Previous issue date: 2015-07-31 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / The root lesion nematode (Pratylenchus brachyurus) is one of the main soybean parasite in Brazil.
Until now, there is no clean and low cost alternative to successfully reduce its population in
contaminated fields. However, recent studies has shown the possibility of inducing the defense
mechanisms in plants as a strategy for the management. The objective of this study was to evaluate
the biochemical response and the development of soybean cultivars when subjected to parasitism
by the nematode Pratylenchus brachyurus and treat with abiotic resistance inducer acibenzolar-S-
methyl (ASM). Soybean was sown in polyethylene bags, and seven days after, the treatments were
inoculated with the nematode. The experiment was conducted in a completely randomized design
in a 5x4 factorial scheme, with eight replications. The treatments consisted of combinations of five
soybean cultivars with: Control (plants not inoculated with the nematode and not sprayed with
ASM), Pb (plants inoculated with the nematode), ASM (plants sprayed with the plant activator)
and Pb + ASM (plants inoculated with the nematode and sprayed with ASM). The treatments with
ASM were sprayed at 14, 21 and 28 days after the nematode inoculation (DAI) and, 24 hours later,
the plant height and the enzymatic activity were evaluated. At 60 DAI it was evaluated the plant
height, the shoots fresh weight (SFW), the roots fresh weigh (RFW), the reproduction factor (Rf)
and the nematode density in the roots. There was higher total protein concentration after the third
ASM application, with higher activity of chitinase (CHI) and β-1,3- glucanase (GLU) in the first
two applications, higher peroxidase (POX) in the second and lipoxygenase (LOX) and
phenylalanine ammonia-lyase (PAL) after the third. The results showed a possible synergistic
effect between the activities of CHI and GLU and antagonistic effect between the POX and the
FAL during the evaluation timings. ASM increased the activity of POX, LOX and FAL in the
absence or presence of the nematode. Activities of CHI and GLU were higher in plants parasitized
by the P. brachyurus. During the three evaluation periods the greatest heights were recorded at 28
DAI with lower height in plants with nematode. There was no statistical difference in height
between treatments. However, there were differences between cultivars. The cultivars BRSGO
8560 RR, Emgopa 313 RR e M-Soy 8360 RR showed greater heights. The plants had higher SFW
and RFW in treatments containing the nematode. In plants treated with ASM the nematode density
was higher, while the Rf was lower. Thus, the results showed that the plant activator ASM and the
nematode P. brachyurus affect the plant development and that the enzymatic activity is influenced
by the number of ASM sprays and by the presence of nematode. The resistance inducer, ASM,
shows potential for reducing the nematode`s reproduction. / O nematoide das lesões radiculares (Pratylenchus brachyurus) é um dos principais fitoparasitas da
sojicultura brasileira. Até o momento, não há uma alternativa limpa e de baixo custo de controle que reduza satisfatoriamente a população desse nematoide nas áreas infestadas ou que proporcione
pleno desenvolvimento das plantas parasitadas. Porém, estudos recentes mostram que há a
possibilidade de induzir as defesas das plantas como estratégia a ser inserida nos programas de
manejo das culturas a fitopatógenos, como os nematoides. Diante do exposto, o objetivo deste
trabalho foi avaliar a resposta bioquímica e o desenvolvimento de cultivares de soja quando
submetidas ao parasitismo pelo nematoide Pratylenchus brachyurus e tratadas com o indutor de
resistência abiótico acibenzolar-S- metil. A soja foi semeada em saquinhos de polietileno e, após
sete dias, os tratamentos foram inoculados com o nematoide. O experimento foi conduzido em
delineamento inteiramente casualizado em esquema fatorial 5x4, com oito repetições. Os
tratamentos foram compostos pela combinação das cinco cultivares de soja com: Testemunha
(plantas não inoculadas com o P. brachyurus e não pulverizadas com o ASM), Pb (plantas
inoculadas com P. brachyurus), ASM (plantas pulverizadas com o indutor de resistência) e
Pb+ASM (plantas inoculadas com o nematoide e pulverizadas com o ASM). Os tratamentos que
receberam ASM tiveram três aplicações (14, 21 e 28 dias após a inoculação do nematoide), sendo a
atividade enzimática avaliada 24 horas após a aplicação do produto. Aos 60 DAI foi avaliado a
altura, a massa fresca da parte aérea (MFPA) e das raízes (MFR), fator de reprodução (FR) e
densidade do P. brachyurus nas raízes. Observou-se maior concentração de proteínas totais após a
terceira aplicação do ASM, com maior atividade da quitinase (QUI) e β-1,3-glucanase (GLU) nas
duas primeiras aplicações, da peroxidase (POX) na segunda aplicação e da lipoxigenase (LOX) e
fenilalanina amônia-liase (FAL) após a terceira aplicação. Os resultados apresentam um possível
efeito sinérgico entre as atividades da QUI e GLU e antagonista entre a POX e a FAL durante
essas épocas. O ASM contribuiu para o incremento da atividade da POX, LOX e da FAL na
ausência ou presença do nematoide e as atividades da QUI e da GLU foram maiores nas plantas
parasitadas pelo P. brachyurus. Não houve diferença estatística para altura entre os tratamentos.
