Les nanoparticules seront sans doute les outils diagnostiques et thérapeutiques de demain. Si ellessont magnétiques, elles sont promises à des applications telles que le renforcement du contraste enIRM, la thermothérapie et la libération contrôlée de médicaments. Les nanomatériaux La1-xSrxMnO3ont été sélectionnés car leur température de Curie (TC) peut être ajustée dans la gamme detempérature thérapeutique. Des particules calibrées en taille et désagrégées ont été élaborées par leprocédé glycine-nitrate (GNP). Les caractérisations chimiques et structurales ont permis de mieuxcomprendre les résultats contradictoires de la littérature concernant la soi-disant dépendance de TCavec la taille des nanoparticules. L’adaptabilité de ces nanoparticules pour des applications enhyperthermie ou en IRM a été confirmée. Enfin, la capacité des nanoparticules à s’échauffer a étéutilisée pour réticuler autour d’elles une couronne de macromolécules thermosensibles selon leconcept nouveau de chimie localement stimulée. / Nanoparticles may be the next generation of diagnostic and therapeutic tools. If they are magnetic,they are dedicated to applications such as MRI contrast agent, thermotherapy and controlled drugrelease. La1-xSrxMnO3 nanoparticles were selected because their Curie temperature (TC) may be tunedwithin the range of therapeutic temperature. Size sorted and disaggregated particles weresynthesized by the Glycine-Nitrate Process. Chemical and structural characterizations allowed abetter understanding of conflicting results found in the literature about the particle size-dependenceof TC. The possibility to use these nanoparticles for hyperthermia and MRI applications has beenconfirmed. Finally, their ability to heat has been used to crosslink thermosensitive macromoleculesall around them according to the new concept of locally stimulated chemistry.

Single-cell tracking of therapeutic cells using Laser Ablation-Inductively Coupled Plasma-Mass Spectrometry

Cellular therapy is emerging as a clinically viable strategy in the field of solid organ transplantation, where it is expected to reduce the dependency on conventional immunosuppression. This has produced a demand for highly sensitive methods to monitor the persistence and tissue distribution of administered cells in vivo. However, tracking cells presents significant challenges. In many cases transplanted cells are autologous with the immune system of the transplant recipient, and hence are invisible to typical methods of detection. To enable their differentiation, the cells must be labelled with a suitable, non-toxic and long lifetime label, prior to their administration to patients. In addition, administered cells represent only a small fraction of the recipient's endogenous cells, which necessitates the use of an extremely sensitive detection method. Laser ablation – inductively coupled plasma – mass spectrometry (LA-ICP-MS) is an exquisitely sensitive analytical technique, capable of imaging trace elements in complex samples, at high spatial resolution.

Synthèse et caractérisation de nanomatériaux hybrides innovants pour le biomédical / Synthesis and characterization of new hybrids nanomaterials for biomedical applications

