Establishing a process for a wetland vegetation rehabilitation and management program focused on reed canarygrass: A Parkland Mews case study

Hence the research objectives of this project were to determine if reed canarygrass abundance has an effect on plant species diversity and assess the effectiveness of novel treatments on reed canarygrass control in a constructed wetland. Four treatments (mowing, herbicide, mowing plus herbicide, and a control) followed by broadcast seeding were applied to regulate growth of reed canarygrass. Principal components analysis, biodiversity measures, and ANOVA were used to identify community composition, quantify biodiversity values and identify treatment differences respectively. Results indicated differences in species composition between east and west blocks of the study site, reed canarygrass abundance appears to keep plant species diversity low, indigenous species were rare, and reed canarygrass was resistant to treatments.The results of this study are not surprising considering there is little evidence that treatments for reed canarygrass control are effective.

The invasion ecology of Didymosphenia geminata

Didymosphenia geminata (Didymo) is a nuisance, bloom forming, invasive freshwater algae. Through an ecological survey I examined habitat associations and effects, identifying bloom preferences for stable, low nutrient rivers, often downstream of still waterbodies; with major observed changes to algal and invertebrate communities. Experimental examination of the functional significance of blooms, provided strong support for the hypothesis that blooms are an adaptive, plastic ecophysiological response to nutrient limitation, maximising internal mat cycling and phosphatase based uptake. Further experimentation identified Didymo absence from higher nutrient waterways, is explained by competition with other algae and suppression of the 'bloom' response. Research suggests Didymo is here to stay, but spread management and mitigation will minimise damage to New Zealand's waterways.

Biological Control of Paropsis charybdis Stål (Coleoptera: Chrysomelidae) and the Paropsine Threat to Eucalyptus in New Zealand

Ineffective biological control of the Eucalyptus pest Paropsis charybdis Stål (Coleoptera: Chrysomelidae: Paropsini) in cold areas of New Zealand was believed to be caused a climatic mismatch of the egg parasitoid Enoggera nassaui Girault (Hymenoptera: Pteromalidae). Two Tasmanian strains of the parasitoid were introduced to test climate-matching theory in 2000, with approximately 7000 wasps released. Establishment of the Florentine Valley strain was detected in 2002 using the Mitochondrial (mtDNA) gene Cytochrome Oxidase I (COI) as a strain specific marker. The hyperparasitoid Baeoanusia albifunicle Girault (Hymenoptera: Encyrtidae) and primary parasitoid Neopolycystus insectifurax Girault (Hymenoptera: Pteromalidae) were detected for the first time in New Zealand.
As paropsines have proven highly invasive internationally, a risk assessment of the paropsine threat to New Zealand was undertaken by evaluating the host range of E. nassaui and a reproductive assessment of 23 paropsine species in the genera Dicranosterna Motschulsky, Chrysophtharta Weise, Paropsis Olivier, Paropsisterna Motschulsky and Trachymela Weise. Enoggera nassaui proved polyphagous, but bioassay results proved that Paropsis species were significantly more susceptible to the egg parasitoid than Chrysophtharta species. Resistance within Chrysophtharta was attributed to spine-like chorion modifications. A COI derived Chrysophtharta phylogeny divided the genus into two distinct groupings, which was supported by chorion morphology.
Paropsine reproductive output was tested for key parameters indicating pest potential. Pest species displayed fecundity exceeding 600 eggs at an oviposition rate above 10 eggs per day⁻¹. Several non-pest species were identified as potential pests based on these parameters. The Chrysophtharta phylogeny suggested a moderate relationship between genetic relatedness and reproductive output. The Acacia defoliating paropsine Dicranosterna semipunctata (Chapuis) was evaluated for its susceptibility to E. nassaui and reproductive output. Egg parasitism occurring in bioassay did not translate into biological suppression following a specifically targeted release of E. nassaui, and the fecundity and oviposition rates fell below the thresholds predicted for a pest paropsine species.
Despite establishment of Tasmanian E. nassaui, hyperparasitism has now rendered this control agent ineffective in New Zealand. Neopolycystus insectifurax offers the best hope for future biological control of paropsine species in New Zealand.

AN ASSESSMENT OF THE INVASIVE POISON HEMLOCK AND ITS INSECT ASSOCIATES IN KENTUCKY

Poison hemlock, Conium maculatum (Apiaceae), is an invasive plant in North America with a unique toxic chemistry. Previous research on this plant has focused on identifying herbivores as potential biological control agents or describing the toxic plant alkaloids. However, none have examined the role of higher trophic levels in the food web surrounding poison hemlock. Generalist predators and food web interactions are an important component of studies investigating invasion effects, as plant or animal introductions can alter ecosystem functioning. In this study, predators in poison hemlock were sampled at the foliar and epigeal levels, resulting in 956 Carabidae and 321 Coccinellidae being collected. Predator connectedness to plant resources was quantified using molecular gut-content and chemical analyses. Foliar Harmonia axyridis (Coccinellidae) contained aphid DNA and plant chemicals, while Harpalus pensylvanicus (Carabidae) only contained alkaloids, suggesting that the ground predators were obtaining plant chemicals via alternative prey. Feeding trials between H. axyridis and their potentially toxic prey, Hyadaphis foeniculi (Aphididae), revealed that the exotic predator shows faster development when consuming aphids from poison hemlock compared to alternative diets. This study reveals that three Eurasian species may be facilitating one another, illustrating the importance of continued examination of invasive species interactions.

