Pentagonal scheme for dynamic XML prefix labelling

In XML databases, the indexing process is based on a labelling or numbering scheme and generally used to label an XML document to perform an XML query using the path node information. Moreover, a labelling scheme helps to capture the structural relationships during the processing of queries without the need to access the physical document. Two of the main problems for labelling XML schemes are duplicated labels and the cost efficiency of labelling time and size. This research presents a novel dynamic XML labelling scheme, called the Pentagonal labelling scheme, in which data are represented as ordered XML nodes with relationships between them. The update of these nodes from large scale XML documents has been widely investigated and represents a challenging research problem as it means relabelling a whole tree. Our algorithms provide an efficient dynamic XML labelling scheme that supports data updates without duplicating labels or relabelling old nodes. Our work evaluates the labelling process in terms of size and time, and evaluates the labelling scheme’s ability to handle several insertions in XML documents. The findings indicate that the Pentagonal scheme shows a better initial labelling time performance than the compared schemes, particularly when using large XML datasets. Moreover, it efficiently supports random skewed updates, has fast calculations and uncomplicated implementations so efficiently handles updates. Also, it proved its capability in terms of the query performance and in determining the relationships. / Libyan government

Developing FlARe: A Chemi-Genetic Maleimide-Based Approach to Fluorogenic Protein Labelling

Studying protein expression, localization, and behaviour within cells is invaluable to understanding biological processes. Fluorescent protein labelling can be used to easily visualize these biomolecules. However, many strategies use large fluorescent proteins or enzyme tags that disrupt the native localization and conformation of proteins of interest. FlARe is a minimalistic, chemi-genetic fluorogenic protein labelling method with two components, the FlARe probe and the dC10 tag. The probe consists of a fluorophore linked to two maleimides, which quench the fluorophore’s emission. They also react with the complementary dC10 peptide tag that is genetically fused to a specific protein and contains two cysteines positioned 10 Å apart. Reaction with dC10 anchors the FlARe probe to the tagged protein and abolishes the maleimides’ quenching mechanisms. This releases the probe’s intrinsic fluorescence, allowing the protein to be visualized by fluorescence. This thesis sets out to complete three objectives: 1) To improve the FlARe probe chemical structure; 2) To implement novel developments of the FlARe protein labelling technique into live mammalian cells; 3) To expand the scope of the FlARe method to include bioconjugation. To make the current FlARe probe design more stable in the aqueous milieu of the cell, a new FlARe probe, KT12, was synthesized. This new probe was more stable than the current YC20 FlARe probe but not more selective. Despite this, valuable insight was made into the mechanisms of maleimide hydrolysis and thiol addition. After establishing that the current methoxymaleimide FlARe probe offered the most selective design, we invested our efforts into implementing the FlARe labelling technique in live mammalian cells. To this end, a recombinant protein carrying different variants of the dC10 tag was successfully labelled with FlARe probes emitting at different wavelengths. Finally, we blended our expertise in probe design and biological implementation to develop a novel fluorogenic linker to form bioconjugates. As a result, a FlARe linker functionalized with both the dimaleimide moiety and an alkyne linker was designed. The successful synthesis and proof-of-principle demonstration of this novel FlARe linker were performed, allowing for the modular formation of fluorogenic bioconjugates. These developments to FlARe advance this minimalistic and fluorogenic protein imaging method. This improves the current capability to fluorescently label proteins in cells in a non-disruptive manner. Thus, complex questions surrounding cell biology can be answered to better understand biological processes.

Hållbarhet är en stark faktor som påverkar både konsumenter och företag i växande utsträckning. Att medvetet konsumera i enlighet med hållbara riktlinjer ligger i många konsumenters intresse, och företag utvecklar kontinuerligt strategier för att bemöta konsumentbehov och höja sin försäljning. I takt med detta ökar utbudet av hållbart märkta produkter och hållbarhetsmärkningar. För konsumenter innebär detta större valmöjligheter men också eventuella svårigheter då hållbarhetsmärkningar ofta upplevs snarlika och svåra att förstå betydelsen bakom, vilket i sin tur kan ha en motverkande effekt på konsumenters köpbeslut. Syftet med denna studie var därför att undersöka hur konsumenter i dagligvaruhandeln ställer sig till svenskt etablerade hållbarhetsmärkningar på likvärdiga produkter, för att utforska hur hållbarhetsmärkningar påverkar konsumenters köpbeslut vid köptillfället. Studien eftersträvade att identifiera de starkaste faktorerna som ligger bakom konsumenters beslut att köpa eller inte köpa hållbart märkta produkter. Avsikten var att ge företag/varumärken en bild av vad konsumenterna behöver för att effektivt vägledas i sitt köpbeslut. Insamling av data genomfördes genom enkäter med kvalitativ karaktär då studien avsåg att belysa konsumentåsikter kopplade till konsumentbeteende. Urvalet som inkluderades i studien utgjordes av individer som köper produkter i dagligvaruhandeln. Analys av data skedde med utgångspunkt i grundad teori där nyckelord och mönster mellan respondenter och svar identifierades och tolkades. Resultatet av studien visade att konsumenter främst påverkas i köpriktning vid köp av hållbart märkta produkter till följd av miljömässiga omvärldseffekter och personliga effekter på den egna hälsan, men även till viss del till följd av omvärldseffekter kopplade till socialt ansvarstagande samt tillgänglighet av information. Bristande tilltro som skapar associationer till opålitlighet samt bristande förståelse till följd av dålig kunskap är de enda faktorerna som har påverkan på köpbeslut i icke-köpriktningen enligt resultatet av studien. Genom att belysa de faktorer som påverkar konsumenter i köpriktning och motverka de faktorer som påverkar konsumenter i icke-köpriktning, kan företag/varumärken uppnå företagsnytta.
