Antioxidant Capacity, Lipid Peroxidation, and Lipid Composition Changes During Long-Term and Short-Term Thermal Acclimation in Daphnia

Examples of phenotypic plasticity—the ability of organisms of identical genotypes to produce different phenotypes in response to the environment—are abundant, but often lack data on the causative physiology and biochemistry. Phenotypes associated with increased protection against or reduced damage from harmful environments may, in fact, be downstream effects of hidden adaptive responses that remain elusive to experimental measurement or be obscured by homeostatic or over-compensatory effects. The freshwater zooplankton crustacean Daphnia drastically increases its heat tolerance as the result of acclimation to high temperatures, an effect often assumed to be based on plastic responses allowing better protection against oxidative stress. Using several geographically distant Daphnia magna genotypes, we demonstrate that the more heat tolerant individuals have a higher total antioxidant capacity (TAC) both in the comparison of heat-acclimated vs. non heat-acclimated females and in the comparison of females to age- and body size-matched males, which show lower heat tolerance than females. However, experimental manipulations of hypothesized antioxidant pathways by either glutathione addition or glutathione synthesis inhibition had no effect on heat tolerance. Lipid peroxidation (LPO), contrary to expectations, did not appear to be a predictive measure of susceptibility to thermal damage: LPO was higher, not lower, in more heat tolerant heat-acclimated individuals after exposure to a lethally high temperature. We hypothesize that LPO may be maintained in Daphnia at a constant level in the absence of acute exposure to elevated temperature and increase as a by-product of a possible protective antioxidant mechanism during such exposure. This conclusion is corroborated by the observed short-term and long-term changes in phospholipid composition that included an increase in fatty acid saturation at 28 °C and up-regulation of certain long-chain polyunsaturated fatty acids. Phospholipid composition was more strongly affected by recently experienced temperature (4-day transfer) than by long-term (2 generations) temperature acclimation. This is consistent with partial loss of thermal tolerance after a short-term switch to a reciprocal temperature. As predicted under the homeoviscous adaptation hypothesis, the more heat tolerant Daphnia showed lower membrane fluidity than their less heat tolerant counterparts, in comparison both between acclimation temperatures and among different genotypes. We conclude that thermal tolerance in Daphnia is influenced by total antioxidant capacity and membrane fluidity at high temperatures, with both effects possibly reflecting changes in phospholipid composition.

Estudo da expressão de genes associados ao perfil de ácidos graxos em bovinos Nelore confinados / Study of the gene expression associated to fatty acid profile in Nellore cattle feedlot

Nos últimos anos tem-se destacado a importância dos ácidos graxos presentes na carne e leite, em especial aqueles que poderiam influenciar a saúde humana. A deposição de gordura, e também sua composição, é resultado da interação entre os fatores genotípicos e fenotípicos. Sendo assim, o contínuo melhoramento genético, com a seleção de determinados genes, pode influenciar na qualidade do produto que chega à mesa do consumidor. Objetivou-se com este estudo avaliar os padrões de expressão dos genes diferencialmente expressos relacionados ao perfil de ácidos graxos da carne de bovinos Nelore confinados, por meio da verificação dos padrões de expressão de genes envolvidos no metabolismo lipídico e na síntese dos ácidos graxos palmítico (C16:0), esteárico (C18:0), oléico (C18:1 cis-9), linoléico (C18:2 cis-9 cis- 12), CLA (C18:2 cis-9 trans-11) e linolênico (C18:3). Foram utilizados 48 bovinos machos inteiros, Nelore, com idade aproximada de 24 meses, dos quais foram coletadas amostras do músculo Longissimus para a realização das análises. Verificou-se um total de 1173 genes diferencialmente expressos entre os grupos de baixa e alta concentração de cada um dos ácidos graxos. Os genes diferencialmente expressos identificados no músculo Longissimus foram ACAT1, ACOX2, ACOT11, ACSM3, ACSS1, AGPAT6, BDH1, CIQTNF3, CYP4B1, DGAT2, FABP3, FABP4, FABP7, GK, IGF2, LCAT, LIPE (HSL), LPL, PLIN1, PLIN5, SCD5 e SLC27A6, que possuem ações nas vias metabólica dos ácidos graxos ou nas vias adjacentes, podendo influenciar sobre o perfil lipídico da carne. Estudos com RNAseq envolvendo o perfil de ácidos graxos na carne ainda são escassos, e em animais zebuínos não há relatos. Desta forma, ainda são necessários mais estudos a fim de esclarecer como determinados genes podem influenciar as características desejadas, como por exemplo, o perfil de ácidos graxos da gordura intramuscular. / Recently, the importance of fatty acids on meat and milk has been highlighted, especially those that could affect human health. The fat deposition, and its composition, is a result of the interaction between genotypic and phenotypic factors. In this way, the continuous genetic improvement, with the selection of some genes, can influence the quality of the product acquired by the consumers. The aim of this study was evaluate the patterns of expression of differentially expressed genes related to the fatty acid profile of Nellore cattle feedlot, by verifying the patterns of expression of genes involved in lipid metabolism and in the synthesis of fatty acids palmitic (C16: 0), stearic (C18: 0), oleic (C18: 1 cis-9), linoleic (C18: 2 cis-cis- 9-12), CLA (C18: 2 cis-9 trans-11) and linolenic acid (C18: 3). Forty-eight bovine, male, Nellore, with an average age of 24 months, from which were collected samples of Longissimus Muscle to carry out the analysis. In this way, further studies are still needed in order to clarify how certain genes can influence the desired characteristics, for instance, the fatty acid profile of intramuscular fat content.

Efeito do teor de fibra e do processamento de milho na qualidade e perfil de ácidos graxos da carne de bovinos Nelore / Effect of roughage level and corn processing on meat quality and fatty acid profile of Nellore bulls

Foi avaliado o efeito do método de processamento do milho (PRC) e do teor de fibra insolúvel em detergente neutro proveniente de forragem (FDNf) na qualidade físico- químicas, analise sensorial e no perfil de ácidos graxos da carne in natura e após cocção de bovinos Nelore em confinamento. Foram utilizados 56 bovinos não castrados da raça Nelore, com peso inicial de 382 kg. Os animais foram distribuídos em baias individuais sendo separados de acordo com o peso vivo inicial, formando assim o delineamento em blocos completos casualizados, em esquema fatorial 2x4. Do total de 112 animais foram selecionados aleatoriamente 56 bovinos perfazendo sete animais de cada tratamento. Foram confinados durante 81 dias, sendo 21 dias de adaptação e 60 dias de tratamento. As dietas de tratamento continham teores de 3, 8, 13 e 18% de fibra insolúvel em detergente neutro proveniente de forragem (FDNf) e dois métodos de processamentos dos grãos de milho, sendo grão úmido ensilado - GU e grão seco moído fino - MM. Os dados foram analisados pelo procedimento MIXED e as medias foram determinadas pelo LSMEANS. As medias foram determinadas pelo LSMEANS. Testes para os efeitos fixos com valores de probabilidade inferiores ou iguais a 0,05 foram considerados significativos. Não foi observado efeito de interação entre FDNf e PRC para as características de qualidade da carne como umidade, força de cisalhamento, pH, cor e características sensoriais. Ao avaliar o teor de EE do músculo Longissimus dorsi houve diferença (P=0,05) entre os métodos de PRC utilizados. Os valores de EE obtidos no músculo L. dorsi dos animais alimentados foram de 1,80 vs. 1,48 para GU e MM, respectivamente. A relação ?6:?3 apresentou decréscimo linear com o aumento no teor de FDNf, mostrando uma relação de 4,24 na dieta com 3% e 3,31 para as dietas contendo 18% de FDNf (P<0,05). A concentração de CLA encontrada na carne de animais alimentados com GU está dentro da faixa recomendada para carne bovina que é de 0,2 a 1,0% do total de ácidos graxos. Houve um aumento no teor de lipídios na carne dos animais que receberam dietas com milho grão úmido ensilado. O aumento no teor de FDNf na dieta resultou em menor relação ?6:?3 na carne, algo desejável sob o ponto de vista da saúde do consumidor. O cozimento da carne não alterou o perfil de ácidos graxos da carne, pode-se então estimar o perfil de ácidos graxos por meio da análise em carnes cruas. / Was evaluated the effect of corn processing method (PRC) and the content of neutral detergent fiber