Development and evaluation of a non-invasive core temperature measurement system

Core body temperature is an important physiological parameter used to identify whether a patient displays a normal, hypothermic, or hyperthermic state. It is routinely monitored during cardiac surgeries and general anesthesia. Currently, the most effective methods for measuring core body temperature are also the most invasive. While select devices have been designed to enable surface recording of internal temperature, none have been implemented in U.S.-based hospitals. The objective of this study was to create a noninvasive core temperature sensor and evaluate its potential of becoming a widely used clinical tool. In tissue phantom and human-based experiments, the prototype performed effectively and posed no safety risk. Provided the prototype can be successfully translated into a more streamlined medical device, it stands to become a staple in operating rooms around the nation. / text

Ecology of the declining Olearia lineata and not-threatened Olearia bullata in human-modified environments and implications for their conservation

Globally, human modification of land and the human-aided introduction of exotic species are considered to be the main drivers behind species decline and extinction. Human colonisation of New Zealand and subsequent development resulted in the conversion of native forests and shrublands into productive land, which together with the introduction of invasive species has caused the decline of many species, and many more are now threatened with extinction. This issue is particularly prevalent in the lowland eastern areas of the South Island, in which there has been intensive land development. One strongly affected group are species within the genus Olearia, in which several species are threatened with extinction, including nationally declining Olearia lineata. In contrast, other species in this genus, such as non-threatened O. bullata, are seemingly not as strongly impacted. My study investigated the mechanisms behind why O. lineata is nationally declining in comparison to the non-threatened congener O. bullata. To do this, I investigated two main research questions; one question investigated the demographic structure of populations to determine if regeneration is occurring and the vegetation composition of the surrounding community. Sampled populations indicated regeneration failure in O. lineata populations but also in O. bullata populations, despite its non-threatened status. I found that a high proportion communities contained exotic grasses, therefore, the second question was how the presence of the exotic grass- Agrostis capillaris, affected recruitment of O. lineata and O. bullata. Two glasshouse experiments investigated how the presence or absence of grass affected germination and growth of seedlings. Both Olearia species were found to germinate in the presence of grass, but seedlings grew significantly better in the absence of grass. Olearia are also important native Lepidoptera hosts as a suite of native moths feed exclusively on this genus. Therefore, a third research question investigated the abundance of larvae and the community composition of Lepidoptera on O. lineata and O. bullata in comparison to another Lepidoptera host Coprosma propinqua. Moth larvae were collected and some species successfully reared to find there was higher larvae abundance on O. lineata and high species overlap between O. lineata and O. bullata with little species overlap between the two Olearia species and C. propinqua. Overall, my research found that exotic invasive grasses potentially cause regeneration failure in both O. bullata and O. lineata. However, the largest impact in the difference of threat status is due to O. lineata populations occurring in the most developed lowland areas of the eastern South Island, whereas O. bullata populations occur in less human modified, higher altitude areas. The further development of these areas through human activity threatens the future persistence of these Olearia species and the future of the Olearia Lepidoptera specialists.

Competition for Conifer Cones as a Potential Mechanism of Endangerment for the Mount Graham Red Squirrel

Non-native species are a major cause of endangerment for native species, but the mechanisms are often unclear. As species invasions continue to rise, it is important to understand how to mitigate this threat. Our field experiment quantified the impact of introduced Abert's squirrels (Sciurus aberti) on rates of food removal within the range of the critically endangered Mount Graham red squirrel (Tamiasciurus hudsonicus grahamensis). In the presence of Abert's squirrels, the time until 50% of cones were removed was 5.55 days faster than when Abert's squirrels were excluded (95% confidence interval 2.25 to 11.63 days). The impact on food availability as a result of cone removal by Abert's squirrels suggests the potential of food competition as a mechanism of endangerment for the Mount Graham red squirrel. Eradication of established non-native populations is rare. Management targeted at specific mechanisms by which non-natives cause endangerment is an essential tool for conservation.

