluronic acid in the form of a mucoid capsule that encapsulates the cell to protect bacteria against the immune system of the host organism. One of the main producers is the bacterial strain Streptococcus equi subsp. zooepidemicus. Contipro a.s. uses the strain CO4A to produce hyaluronic acid in large scale. The production strain was obtained by random mutagenesis by UV light. The aim of the work was to study changes in the genome, which led to a significant increase in hyaluronic acid production, using DNA microarray and real-time PCR (qPCR). The genome of the strain CO4A was sequenced and compared to reference ATCC35246 [1]. The size of the genome is 2,167,251 bp and 83 relevant variants (59 SNV and 34 indels) have been identified. Variants in coding regions were annotated and amino acid sequence changes were determined. In SNV mutations there was a change in the amino acid sequence in 45 cases. The change was identified in every case of indel mutations. The expression level of selected groups of genes was monitored in both strains by the method of DNA microarrays. A cascade of increased expression level of amino sugar metabolism genes leading to the synthesis of UDP-N-acetyl glucosamine was observed in strain CO4A (the increase in expression level of these genes compared to ATCC35246 was on average 28 %). Subsequently, the expression of selected genes was verified by qPCR. There was no significant difference in the expression level of the has operon genes of both strains. The effect of supplementation of the culture medium with N-acetylglucosamine (GlcNAc), which is one of the precursors of HA synthesis, was also studied by qPCR. A positive effect of the supplementation of the culture medium with external GlcNAc in the CO4A strain has been recorded. Also, the supplementation has positive effect on the yield of HA from the medium (increase in yield was on average by 17 %). GlcNAc has been shown to have a positive effect on the yield of HA in ATCC35246 strain as well (increase in yield was 9 % on average), but no significant changes in the expression levels were found in selected groups of genes in ATCC35246.

This diploma thesis studied the interaction of ionic and amphiphilic fluorescent probes with polyelectrolytes. The research of such colloidal system could be helpful in deeper understanding of interactions between polyelectrolytes and surfactants. The aim of this thesis was to determine whether ionic or amphiphilic probes are suitable for this research. Two polyelectrolytes, sodium polystyrene sulfonate and hyaluronan were used in this study. Interactions of polyelectrolytes with fluorescent probes (4-Di-2-Asp, methylene blue, dodecyl acridine orange, DiO and DiA) were studied in this work. Then cationic surfactant cetyltrimethylamonium bromide was added to this system. Samples were studied using fluorescence and UV/VIS spectroscopy. Emission and absorption spectra of these probes were recorded. This experiment was based on hypothesis that due to positive charge these probes will bind to the negatively charged polyelectrolyte. Formation of dimers or other aggegates was presumed for fluorescent probes dodecyl acridine orange and methylene blue. When 4-Di-2-Asp was used,changes in fluorescence intensity and absorption spectra in connection with possible formation of aggregates was supposed to be observed. It should be noted that formation of dimers for 4-Di-2-Asp is not described in literature. The formation of dimers wasn't clearly demonstrated during the measurement. Formation of dimers was observed only with dodecyl acridine orange. Probes DiO and DiA were used only in the study of the interaction with polyelectrolytes. These fluorescent probes showed up as inappropriate, because they adsorbed on tube walls. When cationic surfactant was added to hyaluronan-fluorescent probe system, surfactant pushed away fluorescent probe and bonded electrostatically to negatively charged groups of polyelectrolyte.