No entanto, houve diferença entre as cultivares. A BRSGO 8560 RR, Emgopa 313 RR e M-Soy
8360 RR apresentaram as maiores alturas. As plantas apresentaram maior MFPA e MFR nos
tratamentos contendo o nematoide. Nas plantas tratadas com o ASM a densidade de nematoides foi
maior, enquanto que, o FR foi menor. Sendo assim, os resultados mostram que o indutor e o
nematoide interferem no desenvolvimento da planta e a atividade enzimática é influenciada pelo
número de aplicações do ASM e pela presença do P. brachyurus. O indutor de resistência mostra
potencial para a redução da reprodução deste fitopatógeno.

Efeito do treinamento físico prévio nas alterações de função e estrutura renais provocadas pela administração de adriamicina em ratos / Effects of previous physical training on structural and functional renal disturbances induced by adriamycin in ratsCamila de Mattos Faleiros 03 May 2017 (has links)
A nefropatia induzida por adriamicina (ADR) em ratos é um dos modelos experimentais mais utilizados para o estudo desenvolvimento da doença renal progressiva. Uma dose única deste quimioterápico induz a proteinúria progressiva e irreversível que progride para glomeruloesclerose segmental e focal, com fusão dos processos podais e lesões tubulointersticiais. A lesão das células endoteliais glomerulares precede as alterações dos podócitos na nefropatia induzida pela ADR. A atividade física regular melhora as funções cardíacas e renais em pacientes e animais com doença renal progressiva e pode reduzir ou retardar a progressão da lesão renal. Este estudo avaliou o efeito do treinamento físico prévio na evolução da lesão renal induzida por ADR e a sua relação com o processo inflamatório, a função endotelial e angiogênese. Ratos Wistar submetidos ou não ao treinamento físico receberam ADR (2,5 mg/kg, e.v) ou solução salina fisiológica (SAL). Amostras de sangue e urina foram coletadas 60 dias após as injeções para avaliação da função renal e os rins removidos para estudos histológicos, imuno-histoquímicos, Western blot e de ELISA. Amostras de urina de 24 h, obtidas 7, 30 e 60 dias após a administração de ADR ou SAL, foram utilizadas para avaliação da albuminúria. Os ratos tratados com ADR apresentaram albuminúria progressiva, elevação dos níveis plasmáticos de creatinina e queda da taxa de filtração glomerular (TFG), lesão de podócitos, expansão da área mesangial, alargamento da área intersticial relativa no córtex renal, infiltração de macrófagos, aumento dos níveis de interleucina (IL)-1?, elevação dos níveis urinários do fator de transformação do crescimento ? (TGF-?) e dos níveis urinários de proteína quimiotática de monócitos 1 (MCP-1), diminuição de marcação de aminopeptidase P (marcador de células endoteliais) nos glomérulos e perda de capilares peritubulares corticais, que estavam associados com reduções das expressões do fator de crescimento endotelial vascular (VEGF) e da óxido nítrico sintase endotelial (eNOS) no córtex renal desses animais. Estas alterações foram menos intensas nos ratos que realizaram treinamento físico prévio ao tratamento com ADR. Em conclusão, o pré-condicionamento físico reduziu as lesões renais induzidas pela ADR. Este efeito esteve associado com as reduções do processo inflamatório, das lesões endoteliais e das alterações de fatores relacionados com o processo de angiogênese (VEGF e eNOS) no córtex renal. / Adriamycin (ADR)-induced nephropathy is one of the most experimental models of progressive kidney disease in rats. A single dose of this drug induces progressive and irreversible proteinuria that progresses to focal segmental glomerulosclerosis and tubulointerstitial lesions. The lesion of glomerular endothelial cells precedes the podocyte damage in nephropathy induced by ADR. Regular physical activity improves cardiac and renal functions in patients and animals with progressive renal disease and may reduce or delay the progress of