Bridging

Bridging is considered as a formal, spatial, referential, and tectonic articulation of connectedness between architecture and context. The question is probed through a mixed architectural program situated in the interstice of an urban downtown and residential neighborhood. The architecture originates from singular or hybridized combinations of these characteristics: whereas formal defines the compositional relationships through, for example, orientation, grids, scales, proportions, and contrast or balance among the parts; whereas spatial indicates a gradient of boundaries established through anchoring, intersecting, overlapping, projecting, interlocking, and parallel elements; whereas referential draws connections through an interpretation of distinct characteristics from the present, past, and future environmental context; and whereas tectonic consists of the underlying structure, frame or mass, and materiality without which the formal, spatial, and referential concepts cannot become physical. / Master of Architecture / Bridging is considered as a formal, spatial, referential, and tectonic articulation of connectedness between architecture and context. All of these issues occurred in the modern urban context, which made the "Loci Genius" lose. So how could architects reconstruct and conserve the "Genius Loci." I try to build the temple for people in the high dense city, which help people to find and think for themselves, to keep peaceful. The aim of this work is to map the current development on the Prague cultural scene set in the industrial spaces, it captures in a comprehensive way some common tendencies in the atypical industrial environment. Although there are marginally mentioned various cultural activities, work particularly focuses on the frequent presentation of electronic dance music in abandoned buildings. The work describes the historical development of industrial heritage on the territory of Prague, what kind of cultural businesses in this environment are currently taking part, how do they differ and what do they depend on. After reading this thesis the reader gains insight into the trends currently prevailing on the Prague music scene out of the traditional club environment.

Camundongos foram selecionados geneticamente para alta (AIRmax) ou baixa (AIRmin) resposta inflamatória aguda, sendo utilizados em experimentos de regeneração tecidual. Foi observado que AIRmax apresentam uma regeneração rápida do tecido da orelha em relação aos AIRmin, sugerindo a existência de loci reguladores comuns para ambos fenótipos de inflamação e regeneração do tecido. Neste estudo investigamos o fenótipo inflamatório e o perfil de expressão gênica global em AIRmax e AIRmin durante a fase inicial da regeneração do tecido da orelha. A análise histológica mostrou que AIRmax apresentam uma regeneração completa, com formação de ilhas de cartilagem e glândulas sebáceas no centro da área regenerada. Os AIRmin não apresentam regeneração. Os dados obtidos sobre edema de orelha e níveis de MPO foram maiores em AIRmax quando comparados com AIRmin (P <0,001). A análise de expressão gênica global mostrou 794 genes ativados e 528 genes reprimidos em AIRmax, enquanto 1.086 genes ativados e 1.145 genes reprimidos nos AIRmin, 48 horas após a lesão. AIRmax e AIRmin apresentaram alta modulação de genes sobrerrepresentados em temas biológicos para resposta inflamatória, adesão celular e quimiotaxia (Gene Ontology). No entanto, foi observado uma baixa modulação de genes sobrerrepresentados para o transporte em AIRmax e para taxia, contração muscular e ciclo de ubiquitina em AIRmin. Nas regiões próximas ao QTL anteriormente encontrado no cromossomo 1, foram observados genes diferencialmente expressos como Stat1, Casp8, Hspe1 e Il1r2, enquanto Lect1, Fndc3 e Egr3 foram detectados no cromossomo 14. Os experimentos com qPCR mostram uma alta expressão de Il-1<font face=\"Symbol\">b, Il-8rb e Mmp9 em AIRmax e Cxcl2, Tnfa e Tgfb em AIRmin. Concluímos que os animais AIRmax e AIRmin apresentam (nos cromossomos 1 e 14) genes inflamatórios diferencialmente expressos que podem estar envolvidos nos fenótipos de resposta inflamatória aguda e regeneração do tecido da orelha. / Mice selected for high (AIRmax) or low (AIRmin) acute inflammatory response