Multidimensional Spatial Characterization of Plant Invasions in 'El Pinacate y Gran Desierto de Altar' Biosphere Reserve

Invasive species are considered an agent of ecological change with more significant effects than global warming. Exotic plant invasions threaten biodiversity and ecosystem viability worldwide. Their effects in the Sonoran Desert ecosystems are a growing concern among ecologists and land managers. We hypothesized that highly dynamic desert environments are unstable, therefore more vulnerable to invasion by exotic plant species. To test this hypothesis we used a multidimensional approach to assess the spatial distribution of two exotic species: Brassica tournefortii (Saharan mustard) and Schismus arabicus (Arabian grass), in a portion of 'El Pinacate y Gran Desierto de Altar' Biosphere Reserve (PBR) in northwestern Sonora, Mexico. This approach combined genetic algorithms, geographic information systems, field methods, statistical analysis, and remote sensing modeling at multiple spatial and temporal scales to predict and test the current and potential distribution of the invasives over dynamic landscapes.Predicted probability of invasion was influenced strongly by human factors: Road networks were the strongest predictors of presence, revealing the potential importance of humans as vectors of invasiveness. Dynamic landscapes, associated mostly with vegetation losses, were detected spectrally in the eastern portion of the study area, very likely associated with past agricultural and current grazing activity. Combined models of high probability for invasion by B. tournefortii and S. arabicus over dynamic landscapes were tested against confirmed locations of the invasives and land cover types associated with invasion. Results confirmed the hypothesis of the study and suggest that more dynamic landscapes are more prone to invasion by these two exotic plants in the PBR. B. tournefortii was found associated mostly with landscapes occupied by microphyllous desert scrub and grassland, as well as sarcocaulescent desert scrub. S. arabicus was found more abundantly in the flat low lands occupied by microphyllous and crassicaulescent desert scrub. These relationships cannot, however, be conclusive and require further investigation due to the complex ecology of these invasives.

Validation in-vivo des techniques d’élastographie ultrasonore, invasive et non-invasive, à l’aide d’un modèle porcin

Il est maintenant admis que la composition de la plaque athérosclérotique est un déterminant majeur de sa vulnérabilité à se rompre. Vu que la composition de la plaque affecte ses propriétés mécaniques, l'évaluation locale des propriétés mécaniques de la plaque d'athérome peut nous informer sur sa vulnérabilité. L'objectif est de comparer les techniques d’élastographie ultrasonores endovasculaire (EVE) et non-invasive (NIVE) en fonction de leur potentiel à identifier les composantes calcifiées et lipidiques de la plaque. Les acquisitions intravasculaire et extravasculaire ont été effectuées sur les artères carotidiennes de neuf porcs hypercholestérolémiques à l’aide d’un cathéter de 20 MHz et d'une sonde linéaire de 7.5 MHz, respectivement. Les valeurs de déformation radiale et axiale, rapportés par EVE et NIVE, ont été corrélées avec le pourcentage des zones histologiques calcifiées et lipidiques pour cinq plaques. Nos résultats démontrent une bonne corrélation positive entre les déformations et les composantes calcifiées (r2 = 0.82, P = 0.034 valeur par EVE et r2 = 0.80, P = 0.041 valeur par NIVE). Une forte corrélation entre les déformations axiales et les contenus lipidiques par NIVE (r2 = 0.92, P-value = 0.010) a été obtenue. En conclusion, NIVE et EVE sont des techniques potentielles pour identifier les composants de la plaque et aider les médecins à diagnostiquer précocement les plaques vulnérables. / It is now widely accepted that plaque composition is a major determinant of plaque’s vulnerability to rupture. Since composition of the plaque affects its mechanical properties, the local assessment of mechanical properties of atherosclerotic plaque may inform us about plaque’s vulnerability. The objective is to compare ultrasonic endovascular elastography (EVE) versus non-invasive vascular elastography (NIVE) according to their potential to identify plaque contents. Intravascular and extravascular acquisitions were performed on carotid arteries of nine hypercholesterolemic minipigs with a 20 MHz catheter and a 7.5 MHz standard probe, respectively. Radial and axial strain values, reported by EVE and NIVE respectively, were correlated with histological area of lipid and calcium for five plaques. Our results demonstrate a good positive correlation between strains and calcified contents (r2=0.82, P-value=0.034 by EVE and r2=0.80, P-value= 0.041 by NIVE). Additionally, there is a strong correlation between axial strains and lipid contents by NIVE (r2=0.92, P-value= 0.010). In conclusion, NIVE and EVE are the potential techniques to identify plaque components and to help physicians to early diagnose the vulnerable plaques.