impaired renal function. This study evaluated the effect of previous physical training in renal damage induced by ADR and the role of inflammation, endothelial lesions and angiogenesis in this process. Male Wistar rats submitted or not to previous physical training received ADR (2.5 mg/kg, i.v.) or physiological saline (SAL). Urine and plasma samples were collect 60 days after the injection in order to evaluated the renal function. The kidneys were removed for histological, immunohistochemical, Western blot and ELISA analysis. Twenty-four-hour urine samples were collected to dose albuminuria 7, 30 and 60 days after ADR or SAL injection. ADR-treated rats presented progressive albuminuria, increases in plasma creatinine levels, decreasing glomerular filtration rate (GFR), podocyte damage, mesangial expansion, enlargement of the tubular interstitial relative area of renal cortex, macrophage infiltration, higher interleukin (IL)- 1? levels in renal tissue, urinary transforming growth factor ? (TGF-?) and urinary monocyte chemoattractant protein (MCP)-1, reduction of aminopeptidase P (endothelial cell marker) in the glomeruli and cortical peritubular capillary number. Those were associated with reduction in vascular endothelial growth factor (VEGF) and endothelial nitric oxide synthase (eNOS) expressions in renal cortex. Those alterations were less intense in the animals undergone previous exercise training. In conclusion, physical training prior to ADR injection reduced the renal damage induced by this drug. This effect was related with the reduction of the inflammatory process, endothelial lesions and with the decrease in alterations of factors related to the process of angiogenesis (VEGF and eNOS) in renal cortex.

Análise da influência das forças oclusais e fatores locais na morfologia das lesões cervicais não cariosas: estudo clínico transversal / Influence of occlusal forces and local factors in the morphology of noncarious cervical lesions: cross sectional trialFabrícia Araújo Pereira 01 February 2016 (has links)
Objetivo: Analisar por meio de um estudo clinico transversal a influência das forças oclusais, assim como, de outros possíveis fatores locais na morfologia de lesões cervicais não cariosas (LCNC). Material e Método: Foram avaliados 32 indivíduos que possuíam LCNC nos primeiros pré-molares superiores, totalizando uma análise de 61 dentes, além disso, todos os sujeitos da pesquisa apresentavam todos os elementos dentais. Os mesmos eram pacientes regulares do serviço odontológico da Universidade Federal de Uberlândia, de ambos os gêneros, com idades entre 21 a 64. Para confirmação da morfologia e da severidade (profundidade) das LCNC, foi realizado exame cínico e replica das mesmas. As LCNC foram classificadas em anguladas ou arredondadas e em 3 níveis de severidade. A fim de determinar a presença de forças laterais, a presença de algum tipo de hábito parafuncional, como apertamento e bruxismo, foi analisado por meio de questionário e exame clinico. Também foram analisados, a presença de interferências oclusais (IO) em Relação Cêntrica (RC) e Movimentos excursivos, por meio de exame clinico de manipulação e análise dos modelos de gesso montados em articulador, assim como informações sobre a presença de facetas de desgaste. Além disso, informações sobre o periodonto, também foram coletadas, por meio de exame clinico, da mensuração das recessões gengivais no sentido cérvico-apical e mésio-distal. Assim como, da altura e espessura da cortical óssea vestibular, realizada por meio de Tomografia Computadorizada do tipo Cone-Beam (TCCB). Resultados: Inicialmente, aplicou-se o teste de Shapiro wilk, para avaliar a normalidade dos dados. Para os dados normais, posteriormente, aplicou-se o teste de Kruskall-wallis. Enquanto, que para as variáveis que não apresentaram distribuição normal, foi aplicado o teste Mann- Whitney. Dados gerais da amostra, demonstraram