were used in tissue regeneration experiments. It was observed that AIRmax present faster ear tissue regeneration than AIRmin mice, suggesting the involvement of common regulatory loci for both inflammation and ear tissue regeneration phenotypes. In the study we investigated some inflammatory phenotypes and global gene expression profiles in AIRmax and AIRmin mice during the initial phase of the ear tissue regeneration. The histological analyses showed that AIRmax regeneration was complete, and cartilage islands and sebaceous glands were formed in the middle of the regenerated area. AIRmin mice displayed not regeneration. Ear thickness oedema and MPO levels were higher in AIRmax than AIRmin mice (P< 0.001). Global expression analysis showed 794 activated and 528 repressed genes in AIRmax, while 1086 activated and 1145 repressed genes were observed in AIRmin mice 48 hours after injury. AIRmax and AIRmin mice presented up-regulated genes over-represented in inflammatory response, cell adhesion and chemotaxis biological themes (Gene ontology). However, down-modulated genes were significant over-represented for transportation in AIRmax and for taxis, muscle contraction and cycle ubiquitin in AIRmin mice. In the previously QTL regions detected on chromosome 1 differentially-expressed Stat1, Casp8, Hspe1 and Il1r2 genes were found, while Lect1, Fndc3 and Egr3 were detected on chromosome 14. qPCR experiments showed high expression of Il-1<font face=\"Symbol\">b, Il-8rb and Mmp9 in AIRmax and Cxcl2, Tnfa, and Tgfb1 in AIRmin mice. We conclude that AIRmax and AIRmin mice presented several (some on chromosomes 1 and 14) differentially-expressed inflammatory genes which could be involved in the acute inflammatory response and ear tissue regeneration phenotypes.

Then the DBs of the markers of the Y-STR X-STRs haplotypes were created. Comparison of the obtained data was conducted from allele frequencies (AF), paternity index (PI) by marker and paternity probabilities (PPs) obtained from nine BD population, one of those national (BR), using 241 cases containing the PP 99.99 using 13 loci STRs (CODIS) and 2 more STRs markers of Penta D- and E, granted by the BDs of Núcleo de Pesquisas Replicon, of the Pontifícia Universidade Católica de Goiás (PUC - GO) and LaGene, Laboratório Central de Saúde Pública (SES - GO). 241 cases were analyzed in Trio (alleged father, son and progenitor) and Duo (alleged father and son) situations, using 13 STRs markers (CODIS) by Bayes theorem and the Likelihood Ratio (LR). The results were submitted to the Kruskal-Wallis, chi-square, Spearman correlation and Fishers exact tests and SPSS software (2010). Static analysis of RV cases containing the Trio resulted in 1,148 results PPs 99.99% and 21 results PPs <99.99%, and in a position to Duo 1686 results with PPs 99.99% and 483 results PPs <99.99%. The analysis of the IP average of each marker showed the D21S11 of STRs markers and FGA with the highest power of inclusion and TH01 and D3S1358 with the lowest power of inclusion. The PPs did not show significantly different, containing mostly positive correlation, of moderate to strong, between 8 BDs compared to the BR population databases. This study demonstrated the statistics interference that each allele frequencies DB can have in PP calculations using only 13 loci of genetic, thus making it more significant in cases Duo situation. According to the information available in databases of gene frequencies of the different geographical regions of Brazil, it became possible to conclude that the allele frequencies obtained and the IPs per marker and PPs obtained, suggest strong similarities to those found in the national database. / Para fins de identificação humana na área forense os Estados Unidos criou o Banco de Dados (BD) do Sistema de Índice de DNA Combinado (CODIS), contendo 13 loci: CSF1PO, FGA, TH01, TPOX, VWA, D3S1358, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51 e D21S11. Foi realizado no presente estudo, a comparação dos dados obtidos das frequências alélicas (FA), índices de paternidade (IP) por marcador e probabilidades de paternidade (PPs) obtidas a partir de 9 BD populacionais, sendo 1 nacional (BR), utilizando 241 casos contendo a PP 99,99 com o uso de 13 loci STRs (CODIS) e mais 2 marcadores