from roughage (NDF) in physicochemical quality and sensory analysis in the fatty acid profile of fresh beef and after cooking of Nelore cattle feedlot. We used 56 Nellore bulls with an initial weight of 382 kg. The animals were divided into individual stalls being separated according to initial body weight, thus forming the randomized complete block design in a 2x4 factorial design. Of the total of 112 animals were randomly selected 56 bulls totaling seven animals of each treatment. Were confined for 81 days, being 21 days of adaptation and 60 days of treatment. The treatment diets contained levels of 3, 8, 13 and 18% neutral detergent fiber from roughage (NDFr) and two methods of processing corn grains, being high moisture corn - GU and finely ground dry corn - MM. The data were analyzed using the MIXED procedure and the media were determined by LSMEANS. Tests for fixed effects with probability values less than or equal to 0.05 were considered significant.] There was no interaction effect between NDFr and PRC for meat quality characteristics such as moisture, shear force, pH, color and sensory characteristics. When evaluating the EE content of the Longissimus dorsis there was difference (P=0.05) between the PRC methods used. The EE values obtained in the animals L. dorsi muscle were 1.80 vs. 1.48 for GU and MM, respectively. The concentration of CLA found in animals meat fed with GU is within the recommended range for beef which is 0.2 to 1.0% of total fatty acids. The relationship ?6:?3 displayed a linear decrease with the increase of NDFr level, showing a ratio of 4.24 in the diet with 3% NDFr and 3.31 in diets containing 18% NDFr (P<0.05). The concentration of CLA found in animals meat fed with GU is within the recommended range for beef which is 0.2 to 1.0% of total fatty acids. There was an increase in fat content in the meat of animals fed diets with high moisture corn silage. The increase in the content of FNDr in the diet, resulted in a lower ratio of ?6: ?3 in the flesh, desirable from the point of view of consumer health. The cooking of the meat did not alter the fatty acid profile of meat, can then estimate the fatty acid profile by analyzing in raw meats.

Thus, the fat body, besides acting as a reserve site and synthesis of proteins, carbohydrates and lipids participates in the production of substances with action on the immune system, detoxification, production of eggs, sperm and pheromone. However, the main function of the fat body is the reserve of lipids. The fat body of insects is usually divided into two regions, the visceral fat body, located near the digestive tract, and the parietal fat body, located near the cuticle. The cell types found in the fat body of insects vary, being found only one type in some and more than ten kinds in others. The main cell types found in the fat body of insects are trophocytes, urocytes and oenocytes. The morphology and biochemistry of fat body Lutzomyia longipalpis, the main vector of visceral leishmaniasis in the Americas, was examined by light microscopy, electron microscopy and high-performance thin layer chromatography. Thus, cuts through the abdomen of adult males and females showed that the fatty body is divided into two main components, in accordance with the spatial distribution in the insect's body: one parietal part which is located just under the cuticle and other visceral which is distributed suspended lobes and often associated with tracheas in hemocele. The fat body of L. longipalpis contains only one cell type, trophocyte, which has a large amount of lipid droplets, protein and glycogen granules in their cytoplasm rosettes. The lipid composition varies according to the physiological and insect species. The neutral lipid stored in fat body found more insects is the triacylglycerol. In addition, small amounts of diacylglycerol, steroids, free fatty acids, carotenoids and monoglycerides are carried by lipoforina (major lipoprotein of the insects). The diacylglycerol is derived from triglycerides stored in fat body and is the main form of fatty acid which are recruited to sites of utilization such as flight muscles, for example. Biochemical analysis of the abdominal tergites L. longipalpis males, by high-performance thin layer chromatography, showed the presence of different classes of neutral lipid (mono-, di- and triacylglycerols, fatty acids, cholesterol and esterified cholesterol) and phospholipids (phosphatidylcholine, phosphatidylethanolamine, phosphatidylinositol, lysophosphatidylcholine) in the fat body. Furthermore, the lipid composition of the abdominal tergites varied, with the highest amount of lipids extracted from the fourth tergite, which has pheromone producing gland. Finally, the main neutral lipid extracted from the fat body of L. longipalpis was the triacylglycerol and the main phospholipid was phosphatidylethanolamine. / O corpo gorduroso dos insetos est? envolvido em fun??es de grande import?ncia. Exploring the Interplay of Lipids and Membrane Proteins