que a idade apresentou associação tanto com a morfologia (p = ,016), quanto para a severidade das LCNC (p=,000). O que não foi encontrado para o gênero (morfologia, p =,139; severidade, p =,308), nem para a presença de algum distúrbio gástrico (morfologia, p =,260; severidade, p =,831). Além disso, nenhuma relação foi encontrada entre a preseça de hábitos parafuncionais ou alguma IO, com a morfologia e severidade das LCNC. Todos os sujeitos da pesquisa apresentaram facetas de desgaste, entretanto, a presença de uma ou duas facetas não influenciou a morfologia ou severidade das lesões. Já para a recessão gengival, associação significativa foi encontrada para a morfologia (p =,046). Em relação a altura e espessura da crista da cortical vestibular, a altura foi significante em relação a severidade (p= ,009), e a espessura da crista, apresentou associação com a morfologia (p=,001) e severidade (p=,044). Conclusão: Não houve associação direta entre a morfologia das LCNC e as forças oclusais, entretanto, a idade e o suporte ósseo (menor altura e cortical menos espessa), foram fatores determinantes para a prevalência de LCNC anguladas e mais severas. / Objective: Analyze through a cross clinical study the influence of occlusal forces, as well as other possible local factors in the morphology of cervical lesions noncarious (LCNC). Material and Methods: We evaluated 32 individuals who had LCNC the first premolars, totaling an analysis of 61 teeth, moreover, all the subjects had complete dental arches. They were regular patients of the dental service at the Federal University of Uberlândia, of both genders, ages 21 to 64. To confirm the morphology and severity (depth) of LCNC was held cynical examination and replicates them. The LCNC were classified into angled or rounded and three severity levels. In order to determine the presence of lateral forces, the presence of some kind of habit parafuncional as bruxism and clenching was analyzed by questionnaire and clinical examination. Were also analyzed, the presence of occlusal interferences (OI) in Relation Centrica (CR) and movements excursive through clinical examination manipulation and analysis of plaster models mounted on articulator, as well as information about the presence of wear facets. In addition, information about periodontal were also collected through clinical examination, measurement of gingival recession in cervical-apical and mesiodistally. As well as the height and thickness of the buccal bone cortical held by Computed Tomography Cone-Beam type (TCCB). Results: Initially, we applied the Shapiro Wilk test to assess the normality of the data. For normal data, later applied the Kruskal-wallis test. While that for variables without normal distribution, the Mann-Whitney test was applied. General data of the sample, showed that age was associated with both the morphology (p =. 016), as to the severity of LCNC (p =.000). What was not found for the genre (morphology, p = .139; severity, p = 308), or to the presence of some gastric disorder (morphology, p = .260; severity, p = 831). Furthermore, no relationship was found between the preseça of parafunctional habits or some IO, with the morphology and severity of LCNC. All the subjects showed wear facets, however, the presence of one or two aspects not influence the morphology or severity of the lesions. As for the gingival recession, significant association was found for morphology (p =.046). Regarding the height and thickness of the crest cortical bone, the height was significant in relation to severity (p =.009), and the thickness of the crest, was associated with the morphology (p = .001) and severity (p = .044). Conclusion: There was no direct association between the morphology of NCCL and occlusal forces, however, the age and the bony support (lower thinner height and cortical), were determining factors for the prevalence of NCCL angled and more severe.