Detrital Production in Kelp Beds

Detrital subsidy from highly productive kelp beds and forests to adjacent habitats represents a major form of connectivity between coastal ecosystems that regulates regional patterns of community organization and production. In this thesis, I investigated environmental and biological factors that influence the rate of detrital production in Nova Scotian kelp beds, with emphasis on the role of invasive species in altering these dynamics. The rate of blade erosion of the dominant kelp species (Saccharina latissima and Laminaria digitata) increased significantly with the level of encrustation by the invasive bryozoan Membranipora membranacea and the extent of grazing damage by the native snail Lacuna vincta, and by increased water temperature and site exposure. The rate of detrital production (as dry mass) ranged from 0.5 to 1.71 kg m-2 y-1 across 5 sites, and increased linearly with kelp bed biomass. Spatial variation in the total level of grazing damage on kelp blades by L. vincta was explained in part by a negative relationship with site exposure, and the distribution of grazing along blades was regulated by kelp growth rate and the associated production of grazing-deterrent polyphenolics. Grazing damage by L. vincta that exceeded 0.5 to 1.0% of blade area caused increased rates of erosion during heavy wave action associated with a passing hurricane. The maximum stress before breakage, toughness, and extensibility of blade tissues decreased with the degree of encrustation by M. membranacea or grazing damage by L. vincta, which cause degradation and removal of the outer cell layers of kelp tissues, resulting in stress concentration and breakage at lower force applications than required to break undamaged tissues. The invasive green alga Codium fragile and S. latissima differed with respect to nutritional quality and changes in biochemical composition that occurred over the course of degradation on a sandy bottom adjacent to a kelp bed. Macrofaunal communities colonizing detrital deposits responded to these differences. These findings demonstrate that invasive species can alter the quantity and quality of detritus produced from subtidal kelp beds, and that their community-level effects can extend well beyond the invaded habitats via the export of detritus.

An effect of the invasive Round Goby (Neogobius melanostomus) on the recruitment of unionid mussel Species at Risk (Bivalvia: Unionidae)

I investigated whether Neogobius melanostomus, an invader of biodiversity “hot-spots” in Ontario facilitates or inhibits unionid mussel recruitment by serving as a host or as a sink for their parasitic larvae (glochidia). Infestation and metamorphosis rates of four mussel Species at Risk (Epioblasma torulosa rangiana, Epioblasma triquetra, Lampsilis fasciola, and Villosa iris) and one common species (Actinonaias ligamentina) on N. melanostomus were compared to rates on known hosts in the laboratory. All species successfully infested N. melanostomus, but only E. triquetra, V. iris, and A. ligamentina successfully metamorphosed, albeit at low rates. Neogobius melanostomus collected from areas of unionid occurrence in the Grand and Sydenham rivers exhibited body burdens of 39.4% and 5.1%, respectively. Analyses indicate that N. melanostomus serves more as a sink for glochidia than as a host for unionids, thereby limiting recruitment, which is a novel way by which N. melanostomus is affecting native mussel species.