Dvouohnisková FCS ve výzkumu koloidů / Dual-focus FCS in colloidal research

Tato práce se zabývá studiem fluorescenčně značeného hyaluronanu, konkrétně rhodaminylamino hyaluronátu sodného (Hya-Rh, 40 kDa), pomocí dvouohniskové fluorescenční korelační spektroskopie (2f-FCS). Nejdříve byla prostudována literatura týkající se využití FCS techniky v koloidní chemii a při studiu polymerů, přičemž následně byly shrnuty veškeré poznatky o využití 2f-FCS metody. Na základě prvotních měření byl zjištěn vhodný postup přípravy a způsob uchovávání vzorku Hya-Rh používaném pro následující experimenty. Záhy byly prostudovány možné vlivy koncentrace Hya-Rh na jeho difúzní charakteristiky jak ve vodě, tak ve fyziologickém roztoku. Následně bylo studováno chování Hya-Rh a vliv koncentrace solí alkalických kovů ve vodných roztocích těchto solí a fluorescenčně značeného hyaluronanu. Poté bylo sledováno chování Hya-Rh v závislosti na koncentraci velmi nízkomolekulárního hyaluronanu (VLMW HA, 404 kDa) v čisté vodě i ve fyziologickém roztoku, přičemž získané výsledky byly mezi sebou porovnány. Nakonec bylo využití 2f-FCS metody celkově zhodnoceno a popsáno z hlediska studia chování fluorescenčně značeného hyaluronanu v roztocích.

This thesis is focused on the development and characterization of hydrogels containing humic substances with possible use in cosmetic and pharmaceutical industries. This work follows on the bachelor thesis, theoretical and experimental part is based on already acquired knowledge. The aim of this work was to develop a literature review focusing on the potential use humic gels in practice and also to enhance the consistency of other active ingredients. Based on this literature review, the main aim was to prepare model humic hydrogels with selected active ingredients and their characterization by basic methods of material analysis. These basic methods were especially rheology and visual assessment of consistency during the preparation, then the samples were subjected process of drying and swelling. Experimental results showed considerable influences during the preparation and composition of each sample, which then reflected in their structure and consistency. Furthermore, it was found that the suitable composition and combination of ingredients form hydrogels acceptable characteristics for further use in cosmetic or pharmaceutical applications.

Hyaluronan (HA) is a substance that is commonly used in biomedical applications in the form of hydrogels. One of these biomedical applications is dermatological fillers where HA is cross-linked with 1,4-Butanediol diglycidyl ether (BDDE) to reduce its rapid turnover within tissue. The filler gives a volumetric effect that can fill out wrinkles. This literature study was conducted in collaboration with Galderma to determine if there is research that explores additives to HA hydrogels that give both volumetric and biological effects when applied as filler. Biological effects that improve the skin's appearance and complexion such as a rejuvenation of the skin was preferable. Both polynucleotides and mannitol show great potential to act as additives in injectable hyaluronan hydrogels.  The main effect of polynucleotides (PN) added in hydrogels is that it is collagen stimulating and provides a more natural tissue regeneration. Rheological properties of the filler change with the addition of PN, where elasticity, viscosity and viscoelasticity have been shown to increase. PNs show no toxicity and are considered safe to inject. The study of mannitol has shown that it does not give a volumetric effect after the injected hydrogel has been broken down. What mannitol can help with, is to prolong the life of the hydrogel and reduce the swelling that is a common side effect after an injection. In addition to this, mannitol is a safe substance to inject.

The development of functional hyaluronan hydrogels for neural tissue engineering

Tissue engineers – in order to develop therapies for the treatment of complex neurological injuries and diseases – attempt to recreate elaborate developmental mechanisms in vitro. Neuronal precursor cells are excellent candidates for the study of developmental operations such as cell adhesion, differentiation, and axonal pathfinding. Hyaluronan (HA) is a common polysaccharide that is found extensively throughout the neuronal extracellular matrix (ECM), and can be functionalised and crosslinked to form stable hydrogels that support growing neuronal cells. Hyaluronan hydrogels can be modified chemically and mechanically to mimic the ECM of the developing brain, awarding control over mechanisms such as differentiation and axonal pathfinding. This thesis is concerned with the functionalisation and characterisation of HA hydrogels, ultimately in order to simulate vital properties of the developing brain. Here we show that HA hydrogels can be finely tuned mechanically (by modulating stiffness and viscosity), and chemically, by the conjugation of peptides that mimic the neural cell adhesion molecule (NCAM). NCAM mimics and novel mimics of sialylated NCAM significantly influence the differentiation of NSPCs in 2D and 3D. HA hydrogels successfully support long term culture of neural cells in 3D, and encourage the formation and extension of neurites of several cell types including human, mouse and rat neuronal precursor and stem cells. These results demonstrate for the first time that novel NCAM mimicking peptides can be conjugated to well defined hydrogel matrices that influence intricate developmental behaviours in 3D. Understanding how neural cells form functional networks is essential for the development of clinical approaches that attempt to address the injuries and diseases that affect these systems.