Achados ultrassonográficos em lesões de bifurcação coronária tratadas com stent único versus estratégia com dois stents / Intravascular ultrasound findings in coronary bifurcation lesions treated with single stenting versus double stenting strategiesRicardo Alves da Costa 10 August 2011 (has links)
Estudos comparativos prévios com inclusão de lesões de bifurcação selecionadas sugerem ausência de benefício de implante eletivo de dois versus stent único. No entanto, a aplicabilidade do técnica provisional parece ser dependente da complexidade da lesão de bifurcação. A área luminar da lesão após tratamento percutâneo, conforme medida pelo ultrassom intracoronário, tem demonstrado valor preditivo significante na evolução tardia dos pacientes submetidos a ICP. Os objetivos desta análise foram avaliar as dimensões luminares de lesões de bifurcação coronária complexas tratadas por ICP, conforme a análise pelo ultrassom, e correlacionar os achados com os resultados imediatos e tardios. Também objetivou-se identificar preditores angiográficos e ultrassonográficos de falência do ramo lateral nos diferentes passos do tratamento e seguimento. Entre maio de 2008 e agosto de 2009, 59 pacientes portadores de lesão de bifurcação complexa, com comprometimento significativo (estenose > 50%) dos dois ramos e lesão no ramo lateral extendendo-se além do óstio, foram abordados inicialmente com pré-dilatação do ramo lateral, a qual foi realizada com sucesso (estenose < 50%, fluxo TIMI 3, sem dissecção) em 54 pacientes (91,5%). Esse pacientes foram então randomizados para tratamento com stent único (estratégia provisional) (n = 27) versus stent duplo (n = 27). Durante o procedimento, 6 pacientes alocados no grupo stent único apresentaram falência do tratamento no ramo lateral (estenose residual >50%, fluxo TIMI < 3 ou dissecção), sendo que, 5 pacientes receberam stent adicional no ramo lateral para otimizar o resultado angiográfico. Ao final do procedimento, os valores médios e desvios padrão (DP) da área mínima do lúmen no óstio do ramo lateral (desfecho primário) foram 3,37 (1,62) mm2 no grupo stent único versus 5,50 (1,41) mm2 no grupo stent duplo (p < 0,001), conforme a alocação randômica. No seguimento angiográfico de 9 meses, as taxas de reestenose no ramo lateral foram 21,7% no grupo stent único versus 4% no grupo stent duplo (p = 0,06), sendo que, todas as recorrências envolveram a localização ostial. Considerando-se o tratamento recebido, a taxa de reestenose no ramo lateral foi significantemente maior nos pacientes tratados com stent único versus stent duplo (27,8% versus 3,3%, p = 0,01). Os preditores de falência da estratégia provisional foram: excêntricidade da lesão (p = 0,02), área mínima do lúmen (p = 0,08) e diâmetro mínimo do lúmen no óstio do ramo lateral (p = 0,06), extensão da lesão (p = 0,09) e percentual de estenose (p = 0,07) do ramo lateral. Com relação a reestenose angiográfica no ramo lateral, os preditores foram: área mínima do lúmen no óstio do ramo lateral ao final do procedimento (p = 0,03), tratamento com stent duplo (p = 0,02), diâmetro mínimo do lúmen (p = 0,03) e percentual de estenose (p = 0,02) no óstio do ramo lateral ao final do procedimento, ganho imediato no óstio do ramo lateral (p = 0,09) e diâmetro de referência do ramo lateral (p = 0,03). Estes resultados sugerem que lesões de bifurcação coronária complexas beneficiam-se de tratamento percutâneo com abordagem inicial com estratégia de dois stents, sendo que, tal benefício esteve relacionado a obtenção de maior área luminar no óstio do ramo lateral. / Previous comparative studies including selected bifurcation lesions have shown no advantage of elective double stenting implantation versus single stenting. However, the applicability of the provisional technique appears to be dependent on the bifurcation lesion complexity. The lesion luminal area after percutaneous treatment, as assessed by intravascular ultrasound, has demonstrated significant predictive value in the late follow-up of patients undergoing PCI. The objectives of this analysis were to evaluate the lesion luminal dimensions of complex coronary bifurcation lesions, as assessed by intravascular ultrasound, and to correlate such findings with acute and late outcomes. Also, it was aimed to identify angiographic and intravascular ultrasound predictors of side branch failure