Depuis quelques décennies, les nanomatériaux et tout particulièrement les nanoparticules d’oxyde de fer (magnétite/maghémite) superparamagnétiques ont connus un intérêt croissant en nano-médecine. Leur biocompatibilité et leurs propriétés magnétiques permettent notamment leur utilisation à des fins de diagnostic (IRM, Imagerie optique et nucléaire…) et de thérapie (Hyperthermie, nano vectorisation…). Au cours de cette thèse, la première étape a consisté à étudier en détails l’influence de plusieurs paramètres de synthèse sur les propriétés finales des nanoparticules d’oxyde de fer magnétique. Cette étude avait pour but de développer et d’optimiser une méthode de synthèse dérivée de la méthode de synthèse classique dite de co-précipitation mais modifiée par ajout de ligand citrates au cours de la synthèse. Des nanoparticules d’oxyde de fer d’une taille pouvant être contrôlée entre 4 et 13 nm recouvertes par une couronne de ligands citrates ont ainsi été synthétisées, celles-ci présentent une aimantation à saturation atteignant jusqu’à 75 emu/g d’oxyde de fer qui est une valeur particulièrement haute pour des nanoparticules de cette taille. Tout au long de ce travail, la caractérisation de ces nanoparticules par un panel étendu de techniques (MET, DRX, Mössbauer, IRTF, XPS, mesures magnétiques, DLS …) à permis notamment d’étudier de façon précise les relations existantes entre la taille, le taux de ligand, la composition et les propriétés magnétiques des nanoparticules synthétisées. Ces nanoparticules fonctionnalisées par des citrates ont ensuite été testées en milieu biologique afin d’évaluer leur internalisation dans les cellules et leur cytotoxicité. Dans un deuxième temps, d’autres travaux ont été menés sur les nanoparticules d’oxyde de fer. Notamment le remplacement des ligands citrates par un polymère bio-inspiré pouvant, selon les fonctions chimiques qui lui sont adjointes, avoir de multiples applications dans le domaine biomédical / From decades now, nanomaterials and especially superparamagnetic iron oxide nanoparticles are studied for their numerous applications in nanomedecine area. The biocompatibility and the magnetic properties of such nano-objects allow their utilization for diagnostic (MRI, optical imagery, PET…) and for therapy application (nanovectorization, hyperthermia…) During this thesis work, the first step was to study the influence of several synthesis parameters on the final properties of the magnetic iron oxide nanoparticles. The aim of this study was the development and the optimization of the widely used way of synthesis by co-precipitation modified by a ligand addition during the growth step of the synthesis. Citrate capped iron oxide nanoparticles with a controlled size between 4 and 13 nm have been synthesized, the saturation magnetization of these nanoparticles reach 75 emu/g of iron oxide, this value is particularly high for nanoparticles of such sizes. During this work, the large panel of characterizations performed on these nanoparticles (TEM, XRD, Mössbauer, FTIR, XPS, DLS, Magnetic measurement) allowed to study precisely the relations between size, ligand ratio, composition and magnetic properties of the synthesized nanoparticles. The interaction between the synthesized citrate capped nanoparticles and biological materials such as human cells have been investigated in-vitro notably to evaluate cells internalization and citotoxicity. In a second step, some additional works have been performed on the citrate capped iron oxide nanoparticles in order to replace the citrate ligand by a bio-inspired polymer (poly-oxazoline). This polymer can have multiple biomedical applications depending of the pendent chemical groups that have been fixed on it

Nous avons synthétisé différentes dyades donneurs-accepteurs (D-A) π-conjuguées à base de fullerène pour des applications photovoltaïques dans lesquelles les unités D étaient soit des oligophenylenevinylenes (OPV) soit des oligophenyleneethynylene (OPE) et les unités A étaient le C60 ou un endofullerène du type Y3N@C80. Il y avait une exigence supplémentaire pour nos matériaux, à savoir qu’ils devaient s’auto-organiser en phases liquides-cristallines. Pour ce faire, toutes les unités D contenaient un promoteur mésogène afin d'induire le mésomorphisme de la dyade D- et donc de contrôler la morphologie des couches minces nécessaires à l’élaboraiton des cellules photovoltaïques grâce à une organisation supramoléculaire. En dehors de cela, nous avons étudié l’influence de la nature chimique du donneur (par exemple lyophile ou amphiphile), de la longueur des oligomères et de la multiaddition sur les propriétés photophysiques et sur l'auto-assemblage. Nous avons synthétisé une dyade OPE-Y3N@C80 qui est le premier dérive mésomorphe et photosensible de ce type de métallofullerène endohédral. / We have synthesized different π-conjugated system-fullerene dyads for photovoltaic applications, where the donor units were either oligophenylenevinylene (OPV) or oligophenyleneethynylene (OPE) derivatives and for the acceptor, C60 or Y3N@C80 was used. There was an additional requirement for our materials: liquid crystallinity. All the donor units contained a mesogenic promoter in order to induce mesomorphism in the D-A dyad and to control the morphology of the prepared film through supramolecular organization. Apart from that, we investigated the effect of the chemical nature of the donor moiety (ie. lyophilic or amphiphilic), the oligomeric length and multiaddition on the photophysical properties and on the self-assembly. We have synthesized an OPE-Y3N@C80 dyad which is the first trimetallic nitride template endohedral metallofullerene derivative with mesomorphic and photoactive properties.