STRs do Penta-D e E, cedidos pelos BDs do Núcleo de Pesquisa Replicon, da Pontifícia Universidade Católica de Goiás (PUC-GO) e do LaGene, Laboratório Central de Saúde Pública (SES-GO). Os 241 casos foram analisados nas situações de Trio (suposto pai, filho e genitora) e Duo (suposto pai e filho) usando 13 marcadores STRs (CODIS) através do teorema de Bayes e pela Razão de Verossimilhança (RV). Os resultados foram submetidos aos testes de Kruskal-Wallis, Qui-quadrado, Correlação de Spearman e Exato de Fisher usando o software SPSS (2010). As análises estáticas da RV dos casos contendo o Trio resultaram em 1.148 resultados com PPs 99,99% e 21 resultados com PPs < 99,99%, e em situação de Duo 1.686 resultados com PPs 99,99% e 483 resultados com PPs < 99,99%. As análises das médias dos IPs de cada marcador demonstraram os marcadores STRs do D21S11 e FGA com os maiores poder de inclusão e TH01 juntamente com o D3S1358 os menores poderes de inclusão. As PPs não apresentaram diferenças estatísticas significativas contendo em sua maioria correlações positivas de moderadas a fortes entre os 8 BDs comparados ao BDs populacional do BR. Este estudo demonstrou as interferências estatísticas que cada BDs de frequências alélicas pode exercer nos cálculos de PP quando se utiliza apenas 13 loci para confirmação de vínculo genético, tornando-se assim mais significativas nos casos em situação de Duo. De acordo as informações disponíveis nos BDs de frequências alélicas das diferentes regiões geográficas do Brasil, tornou-se possível concluir, que as frequências alélicas obtidas, bem como os IPs por marcador e as PPs obtidas, sugerem fortes similaridades às encontradas no banco de dados nacional.

Camundongos AIRmax e AIRmin homozigotos para os alelos R e S do gene Slc11a1 foram avaliados para susceptibilidade à artrite induzida por pristane (PIA). A presença do alelo S aumentou a incidência e a gravidade nos AIRmax, sugerindo que o gene Slc11a1, ou outro próximo, esteja interagindo com os loci de resposta inflamatória na modulação de PIA. Para identificar estes loci foram realizados estudos de associação genótipo-fenótipo e de expressão gênica global. Os RNAs das patas dos animais foram isolados após 180 dias da indução por pristane. As análises de expressão gênica global foram realizadas usando a plataforma Codelink (36k genes), cujos resultados foram validados por PCR em tempo real. Os estudos de associação foram realizados através da análise de polimorfismo de microssatélites pelo programa MapManager. Foram identificadas duas regiões nos cromossomos 1 e 11. Um número grande de genes diferencialmente expressos foi verificado nos animais AIRmax SS cujos temas biológicos significativamente sobre-representados foram a resposta inflamatória e quimiotaxia. Os camundongos AIRmax SS também possuem uma ativação maior dos genes Ccl3, Ccl7, C3ar1, Il10, Stat3, Tirap, Trem 1, Trem 3, Mefv, Ptx3, Chi3l3 e Kras. Alguns desses genes co-localizam com regiões previamente mapeadas nos cromossomos 1 e 11. / AIRmax and AIRmin mice homozygous for Slc11a1 R and S allele were evaluated for pristane-induced arthritis (PIA) susceptibility. The presence of S allele increased the incidence and the arthritis severity in ARmax mice, suggesting that Slc11a1 or other closed-linked gene interacts with inflammatory loci to modulate PIA. In order to identify inflammatory modifier loci modulating experimental arthritis development, genotype-phenotype association studies and global gene expression analyses were performed. Mice received i.p. injections of pristane and the paw RNAs were isolated at day 180. Global gene expression analysis was performed on Codelink bioarrays (36k genes) and validated by real time PCR. The microsatellite polymorphism analyses were performed using MapManager program. Two regions on chromosomes 1 and 11 were identified. Higher number of differentially-expressed genes were detected in AIRmax SS subline, which significant over-represented biological themes were related to inflammatory response and chemotaxis. Susceptible AIRmax SS mice also display high up-regulation of Ccl3, Ccl7, C3ar1, Il10, Stat3, Tirap, Trem 1, Trem 3, Mefv, Ptx3, Chi3l3 e Kras genes. Some of them co-localize with previously identified regions mapped on chromosomes 1 and 11.