The interplay between lipids and membrane proteins is known to affect membrane protein topology and thus have significant effect (control) on their functions. In this PhD thesis, the influence of lipids on the membrane protein function was studied using three different membrane protein models. A monotopic membrane protein, monoglucosyldiacylglyecerol synthase (MGS) from Acholeplasma laidlawii is known to induce intracellular vesicles when expressed in Escherichia coli. The mechanism leading to this unusual phenomenon was investigated by various biochemical and biophysical techniques. The results indicated a doubling of lipid synthesis in the cell, which was triggered by the selective binding of MGS to anionic lipids. Multivariate data analysis revealed a good correlation with MGS production. Furthermore, preferential anionic lipid sequestering by MGS was shown to induce a different fatty acid modeling of E. coli membranes. The roles of specific lipid binding and the probable mechanism leading to intracellular vesicle formation were also investigated. As a second model, a MGS homolog from Synechocystis sp. PCC6803 was selected. MgdA is an integral membrane protein with multiple transmembrane helices and a unique membrane topology. The influence of different type of lipids on MgdA activity was tested with different membrane fractions of Synechocystis. Results indicated a very distinct profile compared to Acholeplasma laidlawii MGS. SQDG, an anionic lipid was found to be the species of the membrane that increased the MgdA activity 7-fold whereas two other lipids (PG and PE) had only minor effects on MgdA. Additionally, a working model of MgdA for the biosynthesis and flow of sugar lipids between Synechocystis membranes was proposed. The last model system was another integral membrane protein with a distinct structure but also a different function. The envelope stress sensor, CpxA and its interaction with E. coli membranes were studied. CpxA autophosphorylation activity was found to be positively regulated by phosphatidylethanolamine and negatively by anionic lipids. In contrast, phosphorylation of CpxR by CpxA revealed to be increased with PG but inhibited by CL. Non-bilayer lipids had a negative impact on CpxA phosphotransfer activity. Taken together, these studies provide a better understanding of the significance of the interplay of lipids and model membrane proteins discussed here.

Do total de 112 animais foram selecionados aleatoriamente 56 bovinos perfazendo sete animais de cada tratamento. Foram confinados durante 81 dias, sendo 21 dias de adaptação e 60 dias de tratamento. As dietas de tratamento continham teores de 3, 8, 13 e 18% de fibra insolúvel em detergente neutro proveniente de forragem (FDNf) e dois métodos de processamentos dos grãos de milho, sendo grão úmido ensilado - GU e grão seco moído fino - MM. Os dados foram analisados pelo procedimento MIXED e as medias foram determinadas pelo LSMEANS. As medias foram determinadas pelo LSMEANS. Testes para os efeitos fixos com valores de probabilidade inferiores ou iguais a 0,05 foram considerados significativos. Não foi observado efeito de interação entre FDNf e PRC para as características de qualidade da carne como umidade, força de cisalhamento, pH, cor e características sensoriais. Ao avaliar o teor de EE do músculo Longissimus dorsi houve diferença (P=0,05) entre os métodos de PRC utilizados. Os valores de EE obtidos no músculo L. dorsi dos animais alimentados foram de 1,80 vs. 1,48 para GU e MM, respectivamente. A relação ?6:?3 apresentou decréscimo linear com o aumento no teor de FDNf, mostrando uma relação de 4,24 na dieta com 3% e 3,31 para as dietas contendo 18% de FDNf (P<0,05). A concentração de CLA encontrada na carne de animais alimentados com GU está dentro da faixa recomendada para