Influence of the processes parameters on the properties of the polylactides based bio and eco-biomaterials / Influence des paramètres de procédés sur les propriétés et éco-composites à base de polylactides

Le travail présenté dans ce manuscrit concerne la fabrication de biomatériaux poreux à base d’acide polylactique pour les tissus conjonctifs et calcifiés en utilisant des procédés de chimie verte. Le but de cette thèse est de corréler l’influence de certains paramètres de procédés à la structure morphologique et les propriétés des mousses générées. Nous avons étudié, d’un côté, les effets de mélange d’acide hyaluronique et d’acides polylactiques afin d’améliorer les propriétés d’adhésion de ces biomatériaux. Nos résultats montrent bien une augmentation de l’énergie d’adhésion mais aussi une diminution de la taille équivalente des pores et de la porosité des biomatériaux poreux après moussage par les fluides supercritiques. D’un autre côté, nous avons étudié les effets de mélanges des triphosphates de calcium et d’acides polylactiques en tant que substitut osseux. L’influence d’un ajout de cires en tant qu’agent porogène a été discutée et les méthodes de préparation des pastilles (voie sèche ou humide) ont été analysées. Dans cette optique la fabrication semi-industrielle de biomatériaux poreux a été testée en fixant les paramètres du procédé de moussage par le CO2 supercritique (pression, température et temps de saturation, vitesse de dépressurisation) et nous avons contrôlé les mousses de formulations optimisées en termes de porosité et de distribution des pores. En conclusion, ce travail rend possible d’adapter les paramètres des procédés de CO2 supercritique et de co-broyage aux propriétés des biomatériaux poreux. En perspective, cette ouvre la voie à de nouvelles recherches à la fois dans les domaines des modèles 3D tumoraux et d’ingénierie tissulaire. / The work presented in this manuscript concerns the production of scaffolds based polylactides for connective tissues and bone regeneration by adapting green technology. The aim of this thesis was to correlate the influence of different process parameters on the morphological structures and properties of the scaffold generated. On one hand, we studied effect of the blending of hyaluronic acid and polylactides to enhance the surface adhesion properties of scaffolds. Our results relate to an increase in surface properties but a decrease of equivalent pore size and porosity after foaming scaffolds by supercritical process. Calcium Tri-Phosphate On other hand, we studied the effect of the blending of calcium tri-phosphates and polylactides as bone substitute. Influence of adding wax as porogen agent has been discussed and a comparison between wet and dry methods to generate scaffolds has been analyzed. For this purpose, semi-industrial fabrication of porous biomaterials has been tested by blocking supercritical CO2 parameters (saturation pressure, temperature and time, depressurization rate) and you have control the optimized formulation composite scaffold, in term of porosity and distribution of pores. In conclusion, this work made it possible to adapt the process parameters of supercritical CO2 and co-grinding at the properties of scaffolds. In perspective, this research opens new development ways in scaffolds, in both domains of 3D tumoral model and tissue engineering.

Efeito da associação da triancinolona à viscossuplementação do joelho / Effect of the addition of corticosteroid to viscosupplementation of the knee

