There are eight techniques that the investigator should apply in order to identify suspects or perpetrators of a criminal act. To conduct effective investigation, it is important for the criminal investigators to be familiar with the techniques of identification and the application thereof. The description of offenders plays an important role as far as arresting offenders is concerned. The methods used - or to be used - by investigators to describe a person were also discussed. Establishment of a genetic database and molecular methods for the identification of fish species available on the South African market

Thesis (PhD (Food Sc)--Stellenbosch University, 2011. / ENGLISH ABSTRACT: Consumers have the right to accurate information on the fish products they purchase to enable them to make educated seafood selections that will not endanger their own wellbeing or the wellbeing of the environment. Unfortunately, marine resource scarcity, financial incentives and inadequate or poorly enforced regulations have all promoted the mislabelling of fish species on global markets, the results of which may hold economic, conservation and health consequences. The primary aims of this study were to determine the most commonly available fish species on the South African market, to establish and compare DNA-based methods for the unambiguous identification of these species and to utilise the most applicable methods to evaluate the extent of mislabelling on the local fisheries market. The results from surveys of n = 215 restaurants and n = 200 retail outlets in four South African provinces (Western Cape, Kwa-Zulu Natal, Eastern Cape and Gauteng) indicated that 34 and 70 nominal fish types were available in restaurants and retail outlets, respectively, the most common of which were kingklip, salmon and hake. Over 30% of the fish species being sold were of conservation concern, while several outlets marketed specially-protected, illegal-to-sell species in South Africa. Fish purveyors were poorly equipped to provide information on the identity, origin, production method (farmed/wild) and sustainability of the fish they were selling and the labelling of many packaged fish products was in contravention with South African regulations. Data were published for the first time comparing the efficiency of five methods (urea-SDS-proteinase K, phenol-chloroform, salt extraction, SureFood PREP kit and Wizard Genomic DNA Purification kit) for the extraction of DNA from the muscle tissue of fish species available in South Africa. The SureFood kit was identified as the most suitable method for DNA extraction from fish muscle, extracting significantly (P < 0.05) higher DNA yields than all other methods evaluated and being simple and safe to use. A comprehensive reference library of genetic information was compiled for the first time that contains sufficient DNA sequence data from different mitochondrial DNA loci (16S ribosomal RNA (rRNA), 12S rRNA and cytochrome c oxidase I (COI) genes, as well as the control region) to allow the explicit identification of 53 fish species in South Africa. Although 16S and 12S rRNA gene sequencing allowed the identification of most fish to the genus level, the discrimination of closely-related, congeneric species was problematic when based on these gene regions. Conversely, the vast majority (98%) of fish examined could be readily differentiated by their COI sequences, with only members of the genus Thunnus requiring supplementary control region sequencing for species confirmation. Lastly, sequencing of the COI region was used to show that 9% of fish samples collected from local seafood wholesalers and 31% of samples from retail outlets were mislabelled. This study has established that fish mislabelling is a reality on the South African market and that DNA-based methods should be applied by both industry and regulatory bodies to deter illegal activities and to promote transparency on the domestic fisheries market. / AFRIKAANSE OPSOMMING: Verbruikers het die reg tot akkurate informasie rakende die visprodukte wat hulle aankoop. Molecular genetic strategies to identify Obsessive-compulsive disorder (OCD) and schizophrenia candidate genes in a South African sub-population group

Dissertation (PhD)--University of Stellenbosch, 2007. / ENGLISH ABSTRACT: Obsessive-compulsive disorder is a severe, debilitating psychiatric disorder for which the underlying molecular aetiology still remains unclear. Evidence from family studies have suggested that OCD may be caused by a complex interplay of environmental and genetic factors. In order to identify the genetic factors that mediate OCD susceptibility, several genetic association studies have been undertaken, which have yielded inconsistent findings. Moreover, the majority of these studies have focused on a small number of candidate genes that encode components of the serotonin and dopamine neurotransmitter pathways. However, based on the complexity of clinical manifestations observed in OCD, it is likely that its pathogenesis is mediated by a broader complex of interrelated neurotransmitter systems and signal transduction pathways; consequently there is a need to identify and assess novel candidate genes. One method of identifying such novel OCD candidate genes is by utilising knowledge of diseases with phenomenological overlap with OCD, which lend themselves to better genetic dissection through linkage analysis and animal studies. Genetic loci for such disorders, identified though linkage analysis, could potentially harbour