carne bovina que é de 0,2 a 1,0% do total de ácidos graxos. Houve um aumento no teor de lipídios na carne dos animais que receberam dietas com milho grão úmido ensilado. O aumento no teor de FDNf na dieta resultou em menor relação ?6:?3 na carne, algo desejável sob o ponto de vista da saúde do consumidor. O cozimento da carne não alterou o perfil de ácidos graxos da carne, pode-se então estimar o perfil de ácidos graxos por meio da análise em carnes cruas. / Was evaluated the effect of corn processing method (PRC) and the content of neutral detergent fiber from roughage (NDF) in physicochemical quality and sensory analysis in the fatty acid profile of fresh beef and after cooking of Nelore cattle feedlot. We used 56 Nellore bulls with an initial weight of 382 kg. The animals were divided into individual stalls being separated according to initial body weight, thus forming the randomized complete block design in a 2x4 factorial design. Of the total of 112 animals were randomly selected 56 bulls totaling seven animals of each treatment. Were confined for 81 days, being 21 days of adaptation and 60 days of treatment. The relationship ?6:?3 displayed a linear decrease with the increase of NDFr level, showing a ratio of 4.24 in the diet with 3% NDFr and 3.31 in diets containing 18% NDFr (P<0.05). The concentration of CLA found in animals meat fed with GU is within the recommended range for beef which is 0.2 to 1.0% of total fatty acids. There was an increase in fat content in the meat of animals fed diets with high moisture corn silage. The increase in the content of FNDr in the diet, resulted in a lower ratio of ?6: ?3 in the flesh, desirable from the point of view of consumer health. The cooking of the meat did not alter the fatty acid profile of meat, can then estimate the fatty acid profile by analyzing in raw meats.

L'incorporation d'ingrédients bioactifs dans les produits alimentaires est en plein essor. Il a été démontré que ces ingrédients possèdents des propriétés biologiques importantes permettant l'amélioration de la santé et la prévention des maladies dites de civilisations. Toutefois, l'ajout de ces molécules bioactives est dans la plupart des cas impossible ou insuffisant, du fait que ces composés ne sont que peu solubles dans les systèmes aqueux et présentent i) une stabilité limitée contre les dégradations chimique ou physique, ii) une libération non contrôlée ou une faible biodisponibilité. Face à ces contraintes, les recherches actuelles visent à élaborer des systèmes d'encapsulation efficaces pour résoudre ces problèmes de formulation. Dans notre étude, deux représentants d'ingrédients alimentaires ont été choisi : le sesquiterpène nérolidol (Ner) et le flavonoïde quercétine (Quer) présentant diverses activités biologiques mais des propriétés physicochimiques problématiques. Ainsi, l'objectif de notre travail a été d'encapsuler ces composés actifs dans les cyclodextrines (CDs), les liposomes conventionnels (LCs) et le système mixte cyclodextrine-liposomes (DCLs) afin de développer des systèmes naturels et éco-compatibles ayant des applications potentielles dans les domaines alimentaires.Trois axes ont été abordés. Le premier axe a porté sur la préparation et la caractérisation des complexes d'inclusion CD/invité en solution et à l'état solide. Les techniques de spectroscopie UV-visible, Chromatographie Liquide à Haute Performance (CLHP), Carbone Organique Total (TOC), ¹H Résonance magnétique nucléaire (RMN), 2D ROESY RMN et de la modélisation moléculaire ont été utilisées comme outils pour la caractérisation des complexes obtenus. Des études de phase de solubilité ont également été réalisées. Le deuxième axe a porté sur la préparation des LCs et DCLs par la méthode d'injection éthanolique et leur caractérisation. Les préparations des LCs encapsulant la quercétine a été réalisée à partir de phospholides naturels de jaune d'oeuf (Lipoid E80) et de soja insaturés (Lipoid S100) ou saturés (Phospholipon 90H) afin d'étudier l'effet de la composition lipidique que les caractéristiques des liposomes. La formulation optimale a été par la suite appliquée pour préparer des LCs encapsulant le nérolidol et des DCLs encapsulant les deux molécules. Ce dernier est produit par l'incorporation des complexes d'inclusion HP-β-CD/Ner (à différents rapport moléculaire CD:Ner) et SBE-β-CD/Quer dans la cavité aqueuse des liposomes. Le dernier axe a été