O presente estudo destinou-se a avaliar se os resultados clínicos iniciais da viscossuplementação poderiam ser melhorados com a adição de corticosteróide. As injeções intra-articulares são usadas há muitos anos no tratamento da osteoartrite dos joelhos, principalmente com suspensões cristalinas de corticosteróides. A viscossuplementação é uma intervenção relativamente nova, atualmente recomendada no tratamento da osteoartrite. Trata-se da injeção de ácido hialurônico exógeno em articulações diartrodiais, visando, além de restaurar as propriedades reológicas do líquido sinovial, efeitos modificadores da doença osteoartrite. Revisões sistemáticas mostram que a melhora clínica ocorre em duas a cinco semanas após a viscossuplementação. Comparando-se a viscossuplementação com a injeção intraarticular com corticosteróides, dados recentes sugerem maior eficiência no alívio da dor nas quatro primeiras semanas após a infiltração com corticosteróides, similaridade dos procedimentos ao redor da quarta semana e melhores resultados com a viscossuplementação após a oitava semana. Este inicio de ação mais tardio, associado a relatos de sinovite reacional após a viscossuplementação podem desencorajar médicos e pacientes ao uso desta modalidade de tratamento. No presente estudo foram avaliados 104 pacientes em tratamento para osteoartrite do joelho no grupo de doenças osteometabólicas do Instituto de Ortopedia do Hospital das Clínicas da FMUSP. Os pacientes foram randomizados em dois grupos. Um dos grupos foi denominado VS e recebeu uma única injeção intra-articular de 6ml de Hylan GF-20 (Synvisc One®-Genzyme) no joelho estudado. O segundo grupo foi denominado VS+T e recebeu uma injeção intra-articular de 6ml de Hylan GF-20 (Synvisc One®-Genzyme) mais 1ml (20mg) de Hexacetonido de Triancinolona (Triancil®-Apsen). Foram aplicados a escala visual analógica de dor (EVA) e os questionários de WOMAC e Lequesne uma semana antes da injeção e após uma, quatro, 12 e 24 semanas. Os dois grupos com 52 pacientes cada eram homogêneos. Na primeira semana, o WOMAC e a EVA apresentaram melhores resultados no Grupo VS+T (p < 0,01) em relação ao Grupo VS. Na quarta semana não houve diferença entre os grupos. Ambos apresentaram resultados similares nas semanas 12 e 24. Concluiu-se que a adição de hexacetonido de triancinolona melhorou os resultados clínicos da viscossuplementação no curto prazo, sem interferir nos resultados a longo prazo ou na incidência de efeitos adversos / The present study aims to assess if the initial results of viscosupplementation can be improved by the addition of corticosteroid. Intraarticular injections have been used for many years to treat arthritis and other painful articular disorders, mainly using long-lasting crystalline corticosteroid suspensions. Viscosupplementation is a relatively new intervention that is now widely used and recommended for the treatment of knee osteoarthritis. It is comprised of the injection of exogenous hyaluronic acid in diarthrodial joints, in order to restore the rheological properties of synovial fluid and also to promote osteoarthritis disease-modifying effects. Several placebo-controlled studies reported that clinical improvement began only within two to five weeks after viscosupplementation. When comparing viscosupplementation versus intraarticular injection of corticosteroid, recent data suggest that from baseline to week four, intraarticular steroid were more effective for pain relief. By the fourth week, however, both provided similar relief, but beyond the eighth week, hyaluronic acid provided greater pain reduction. The mechanism of action of hyaluronic acid, with delayed onset of pain/functional improvement, combined with reports of reactional sinovitis may discourage physicians and patients regarding this treatment modality. The present study evaluated 104 patients receiving usual care for knee osteoarthritis at the University of São Paulo Medical Center. Patients were randomized to receive either a single intra-articular injection of 6ml of Hylan GF-20 (Synvisc One®-Genzyme) (Group VS) or a single intra-articular injection of 6ml of Hylan GF-20 (Synvisc One®-Genzyme) plus 1ml (20mg) of Triamcinolone Hexacetonide (Triancil®-Apsen) (Group VS+T). VAS, WOMAC and Lequesne questionnaires were applied one week prior the injection, and after one, four, 12 and 24 weeks. The two groups with 52 patients each were homogeneous. At week one, WOMAC and VAS showed better results for Group VS+T compared to Group VS (p < 0,05). At week four the scores did not show statistically significant differences. The groups showed similar results at weeks 12 and 24. In conclusion, the addition of triamcinolone improved first-week symptom and functional scores of viscosupplementation, but not beyond. It did not seem to alter the likelihood of adverse effects
