The thesis is focused on the aesthetic value of the landscape. The literature review is clarified a few important concepts that are necessary to understand the issue. The aim of this work is to capture the importance of landscape in terms of aesthetics. In this work was developed a questionnaire that was supposed to find out how the landscape is perceived by residents. Upon completion of the research, questionnaires were comprehensively evaluated and was conducted a case study on the town of Rosice. Based on the results and their mutual comparison it is possible to evaluate how the general public perceives selected landscape elements and how it operates landscape in which they live. As part of the work was also to determine which of the landscape values deemed important to preserve for future generations and identify problems that threaten the landscape around them.

PhD thesis deals with the impact of natural elements and structures in the image of the city. Topic is on the edge of urbanism, urban composition, aesthetics and landscape protection. This topic is often mentioned in relation with the identification and evaluation of landscape character. Landscape character significantly affects the urban structure of a city -- foundation of the city and functional layout; It is also reflected in the spatial and urban composition. Image of the city can be analyzed from different perspectives, just as it is possible to analyze the urban composition. Image of the city is solved by number of methodologies and PhD theses, although there are rather focused on the landscape or the town silhouette in surrounding landscape. This PhD thesis is focused on natural elements and structures in the city. So it is not about a city in a surrounding landscape but about townscape. It analyzes the image of the city and follows the methodology of Michael Hexner -- it evaluates the image of the city with regard to relationship of urban structures and natural elements with respect to their visual expression and visual appeal. Conclusions of the work define a relationship between the application of natural elements in the image of the city and its positive acceptance, perception and evaluation. The thesis is mainly focused on assessment of an aesthetic and compositional relationship of natural elements inside the city.

Poetika a malebnost v krajině / Poetics and picturesqueness in landscape

This thesis offers a compact insight into landscape, its poetics and picturesqueness. It mentions not only basic landscape definition and landscape character but also its structures and typologies. Next, it describes how a person perceives the landscape trough centuries. The next chapter is devoted to Karel Hynek Mácha who was not only a famous poet and representative of Romanticism but also a painter and traveler. Thesis maps poetics and picturesque aspects trough poems, notes and sketches. Author then follows Karel Hynek Mácha. The main aim was to perceive the landscape differently. Main focus is on the poetic and picturesqueness only trough her own eyes, paper and pencil without modern technology and just feel the magic of the moment. Literary research is followed by a project which is situated near Karlovy Vary in the west part of the Czech Republic. Concretely is the landscape located nearby Andělská Hora. First, the thesis describes the character of the area by the help of analysis. There are historical analysis, actual condition, natural condition and trees inventory. Analysis is followed by the project which first contains general concept and its basic principles. Project also contains drawing of the individual sites. The project is aimed at both landscape solutions and restoration of small sacral architecture and other elements that will not only contribute to greater scenic beauty, but also support the genius loci of the local landscape. Proposed elements not only enrich the local landscape and enhance tourist's stay, but also get them to think. It is important to keep in mind the historical legacy and ancestral land.

Genetic mapping of QTL for Fusarium head blight resistance in winter wheat cultivars Art and Everest

Master of Science / Agronomy / Allan Fritz / Fusarium head blight (FHB) is a fungal disease, mostly commonly associated with F. graminearum, which affects cereal crops such as wheat resulting in substantial yield losses and reductions in grain quality. The onset of the disease can occur rapidly when warm, wet or humid weather coincides with flowering in the spring. The pathogen also produces mycotoxins such as deoxynivalenol (DON) that accumulate in the grain and can be toxic to humans and animals. This results in additional economic losses as contaminated grain must be discarded or blended to reduce the amount of toxin in order to meet federal regulatory limits. Development and deployment of resistant cultivars has proved to be an effective method to combat the disease, and many resistant sources have been reported in the literature with the majority of major resistance coming from Chinese landraces. Transferring resistance from these sources into cultivars adapted to the U.S. has been a slow process due to linkage of FHB resistance genes with poor agronomic traits. Therefore, it is important for breeders to search for sources of resistance in native material adapted to their local conditions. In this study, we aimed to identify quantitative trait loci (QTL) for resistance to spread of FHB within the head (Type II resistance), accumulation of DON toxin in grain (Type III resistance), and resistance to kernel infection (Type IV resistance). Plant material consisted of 148 doubled haploid (DH) lines from a cross between the two moderately resistant hard red winter wheat (HRWW) cultivars Art and Everest. The study was conducted for two years using a point inoculation technique in a greenhouse in Manhattan, KS. Three QTL conferring resistance to FHB traits were detected on chromosomes 2D, 4B, and 4D. The QTL on chromosomes 4B and 4D overlapped with the major height genes Rht1 and Rht2, respectively. Plant height has shown previous associations with FHB, though the underlying cause of these associations is not well understood. The majority of results have reported increased susceptibility associated with shorter plant types; however, in this study, the haplotype analysis for the Rht-B1 and Rht-D1 loci showed an association between the dwarfing alleles and increased resistance to FHB. This suggests either pleiotropic effects of these loci or perhaps linkage with nearby genes for FHB resistance. Markers close to the peaks of the FHB resistance QTL have the potential for Kompetitive Allele Specific PCR (KASP) marker development and subsequent use in marker assisted selection (MAS) to help improve overall FHB resistance within breeding programs.

Use of tRNA Gene Probes to Identify Polymorphic Loci in the Bovine Genome

A 30-mer oligonucleotide probe encoding the "A box" and anticodon loop regions of a human glycine tRNA gene was used to isolate a 581bp DNA fragment from a bovine genomic DNA library. Although the cross-hybridizing segment of DNA was found not to encode any tRNA gene or pseudogene, a region with homology to the "C-element" of the "BOV-tA" type Alulike artiodactyl retroposons was identified. This cross-hybridization was determined to be the result of conserved RNA polymerase III promoter elements in the probe portion of the tRNA gene and these repetitive elements. A microsatellite repeat (TC) was also found associated with this element. Future screening for bovine tRNA genes will require the use of a) longer probes and higher stringency hybridization conditions or b) the simultaneous screening with probes from the 5' and 3' ends of the gene which avoid the conserved Pol III promoter boxes.