Semi-automated immunohistochemical staining of the VEGF-A-protein for clinical use and the identification in NHG-graded breast carcinoma

Angiogenesis has a crucial influence on tumour development and identification of microvessels in malignant breast cancer tissue is an indicator for worse prognosis. Angiogenesis is partially governed by the family of vascular endothelial growth factors (VEGF) and their receptors, by which the VEGF-A-protein seems to be the most important factor.     The aims of this work were to first establish a method for immunohistological (IH)-staining of the VEGF-A-protein for clinical use and then to label and evaluate the expression of this protein in 31 Nottingham Histology Graded (NHG I-III) breast carcinoma.      Formaldehyde-fixated tissues from invasive breast neoplasms and control tissues were labelled with monoclonal antibodies against VEGF-A and CD31-proteins using a semi-automated IH-system from Ventana BenchMark. Positively stained vessels were counted from digital copies of microscopic pictures related to mm2 tissue.     A method of IH-labelling with VEGF-A protein was successfully established before staining of the breast tissue and in 19 of the 31 breast cancers. Vessels were counted for both antibodies. The VEGF-A-antibody stained 2.7 ± 2.3 (mean ± SD) vessels/mm2 and the CD31-antibody stained 27.3 ± 19.3 in the breast carcinoma tissue. The percent of VEGF-A-stained vessels in relation to CD31-stained were 7.6% in the NHG-I- (n=3), 7.8% in the NHG-II- (n=10) and 15.0% in the NHG-III-group (n=6).     The results demonstrate that increased NHG-grade and lower differentiation can be associated with higher percent of vessels expressing VEGF-A-protein. The result was not statistically certified because of the small number of stained breast cancers and additional investigations are recommended before clinical use.

Adolescent Perceptions and Attitudes towards Invasive Species and Nature

Invasive species are one of many important environmental issues facing Canadians today. A great deal of research has explored both the scientific and social aspects of invasive species. However, the cumulative research has not yet thoroughly explored people’s thoughts and feelings about, or perceptions of, invasive species and the influence these may have on management of, or policy decisions regarding, invasive species. This thesis research project was designed to assess the attitudes and perceptions that high school students have towards invasive species and to determine to what extent learning about invasive species alters their connection with nature. The study group was comprised of students from four Grade 11 Environmental Science classes from three high schools within the city of Guelph, Ontario. The students received regular classroom instruction from their teachers covering the course content, including invasive species. Students also made weekly visits to a local nature centre, providing them with hands-on learning experiences related to the course content. Students responded to an 80-question survey that assessed their knowledge of local invasive species and attitudes towards them as well as students’ connection to nature. Their connection to nature was assessed using a modified version of the Connectedness to Nature Scale (Mayer & Frantz, 2004). The surveys were administered by the classroom teachers in October, 2010, prior to the presentation of instructional material covering invasive species and then again in December, 2010 when the presentation of instructional material concerning invasive species was complete. The results showed that the students’ knowledge of invasive species upon entering the course was quite low and each of the classes witnessed a significant increase in knowledge. The survey results did not reveal any change to the students’ connection to nature; however, they did reveal three underlying themes in students’ attitudes towards invasive species: a concern about invasive species as a threat or problem; feelings of acceptance towards invasive species; and feelings of anxiety about invasive species. These attitudes remained relatively unchanged after the course.

Investigating the efficacy of voluntary initiatives for reducing horticultural introductions of invasive species

The horticultural industry is responsible for approximately half of the invasive plant introductions in North America. To reduce these introductions, voluntary initiatives are preferred over government regulations. This thesis aims to evaluate the effectiveness of two types of voluntary initiatives. At the gardener level, I investigated the effectiveness of alternative species promotion campaigns, called “Grow Me Instead” programs. Adult gardeners visiting the Royal Botanical Gardens in Hamilton, Ontario, as well as customers at two garden centres, participated in a conjoint analysis which measured their preferences for various traits of potential ground cover species. Results showed that gardeners generally prefer plant species having invasive characteristics, suggesting these programs may not be as effective as initially believed. At the retailer level, this study aimed to build upon the work done by Burt and colleagues (2007) to obtain further understanding of the relative strength of internal (ethical motivations) and external (legislation, stakeholder pressures and economic opportunities) factors for motivating participation in voluntary initiatives. Telephone interviews were conducted with 30 industry professionals from southern Ontario to assess their adoption of the St. Louis Voluntary Codes of Conduct. Results found that participation rates of industry professionals in southern Ontario were lower for every specific initiative than those interviewed by Burt et al. (2007). Industry professionals presently experience the most pressure to participate from a sense of personal responsibility and the desire to create a green business image. Pressure was significantly higher from these sources than from pressure from employees. Together these two studies identified several barriers to the efficacy of voluntary initiatives as well as some reasons for optimism. To ensure the success of future voluntary initiatives, efforts must be made to encourage these two groups to work together. Understanding how both retailers and gardeners respond to voluntary initiatives will assist in the development of more effective programs and lead to fewer horticultural invasive species introductions in the future.