Les protocoles de détection de QTL varient selon le modèle étudié, car ils dépendent de nombreux paramètres. Cette thèse s’est intéressée à la façon d’adapter ces protocoles à travers deux exemples de recherches de QTL chez la Poule, en utilisant deux méthodes statistiques différentes : maximum de vraisemblance (MV) et régression linéaire (RL), qui ont été comparées. Dans un premier temps, des QTLs de résistance au portage de salmonelles ont été identifiés, d’effets faibles et dont les positions varient selon la méthode utilisée. Dans un deuxième temps, des QTLs de caractères de digestibilité et d’anatomie du tube digestif ont été identifiés, avec des résultats semblables avec les deux méthodes. De nombreux QTLs d’effets faibles à modérés ont été identifiés. Les résultats de cette thèse montrent que la comparaison des deux méthodes est toujours utile et car dans certaines conditions les résultats obtenus diffèrent entre les deux méthodes. / The QTL detection protocols vary depending on the model studied, because they depend on many parameters. This thesis has focused on how to adapt these protocols through two examples of QTL detection in Chicken, using two different statistical methods: maximum likelihood (ML) and linear regression (LR), which results were compared on two examples. Initially, QTLs controlling resistance to Salmonella carrier-state have been identified, of small effects and whose positions vary according to the method. In a second step, QTLs controlling digestibility and anatomy of the gastro-intestinal tract were identified with similar results for both methods. Many QTLs of small to moderate effects were identified. The results of this thesis show that the comparison of the two methods is always helpful as under certain conditions the results may vary with the method.

Caractérisation des déterminants génétiques et moléculaires liés à la résistance au dépérissement bactérien chez l'abricotier et analyse des risques associés / Caracterization of genetic and molecular determinants of resistance to bacterial canker in apricot and analysis of the associated risks

Parmi les Prunus, genre botanique d’intérêt économique important, l’abricotier (Prunusarmeniaca L.) est une culture emblématique du Bassin Méditerranéen. Il y est soumis à des contraintes biotiques importantes, parmi lesquelles le dépérissement bactérien, causé par Pseudomonas syringae (Psy), peut mener à des phénomènes de mortalité en verger au niveau des régions à hivers froids et humides. La mise en évidence de différences variétales en verger offre potentiellement des perspectives de contrôle de la maladie à travers le levier génétique. Aussi, ce travail de thèse avait pour principaux objectifs (i) d’identifier chez la plante des régions génomiques liées à la résistance partielle à la bactérie et (ii) d’étudier un plan factoriel d’interaction entre les diversités de la plante et de la bactérie (GxG) afin d’apprécier la généricité de la résistance et sa durabilité. Afin de répondre au premier objectif, deux approches complémentaires ont été mobilisées : une cartographie de QRLs (Quantitative Resistance Loci) sur quatre populations biparentales dont trois sont issues du croisement avec un géniteur commun ainsi qu’une analyse d’association sur une core-collection. Les données phénotypiques mobilisées correspondent à des symptômes issus d’inoculations contrôlées ainsi que des notes de mortalité obtenues suite à infection naturelle en verger. Ces deux approches (analyse de liaison et d’association) ont permis de mettre en évidence 22 QRLs de résistance, parmi lesquels seuls 2 QRLs sur les chromosomes 6 et 7 colocalisent entre les deux approches. Deux régions majeures détectées en étude d’association sur les chromosomes 5 et 6 se sont révélées être en déséquilibre de liaison et contrôlent près de 26 et 43% de