throughout the procedural steps and follow-up. Between may 2008 and august 2009, 59 patients with complex bifurcation lesions, including significant involvement (> 50% stenosis) of both branches and side branch lesion length extending from its ostium, were approached initially with side branch predilatation, which was successful (< 50% stenosis, TIMI 3 flow, no dissection) in 54 patients (91.5%). These patients were then randomized for treatment with single stenting (provisional strategy) (n = 27) versus double stenting (n = 27). During procedure, 6 patients allocated in the single stenting arm presented side branch failure (> 50% residual stenosis, TIMI flow < 3 or dissection), given that 5 patients received an additional stent in the side branch in order to optimize the angiographic result. At final procedure, the mean value and standard deviation (SD) for minimum lumen area at the side branch ostium (primary endpoint) were 3.37 (1.62) mm2 in single stenting versus 5.50 (1.41) mm2 in double stenting (p < 0.001), according to the randomized allocation. In the angiographic follow-up at 9 months, the restenosis rates in the side branch were 21.7% in single stenting versus 4% in double stenting (p = 0.06), given that all recurrences involved the ostial location. Considering the treatment received, the side branch restenosis rate was significantly increase among patients treated with single stenting versus double stenting (27.8% versus 3.3%, p = 0.01). The predictors of provisional strategy failure were: lesion eccentricity (p = 0.02), minimum lumen area (p = 0.08) and minimum lumen diameter in the side branch ostium (p = 0.06), lesion length (p = 0.09) and percent diameter stenosis (p = 0.07) of the side branch. Regarding angiographic restenosis in the side branch, predictors were: minimum lumen area in the side branch ostium at final procedure (p = 0.03), treatment with double stenting (p = 0.02), minimum lumen diameter (p = 0.03) and percent diameter stenosis (p = 0.02) in the side branch ostium at final procedure, acute gain at the side branch ostium (p = 0.09) and side branch reference diameter (p = 0.03). These results suggest that complex coronary bifurcation lesions may benefit from a primary percutaneous approach with double stenting strategy, given that most such benefit was associated with a larger lumen area obtained at the side branch ostium.

Toyoi sobre a proteção e a sobrevivência de camundongos contra a colonização intestinal, translocação e lesões intestinais após desafio com S. Typhimurium. Os animais foram alimentados 20 dias com rações contendo S. boulardii e B. Toyoi, administrados individualmente ou em associação, nas concentrações de 8 e 7 log UFC. g-1 de ração, respectivamente. Animais do grupo controle receberam ração comercial sem probióticos. No 10º dia após o início da administração dos probióticos, os animais foram desafiados por gavagem com 5,17 log UFC de S. Typhimurium (DL50). Proteção contra colonização de S. Typhimurium foi avaliada após eutanásia dos camundongos que sobreviverem após 10 dias, através de contagens de células viáveis do patógeno presentes nas fezes e intestino delgado. Para a avaliação da ocorrência de translocação, 2/3 dos órgãos (fígado e baço) foram macerados, ressuspensos em 1 mL de solução salina a 0,9% estéril e contagens de células viáveis do patógeno, em UFC.ml-1, foram realizadas em placas de XLD. Para a avaliação da ocorrência de lesão, 1/3 do intestino delgado foi avaliado através de histopatologia. O experimento foi repetido duas vezes. S. boulardii não protegeu os camundongos da morte induzida por S. Typhimurium e da ocorrência de lesões intestinais. Não houve sinergismo entre os dois micro-organismos avaliados. Entretanto, S. boulardii, assim como B. Toyoi reduziram a eliminação de S. Typhimurium pelas fezes e preveniram a translocação do patógeno para o fígado dos camundongos que sobreviveram ao desafio. B. Toyoi protegeu os animais da morte induzida pela infecção com S. Typhimurium e da ocorrência de lesões e o efeito obtido pela associação, ocorreu, provavelmente, devido somente ao efeito de B. Toyoi. Assim, novos estudos serão realizados para avaliar os efeitos da associação destes probióticos frente ao desafio com S. Typhimurium e para avaliar as causas do efeito antagônico apresentado entre eles sobre a persistência deste patógeno nas fezes dos