Ce manuscrit de thèse présente la synthèse de nanoparticules d’oxyde de fer superparamagnétiques couramment appelées SPIONs servant d’agents de contraste pour l’imagerie par résonance magnétique (IRM) et la génération de chaleur pour la thérapie cellulaire par hyperthermie induite par champ magnétique radiofréquence (HMRF). Le contrôle des tailles et de la distribution en tailles des SPIONs et donc de leurs propriétés magnétiques a été obtenu en utilisant un copolymère arborescent G1 (substrat de polystyrène branché en peigne noté G0, greffé avec des groupements pendants poly(2-vinyle pyridine) ) comme milieu « gabarit », tandis que la stabilité colloïdale et la biocompatibilité des SPIONs ont été apportées par un procédé de poly-complexation ionique grâce à un copolymère double-hydrophile acide polyacrylique-bloc-poly(acrylate de 2-hydroxyéthyle) PAA-b-PHEA. / This Ph.D. dissertation describes the synthesis of superparamagnetic iron oxide nanoparticles (SPIONs) designed to serve as magnetic resonance imaging (MRI) contrast agents and for heat generation in cellular radiofrequency magnetic field hyperthermia (MFH) treatment. Control over the size and size distribution of the iron oxide nanoparticles (NPs), and thus over their magnetic properties, was achieved using a G1 arborescent copolymer (comb-branched (G0) polystyrene substrate grafted with poly(2-vinylpyridine) side chains, or G0PS-g-P2VP) as a template. Good colloidal stability and biocompatibility of the SPIONs were achieved via the formation of polyion complex (PIC) micelles with a poly(acrylic acid)-block-poly(2-hydroxyethyl acrylate) (PAA-b-PHEA) double-hydrophilic block copolymer.

Synthesis, Characterization and Structure-Property Relationships of Polymer-Stabilized Nanoparticles Containing Imaging and Therapeutic Agents