Invasive plant influence on the native grass community of the White Lake Basin, British Columbia

Invasive plants can have significant ecological effects. Cheatgrass (Bromus tectorum) is of particular concern in North America, where its competitive nature can seriously degrade natural grasslands. This study, conducted in the White Lake Basin region of British Columbia, investigated the impact of cheatgrass on native plant diversity and the relationship between diffuse knapweed (Centaurea diffusa) and introduced grass cover. The study also analysed the association of cheatgrass with other grass species to provide insight in selecting grass seed composition for seeding after habitat disturbance. Results indicated higher native grass diversity in plots without cheatgrass and in plots containing bluebunch wheatgrass (Pseudoregneria spicata). While diffuse knapweed decreased during the study period, due to biological control, invasive grasses increased. Native grass species positively associated with cheatgrass include needle and thread grass (Hesperostipa comata), Sandberg bluegrass (Poa secunda), and sixweeks fescue (Vulpia octoflora), indicating potential for seeding in disturbed areas prone to cheatgrass.

Life-History Traits Of Chrysomya rufifacies (Macquart) (Diptera: Calliphoridae) And Its Associated Non-Consumptive Effects On Cochliomyia macellaria (Fabricius) (Diptera: Calliphoridae) Behavior And Development

Blow fly (Diptera: Calliphoridae) interactions in decomposition ecology are well studied; however, the non-consumptive effects (NCE) of predators on the behavior and development of prey species have yet to be examined. The effects of these interactions and the resulting cascades in the ecosystem dynamics are important for species conservation and community structures. The resulting effects can impact the time of colonization (TOC) of remains for use in minimum post-mortem interval (mPMI) estimations. The development of the predacious blow fly, Chrysomya rufifacies (Macquart) was examined and determined to be sensitive to muscle type reared on, and not temperatures exposed to. Development time is important in forensic investigations utilizing entomological evidence to help establish a mPMI. Validation of the laboratory-based development data was done through blind TOC calculations and comparisons with known TOC times to assess errors. A range of errors was observed, depending on the stage of development of the collected flies, for all methods tested with no one method providing the most accurate estimation. The NCE of the predator blow fly on prey blow fly, Cochliomyia macellaria (Fabricius) behavior and development were observed in the laboratory. Gravid female adult attraction was significantly greater to resources with predatory larvae rather than prey larvae and oviposition occurred on in the presence of heterospecific (predatory) and conspecific larvae equally. However, the life stages necessary for predation to occur never overlapped and so these results may not be as surprising as they seem. Conversely, exposing prey larvae to predator cues through larval excretions/secretions led to larger prey larvae and faster times to pupariation when appropriate life stages overlapped. Differences in size and development times of prey larvae in the presence of predatory cues could lead to errors when estimating the mPMI. These data also partially explain the ability of C. macellaria to survive in the presence of Ch. rufifacies. Colonization of a resource with late instar Ch. rufifacies enhanced development and size of resulting larvae indicating that lag colonization, rather than being a primary colonizer, could become an alternate strategy for C. macellaria to survive the selective pressures of the predator, Ch. rufifacies. The differing effects of temperature on Ch. rufifacies and C. macellaria may also lend an advantage to C. macellaria over the predacious Ch. rufifacies in an environment with variable temperatures unlike what Ch. rufifacies is adapted for.

An Augmented Virtuality Navigation System for Arthroscopic Knee Surgery

Arthroscopic knee surgery can be challenging because there is no intuitive relationship between the arthroscopic image, shown on a screen above the patient, and the camera in the surgeon's hand. As a result, arthroscopic surgeons require extensive training and experience. This thesis describes a computer system to help improve target acquisition in arthroscopy by visualizing the location and alignment of an arthroscope using augmented virtuality. A 3D computer model of the patient's joint (from CT) is shown, along with a model of the tracked arthroscopic probe and the projection of the camera image onto the virtual joint. We performed a user study to determine the effectiveness of this navigated display; the study showed that for novice residents, the navigated display improved target acquisition. However, residents with at least two years of experience performed worse.