novel OCD candidate genes, while genes implicated through animal models may lead to the identification of additional susceptibility genes through delineation of pathways by, for instance, interactome analysis. One such disorder is schizophrenia, which manifests overlap in both symptoms and brain circuits with OCD. In schizophrenia, in addition to several case-control association studies having been performed, linkage data, studies of chromosomal aberrations and animal models have led to the identification of many chromosomal regions that may contain genes involved in its aetiology and thus may also contain OCD candidate genes. In the present investigation, this approach was employed using previously reported schizophrenia susceptibility loci to identify novel OCD candidate genes. All genes residing in each of these loci were catalogued and individually analysed using a battery of bioinformatic techniques in order to assess their potential candidature for OCD susceptibility. These analyses yielded 13 credible OCD candidate genes.Additional candidates were sought using information regarding a well-defined schizophrenia animal model, the heterozygous reeler mouse, that exhibits neurodevelopmental, neuroanatomical and behavioural abnormalities, similar to those displayed by patients with schizophrenia. The phenotype of these mice is caused by a mutation in Reln, which encodes reelin, a large extracellular matrix protein that plays a pivotal role in the ordered migration of neurons during the development of laminar brain structures. The fact that both reelin protein and mRNA levels have been shown to be reduced in post-mortem brain sections of schizophrenic patients, coupled with the observed behaviour and neurochemical similarities between the heterozygous reeler mouse and schizophrenic patients suggests that reelin may be involved in the pathogenesis of schizophrenia and hence also OCD. Furthermore, genes encoding proteins that interact with reelin may thus also be considered plausible candidate genes for both schizophrenia and OCD. For this reason, novel reelin-interacting proteins were sought using the N-terminal reeler-domain of reelin, a domain only found in proteins involved in neuronal migration, as “bait” in a yeast twohybrid screen of a foetal brain cDNA library. Putative reelin ligands were subsequently reevaluated using co-immunopreciptitation and mammalian two-hybrid analysis to corroborate the yeast two-hybrid findings. Results of these analyses showed that WDR47, a WD40-repeat domain protein, interacts with reelin via its reeler-domain; therefore, the gene encoding this ligand protein, as well as RELN itself, was also considered a credible OCD candidate gene. Each of the candidate genes identified using the afore-mentioned strategies were assessed for their potential role in the aetiology of OCD by case-control association studies of a cohort of Afrikaner OCD patients and control individuals. Statistically significant associations were detected for two genes, DLX6 and SYN3, with the disorder. These associations are exciting as they may point to novel mechanisms involved in OCD development. The identification of WDR47 as a novel reelin-interacting protein has significant implications for our understanding of reelin-dependant signalling. Using this protein as the starting point, further novel components of the reelin signalling pathway may be unravelled, an investigation which may lead to the identification of novel roles for reelin in neurodevelopment. Such novel components may, of course, also be considered OCD and schizophrenia candidate genes, which may, in turn, augment the existing knowledge of the pathophysiologies of OCD, schizophrenia and other neurodevelopmental disorders. Taken together, the current study yielded exciting results that warrants follow-up investigation in future. The identification of DLX6 and SYN3 as novel OCD susceptibility genes as well as the identification of WDR47 as a reelin-interacting protein may provide investigators with alternative avenues of research into potential pathological mechanisms involved both in OCD and schizophrenia, which may ultimately lead to alternative pharmacotherapy. / AFRIKAANSE OPSOMMING: Obsessiewe kompulsiewe steuring (OKS) is `n ernstige, verswakkende psigiatriese steuring waarvan die onderliggende molekulêre etiologie steeds onbekend is. Bewyse verkry vanuit familiestudies het voorgestel dat OKS moontlik veroorsaak word deur `n komplekse interaksie van omgewings en genetiese faktore. Om die genetiese faktore te identifiseer wat OKS vatbaarheid veroorsaak, is `n hele aantal genetiese assosiasie studies onderneem, wat teenstrydige resultate gelewer het. Wat meer is, die grootste hoeveelheid van hierdie studies het gefokus op `n klein aantal kandidaatgene wat vir komponente van die serotonien en dopamine neurotransmittor weë enkodeer. Dit is egter, gebaseer op die kompleksiteit van die kliniese manifestasies wat waargeneem word in OKS, heel moontlik dat die patogenisiteit van die siekte bemiddel word deur `n breër kompleks van interverwante neurotransmittor sisteme en seintransduksie weë. Die feit dat daar aangetoon is dat beide reelin protein en bRNS vlakke verlaag is in post-mortem brein seksies van skisofrenie pasiënte, gekoppel aan die gedrags- en neurochemiese ooreenkomste wat gesien word tussen heterosigotiese “reeler” muise en skisofrenie pasiënte, stel voor dat reelin betrokke is by die patogenese van skisofrenie en dus ook OKS. Vir hierdie rede is nuwe proteïene gesoek wat `n interaksie