animais e sobre a translocação para o fígado. / This study aimed to evaluate the effect of the association between the Saccharomyces boulardii and Bacillus cereus var. Toyoi probiotics on the survival and protection of mice against the occurrence of intestinal colonization, translocation and intestinal lesions after the challenge with S. Typhimurium. The animals were fed for 20 days with diet containing S. boulardii and B. Toyoi, administered individually or in association, in the food concentrations of 8 and7 log CFU. g-1, respectively. The control group animals were fed with commercial food without probiotics. On the 10th day after the start of administration of probiotics, the animals were challenged by the gavages with 5.17 log CFU of S. Typhimurium (LD50). Protection against colonization of S. Typhimurium was evaluated after euthanasia of the mice, which survived after 10 days, through the viable cell counts of the pathogen presented in the small intestine and stool. To evaluate the incidence of translocation, 2/3 of the organs (liver and spleen) were macerated, and resuspended in 1 ml of saline solution in 0.9% sterile, and viable cell counts of the pathogen were performed. To evaluate the occurrence of injury, 1/3 of the small intestine was evaluated through histopathology. The experiment was repeated twice. S. boulardii did not protect the mice from induced death by S. Typhimurium and the occurrence of intestinal lesions. For this reason, it was not possible to assess the occurrence of synergism between the two micro-organisms evaluated. However, S. boulardii as well as B. Toyoi reduced the elimination of S. Typhimurium through the stool and prevented the translocation of the pathogen to the liver of mice that survived the challenge. B. Toyoi protected animals from induced death through the infection with S. Typhimurium and through the occurrence of injuries, and the effect obtained by the association was, probably, due only by the effect of B. Toyoi. Thus, further studies will be conducted to evaluate the effects of the combination of these probiotics facing the challenge with S. Typhimurium and to assess the causes of the antagonistic effect presented among them on the persistence of this pathogen in the stool of animals and on the translocation to the liver.

Traumatic damage to the cervical spinal cord has a dramatic impact on determinants affecting cardiovascular fitness. In tetraplegia, in addition to diminished skeletal muscle function, impaired sympathetic cardiovascular innervation together with a dysfunction of catecholamine release is typically present. Individuals with tetraplegia are at a high risk of long-term inactivity, which increasingly contributes to the development of common civilization diseases. The aim of the current dissertation project is to investigate the cardiovascular fitness in males with cervical spinal cord injury (n=20) as compared to able- bodied males (n=27). Both groups completed exercise testing on an arm-crank ergometer. Arterial blood pressure response to exercise, peak oxygen consumption and peak heart rate were the primary outcome variables. In the first part of the study, a graded maximal arm- crank ergometer test to the subjective exhaustion was performed for the inter-group peak exercise comparison. The peak power dramatically differs between the groups and could be an important confounding factor limiting the evaluation of a physiologic response related to the equivalent physical loading. Noradrenergic Fiber Sprouting in the CerebellumKostrzewa, Richard M., Harston, Craig T., Fukushima, Hideki, Brus, Ryszard 01 January 1982 (has links)
In order to attain a better understanding of the sprouting response of noradrenergic fibers in the central nervous system (CNS), noradrenergic innervation to the cerebellum was observed by the glyoxylic acid method after a variety of manipulations and in a genetic variant of mouse classified as "Purkinje cell degeneration" (pcd/pcd). It has been found that a midbrain lesion in rats at birth will result in a collateral sprouting response of noradrenergic fibers in the cerebellum at 8 weeks, as indicated by the increased number of histofluorescent fibers observed in the molecular layer of the cerebellar cortex. Another procedure, treatment of neonatal rats with nerve growth factor alone appears to produce a temporary stimulation