la variation des symptômes. Deux mécanismes complémentaires reposant sur le blocage de l’infection de Psy et sur la limitation de la progression locale de la bactérie dans les tissus ont été mis en évidence à travers la détection de QRLs sur les chromosomes 3, 6, 8 d’une part et 1,4et 6 d’autre part. Le second objectif a été abordé grâce à une étude d’un plan factoriel d’interaction entre 20 accessions d’abricotier et 9 souches de Psy, échantillonnées d’après la connaissance de l’épidémiologie de la maladie en verger. L’analyse statistique de ce dispositif mis en œuvre à la fois en verger et en laboratoire a démontré la prédominance de l’effet du facteur souche dans la variabilité des symptômes étudiés et la très faible importance du facteur d’interaction GxG, indiquant une potentielle généricité des facteurs de résistance et des perspectives favorables à leur durabilité en verger.Les résultats issus de cette thèse contribuent à offrir une meilleure compréhension des mécanismes de résistance partielle au dépérissement bactérien de l’abricotier et fournissent des marqueurs et haplotypes, potentiellement mobilisables dans le cadre de programmes d’innovation variétale. / Within the genus Prunus, that contains highly valuable species, apricot (Prunusarmeniaca L.) is an emblematic Mediterranean crop. But apricot cultivation is constrainedby many biotic stresses, among which bacterial canker caused by Pseudomonas syringae(Psy) is particularly severe and can lead to the death of the trees in regions with humidand cold winters. Differences of susceptibilities have been observed between cultivars inorchards and create opportunities for disease management through genetic improvement.This thesis aimed to (i) identify genetic determinants linked to partial resistance to thebacterium and to (ii) study a factorial interaction design between both diversities of theplant and the pathogen (GxG interaction) in order to assess resistance genericity anddurability. With regard to the first objective, two complementary approaches were used :QRL (Quantitative Resistance Loci) mapping over four biparental progenies, amongwhich three were obtained with a cross involving a common genitor, and a genome-wideassociation study on a core-collection. The phenotypic data mobilized in this work rely onsymptoms issued from controlled inoculations and on mortality notations followingnatural infections in the orchard. These approaches led to the detection of 22 QRLs amongwhich only 2 QRLs, located on chromosomes 6 and 7, co-localized between the twomethods. Two main regions detected in the association study, over the chromosomes 5and 6, appeared to be in linkage disequilibrium and controlled 26 and 43% of the variationof the symptoms. A complementarity between two mechanisms, one that involves blockingthe infection of Psy and the other that limits bacterial mobility in the tissues has beenrevealed through the detection of QRLs over chromosomes 3, 6, 8 for one mechanism and1,4, 6 for the other, respectively. The second objective was fulfilled with a study of afactorial interaction design between 20 apricot accessions and 9 Psy strains, which weresampled according to the previous knowledge of the disease epidemiology in the orchard.Statistical analyses of phenotypic data obtained both from the orchard and a laboratorytest showed a clear predominance of the strain effect on symptom variability and a weakimportance of the GxG interaction factor. This last result highlighted a potentialgenericity of the resistance factors and favorable perspectives of durability in the orchard.The results issued from this thesis contribute to a better understanding of the mechanismsunderlying partial resistance of apricot to bacterial canker. Moreover, it provide markersand haplotypes of interest which could be mobilized in breeding programs.