met reelin toon, deur gebruik te maak van die N-terminale reeler-domein van reelin, `n domein wat slegs gevind word in proteïene wat betrokke is by neuronale migrasie, as “aas” in `n gis-twee-hibried sifting van `n fetale brein cDNS biblioteek. Vermeende reelin ligande is vervolgens herevalueer deur gebruik te maak van koimmunopresipitasie en soogdier twee-hibried analises om die gis-twee-hibried bevindings te bevestig. Resultate van hierdie analises het getoon dat daar interaksie is tussen WDR47, `n WD40-herhalingsdomein protein, met reelin via sy reeler-domein. Statisties-betekenisvolle assosiasies met die versteuring is vasgestel vir twee gene, DLX6 en SYN3. Hierdie assosiasies is opwindend aangesien hul nuwe meganismes betrokke by OKS ontwikkeling mag aantoon. Die identifikasie van WDR47 as ‘n nuwe protein wat interaksie met reelin vertoon, het betekenisvolle implikasies vir die verstaan van reelin-afhanklike seining. Enhancing the validity of the SAPS gold fingerprinting database through improved sample-collection techniques

This study's main focus was to analyse the methodology used by gold mines when obtaining reference samples required for gold fingerprinting analysis. Such methodology is also regulated by legislation in that in terms of the Precious Metals Act 37 of 2005 sect 22(1), "Any producer or any person who imports precious metals must submit to the Forensic Science Laboratory (FSL) of the South African Police Service (SAPS) such specimens of any precious metals produced or imported by him or her as may be prescribed". The gold mines are producers of precious metals, specifically gold, and they are therefore required to submit reference samples, which are then used by the SAPS FSL to maintain a database of gold samples. This database can then be used for comparison purposes during the forensic science of gold fingerprinting. The supporting regulations of the Precious Metals Act, Section 19 stipulate seven requirements for the submission of reference samples and this study intends to give guidelines to security officials employed on the gold mines of South Africa to ensure that these requirements are met and that the validity of the SAPS gold fingerprint database is enhanced through improved sample-collection techniques. The use of gold fingerprinting is part of the forensic investigation process used when trying to establish the origin of gold confiscated as an exhibit during police and mining security crime operations and recoveries, and this allows for victim identification in the resultant criminal trial and proper disposal of recovered gold-bearing materials that are recovered. It is important for the mine security official responsible for reference sample collection to keep in mind that the methodology used when obtaining these samples could be challenged in court and that, therefore, definitive guidelines should be followed to ensure that the most representative samples are collected and that the collection and handing over of samples meet chain of evidence requirements. / Criminology / M.Tech (Forensic Investigation)

The use of the Automated Fingerprint Identification System to improve the quality of service rendered by the South African Police Service on the East Rand

The globalisation process that drives the progression and trans-national nature of crime requires that the police should use sophisticated and/or state-of-the art technologies to help them to combat criminality. The use of technology by the police is thus viewed as one of the appropriate responses to deal with the threats posed by crime. In an attempt to ascertain the significance of technology in police work, this research examined the impact that the use of biometric technology such as the Automated Fingerprint Identification System, has had on the quality of seiVices rendered by the South African Police Service. Following the police's task of identifying criminal fingerprints, the problem encountered in this research was that, ever since the introduction of the Automated Fingerprint Identification System, no research had been conducted to assess the impact thereof on the level and quality of seiVices rendered by the South African Police SeiVice. This problem was further intensified by the fact that there was no framework to assess the impact that the utilisation of the Automated Fingerprint Identification System has had on police work in South Africa. Consequently, the problems that were identified in this research compromised not only the quality of seiVices rendered by the police, but also the safety and security within communities. The findings in this research showed that the utilisation of biometric technologies such as the Automated Fingerprint Identification System have helped the police to accurately identify and arrest criminal suspects. However, some of the conclusions reached suggested that the quantitative aspect of the utilisation of biometric systems and the elated portable apparatuses within the South African Police Service was emphasised more than the qualitative aspect thereof. The emphasis was evident in the statistics regarding the use of the Automated Fingerprint Identification System, which focused primarily on police productivity and to a lesser degree on accountability related issues. This study espoused a qualitative paradigm and the investigations in the research were focused on the East Rand policing precinct. The participants who informed this research were selected from the research population using random and purposive sampling