The controllable design of magnetic nanocarriers is essential for advanced in vivo applications such as magnetic resonance image-guided therapeutic delivery and alternating magnetic field-induced remote release of drugs. This work describes the fabrication of polymer-stabilized nanoparticles encapsulating imaging and therapeutic agents and delineates relationships among materials parameters and response. The effect of aggregation of magnetic iron oxide nanoparticles in aqueous suspension was characterized using a well-defined core-corona complex comprised of a superparamagnetic magnetite nanoparticle stabilized by terminally-anchored poly(N-isopropylacrylamide) (PNIPAM) corona. The modified Vagberg density distribution model was employed to verify that the complexes were individually dispersed prior to aggregation and was found to accurately predict the intensity-weighted hydrodynamic diameter in water. Aggregation of the complexes was systematically induced by heating the suspension above the lower critical solution temperature (LCST) of the polymer, and substantial increase in the NMR transverse relaxation rates was noted. Controlled clusters of primary iron oxide nanoparticles stabilized by the biodegradable block copolymer, poly(ethylene oxide-b-D,L-lactide) were fabricated by a scalable, rapid precipitation technique using a multi-inlet vortex mixer. Quantitative control over iron oxide loading, up to 40 wt%, was achieved. Correlations between particle parameters and transverse relaxivities were studied within the framework of the analytical models of transverse relaxivity. The experimental relaxivities typically agreed to within 15% with the values predicted using the analytical models and cluster size distributions derived from cryo-transmission electron microscopy. Hydrophilic-core particles assembled using the poly(ethylene oxide-b-acrylate) copolymer and at similar primary nanoparticle sizes and loadings had considerably higher transverse (r2) and longitudinal (r1) relaxivities, with r2s approaching the theoretical limit for ~ 8 nm magnetite. Block copolymer nanoparticles comprised of poly(D,L-lactide) and poly(butylene oxide) cores were utilized to encapsulate the poorly water-soluble antiretroviral drug, ritonavir, at therapeutically-useful loadings. Controlled size distributions were achieved by incorporation of homopolymer additives, poly(L-lactide) or poly(butylene oxide) during the nanoparticle preparation process. Nanoparticles either co-encapsulating a highly hydrophobic polyester poly(oxy-2,2,4,4-tetramethyl-1,3-cyclobutanediyloxy-1,4-cyclohexanedicarbonyl) within the core or possessing crosslinkable groups around the core were also successfully fabricated for potential sustained release of ritonavir from block copolymer carriers. / Ph. A teragnóstica de doenças tem sido extremamente marcada nos últimos anos por nanomateriais formados pela conjugação de nanopartículas a biomoléculas, pois a aplicação de tecnologias baseadas em materiais na dimensão nanométrica é capaz de aumentar a seletividade, sensibilidade e praticidade dos métodos atualmente empregados, ou mesmo criar novos métodos de diagnóstico e tratamento de doenças. Dentre os vários tipos de nanomateriais desenvolvidos, aqueles baseados em nanopartículas de ouro ou nanopartículas magnéticas apresentam propriedades químicas e físicas diferenciadas que propiciam novas possibilidades. Por exemplo, a presente tese demonstrou que nanopartículas superparamagnéticas são excelentes agentes de contraste em exames de imagem por ressonância magnética (IRM) por serem mais seguros, apresentarem melhor contraste nas imagens e possibilitarem direcionar/concentrar o material em tecidos ou tumores através de um gradiente de campo magnético aplicado. Foram feitos numerosos ensaios de toxicidade tanto in vitro quanto in vivo para assegurar a segurança da aplicação de nanopartículas no organismo, cujo potencial de uso somente se tornará uma realidade caso os nanomateriais se mostrem não tóxicos e biocompatíveis. Apesar dos significativos avanços na área da aplicação desses nanomateriais, não foram encontrados na literatura modelos capazes de explicar ou prever por quais sítios de ligação devem ocorrer as interações proteína-nanopartícula, como também não foram encontrados estudos sistemáticos acerca dos fatores que determinam a estabilidade e a funcionalidade dos nanobioconjugados (NBCs). Assim, nesta tese buscamos compreender os fatores responsáveis pela ligação/adsorção das proteínas nas nanopartículas de ouro e sua influência sobre a estabilidade das suspensões e a funcionalidade das proteínas. Desta forma, foram obtidos NBCs com propriedades adequadas para o desenvolvimento ou aprimoramento de ensaios de diagnóstico e até para o tratamento de doenças. Foi demonstrado o potencial das nanopartículas de ouro para melhorar a performance de imunoensaios do tipo ELISA, mas também podem ser utilizadas para o desenvolvimento de métodos de diagnóstico, explorando as propriedades plasmônicas das nanopartículas de ouro acopladas a técnicas como SERS, SPR e microscopia Raman confocal. / Theranostics has been intensively pursued in recent years using hybrid materials based on nanoparticles conjugated with biomolecules. This is an interesting strategy to increase the selectivity and sensitivity, as well as to improve the currently used methods facilitating their use or creating new ones. Among the various types of nanomaterials, those based on gold and magnetic nanoparticles exhibit interesting chemical and physical properties in the biological environment, differing from that of free drugs or current explored in assay methods. For example, superparamagnetic nanoparticles are excellent contrast agents for magnetic resonance image (MRI) diagnostics because they are safer, present a better contrast efficiency for imaging and can be magnetically accumulated in tissues or tumors using a magnetic field. Numerous in vitro and in vivo toxicity assays were performed to ensure the safety for medical applications. Clearly, these type of applications only will be realized if nanomaterials prove to be nontoxic and biocompatible. This imply an strict control on their structure and composition. However, despite the significant advances in the development of such nanomaterials, there were not found in the literature model systems explaining or that can be used to predict by which sites the protein-nanoparticle binding should take place. In addition, no systematic studies on the factors determining the stability and the functionality of nanobioconjugates (NBC) were found. These nanomaterials can improve the ELISA immunoassay, or other diagnosis methods can be developed by using the gold nanoparticles plasmonic properties in association with SERS, SPR and confocal Raman microscopy techniques.