It was observed that AIRmax present faster ear tissue regeneration than AIRmin mice, suggesting the involvement of common regulatory loci for both inflammation and ear tissue regeneration phenotypes. In the study we investigated some inflammatory phenotypes and global gene expression profiles in AIRmax and AIRmin mice during the initial phase of the ear tissue regeneration. The histological analyses showed that AIRmax regeneration was complete, and cartilage islands and sebaceous glands were formed in the middle of the regenerated area. AIRmin mice displayed not regeneration. Ear thickness oedema and MPO levels were higher in AIRmax than AIRmin mice (P< 0.001). Global expression analysis showed 794 activated and 528 repressed genes in AIRmax, while 1086 activated and 1145 repressed genes were observed in AIRmin mice 48 hours after injury. AIRmax and AIRmin mice presented up-regulated genes over-represented in inflammatory response, cell adhesion and chemotaxis biological themes (Gene ontology). However, down-modulated genes were significant over-represented for transportation in AIRmax and for taxis, muscle contraction and cycle ubiquitin in AIRmin mice. In the previously QTL regions detected on chromosome 1 differentially-expressed Stat1, Casp8, Hspe1 and Il1r2 genes were found, while Lect1, Fndc3 and Egr3 were detected on chromosome 14. qPCR experiments showed high expression of Il-1<font face=\"Symbol\">b, Il-8rb and Mmp9 in AIRmax and Cxcl2, Tnfa, and Tgfb1 in AIRmin mice. We conclude that AIRmax and AIRmin mice presented several (some on chromosomes 1 and 14) differentially-expressed inflammatory genes which could be involved in the acute inflammatory response and ear tissue regeneration phenotypes.

Camundongos AIRmax e AIRmin homozigotos para os alelos R e S do gene Slc11a1 foram avaliados para susceptibilidade à artrite induzida por pristane (PIA). A presença do alelo S aumentou a incidência e a gravidade nos AIRmax, sugerindo que o gene Slc11a1, ou outro próximo, esteja interagindo com os loci de resposta inflamatória na modulação de PIA. Para identificar estes loci foram realizados estudos de associação genótipo-fenótipo e de expressão gênica global. Os RNAs das patas dos animais foram isolados após 180 dias da indução por pristane. As análises de expressão gênica global foram realizadas usando a plataforma Codelink (36k genes), cujos resultados foram validados por PCR em tempo real. Os estudos de associação foram realizados através da análise de polimorfismo de microssatélites pelo programa MapManager. Foram identificadas duas regiões nos cromossomos 1 e 11. Um número grande de genes diferencialmente expressos foi verificado nos animais AIRmax SS cujos temas biológicos significativamente sobre-representados foram a resposta inflamatória e quimiotaxia. Os camundongos AIRmax SS também possuem uma ativação maior dos genes Ccl3, Ccl7, C3ar1, Il10, Stat3, Tirap, Trem 1, Trem 3, Mefv, Ptx3, Chi3l3 e Kras. Alguns desses genes co-localizam com regiões previamente mapeadas nos cromossomos 1 e 11. / AIRmax and AIRmin mice homozygous for Slc11a1 R and S allele were evaluated for pristane-induced arthritis (PIA) susceptibility. The presence of S allele increased the incidence and the arthritis severity in ARmax mice, suggesting that Slc11a1 or other closed-linked gene interacts with inflammatory loci to modulate PIA. In order to identify inflammatory modifier loci modulating experimental arthritis development, genotype-phenotype association studies and global gene expression analyses were performed. Mice received i.p. injections of pristane and the paw RNAs were isolated at day 180. Global gene expression analysis was performed on Codelink bioarrays (36k genes) and validated by real time PCR. The microsatellite polymorphism analyses were performed using MapManager program. Two regions on chromosomes 1 and 11 were identified. Higher number of differentially-expressed genes were detected in AIRmax SS subline, which significant over-represented biological themes were related to inflammatory response and chemotaxis. Susceptible AIRmax SS mice also display high up-regulation of Ccl3, Ccl7, C3ar1, Il10, Stat3, Tirap, Trem 1, Trem 3, Mefv, Ptx3, Chi3l3 e Kras genes. Some of them co-localize with previously identified regions mapped on chromosomes 1 and 11.