techniques. In the same way, the data that informed this research was collected using semi-structured interviews and an in-depth review of the literature on policing and its utilisation of the different technologies. The research espoused social constructivism and phenomenology as the philosophical worldviews of choice to form constructivist phenomenology and to attain theoretical triangulation. Moreover, evaluation research and a case study were espoused as the research designs of choice, to attain methodological triangulation. The various triangulation techniques espoused in this research project were critical in attaining quality assurance in terms of the research methodology applied. In the same way, a myriad of qualitative techniques such as dependability, transferability, confimability and credibility were also employed in order to ensure that the integrity of the data that was collected in this study remained intact. Finally, the data that was collected to inform this research was analysed using the qualitative content analysis technique. The use of the Automated Fingerprint Identification System to improve the quality of service rendered by the South African Police Service on the East Rand

The globalisation process that drives the progression and trans-national nature of crime requires that the police should use sophisticated and/or state-of-the art technologies to help them to combat criminality. The use of technology by the police is thus viewed as one of the appropriate responses to deal with the threats posed by crime. In an attempt to ascertain the significance of technology in police work, this research examined the impact that the use of biometric technology such as the Automated Fingerprint Identification System, has had on the quality of seiVices rendered by the South African Police Service. Following the police's task of identifying criminal fingerprints, the problem encountered in this research was that, ever since the introduction of the Automated Fingerprint Identification System, no research had been conducted to assess the impact thereof on the level and quality of seiVices rendered by the South African Police SeiVice. This problem was further intensified by the fact that there was no framework to assess the impact that the utilisation of the Automated Fingerprint Identification System has had on police work in South Africa. Consequently, the problems that were identified in this research compromised not only the quality of seiVices rendered by the police, but also the safety and security within communities. The findings in this research showed that the utilisation of biometric technologies such as the Automated Fingerprint Identification System have helped the police to accurately identify and arrest criminal suspects. However, some of the conclusions reached suggested that the quantitative aspect of the utilisation of biometric systems and the elated portable apparatuses within the South African Police Service was emphasised more than the qualitative aspect thereof. The emphasis was evident in the statistics regarding the use of the Automated Fingerprint Identification System, which focused primarily on police productivity and to a lesser degree on accountability related issues. This study espoused a qualitative paradigm and the investigations in the research were focused on the East Rand policing precinct. The participants who informed this research were selected from the research population using random and purposive sampling techniques. In the same way, the data that informed this research was collected using semi-structured interviews and an in-depth review of the literature on policing and its utilisation of the different technologies. The research espoused social constructivism and phenomenology as the philosophical worldviews of choice to form constructivist phenomenology and to attain theoretical triangulation. Moreover, evaluation research and a case study were espoused as the research designs of choice, to attain methodological triangulation. The various triangulation techniques espoused in this research project were critical in attaining quality assurance in terms of the research methodology applied. In the same way, a myriad of qualitative techniques such as dependability, transferability, confimability and credibility were also employed in order to ensure that the integrity of the data that was collected in this study remained intact. Finally, the data that was collected to inform this research was analysed using the qualitative content analysis technique. The research is concerned with how the post-mortem report can be used as a source of information in the investigation of murder cases to identify crime, individual crimes and trace suspects, etc. The understanding of the post-mortem report, and the information which can be obtained from it, will assist the police investigators to handle murder cases in a professional way. The purpose of this research was to evaluate the existing manner in which police investigators use post-mortem reports in their investigations, with the intention of determining its strengths and weaknesses, and considering how the usage can be improved. Secondly, the researcher wanted to explore how investigators use postmortem reports as a source of information in their investigations. To accomplish this, the researcher perused international and national literature in an attempt to explore the field. The researcher wanted to apply the new research knowledge in order to develop good practice in the field.