The distribution of Dreissena and other benthic invertebrates in Lake Erie, 2002.

A lake-wide benthic survey of Lake Erie during summer 2002 indicated that Dreissena bugensis is the dominant dreissenid in Lake Erie, especially in the east basin where this species was found at every station but no Dreissena polymorpha were collected. Mean (±SD) densities of dreissenid mussels were comparable between the west (601±2,110/m2,n=49) and central (635±1,293/m2; n=41) basins, but were much greater in the east basin (9,480±11,173/m2;n=17). The greater variability in mussel density among stations and replicate samples in the central and west basins than in the east basin is attributable to the preponderance of fine-grained substrata in the nearshore, higher episodic rates of sediment deposition and periodic hypoxia in bottom waters. Although there was little change in lake-wide mean dreissenid densities between 1992 and 2002 (declining from ca. 2,636 individuals/m2 to 2,025 individuals/m2), basin-averaged shell-free dry tissue mass increased by almost four-fold from ca. 6.8±15.6 g /m2 to 24.7±71.3 g/m2 in the same interval. Up to 90% of this biomass is in the eastern basin. Other changes in 2002 include the virtual absence of mussels in the 3 to 12 mm size range, probably because of predation by round gobies, and an increase in the average size of mature mussels. The substantial changes observed between 1992 and 2002 suggest that dreissenid populations in Lake Erie were still changing rapidly in abundance and biomass, as well as species composition. The results of this survey suggest that a direct link between Dreissena spp. and hypolimnetic hypoxia in the central basin is unlikely. The dominant organisms of Lake Erie in 2002 were D. rostriformis bugensis (38%), Oligochaeta (33%), Chironomidae (18%), Sphaeriidae (2.7%), Amphipoda (2.3%) and Hydrozoa (2.2%). Mean invertebrate density was greater in the east basin, especially on hard substrates, than either the west or central basin. In the central basin, sites ≥5m supported greater numbers of organisms, than shallow (≤2m) sites in the nearshore wave zone. The greatest number of taxa were observed in the central basin, likely a resutlt of greater sampling effort there. Gammarus fasciatus comprised 80% of all amphipods, being most abundant on Dreissena-dominated hard substrates in the east basin. The introduced species, Echinogammarus ischnus occured at only 11 of 69 sites, and was the only amphipod found at 4 east basin sites, but at relatively low densities. Diporeia were not found in our survey. Hexagenia was collected at only 4 stations, all in the west basin. Chironomids were dominated by Tanytarsus and Chironomus, with Procladius, Dicrotendipes and Polypedilum also being relatively common. Oligochaete Trophic Index for 2002 indicates a similarly mesotrophic condition throughout the lake and marginal nutrient enrichment of sediments between years 1979-2002. Multivariate ordination of community data indicates clear separation of sites by year and basin as expected given the extirpation of Diporeia and the introduction of Dreissena and E. ischnus, but also reveals subtle changes in benthic structure over the last 2 decades. The benthic community of Lake Erie in 2002 does not likely represent an equilibrium condition.

An Inventory and Recommendations for the Management of High Priority Invasive Alien Plants in Point Pelee National Park

Point Pelee National Park in Ontario, Canada has been affected by a long history of human activity. This activity has encouraged the establishment of approximately 276 exotic invasive plant species. These plants decrease biodiversity and effective function of ecosystems within the Park. A 5 Year Exotic Plant Species Management Plan for the Park was written in 1990. Since that time inventories have been done on some of the species but a monitoring of all the high priority exotic invasive plants has not. As part of this study an inventory of the high priority exotic invasive plants and their spatial extent in the Park, was created. From May to September, 2011 a comprehensive inventory took place within a 5.5km stretch in the terrestrial area at the southern end of the Park. Systematic belt transects were performed, on foot, from west to east at 100 m intervals. Within the quadrats percent cover of each plant species was determined. The data collected in the field were compiled using geographic information systems (GIS), resulting in maps of the extent of the most abundant species studied. Analysis as part of this study included using the data to determine which plants and areas are higher priorities for management within the Park. Quadrats were analysed for diversity using the Simpson Index and, since the data was non-parametric, comparisons were made across diversity and native richness using the Kruskal Wallis test. Alliaria officinalis is the only non-native species that is widespread within the study area. Other non-native species with a high potential for invasiveness were observed but only consisted of a few individuals along roads and paths. Some non-native species were present along or near roads and paths in denser patches (17-100% plot coverage). Recommendations include removal of some non-native species deemed to be a potential threat to native richness and diversity, followed by re-vegetation with native species, and continued monitoring. Future restoration efforts are best directed at the area around DeLaurier, along west beach and at the Tip. These areas have the lowest diversity and native richness and therefore need the most improvement.