of noradrenergic fiber growth in the cerebellum, as observed by the histofluorescent method, although the innervation at 6 weeks or later is ultimately unchanged from the control group. In contrast, NGF (500 units) given to rats in combination with 6-hydroxydopa (6-OHDOPA) (60 μg/g IP) at 3 days postbirth produces a hyperinnervation of the cerebellum by noradrenergic fibers by 2 weeks of age and until at least 8 weeks of age. A third procedure, locus coeruleus implantation, was generally unsuccessful using the procedures described, since the implant was usually non-viable after several days. In a few instances where histofluorescent nuclei were found within the implant, there was an abundance of histofluorescent fibers within and adjacent to the implant, with fibers appearing to grow into host cerebellum. In the final procedure, it was noted that the density of noradrenergic input to the molecular layer of the cerebellar cortex was markedly increased in a genetic mutant mouse, classified as "Purkinje cell degeneration" (pcd/pcd), which is characterized by the absence of Purkinje cells of the cerebellum in adulthood. However, because of the tissue shrinkage that occurs after loss of Purkinje cells during postnatal development, it is unclear as to whether this observation represents hyperinnervation or a normal complement of fibers in a smaller brain space. The above procedures demonstrate the plasticity of noradrenergic fibers in neonatal cerebellum, a brain region that undergoes considerable postnatal development. The cerebellum is thought to be a good site for studying development/ regeneration/sprouting of noradrenergic fibers in particular, and central axonal processes in general.

Aims: Noninvasive fractional flow reserve (FFR) measurement with computed tomography (FFRCT) is a newly described method for assessing functional significance of coronary disease. The objective of this metaanalysis is to determine the diagnostic performance of FFRCTin the assessment of hemodynamically significant coronary artery stenosis. Methods: PubMed and the Cochrane Center Register of Controlled Trials were searched from January 2000 through February 2015. Six original studies were found comparing FFRCTto invasive FFR in evaluating hemodynamic significance of coronary lesions (1354 vessels; 812 patients). Lesions were considered hemodynamically significant if invasive FFR was 0.80 or less. FFRCTused the same cutoff as invasive FFR to be considered as a positive test. Sensitivity, specificity, positive and negative likelihood ratios, and diagnostic odds ratio were calculated. Results: One-third of the lesions (n=443) were hemodynamically significant. The pooled per-vessel analysis showed that the sensitivity, specificity, negative and positive likelihood ratios, and diagnostic odds ratio of FFRCTto diagnose hemodynamically significant coronary disease were 0.84 [95% confidence interval (CI):0.80-0.87], 0.76 (95% CI: 0.73-0.79), 0.22 (95% CI: 0.17-0.29), 3.48 (95% CI: 2.21-5.47), and 16.82 (95% CI: 8.20-34.49), respectively. Conclusion: The results of this meta-analysis demonstrate that FFRCTresults correlate closely with invasive coronary angiography and FFR measurement. It is a feasible noninvasive method to assess hemodynamic significance of coronary lesions in patients with stable coronary artery disease.

Axonal Regeneration in the Sensory Dorsal Column PathwayHagg, Theo 06 February 2015 (has links)
This review provides a short historical background to the field of axonal regeneration and discusses the advances made in over 100 studies between 2007 and 2012 in understanding the molecular mechanisms underlying the conditioning lesion and regeneration of primary sensory axons in the dorsal columns of the spinal cord. Treatment strategies to stimulate axon growth and reinnervation of the spinal cord through the dorsal root entry zone and of the dorsal column nuclei in the medulla are highlighted. Major breakthroughs have been made, e.g., reinnervating the nucleus gracilis in the medulla using neurotrophic factor gradients and grafts as relays and identifying chondroitin sulfate proteoglycan receptors. The experimental accessibility of the dorsal column axons has also resulted in new technological advances, including live imaging. Last, future directions are discussed, including some challenges of translation to humans.