orienté vers l'évaluation de l'effet de l'encapsulation sur les propriétés physicochimiques du nérolidol et de la quercétine (libération in vitro, photostabilité, stabilité dans les milieux gastro-intestinales, stabilité de stockage) et leur activité antioxydante. Les résultats ont montré que les CSs ont été capables d'encapsuler les composés actifs étudiés, d'augmenter leur solubilité, leur photostabilité ainsi que leur activité antioxydante. En outre, les liposomes à base de Lipoid E80 ont été trouvés majoritairement de taille nanométrique et ont conféré aux molécules une efficacité d'encapsulation (EE) élevée ainsi qu'une meilleure stabilité par rapport aux deux autres types de liposomes. De plus, la taille des DCLs ains que leur EE ont été prouvées dépendante du rapport moléculaire CD:invité. Par rapport aux LCs, les DCLs ont assuré une libération prolongée du nérolidol, ont augmenté la photostabilité des composés et la stabilité de la quercétine dans les milieux biologiques. Les résultats de cette étude suggèrent que ces systèmes peuvent être considérés comme outils prometteurs pour l'optimisation des formulations alimentaires incorporant le nérolidol et/ou la quercétine. / Phytochemicals are widely distributed secondary metabolites, divided into three major classes : terpenoids, flavonoids and alkaloids. They are shown to possess important biological properties such as anti-cancer, anti-inflammatory and anti-microbial properties. Therefore, increasing the use of these bioactive molecules in food products may reduce the risk of widespread diseases referred to as "diseases of civilization". However, their low solubility, susceptibility to degradation and their rapid release reduce their bioavailability in the human body and thus their biological effect. To solve the aforementioned physicochemical drawbacks, encapsulation systems were developed to allow the incorporation of phytochemicals in food. In this study, two food ingredients : the sesquiterpene nerolidol and the flavonol quercetin were selected du to their potent biological activities but their problematic physicochemical properties.Therfore, the aim of this work was to encapsulate these molecules into cyclodextrins (CDs), conventional liposomes (CLs) and the double systeme drug-in-cyclodextrin-in-liposomes (DCLs), in order to develop nztural and biocompatible formulations that may find applications in food fields. This project was built around three main research axes. The first part dealt with the preparation and the characterization of CD/guest inclusion complexes both in solution and in solid state. Characterizations were performed with UV-visible spectroscopy, High Performance Liquid Chromatography (HPLC), Total Oragnic Carbon (TOC), ¹H NMR, 2D ROESY NMR, and molecular modeling. These investigations were complemented with phase solubility studies.The second axis addressed the preparation of CLs ans DCLs by ethanol injection method and characterization of the vesicles. CLs encapsulating quercetin were prepared from three different types of phospholipids (Lipoid E80, Lipoid S100, Phospholipon 90H) in order to study the effect of lipid composition on the characteristics of liposomes. The optimal formulation was then selected to prepare nerolidol loaded-CLs and DCLs encapsulating the two compounds. HP-β-CD/Ner (at different CD:Ner molar ratios) and SBE-β-CD/Quer inclusion complexes were used as the aqueous phase in the DCL system. The last part focused on the effect of encapsulation on the physicochemical properties of nerolidol and quercetin (in vitro release, photostability, stability in gastro-intestinal fluids, storage stability) and their antioxidant activities. Results demonstrated that CDs could successfully encapsulate bioactive compounds, enhance their solubility , photostability and antioxidant activity. Furthermore, Lipoid E80-liposomes were nanometric in size, exhibited a high entrapment efficiency and higher stability in comparison to the other formulations. Moreover, CD:guest molar ratio influenced the size of DCLs and their encapsulation efficiency. When compared to CLs, DCLs extended the release of neridol, enhanced the photostability of both compounds ans increased the stability of quercetin in biological fluids. These results could be considered as a promising tool to achieve an optimized and efficient formulation incorporating nerolidol and quercetin in food industry.
